1.Mechanism of growth hormone inhibiting of alveolar type Ⅱ epithelial cells in rats

Objective To investigate the mechanism of growth hormone inhibiting IPS-induced apoptosis of alveolar type Ⅱ epithelial cells in rats. Method Isolated and purified AEC Ⅱ cells of SD rats were divided into 5 groups,8 duplicate wells in each group. Group I served as control group; group Ⅱ:LPS 10 ug/ml;group Ⅲ:LPS 10 ug/ml + GH 50 ng/ml;gronp IV :LPS 10 ug/ml + GH 100ng/ml; group V: LPS 10 ug/ml + GH 200 ng/ml. LPS was finally added into wells in group Ⅱ~V . After the cells were incubated for 24 hours, the apoptosis rate and necrosis rate of AEC Ⅱ cells stained with Annexin V/PI were detected by flow cytometry and Fas protein of AEC Ⅱ cells were measured by immunocytochemistry. Results (1) The apoptosis rate and necrosis rate of AECⅡ cells in group Ⅱ,Ⅲ, Ⅳ and V were significantly hitOer than those in group Ⅰ( qapoptosis rate Ⅰ, Ⅱ =12.26,qnecroeis Ⅰ,Ⅱ=18.34, qapoptosisⅠ.Ⅱ=9.63,qnecrosisⅠ,nⅡ=5.75,qapotosisⅠ,Ⅳ= 9.15,qnecrosisⅠ,Ⅳ= 5.39, qapotosisⅠ,Ⅴ = 10.87, qnecrosisⅠ,Ⅴ = 5.91, P 0.05), but lower in group Ⅲ,IV and V than those in group Ⅱ(qapoptosis Ⅱ,Ⅲ= 15.24, qpecrosisⅡ,Ⅲ=16.38, qapoptosisⅡ.Ⅳ = 15.95,qnecrosisⅡ.Ⅳ=16.95, qapoptosis rate Ⅱ,Ⅴ=14.57, qnecrosisⅡ.Ⅴ = 15.61,P<0.05). (2)The positive rate of Fas expression on AEC Ⅱ cells in group Ⅱ,Ⅲ, Ⅳ and V was obviously higher than that in group Ⅰ. ( q Ⅰ.Ⅱ=35.67, qⅠ ,Ⅲ=14.32, qⅠ,Ⅳ = 13.87, qⅠ.Ⅴ=26.16, P<0.05), but lower in gronpⅢ ,Ⅳ and Ⅴ than that in gronp Ⅱ(qⅡ,Ⅲ=12.54, qⅡ,Ⅳ = 13.02, qⅡ,Ⅴ =6.96, P<0.05). Conclusions GH can probably de-crease the apoptosis of AEC Ⅱ cells by inhibiting Fas expression.
2.Clinical study on correlation between the level of CD4+CD25+regulatory T cells in peripheral blood and APACHE Ⅱ scores in patients with sepsis

Objective To evaluate the correlation between the level of CD4+CD25+regulatory T cells in periphend blood and disease severity in patients with sepsis.Methods Thirty-six septic patients and 5healthy controls were enrolled.Septic patients were divided into sepsis group(n=10),severe sepsis group (n=15)and septic shock group(n=11).The lymphocyte was seperated from peripheral blood and marked by PE-CD4 and FTTC-CD25 monoclonal antibody,the level of CD4+CD25+ regulatory T cells was detected by flow cytometry,and the clinical data of APACHE Ⅱ scores of septic patients was considered in 24 hours.The correlation between level of CD4+CD25+ regulatory T cells in peripheral blood and APACHE Ⅱ scores in septic patients was analyzed.Results Compared with the healthy controls [(5.48±0.98)%],the level of CD4+ CD25+ regulatory T cells in sepsis group(10.31±2.32)%,severe sepsis group(14.27 43.33)%,septic shock group(15.32±3.98)% had a significant increase(P<0.05 or<0.01).The log value of regulatory T cells in each group correlated positively with the APACHE Ⅱ scores(r=0.829,P=0.032;r=0.868,P=0.021;r=0.913,P=0.009),and the total coefficient of correlation was 0.903(P=0.013).Conclusion The level of CD4+CD25+ regulatory T cells in peripheral blood in septic patients has an abnormal increase,and their levels are related with the severity of disease.
3.Morphological and Chemical Variation of Prunella vulgaris Populations from Different Locations in China

