Objective The effect of different doses of ethylnitrosourea(ENU)and cyclophosphamide(CP)on the loss rate of CD59 on peripheral blood erythrocytes was explored to optimize the detection method of Pig-a gene mutation. Methods According to the weight and loss rate of CD59 on peripheral blood erythrocytes, rats were divided into 4 groups:the control group,CP 40 mg/kg group,ENU 10 mg/kg group and ENU 40 mg/kg group(n=6). The control group was injected i.p. with PBS,other groups were injected i.p. with corresponding solutions. The body weight of rats on days 0,7,14,21, 28, 42 and 56 were recorded. At the same time, blood samples were collected and incubated with antibodies, and the loss rate of RBCCD59-was detected by flow cytometry. Results Compared with the control group, at different time points, the body weight and weight gain of ENU 10 mg/kg group and ENU 40 mg/kg group had no statistically significant difference(P > 0.05),while those in the CP 40 mg/kg group were significantly decreased(P <0.05). The loss rate of RBCCD59-was significantly increased in the CP 40 mg/kg group at 28,42 and 56 days, ENU 10 mg/kg group at 42 and 56 days, and ENU 40 mg/kg group at 7,14,21,28,42 and 56 days,(P < 0.05). The results showed a dose-response relationship. Conclusions Under the conditions of this Pig-a mutation detection method,ENU is superior to CP on raising loss rate of RBCCD59-,ENU 40 mg/kg is better than 10 mg/kg,and 28 days is suitable as the test period.

OBJECTIVE:
To study the changes of vestibular function in patients with diabetes mellitus and its clinical significance.
METHOD:
Electronystagmography (ENG) was used to examine 76 patients with diabetes mellitus and 60 healthy adults subjects. After clinical detection of vestibular function including spontaneous nystagmus, positional test, head shaking nystagmus, neck torsion test, caloric test, and sensory organization tests which consist of gaze, saccade and smooth pursuit test, the results of these two groups were recorded for qualitative and quantitative statistical analysis.
RESULT:
The rate of vestibular dysfunction in patients with diabetes mellitus were 68.4%. and that of the controls were 8.3%. There was significant difference between these two groups (chi2 = 15.472, P < 0.01).
CONCLUSION
Vertigo or dizziness occurred in patients with diabetes mellitus might be related to vestibular dysfunction. ENG test could be used as one of the objective clinical examinations in patients with diabetes mellitus.

Objective:To investigate the clinical features of frequent acute exacerbations of chronic obstructive pulmonary disease(COPD)in the elderly. Methods:A total of 93 elderly patients with stable COPD who were followed up in Bethune Hospital Affiliated to Shanxi Medical University from August 2015 to September 2016 were retrospectively analyzed. They were divided into the frequent acute exacerbation group(n=45)and the non-frequent acute exacerbation group(n=48). Data on the body mass index(BMI), lung function, the smoking index(package year), the respiratory status questionnaire and serum creatinine levels were collected.Logistic regression analysis was used to analyze the influencing factors for the frequency of acute exacerbations of COPD in elderly patients. Results:The BMI was lower [19.59(18.08-23.50)kg/m 2vs. 23.36(21.14-25.46)kg/m 2, Z=8.898, P=0.003], while scores of the modified Medical Research Council(mMRC)dyspnea scale, COPD Assessment Test(CAT)and smoking index were higher [3.00(1.00-3.00) vs. 1.00(0.00-1.00), 15.00(8.50-17.50) vs.10.00(6.00-13.75), 60.00(30.00-80.00)package years vs.37.50(3.75-60.00)package years, Z=17.671, 7.318 and 6.589, P<0.001, 0.007 and 0.010]in the frequent acute exacerbation group than in the non-frequent acute exacerbation group. The glomerular filtration rate(GFR)and percentage of predicted forced expiratory volume in the first second were lower in the frequent acute exacerbation group than in the non-frequent acute exacerbation group [105.13(89.84-114.69)ml·min -1·1.73m -2vs.121.74(93.89-142.02)ml·min -1·1.73m -2, 40.10(31.40-56.00)% vs.70.65(47.36-85.36)%, Z=6.071 and 17.814, P=0.014 and<0.001]. Logistic regression analysis showed that the odds ratios( OR)with 95% confidence interval(95% CI)of the smoking index and GFR were 1.025(1.006-1.044)and 0.957(0.927-0.995)( P=0.009 and 0.025), respectively, indicating that with the increase of smoking index and the decrease of GFR, the risk of frequent exacerbations of COPD was increased. Conclusions:There are phenotypic differences between elderly COPD patients with frequent acute exacerbations and without frequent acute exacerbations.Elderly COPD patients with frequent acute exacerbations have a lower BMI index, a higher smoking index, more clinical symptoms, worse pulmonary function, and are prone to early kidney injury.

