1.Effect of Helicobacter pylori eradication on childhood acute idiopathic thrombocytopenic purpura.

OBJECTIVETo evaluate the effect of Helicobacter pylor (Hp) eradication in children with acute idiopathic thrombocytopenic purpura (ITP).
METHODSNinety-three children with acute ITP and Hp infection were divided into two groups and treated with prednisone and Hp eradication (group A, 51 cases) or with prednisone without Hp eradication (group B, 42 cases).
RESULTSThe Hp eradication rate was 94.1% in group A. No difference was found in the therapeutic effects on IPT between the two groups, but the recurrence rate in one year in group A was significantly lower than that in group B.
CONCLUSIONNHp eradication does not obviously enhance the therapeutic effect on childhood acute ITP, but can decrease the relapse rate in one year. HP eradication therapy is recommended in children with acute ITP and Hp infection.

2. Effect of Electroacupuncture at Different Acupoints on Steroid Hormones and Ovarian Androgen Receptor Expression in Polycystic Ovary Syndrome Rats

OBJECTIVE: To observe the effect of electroacupuncture (EA) at different acupoints on hyperandrogenism and ovarian androgen receptor (AR) expression in rats with polycystic ovary syndrome (PCOS), so as to explore its mechanisms underlying improvement of PCOS. METHODS: Sixty female SD rats were randomly divided into control, model, Zusanli (ST 36), Sanyinjiao (SP 6), Guanyuan (CV 4) and comprehensive groups (10 rats/group). The PCOS model was established by giving (gavage) the animals with Letrozole solution (1.0 mg/kg), once daily for 21 consecutive days. EA stimulation was applied to ST 36, SP 6, or CV 4 respectively, once daily for 14 days. The comprehensive group was treated by EA treatment of the abovementioned three acupoints in the meantime. Before and after the treatment, the body mass and the ovary weight were measured. The morphological structure of the ovary was observed under microscope after H.E. staining. The levels of serum hormone and sex hormone binding globulin (SHBG) were assayed by ELISA. Free androgen index (FAI) was calculated and the expression of AR in the ovary was detected by immunohistochemistry. RESULTS: After modeling, the body mass, ovary weight, serum testosterone (T) and FAI levels and the expression of AR protein in late ovarian follicles were significantly increased (P<0.01, P<0.05), and serum estradiol (E2) and SHBG contents were significantly decreased relevant to the control group (P<0.01). Following EA intervention and compared with the model group, the body weight of each EA group was significantly reduced (P<0.05), the ovarian pathological changes were improved, and serum T and FAI levels were significantly decreased (P<0.01), and serum E2 and SHBG contents significantly increased in the ST 36, CV 4 and comprehensive groups (P<0.01, P<0.05). The ovary weight of only CV 4 group was significantly lower than that of the model group (P<0.01). The expression of AR protein in late follicles of both CV 4 and SP 6 groups was markedly decreased (P<0.05, P<0.01). CONCLUSION: EA at ST 36, SP 6, and CV 4 all can improve the hyperandrogenism and ovarian polycystic morphological changes of PCOS rats, while different acupoints have different functional characteristics.

Cancer patients will have the feeling of become a burden to others in the treatment process, this feeling is called self-perceived burden. self-perceived burden will add to the psychological burden of patients, influence the patient's quality of life and treatment decisions for cancer treatment and recovery. In this article, by analyzing the domestic and foreign scholars on the research of self-perceived burden of cancer patients, highlight the patients self-perceived burden level and influencing factors, provide basis for clinical nursing workers to improve psychological status in cancer patients.
4.Effect of resveratrol in regulating proliferation and apoptosis of rectal cancer cells via up-regulating PTEN.

To analysis the relative expression of PTEN(phosphatase and tensin homology deleted on chromosome ten) in rectal cancer cell lines and the effects of resveratrol on cell proliferation and apoptosis of rectal cancer cells in vitro. Western blot was used to assess the protein expression of PTEN in rectal cancer cell lines and non-malignant colon epithelial cell line. After treating human rectal cancer cell line SW480 with different concentrations of resveratrol, the cellular proliferation was detected by cell counting Kit-8(CCK-8) assay. The apoptotic rate was analyzed by flow cytometry. The protein expressions of Caspase-3, PTEN, p53 and phosphorylase AKT(p-AKT) were detected by Western blot. shRNA(short hairpin RNA) was built to down-regulate PTEN expression in SW480 cells, and detect whether the own-regulated PTEN expression impact resveratrol's effect in growth inhibition and apoptosis induction of in vitro rectal cancer cells. Our results demonstrated that the relative level of PTEN was markedly decreased in Caco-2 and SW480, with statistically significant differences(P<0.001). Resveratrol inhibited the proliferation of rectal cancer cells markedly, and triggered apoptosis of SW480 cells. The protein expressions of Caspase-3, PTEN and p53 were all increased after being treated with resveratrol, while the expression of p-AKT was decreased after the treatment. The expression of endogenous PTEN was significantly decreased in shRNA PTEN-infected SW480 cells. Meanwhile, shRNA PTEN could obviously reduce SW480 cells' anti-proliferative and apoptotic effects. The experimental results suggested that resveratrol can be used as an anti-proliferative and apoptosis induction agent in rectal cancer through up-regulation of PTEN.
5.Tetramethylpyrazine accelerated spinal cord repair through regulation of caspase-3 and neurofilament protein expression: an acute spinal cord injury model in rats.