Objective To investigate the variation of chemical characteristics with environmental factors and establish a relationship betweem morphological characters and chemical composition of Prunella vulgaris collected in different areas of China.Methods Twelve phenotypic traits and three chemical compositions were assessed in 28 populations of P.vulgaris collected from different locations in China.Results The variability ranges observed at phenotypic and chemical levels were polymorphic.According to the morphological traits,28 populations of P.vulgaris could be grouped into six clusters,and two morpho-types could be clearly distinguished.Perceptible differences could be discerned in the plant height,leaf length,corolla length,calyx length,fruiting spikes length,and maturity period.Based on three kinds of components including ursolic acid,total flavonoids,and total polysaccharides,all populations could be identified as four types.Cluster Ⅳ showing high content of ursolic acid,total flavonoids,and total polysaccharides could be utilized to develop superior derivatives.Conclusion The variation of chemical characteristics is influenced by the genetic and environmental factors,such as soil,climate,longitude,and altitude.It provides a solid basis for efficiently evaluating qualities and establishing good agricultural practices for P.vulgaris.
4.Textual research on original plant and dietotherapy history of Prunella vulgaris.

OBJECTIVETo investigate the original plant and dietotherapy history of Prunella vulgaris by textual researches, and provide a theoretical basis for clinical medication, exploitation and protection of wild resources.
METHODThe ancient literatures in the graphic, geographical distribution and morphological description of P. vulgaris were used in this studied.
RESULTThe varieties of P. vulgaris for therapeutic were no confusion. However, there has existed confusion between P. vulgaris and its confusable species (Ajuga ciliate, A. decumbens and Lagopsis supina) with appellations, morphological description and attached drawing of original plant.
CONCLUSIONThe original plant is originated from two species (P. vulgaris, P. asiatica). P. hispida was used as a certified medicinal material in Yunnan province from Ming Dynasty to modern time. The dietotherapy history of P. vulgaris in China can be traced back to Ming Dynasty or before.

5.Establishment of methods for producing rat models of sepsis-associated encephalopathy

Objective To establish sepsis-associated encephalopathy (SAE) animal models by using neurobiology score,electroencephalography (EEG),somatosensory evoked potentials in order to provide evidence for early clinical diagnosis of SAE.Methods A total of 30 rats were weighted,numbered,and monitored with EEG electrodes 10 days before modeling.Ten days later,rats were weighted,numbered,and divided randomly (random number) into groups.Rat models of sepsis were made by cecal ligation and puncture (CLP).The changes of their neurological behaviors were observed and EEG was used to monitor at 4,6,8,12 and 24 hours after CLP.The changes of EEG waveform and somatosensory evoked potentials were analyzed and recorded.Rat models of sepsis were divided into sepsis + non-SAE group and SAE group based on the presence or absence of EEG or somatosensory evoked potentials changes ~thin 24 hours.Rats were sacrificed 24 hours later,and histopathological changes of brain tissue were observed under electronic microscopy.Thus,the feasibility of establishing early SAE animal model by monitoring the changes of neurological behaviors,EEG and somatosensory evoked potentials was evaluated.Results SAE could be early diagnosed by using neurobiology score,reduced α wave and markedly increased δ wave on EEG,reduced amplitudes of evoked potentials P1,and significantly prolonged latency of S-P1 and NI-P1.In survived septic rats,6 had changes on neurological behaviors,EEG and somatosensory evoked potentials,and thus were diagnosed as SAE.The incidence of SAE was 46%.Conclusions SAE can be diagnosed in early stage by using neurobiology score,EEG and somatosensory evoked potentials,confirming the SAE rat models to be successfully established.
6.Mental Health Status of Old People in Endowment Institutions and the Influencing Factors in Shandong Province

Objective:To study the mental health of old people who live in endowment institutions in Shandong Province and its influencing factors. Methods: A sample of 2200 old people over 60 years old was selected with the stratified random sampling method. They were investigated using the self-designed questionnaires, symptom checklist (SCL-90), and Social Support Rating Scale. A total of 2013 questionnaires were recovered effectively (91. 5%), and 271 questionnaires were analyzed in this study. Results:41. 7% of old people in endowment insti-tutions in Shandong Province is positive in SCL -90 . Multivariate analysis showed that self -care ability ( P =0 . 045 ) and type of chronic disease ( P=0 . 025 ) associated with mental health problems of old people in Shandong Province. Conclusion:The mental health status of old people in endowment institutions in Shandong Province was not optimistic and its influencing factors included self-care ability and the type of chronic disease. It should take measures to promote successful aging in Shandong Province.
7.Distribution and drug resistance of Acinetobacter baumannii in our hospital during 2012-2013