Objective:To investigate the effect of 125I-RSOAds-hTERT/PSA oncolytic adenovirus on targeted therapy of prostate cancer and its effect on tumor microenvironment. Methods:125I-RSOAds-hTERT/PSA ( 125I-virus complex) oncolytic adenovirus was constructed by PCR amplification and double restriction enzyme ligation. TUNEL staining, flow cytometry and Caspase-3 immunoblotting assay were used to detect the killing effect of 125I-RSOAds-hTERT/PSA oncolytic adenovirus on prostate cancer cells in vitro and in vivo, respectively. To explore the effect of 125I-virus complex on tumor tissue cytokine secretion levels, interleukin 2 (IL-2), IL-10, tumor necrosis factor-α (TNF-α) and interferon-γ (IFN-γ) in the culture supernatant of human prostate cancer cell line PC3, mouse prostate adenocarcinoma cell line RM-1, and mice serum were detected by ELISA. We explored the regulation of 125I-virus complex on the expression of CD24, CD44 and prostate stem cell antigen (PSCA) in prostate tumor tissues and tumor cells through immunohistochemistry. Meanwhile, the expression levels of CD32 and vascular endothelial growth factor (VEGF), as well as CD4+ , CD8+ and macrophage infiltration in tumor tissue were detected through immunofluorescence experiments. Results:125I-virus complex oncolytic adenovirus significantly increased tumor cell apoptosis in vitro and in vivo that was significantly higher than that of 125I group and virus complex group. Meanwhile, IL-2 ( t=-183.30, -38.20, P<0.05), IL-10 ( t=113.80, 92.71, P<0.05), TNF-α ( t=-73.20, -73.91, P<0.05), IFN-γ ( t=-65.37, -139.70, P<0.05) increased in vitro and in vivo. 125I-virus complex reduced the expression of CD24, CD44 and PSCA in tumor cells and tumor tissue, reduced the weight of tumor tissue, inhibited angiogenesis of tumor tissue ( t=8.55, P<0.05), and regulated the immune response in tumor tissue. Conclusions:125I-virus complex targeting prostate cancer can significantly kill cancer cells, reduce the weight and angiogenesis of tumor, and improve tumor microenvironment.

AIM:To test and compare the perceptual eye position and fixation stability of adolescents with emmetropia and adolescents with low myopia, investigating the characteristics of the perceptual eye position and fixation stability of adolescents with low myopia.METHODS: Cross-sectional study. A total of 132 adolescents(264 eyes)who visited in the ophthalmology clinic of our hospital from April to December 2023 were randomly selected as the research subjects. Participants were categorized into normal control group(n=45, 90 eyes), simple low myopia group(n=45, 90 eyes)and low myopia with anisometropia group(n=42, 84 eyes)according to their refractive status and were underwent assessments for perceptual eye position and fixation stability.RESULTS: Compared with the normal control group, the static and dynamic horizontal perceptual eye position deviation of the simple low myopia group and the low myopia with anisometropia group were significantly increased(P<0.05). Compared with the simple low myopia group, the static and dynamic horizontal perceptual eye position deviation of the low myopia with anisometropia group were significantly increased(P<0.05). There was no significant difference in static and dynamic vertical perceptual eye position deviation among the three groups(P>0.05); compared with the normal control group, the horizontal and vertical fixation stability of the simple low myopia group and the low myopia with anisometropia group were significantly worse(all P<0.01), but there was no differences in the simple low myopia group and the low myopia with anisometropia group(P >0.05).CONCLUSION: Abnormalities are observed in perceptual eye position and fixation stability function in adolescents with low myopia compared with those adolescents with emmetropia, even at best corrected visual acuity. The occurrence of anisometropia could lead to an increased degree of horizontal perceptual eye position displacement.
