1.EFFECT OF RSV M_(2-1) GENE EXPRESSION ON THE GROWTH OF HUMAN LUNG ADENOCARCINOMA PAa IN VIVO AND VITRO

To explore the the effect of RSV M 2-1 gene expression on the growth of human lung adenocarcinoma PAa cell line, the recombinant RSV M 2-1 gene eukaryotic plasmid PXJ 41/ M 2-1 was transfected to the human lung adenocarcinoma PAa cell line. Expression of the M 2-1 gene was examined by RT PCR and Western blot. The growth of the human lung adenocarcinoma PAa cell was observed by MTT curve, flow cytometry, the capacity of inherence, colony forming units and inoculation of nude mouse. The results showed that ①The desired fragments of M 2-1 gene were digested by restriction enzyme and RT PCR, respectively. ②The bands of M 2-1 gene protein were found by Western blot. ③ The ratio of transfected PAa cells in S phase of cell cycle was decreased, but increased in G 2 /M phase. Colony forming efficiency and units of the transfected PAa cell were increased, but the capacity of its inherence was decreased. ④Tumorogensising time of the transfected PAa cell in nude mouse was delayed, but its growing speed was increased. The tumor tissue transformed to some extent from adenocarcinoma to squamous carcinoma was found in the nude mouse. It is suggested that the M 2-1 gene may promote the growth of PAa cells, which may implicate some relationship between RSV and lung cancer.

Objective To evaluate the effect of direct stenting on no-reflow in patients with ST-segment elevation (STEMI) acute myocardial infarction. Methods A total of 157 patients with STEMI received direct stenting (DS,n=85) or conventional post-dilation stenting (CS,n=72). The time of X-ray exposure, the amount of contrast media consumed, the TIMI grading after stenting and ST-segment changes of electrocardiogram were compared between two groups. Objective To evaluate the effect of injecting UK and Herbesser into the target coronary artery where no-reflow happened during percutaneous coronary intervention. Methods Among the 820 acute coronary syndrome (ACS) patients who underwent PCI from Jan. 1990 to Apr. 2004, no-reflow phenomenon occurred in 86 patients. Three hundred cases with acute myocardial infraction (AMI) underwent primary PCI, no-reflow phenomenon occurred in 50 cases. In the other 520 unstable angina pectoris (UAP) cases, no-reflow phenomenon occurred in 36 cases during PCI. All the 86 patients with no-reflow were given intra-coronary arteries nitroglycerin but 78 of them showed no improvement in coronary perfusion and were in turns devided into 3 groups randomly. Group A (n=26) was given intra-vessel UK and Herbesser alternately. The maximan dosage of UK was 2?105 U kIU and 2 mg for Herbesser. Group B (n=24) was given intra-coronary Herbesser 500 ?g each time until the maximum amount reached 2 mg. Group C (n=28) was given UK which maximun dosage was 2?105 U. 8.Three-dimensional transvaginal ultrasonography in diagnosis of intrauterine adhesion

Seventy eight patients suspected of intrauterine adhesion (IUA) were scanned with two-dimensional (2D) and three-dimensional (3D) transvaginal uhrasonography, the results were analyzed with hysteroscopic findings as the gold standard. The sensitivity, specificity, accuracy, positive predictive value and negative predictive value of three-dimensional transvaginal ultrasonography in diagnosis of IUA were 97% (58/60), 18/18, 97% (76/78), 100% (58/58) and 90% (18/20). Three-dimensional transvaginal ultrasonography was more accurate in diagnosis of IUA than two-dimensional ultrasonography ( P < 0. 01 ),and the IUA classification and grading with three-dimensional transvaginal ultrasonography was better correlated with hysteroscopic findings.

Objective To explore the prognostic value for cardiovascular events of elevated NT-proBNP in coronary heart disease(CHD) patients with metabolic syndrome(MS).Methods The level of NT-proBNP was measured in 165 CHD patients with MS diagnosed by coronary angiography.After 2 years follow-up,we recorded the corresponding cardiovascular events in those patients.Results The NT-proBNP level was much higher in ACS group than that in SCHD group( P < 0.01 ).There were totally 45 events after 2 years.After adjustment of traditional risk factors including hypertension,diabetes,smoking history and high sensitivity C reactive protein( hs-CRP),Cox proportional hazard analysis indicated that high NT-proBNP level( > 111.58ng/L)was an independent risk factor for future events (relative risk 2.67,P <0.01 ).Conclusion Elevated NT-proBNP level might be one of the independent risk factor for cardiovascular events in CHD patiens with MS.
10.Fas ligand and Anti-human DR5 monoclonal antibody induce tumor cell lines apoptosis

Objective:To study the cytotoxic effects on tumor cell lines Hela,BGC823,MCF-7,L342,H9101,D6 induced by Fas ligand and anti-human DR5 monoclonal antibodies(Anti-DR5 mAb) and the underlying mechanism.Methods:Fas/DR5 mRNA were detected by RT-PCR. Cytotoxicity exerted by FasL/Anti-DR5 mAb on tumor cell lines was measured by MTT colorimetry and the induced apoptosis was determined by agarose gel electrophoresis.Results:The expression of DR5 on BGC823 and Hela cells were higher and DR5 didn’t express in D6. The expression of Fas on H9101 and L342 were higher and Fas didn’t express in D6. Cell line H9101 and L342 were sensitive to Anti-DR5 mAb and FasL in a dose dependent manner; Cell line MCF-7 and BGC823 were sensitive to FasL but were partially sensitive to Anti-DR5 mAb; Cell line Hela was partially sensitive to FasL but was sensitive to Anti-DR5 mAb; Cell line D6 was insensitive to two apoptosis inductions.Conclusion:Apoptosis induced by Fas ligand and Anti-DR5 mAb vary among tumor cell lines. The underlying mechanism may be relevant to Fas/DR5 mRNA expression,the release of Caspase-8 and Bcl-2.