Objective To understand the clinical distribution and drug resistance of Acinetobacter baumannii during 2012-2013 to provides the theoretical basis for clinical rational drug uses and the prevention and control of hospital infection.Methods To un-derstand the clinical distribution and drug resistance of Acinetobacter baumannii during 2012-2013 to provides the theoretical basis for clinical rational drug uses and the prevention and control of hospital infection.Results The main sources of specimens were sputum(88.7%),followed by secretions (4.6%).Acinetobacter baumannii infection occurred mainly in emergency ICU,surgical ICU,nerve internal medicine department,etc.Acinetobacter baumannii had the high sensitivity to carbapenem.Multiple drug resist-ance was decreased from 47.2% in 2012 to 46.4% in 2013,pan-drug resistance was decreased from 33.3% in 2012 to 27.8% in 2013.Conclusion Multiple drug resistance of Acinetobacter baumannii is serious,there is an upward trend in resistance to common-ly used antibacterial drugs,clinic should reasonably use antibacterial drugs based on the results of drug susceptibility test.
8.Textual research on change of medicinal parts and herbal medicine of Prunella vulgaris.

OBJECTIVETo investigate the herbal medicine of Prunella vulgaris by textual researches, and provide a theoretical basis for clinical medication, exploitation and protection of wild P. vulgaris resources.
METHODTextual research on medicinal works of past dynasties and field work were adopted.
RESULTThe natural distributions of P. vulgaris were concentrated distribution in Sichuan province, Huaihe river basin, and Middle-Lower Yangtze river valley in Chinese history. The indications of P. vulgaris in ancient and modern times were basically identical. While there were difference between the medicinal parts, harvest period and processing methods existed difference between ancient and modern.
CONCLUSIONThree periods that whole grass of P. vulgaris as medicinal parts (from late Ming dynasty to late Qing dynasty and early stage of Republic of China), both whole grass and spicas as medicinal parts (from mid-term Republic of China to 1963), and the semi-maturity or maturity of spicas as medicinal parts (from 1963 to today) existed. The processing method for medicinal parts of P. vulgaris adopted sun drying and shady drying in ancient China, but only the sun drying was only used in modern times.

9.Virulence gene detection and homology analysis of Klebsiella pneumoniae inducing liver abscess

ObjectiveTo investigate the serotype and virulence genes of Klebsiella pneumoniae capsule for in-hospital liver abscess, as well as the homology of these strains. MethodsA total of 26 non-repetitive strains of Klebsiella pneumoniae isolated from the patients with liver abscess in The First Affiliated Hospital of Jiamusi University from October 2018 to October 2019 were collected. These strains were identified to be Klebsiella pneumonia by Vitek-2 Compact automatic microbiological analyzer. The string test was performed for these strains; PCR was used to determine the major capsular serotypes and related virulence genes; multi-locus sequence typing was used to analyze the homology of the strains. The Fisher’s exact test was used for comparison of categorical data between groups. ResultsThe positive rate of all 26 strains of Klebsiella pneumoniae was 100% in the string test, with 17 strains of K1 type, 5 strains of K2 type, 1 strain of K5 type, 1 strain of K57 type, and 2 strains with unknown serotype. The virulence genes rmpA, aero, and ureA had a positive rate of 100% (26/26); uge and mrkD had a positive rate of 96.2% (25/26); fimH had a positive rate of 80.8% (21/26); iucB had a positive rate of 73.1% (19/26); wcaG, magA, and kfu had a positive rate of 65.4% (17/26); allS had a positive rate of 61.5% (16/26); kpn had a positive rate of 30.8% (8/26); iroNB had a positive rate of 7.7% (2/26). The cf29a gene was not detected; wcaG, magA, and allS were only detected in K1 serotype; uge was not detected in K57 serotype. Multi-locus sequence typing found 17 trains with ST23 type, 3 strains with ST86 type, 2 strains with ST65 type, 2 strains with ST1934 type, 1 strain with ST485 type, and 1 strain with ST592 type. ConclusionK1 and K2 serotypes are the main serotypes in the strains in this experiment, and ST23 type is the main sequence type for infection in our hospital.
10.Analysis of the Influencing Factors and Countermeasures on the Subjective Well-being of Female Elderly in Urban Community

Objective:To explore the subjective well-being and its influencing factors of female elderly in ur-ban community in Shandong province. Methods: Used the multi-stage stratified sampling method to extract 449 elderly women in 6 communities of 18 cities in Shandong Province. The basic situation and Subjective well-being of their were investigated by using the self-designed questionnaire and Memorial University of Newfoundland Scale of Happiness ( MUNSH) . Results:The economic situation, the number of children, living conditions, self-care ability, chronic disease severity, and other variables have a significant effect on the subjective well-being of fe-male elderly. Conclusions:To improve the economic situation of the urban community elderly women, pay atten-tion to their physical and mental health level, improve the pension and health insurance system, intensify the con-struction of old-age care institutions, playing the roles of family, children, society in the network to support the community elderly women society, can significantly improve the community elderly women′s subjective well-be-ing.
