1.Studies on diversity of diosgenin in different part of rhizome of Dioscorea zingiberensis in various growing period

ObjectTo study the accumulation and distribution of dioscin in the different part of rhizome of Dioscorea zingiberensis C. H. Wright and the differences of its diosgenin. Methods Histochemistry has been used to study the accumulation and distribution of dioscin in the different part of rhizome and diosgenin was determined by HPLC. Results Dioscin distributed mainly in ground tissue. The amount of dioscin accumulated in the plant part from the region distributed with small vascular bundle (SVB) is the most abundant, and the diosgenin is also the highest in the same region; both of them in the region with no vascular bundle (NVB) are next to that of SVB; the amount of dioscin accumulated in the region distributed with big vascular bundle (BVB) is the lowest, and the diosgenin is the lowest in the same region too. Conclusion Diosgenin in every region of the three of male rhizome is higher than that of female rhizome and diosgenin in every region of the three of monoecism is between those of male and female rhizome.
2.INHIBITORY EFFECTS OF INDOMETHACIN AND MELOXICAM ON NF-κB IN MOUSE PERITONEAL MACROPHAGES

AIM To study the inhibitory effects of indomethacin and meloxicam on NF-κB from lipopolysaccharide (LPS) stimulated peritoneal macrophages of mice. METHODS NF-κB was measured with the method of electrophoretic mobility shift assay (EMSA). RESULTS After induction by LPS at the concentrations of 1 and 3 μg.mL-1, the NF-κB content of the mouse peritoneal macrophages increased markedly. Indomethacin and meloxicam, at the concentrations of 10-7-10-5 mol.L-1, decreased the activation of NF-κB at the concentrations of 1 and 3 μg.mL-1 in activated mouse peritoneal macrophages induced with LPS at the concentrations of 1 and 3 μg.mL-1. CONCLUSION The inhibitory effects of indomethacin and meloxicam on NF-κB activation may be one of their mechanisms of antiinflammatory actions.
4.The application study of multi-b value diffusion weighted imaging in preoperative histological grading of supratentorial glioma

Objective To investigate the optimal b value of diffusion-weighted imaging (DWI)in predicting the grade of cerebral gliomas.Methods 38 patients with pathologically-proved brain gliomas (24 high-grade gliomas and 14 low-grade ones)were studied retrospectively.All patients received conventional MRI and DWI examination with three different b values (1 000 s/mm2 ,2 000 s/mm2 and 3 000 s /mm2 ).The signal features of the tumor on DWI with three different b values were evaluated and compared.The minimum ADC values of the tumors were calculated and compared between each other.Through drawing the ROC curve of different b values, the best diagnostic threshold was found.The sensitivity and specificity in predicting the grade of brain gliomas were assessed using Chi-square test.Results On DWI with b value of 3 000 s/mm2 ,91.6% (22/24)of high-grade tumors showed hyper-intensity, while 85.7%(12/14)of low-grade tumors presented hypo-intensity signals.Regarding hyper-intensity signals as diagnostic criterion for high-grade tumors,the sensitivity and specificity were 91.6% and 100% respectively.When the b value was similar,the minimum ADC value of high-grade glioma was significantly lower than that of low-grade glioma,and statistically significant differences in differentiating high-grade glioma from low-grade glioma existed among three b values (P <0.05).When the ADC 3 000 value<0.74×10 -3 mm2/s was regarded as a standard for the identification of high-grade and low-grade glioma,the sensitivity and specificity were 100% and 87.3%respectively.Conclusion DWI with high b value is more useful than standard moderate b value in preoperative grading the gliomas.When DWI with b value of 3 000 s/mm2 is used,the minimum ADC value will provide quantitative indicators in preoperatively precisely predicting grading glioma.
5.Reason Analysis and Countermeasures of 747 Cases of Medical Safety (Adverse)Events

