2.The application of case-based learning with multimedia teaching in the clinical teaching of pediatric surgery

Objective To explore the application of case based learning(CBL)combined with multimedia teaching in the clinical pediatric surgery.Methods 144 five-year program students of Pediatrics in grade 2009 were randomly and evenly divided into the experimental group using CBL with multimedia teaching,and the control group using multimedia teaching mode.After the end of the course the students of the two groups took clinical teaching knowledge test.The evaluation scores were shown by mean ± standard deviation and comparison was made between the two groups with t test.At the same time,teaching satisfaction survey was conducted among the students of the experimental group.Results The usual results,the theoretical scores and total scores of the students in the experimental group were higher than those of the control group students in the operation skills,clinical thinking,and there was statistically significant difference in P<0.01.(P=0.003,P=0.002,P=0.008,P=0.009,P=0.008).The questionnaire survey shows that compared with traditional multimedia teaching,CBL combined with multimedia teaching can preferably culture students' various abilities and is beneficial to learning.Condusion The use of CBL combined with multimedia teaching can affirmatively improve the clinical teaching effect in the clinical pediatric surgery,and it deserves to be generalized in the future teaching.
3.Effects of Plasmid Fibroblast Growth Factor-2 Magnetic Chitosan Gelatin Microspheres on Proliferation and Differentiation of Mesenchymal Stem Cells.

The purpose of this study is to investigate the effect of superparamagnetic chitosan FGF-2 gelatin microspheres (SPCFGM) on the proliferation and differentiation of mouse mesenchymal stem cells. The superparamagnetic iron oxide chitosan nanoparticles (SPIOCNs) were synthesized by means of chemical co-precipitation, combined with FGF-2. Then The SPCFGM and superparamagnetic chitosan gelatin microspheres (SPCGM) were prepared by means of crosslinking-emulsion. The properties of SPCFGM and SPIONs were measured by laser diffraction particle size analyser and transmisson electron microscopy. The SPCFGM were measured for drug loading capacity, encapsulation efficiency and release pharmaceutical properties in vitro. The C3H10 cells were grouped according to the different ingredients being added to the culture medium: SPCFGM group, SPCGM group and DMEM as control group. Cell apoptosis was analyzed by DAPI staining. The protein expression level of FGF-2 was determined by Western blot. The proliferation activity and cell cycle phase of C3H10 were examined by CCK8 and flow cytometry. The results demonstrated that both of the SPIOCNs and SPCFGM were exhibited structure of spherical crystallization with a diameter of (25 ± 9) nm and (140 ± 12) μm, respectively. There were no apoptosis cells in the three group cells. Both the protein expression level of FGF-2 and cell proliferation activity increased significantly in the SPCFGM group cells (P < 0.05). The SPCFGM is successfully constructed and it can controlled-release FGF-2, remained the biological activity of FGF-2, which can promote proliferation activity of C3H10 cells, and are non-toxic to the cell.

4.Therapeutic comparison of titanium elastic nailing and plating in treatment of both radius and ulna fracture in children

Objective To compare the effect of titanium elastic nailing (TEN) and plating in treatment of both radius and ulna fractures in children older than 10 years.Methods This retrospective analysis comprised 38 children (50 sides) of both radius and ulna fractures treated with TEN (30sides) or plate (20 sides) between 2009 and 2014.Parameters compared between the two fixation methods were operation time,ratio of open/close reduction,time to internal fixation removal,incidence of postoperative complications,and bone union time.Anderson score was used to evaluate the treatment results.Results Operation time [(94.3 ± 39.5) min vs (120.5 ± 45.4) min],time to internal fixation removal [(210.8 ± 84.1) d vs (264.4 ± 88.9) d],and ratio of open/close reduction (20/10 vs 20/0)varied significantly between TEN and plating groups (P < 0.05 or 0.01).Whereas,no statistically significant differences were noted in regard to complication rate,bone union time,and effect evaluation.Conclusion For fracture of the radius and ulna in children older than 10 years,TEN should be used for closed type without obvious displacement,but plate fixation is associated with lower incidence of fracture malunion and better outcome in treatment of open type with obvious displacement.
5.Application of femoral neck anteversion′s correction in the treatment of children′s developmental dysplasia of the hip

