1.THE SEPARATION OF HYDROCARBONS AND GLYCERIDES IN LIVER OIL OF MUSTELUS GRISEUS BY HPLC

A new method of the separation of hydrocarbons and glycerides in liver oil of Mustelus jriseas was designed. The Waters Prep-500A Liquid Chromatograph with silica gel column and differential refractive index detector was used. The mobile phase was petroleum ether and ethyl acetate. The pure hydrocarbons' and glycerides were obtained from the liver oil.
2.Separation and identification of squalene in liver oil from Mustelus griseus

The pure squalene(99.9%) was separated from liver oil of Mustelus griseus(Pietschmann) by TLC on silver nitrate-impregnated silica gel. The structure of squalene was confirmed by MS, ~1H-NMR and ~(13)C-NMR techniques.
3.Pigmented Villonodular Synovitis of the Knee Joint:Imaging Manifestations

Objective To explore the imaging characteristics of pigmented villonodular synovitis(PVNS) of knee joint.Methods X-ray,CT and MR imaging features of PVNS of knee confirmed by operation,arthroscope and pathology were retrospectively analysed with literatures review.Results X-ray could only show the swelling of the joint cpasules,the destruction,defect and sclerosis of the bone under the articular surface.CT could show the soft tissue nodules in joint cavity projected from joint capsule.MRI was sensitive in showing the thickened synovium of joint and joint effusion,the soft tissue nodules and thickened synovium of joint were iso-or low signal intensity on T1WI,isointensity in one and low or slight hyperintensity in 4 on T2WI.Multiple haemosiderin deposit nodules were seen in the thickened synovium,which were low signal intensity on T1WI and T2WI in 4 cases,and the nodules were enhanced obviously on contrast-enhanced MR images.Conclusion MRI possesses obviously in diagnosis of PVNS of knee joint than X-ray and CT,which is helpful in diagnosis and differential diagnosis of PVNS of knee joint.
4.An evaluation of isoptin for kidney transplant recipients

Objective To evaluate the use of isoptin in kidney transplants recipients. Methods The graft acute rejection rate,the cyclosporin A daily dose and its trough level and toxicity,the blood pressure and cardiovascular complications,the serum creatinine and blood sugar were studied in cadaveric kidney transplant recipients both with and without the additional use of isoptin. Results With the additional use of isoptin in 44 cadaveric kidney transplant recipients,the acute rejection rate has not increased.After using isoptin for 2 weeks,CsA blood level has been increased 25.5% whereas the CsA dose been cut down 32.6 %.So,drug expense dropped about 30.0% and CsA toxicity (hand tremble,insomnia) was slightly decreased. Conclusions Isoptin does not inecease the graft acute rejection rate yet makes the CsA dose and its toxicity less.
5.Clinical significance of microRNA-155 peripheral blood and CD4+T cytological test for patients with atopic dermatitis

Objective:To study the expression level of peripheral blood mircoRNA-155 and the variation characteristics of CD4+T cell percentage and to expound the clinical significance of mircoRNA-155 quantitative test and CD 4+T cell percentage cytological test.Methods:Fluorescent quantitative PCR had been used to detect the expression level of mircoRNA -155 in the peripheral blood.Flow cytometry had been used to detect the cell percentage of the peripheral blood T lymphocyte subset CD 3+T cell,CD4+T cell and CD8+T cell.The mircoRNA-155 expression levels and the cell percentages of CD 4+T were tested respectively in the selected 40 patients with atopic dermatitis ,30 patients with skin eczema and 30 healthy controls.Finally,the differences of the above groups were counted and analyzed .Results:The relative expression levels of mircoRNA-155 of the atopic dermatitis group were higher than those of the eczema group and the normal control group ( P<0.05 ).The CD4+T cell percentage of the atopic dermatitis group was higher than those of the eczema group and the normal control group ( P<0.05 ) .Conclusion: The expression level of the peripheral blood mircoRNA-155 in the patients with atopic dermatitis is increased significantly and CD 4+T cell percentage in the patients is raised as well.Consequently ,the clinical detection of miRNA-155 and CD4+T cell shall be of great significance .
6.Treatmant of no-reflow phenomenon during percutaneous coronary intervention