OBJECTIVE:To provide reference for avoiding the recurrence of medical safety(adverse)events or inducing more severe medical accident. METHODS:The medical safety(adverse)events reported by a class A grade 3 hospital from July 2013 to March 2016 were studied retrospectively,and excluded purely adverse drug reactions and transfusion reactions to analyze the rea-son and characteristics. RESULTS:Of 747 events,there were 578 medical,nursing and drug-related medical safety (adverse) events,accounting for 77.38%;among 175 medical related medical safety(adverse)events,top 3 reasons were basic skills defi-ciencies,defects in the system processes,inadequate communication and education, accounting for 32.00%,24.00%,19.43%. The occurrence of medical safety (adverse) events were closely related to working life and qualifications;among 578 medical, nursing and drug-related medical safety (adverse) events,the incidence of adverse events was 71.11% in 5-year working life group,and that was 73.18% in junior titles group. CONCLUSIONS:Various professionals should strictly implement procedure and regulations,strengthen communication and education in daily work,and improve professional knowledge and skills so as to prevent the occurrence of adverse event completely.
6.Detection of virus from throat swabs of patients with respiratory tract in-fection

Objective To investigate the detection of respiratory tract virus from throat swabs of patients with lower respiratory tract infection(LRTI)in a hospital,so as to provide reference for clinical diagnosis and treatment. Methods Throat swabs of hospitalized children with LRTI between May 2103 and April 2014 were detected by di-rect immunofluorescence assay,infection/carriage of respiratory syncytial virus(RSV),adenovirus(ADV),influen-za virus A(IV-A),IV-B,parainfluenza virus 1(PIV-1),PIV-2,and PIV-3 in children were surveyed.Results Of 8 425 throat swab specimens of hospitalized patients,1 879 were detected virus,the total detection rate was 22.30%,detection rate of RSV was the highest (11 .31 %),followed by PIV-3(4.37%).There was no significant difference in the total detection rate of respiratory tract virus between children of different genders (P =0.051).Of all age groups,detection rate of RSV in infants and young children was the highest,the detection rates of 7 kinds of virus from preschool children were approximate,detection rate of ADV from primary school children was the high-est (37.50%).In spring and winter,children mainly infected with RSA,accounting for 62.82% and 69.88% re-spectively;in summer,PIV-3 was the predominant virus,accounting for 41 .47%,in autumn,there was little difference in the percentage of each virus.Conclusion The detection rate of 7 kinds of virus isolated from throat swabs of hospitalized children with LRTI is related to age and season;the detection rate of 7 kinds of virus from in-fants and young children is high,RSA is the main virus.
7.Effect of Ginkgo Biloba extract on the expression of Activin A during the process of hepatic fibrosis

AIM:Activin A,a member of transforming growth factor superfamily,is the negative regulator factor in liver regeneration. In this study,the effects of extract of Ginkgo Biloba on hepatic fibrosis and the expression of Activin A in rats with cirrhosis were investigated. METHODS:The experiment was performed at in the Central Laboratory of Wuhan First Hospital from September 2005 to December 2006. ①Thirty-six male SD rats of(160?20) g were randomized into 3 groups:control group,model group and treatment group. ② Except the rats in the control group,others were intraperitoneally injected with 500 mL/L CCl4 for 8 weeks to establish models of hepatic fibrosis. Meanwhile,the extract treatment group was infused with the extract of Ginkgo Biloba(Chinese drugs preparation laboratory of Wuhan First Hospital,detected by Hubeu Wushi Medicine Industry Co.,Ltd. No. 02-391) daily for 8 weeks. ③After administration,all anesthetized rats were sacrificed. Blood samples were collected for the determination of liver function biochemical indexes. Liver tissue samples were used for histopathological examinations. The expression of Activin A was determined by immunohistochemistry and RT-PCR. RESULTS:All 36 rats were involved in the final analysis. ①The liver function in extract treatment group was significantly improved compared with that in model group. ②The grade of fibrosis in extract treatment group were remarkably lower than that in model group under light microscope. ③The positive staining of Activin A in treatment group was significantly reduced compared with model group. ④The expression of Activin A mRNA in extract treatment group was significantly reduced compared with model group. CONCLUSION:Extract of Ginkgo Biloba can effectively decrease the expression of Activin A in rats with hepatic fibrosis caused by CCl4,and lessen the degree of hepatic fibrosis.
8.Preparation and Application of Chlorogenic Acid Imprinted Solid Phase Extraction Materials Based on Multiwalled Carbon Nanotubes