Objective To discuss application of femoral neck anteversion′s correction in the treatment of children′s developmen-tal dysplasia of the hip .Methods 98 children diagnosed DDH from Jan 2006 to Jan 2010 were treated and followed up ranging from 12 to 48 months ,average 32 months .Before the operations ,FNA index were evaluated by X-ray and thin slice hip joint scan and 3D reconstruction to guide the rotary osteotomy .After the operations ,FNA index were rechecked .Results According to Mckay′s clas-sification ,clinical assessment was performed .84 hips were evaluated as excellent (73 .68% ) ,20 hips as good(17 .55% ) ,10 hips as acceptable(8 .77% ) ,and no poor hip (0% ) was reported .FNA decreased averagely 16 .3° ± 2 .0° .FNA had significant difference with themselves before the operations(P<0 .01) and had no significant difference with the normal side(P>0 .05) .Conclusion The change of FNA is one of the most important pathological changes .FNA′s correction is one of the most valuable to guarantee success operation .
6.Serial Expression of the Truncated Fragments of the Nucleocapsid Protein of CCHFV and Identification of the Epitope Region

The Crimean-congo hemorrhagic fever virus(CCHFV)is a geographically widespread fatal pathogen.Identification of the epitope regions of the virus is important for the diagnosis and epidemiological studies of CCHFV infections.In this study,expression vectors carrying series truncated fragments of the NP(nueleoeapsid protein)gene from the S fragment of CCHFV strain YL04057 were constructed.The recombinant proteins were expressed in E.coli and purified for detection.The antigenic of the truncated fragments of NP was detected with a polyclonai serum(rabbit)and 2 monoclonal(mAbs)(14B7 and 43E5)against CCHFV by Western-blot analyses.The results showed that the three expressed constructs,which all contained the region 235AA to 305AA could be detected by mAbs polyclonal serum.The results suggest that region 235-305 aa of NP is a highly antigenic region and is highly conserved in the NP protein.
7.Effects of light on flavonoids accumulation and related gene expression in suspension cultures of Scutellaria baicalensis.

OBJECTIVETo investigate the effects of light on baicalin,baicalein accumulation and related genes expression in suspension cultures of Scutellaria baicalensis.
METHODThe content of baicalin, baicalein in suspension cultures of S. baicalensis was determined by HPLC. The related genes expression was analyzed by semi-quantitative PCR.
RESULTActive ingredients in suspension cultures of S. baicalensis were promoted significantly by light, PAL gene transcription under the conditions of light was higher than that under the dark conditions, the UBGAT gene expression levels was significantly related to the baicalein (r = -0.995).
CONCLUSIONActive ingredients and related genes (PAL, UBGAT) in suspension cultures of S. baicalensis were promoted significantly by light.

Objective To explore the epidemic situation of animal plague in Junggar Basin natural plague foci.Methods Data on epidemics of plague and on population involved,as well as results on antibodies and pathogens,were analyzed.Samples on animals and vectors were collected from 18 counties in Junggar Basin plague natural foci between 2007 and 2016.Results The density of Rhombomys (R.) opimus was temporally fluctuant,from 2.1/hm2 to 22.6/hm2 respectively.However,the spatial distribution appeared asymmetrical,with the highest seen in Kelamayi and Wumuqimidong counties,as 14.2/hm2 and 13.0/hrn2 respectively.Rates of capture on nocturnal rodents were from 4.2% to 10.1%,with the highest rate as 10.1% in 2014.Meriones meridianus appeared the dominant species in the nocturnal community of rodents,which accounted for 81.9%.Regarding the spatial and temporal distributions,rates ofR.opimus with fleas appeared fluctuant,with an average rate as 90.7% and the average total flea index was 10.44.In flea community of R.opimus,Xenopsylla (X.)skrjabini was found the dominant species,popular in distribution and accounted for 47.8%.The average rate of nocturnal rodents with flea was 20.2%,with total flea index as 1.20 and the dominant fleas were X.conformis conformis and Nosopsyllus laeviceps.A total of 13 species with 9 087 serum samples from rodents were detected as having Y.pestis antibody by IHA,with 617 positive samples.Of them,the positive rate of having R.opimus appeared the highest (9.4%),followed by D.sagitta (1.1%).Spatially,two clustered areas were found,with one in the eastern Junggar Basin from Changji to Mulei county,with the antibody positive rates ofR.opimus as 14.3%.The other one was in the central area of Junggar Basin,including Kelamayi,Shawan and Wusu counties,with the antibody positive rate as 13.6%.The prevalence of plague on R.opimus was fluctuant,with the lowest seen in 2008,with the average antibody positive rate of R.opimus as 1.0% and the highest as 19.3% in 2013.A total of 18 strains were isolated from 2007 to 2016.However,most of the strains were isolated from R.opimus and parasitic fleas,accounted for 8/9,in Kelamayi,Wulumuqi-midong and Jimusaer,respectively.Conclusions Complex ecosystem was seen in the Junggar Basin natural plague foci,with the multiple composition of species and different community structure of hosts and vectors,plus the flustering prevalence.Animal plague was seen in the whole region with succession of the plague nature foci.Passive transmission of plague between human beings and animals through close contacts was seen which was driven by economic benefits to some degree.