Objective To evaluate the effect of injecting UK and Herbesser into the target coronary artery where no-reflow happened during percutaneous coronary intervention. Methods Among the 820 acute coronary syndrome (ACS) patients who underwent PCI from Jan. 1990 to Apr. 2004, no-reflow phenomenon occurred in 86 patients. Three hundred cases with acute myocardial infraction (AMI) underwent primary PCI, no-reflow phenomenon occurred in 50 cases. In the other 520 unstable angina pectoris (UAP) cases, no-reflow phenomenon occurred in 36 cases during PCI. All the 86 patients with no-reflow were given intra-coronary arteries nitroglycerin but 78 of them showed no improvement in coronary perfusion and were in turns devided into 3 groups randomly. Group A (n=26) was given intra-vessel UK and Herbesser alternately. The maximan dosage of UK was 2?105 U kIU and 2 mg for Herbesser. Group B (n=24) was given intra-coronary Herbesser 500 ?g each time until the maximum amount reached 2 mg. Group C (n=28) was given UK which maximun dosage was 2?105 U. Objective To explore the effect of oral care on the prevention of ventilator associated pneumonia in patients with severe traumatic brain injury by aspiration of subglottic secretion combined with 2% acetic acid chloride solution. Methods A total of 100 cases of ICU patients with mechanical ventilation for longer than 48 hours were divided into group B and group C using a random number table method according to tracheotomy mechanical ventilation time sequence number from January 2014 to June 2015. Another 50 ICU patients in group A (as the case for retrospective analysis, from April to November , 2013) were treated by routine tracheal suction, normal saline cotton for oral care, group B were treated with aspiration of subglottic secretion, normal saline cotton for oral care, group C were treated with aspiration of subglottic secretion combined with 2% acetic acid chloride solution for oral cleaning. And then VAP occurrence rate, plaque index, plaque clearance were evaluated. Results The incidence rates of early onset VAP in three groups were 14% (7/50), 8% (4/50), 4% (2/50), and incidence rates of late onset VAP were 22% (11/50),12% (6/50), 8% (4/50) respectively, the differences were significant (χ2=3.65, 3.88,P<0.01). The plaque index and plaque clearance rates of the three groups on the third day were 2.14± 0.35, (40.12 ± 13.11)%, 2.10 ± 0.33, (39.17 ± 14.21)%, 1.03 ± 0.24, (60.12 ± 15.07 )%(F=12.21,χ2=3.82,P<0.01);while on the 7th day, they were 1.76 ± 0.47, (41.12 ± 13.34)%, 1.80 ± 0.32, (39.37 ± 12.21)%, 0.99 ± 0.15, (60.33 ± 14.25)%(F=13.14,χ2=4.20,P<0.01). Conclusions The incidence rate of VAP can be decreased by the combination of aspiration of subglottic secretion and oral cleaning with 2%acetic acid chloride solution.
8.Fas ligand and Anti-human DR5 monoclonal antibody induce tumor cell lines apoptosis

Objective:To study the cytotoxic effects on tumor cell lines Hela,BGC823,MCF-7,L342,H9101,D6 induced by Fas ligand and anti-human DR5 monoclonal antibodies(Anti-DR5 mAb) and the underlying mechanism.Methods:Fas/DR5 mRNA were detected by RT-PCR. Cytotoxicity exerted by FasL/Anti-DR5 mAb on tumor cell lines was measured by MTT colorimetry and the induced apoptosis was determined by agarose gel electrophoresis.Results:The expression of DR5 on BGC823 and Hela cells were higher and DR5 didn’t express in D6. The expression of Fas on H9101 and L342 were higher and Fas didn’t express in D6. Cell line H9101 and L342 were sensitive to Anti-DR5 mAb and FasL in a dose dependent manner; Cell line MCF-7 and BGC823 were sensitive to FasL but were partially sensitive to Anti-DR5 mAb; Cell line Hela was partially sensitive to FasL but was sensitive to Anti-DR5 mAb; Cell line D6 was insensitive to two apoptosis inductions.Conclusion:Apoptosis induced by Fas ligand and Anti-DR5 mAb vary among tumor cell lines. The underlying mechanism may be relevant to Fas/DR5 mRNA expression,the release of Caspase-8 and Bcl-2.
9.The protective effects of Qiqiong Capsule on focal cerebral ischemia/reperfusion injury

ObjectiveTo study the protective effects of Qiqiong(QQJN) on focal cerebral ischemia/reperfusion injury and its mechanism. MethodsMiddle cerebral artery occlusion(MCAO) was used to make focal cerebral ischemia/reperfusion model by intravascular nylon filament occlusion. The protective effects of QQJN were evaluated by investigating neurological function score, percentage of cerebral infarction, pathomorphology of brain, the activity of SOD and the content of MDA in hrain tissue,thrombogenesis and platelet aggregation in vitro. ResultsCompared with model group, QQJN(4.4、8.8g/kg)could decrease the neurological score in 8 and 22h after reperfusion, reduce the percentage of cerebral infauction,improve pathomorphology of brain, decrease the length, wet weight and dry weight of thromb and inhibit platelet aggregation. ConclusionQQJN had protective effects on focal cerebral ischemia/reperfusion injury. The role of anti-injury of free radicals,inhibit thrombogenesis and platelet aggregation should contribute to its neuroprotective effects.
10.Purification and characterization of esterase from Morganella morganii ZJB-09203.

Enantioselective hydrolysis of 2-carboxyethyl-3-cyano-5-methylhexanoic acid (CNDE) is the key step in chemoenzymatic synthesis of pregabalin. We purified an intracellular carboxyl esterase from Morganella morganii ZJB-09203, which exhibited high enantioselectivity and activity towards CNDE. The carboxyl esterase was purified to electrophoretic homogeneity by ammonium sulfate fraction precipitation, Phenyl Sepharose 6 FF hydrophobic interaction chromatography, anion exchange with DEAE Sephadex A-50 and Bio-Scale CHT column. The purified enzyme was a monomer with molecular mass of 68 kDa determined by SDS-PAGE and gel chromatography. Substrate specificity of the enzyme towards p-nitrophenyl esters suggested that the purified enzyme was an esterase. The optimal reaction pH for CNDE hydrolysis was 9.0, and optimal temperature was 45 degrees C. The esterase was stable between pH 7.0 and 9.0, and at 40 degrees C. The enzyme activity was enhanced by Ca2+, Cu2+ and Mn2+, whereas strongly inhibited by Co2+, Fe3+, Ni2+ and EDTA. Meanwhile, we investigated the kinetic parameters of the esterase towards p-nitrophenyl esters and effect of CNDE concentration on conversion. The present study reported the esterase capable of stereospecific hydrolysis of CNDE for the first time. Our research will provide foundations for industrial production of Pregabalin using the new biocatalyst.