A novel molecularly imprinted polymers (MIPs) was prepared based on multiwalled carbon nanotubes (MWNTs) by using chlorogenic acid as template, methacrylic acid(MAA) as the functional monomer, and ethylene glycol dimethacrylate (EGDMA) as the cross-linker. Infrared spectroscopy, scanning electron microscopy, and thermogravimetric analysis were employed to characterize the composite structure and morphology, the results showed that a stable imprinted layer was grafted on the MWNTs with the average thickness of 30-40 nm. The dynamic adsorption and static adsorption experiments were also executed and the results showed that two kinds binding sites with the maximum apparent binding constant 21.5 μmol/g and 32.7 μmol/g existed v.i. between the imprinted polymers and chlorogenic acid. As solid phase extraction (SPE) materials, the separation and enrichment of chlorogenic acid from honeysuckle were successful with a enrichment factor of 25.
9.Activation of sonic hedgehog signaling pathway in S-type neuroblastoma cell lines.

The effects of Sonic hedgehog (Shh) signaling pathway activation on S-type neuroblastoma (NB) cell lines and its role in NB tumorigenesis were investigated. Immunohistochemistry was used to detect the expression of Shh pathway components-Patched1 (PTCH1) and Gli1 in 40 human primary NB samples. Western blotting and RT-PCR were used to examine the protein expression and mRNA levels of PTCH1 and Gli1 in three kinds of S-type NB cell lines (SK-N-AS, SK-N-SH and SHEP1), respectively. Exogenous Shh was administrated to activate Shh signaling pathway while cyclopamine was used as a selective antagonist of Shh pathway. S-type NB cell lines were treated with different concentrations of Shh or/and cyclopamine for different durations. Cell viability was measured by using MTT method. Apoptosis rate and cell cycle were assayed by flow cytometry. The xenograft experiments were used to evaluate the role of Shh pathway in tumor growth in immunodeficient mice. High-level expression of PTCH1 and Gli1 was detected in both NB samples and S-type NB cell lines. Cyclopamine decreased the survival rate of the three cell lines while Shh increased it, and the inhibition effects of cyclopamine could be partially reversed by shh pre-treatment. Cyclopamine induced the cell apoptosis and the cell cycle arrest in G(0)/G(1) phase, while Shh induced the reverse effects and could partially prevent effects of cyclopamine. Cyclopamine could also inhibit the growth of NB in vivo. Our studies revealed that activation of the Shh pathway is important for survival and proliferation of S-type NB cells in vivo and in vitro through affecting cell apoptosis and cell cycle, suggesting a new therapeutic approach to NB.
10.Effects of rhein lysinate on the expressions of TNF-α,IL-6 and NF-κB in the kidney tissue of SAMP 1 0 mice

Objective To investigate the effects of rhein lysinate (RHL)on the expressions of TNF-α,IL-6 and NF-κB in the kidney tissue of senescence accelerated mouse prone 10 (SAMP 10)mice.Methods We selected 1 8 male mice (SAMP 1 0 )aged 7 months for the study and randomly divided them into blank control group and groups of different concentrations of RHL;six senescence accelerated mouse resistance 1 (SAMR 1 )served as the young control group.After 6 weeks’ treatment,HE staining was used to detect the pathological changes of the kidney.The expressions of TNF-α,IL-6 and NF-κB at the protein level were detected by immunohistochemistry and Western blotting.Results RHL treatment did not affect the body weight of SAMP 10 mice (P>0.05 ). Compared with SAMR 1 mice, contracted and destroyed renal glomeruli and infiltration of mononuclear macrophages were observed in control SAMP10 mice.However,this pathological process was blocked by RHL (25 mg/kg and 50 mg/kg ) treatments. In addition, the overexpressions of TNF-α, IL-6 and NF-κB and the phosphorylation of NF-κB in the kidney tissue of SAMP 10 mice could be inhibited by RHL treatments (P<0.05). Conclusion RHL inhibits the inflammatory reaction of the kidney tissue,which may be one of the mechanisms by which RHL exerts its kidney-protecting and anti-aging effects.