OBJECTIVE:
To investigate the effect of tetramethylpyrazine on spinal cord repair through hypothesized pathways including regulation of caspase-3, neurofilament protein NF-L, NF-H, and NF-M expression.
METHODS:
Allenos weight drop method was used to establish acute spinal cord injury (SCI) rat model at T10 section and was conducted in 80 adult Sprague Dawley rats that were subsequently divided into pseudo-therapy group (sodium chloride group, n=40) and tetramethylpyrazine group (treatment group, n=40). Eight normal rats were included into the pseudo-surgery normal group (n=1 for each time point). The function of spinal cord was evaluated with animal behavioral scores by measuring modified Rivilin loxotic plate degree and counting BBB score at 1, 3, 7, 14, and 21 d postoperatively. The injured spinal cord tissue samples were harvested at 1 h, 3 h, 6 h, 1 d, 3 d, 7 d, 14 d, and 21 d postoperatively (n=5 for each time point) for the preparation of continuous histological sections that were analyzed the expression of caspase-3,NF-L,NF-H, and NF-M by immunohistochemistry method.
RESULTS:
At postoperative Day 7, 14, and 21, animal behavioral scores revealed higher modified Rivlin loxotic plate degree and BBB scores in the treatment group when compared with those of the control group (P<0.05). The cells in the injured spinal cord tissue expressed significantly less caspase-3 and significantly more NF (NF-L, NF-H and NF-M) in the treatment group than those in the control group at Day 3, 7, and 14 (P<0.05). There was positive correlation between modified Rivlin loxotic plate degree and NF expression, BBB score and NF expression; and negative correlations between BBB score and caspase-3 expression, caspase-3 expression and NF expression.
CONCLUSION
Tetramethylpyrazine improves spinal cord healing through regulation of caspase-3 and neurofilament protein expression.

6.Study on the treatment of cerebral infarction with Chinese medicine Shenmajingfu granule by network pharmacology

Objective To explore the material basis and mechanism of the Chinese medicine Shenmajingfu granules in the treatment of cerebral infarction. Methods The potential active ingredients and targets of Shenmajingfu granules were retrieved through TCMSP, ETCM database and TCM Database. The related target genes of cerebral infarction were searched from OMIM database. The common targets of Shenmajingfu granules and cerebral infarction were obtained by the intersection method. Cytoscape was used to construct active components of Shenmajingfu granules-targets network. Protein-protein interaction network was constructed by STRING software. DAVID database was used for GO and KEGG enrichment analysis. Results The 183 potential active ingredients of Shenmajingfu granules were screened out. 1785 potential targets were screened in the TCMSP database, including 30 targets related to cerebral infarction. These target genes were mainly involved in the inflammatory response and apoptosis process, involving the TNF signaling pathway, HIF-1 signaling pathway and NF-κB signaling pathway. Conclusion The therapeutic effect of Shenmajingfu granules on cerebral infarction may be related to the regulation of inflammatory response, improvement of impaired neurological function and protection of cerebral ischemia-reperfusion injury.
7.Inspiration of The Inner Canon of Yellow Emperor (《黄帝内经》) to the Construction of the Theory of "Middle Jiao Deficiency Causes Seminal Changes"

Based on the theory of spleen-stomach in The Inner Canon of Yellow Emperor (《黄帝内经》), the theory of "middle jiao deficiency causes seminal changes" focuses on the ways in which the spleen-stomach deficiency affect male seminal disorders, as well as the ideas and methods of treating seminal disorders from the perspective of the spleen and stomach. Based on the discussion of spleen and stomach in The Inner Canon of Yellow Emperor, the theoretical connotations of “middle jiao deficiency causes seminal changes” are clarified in terms of the scope of the disease, the etiology of the spleen deficiency, the pathogenesis changes, and the treatment of the disease. It is believed that the connotation of "semen" includes the semen itself and the semen chamber as a whole, and that the core pathogenesis of "seminal changes" is the deficiency of spleen and stomach, which also involves kidney deficiency. Taking the theory of "middle jiao deficiency causes seminal changes" as a guide, the treatment idea of "regulating middle jiao and rectifying essence" was established, with the key of tonifying the middle jiao and regulating transportation and transformation of spleen and stomach, and the aim of regulating essence chamber and promoting the metabolism of turbid substances and the production of essence. It is divided into three methods, including tonifying, transporting, and clearing the middle jiao to rectify semen, so as to providing new ideas for the treatment of male seminal disorders from the spleen and stomach.
8.Anti-inflammatory and analgesic study of Yinlian Tongfeng granules on adjuvant arthritis rats

Objective To investigate the relieving effect of Yinlian Tongfeng granules on adjuvant arthritis rats. Methods A complete Freund adjuvant model was built for this study. 48 rats were randomly divided into control group, model group, Yinlian Tongfeng granules high, medium and low dose group (15.4g/kg, 7.7g/kg, 3.8g/kg). After the drug intervention, arthritis index, ankle swelling rate, formaldehyde induced pain score were recorded. Elisa method was used to detect IL-4, IL-10 and the content of PK1, PK2. The pathological changes of toe pad in rats were observed. Results Compared with the model control group, Yinlian Tongfeng granules groups significantly inhibited the ankle swelling, reduced the pain reaction caused by formaldehyde (P<0.01), significantly increased the content of anti-inflammatory factors IL-4 and IL-10 (P<0.01), and decreased the content of PK1 and PK2 (P<0.01). Toe edema and lymphocyte infiltration were alleviated. Conclusion Yinlian Tongfeng granules have a significant inhibitory effect on adjuvant arthritis, suggesting that it has a potential therapeutic application for rheumatoid arthritis.
9.Scientific Connotations of "Reinforcing Earth for Generating Metal" in the Treatment of Lung Cancer from the Perspective of the Microecology of Intestinal Microbiota

"Reinforcing earth for generating metal" is based on the treatment principle of the five-element theory of traditional Chinese medicine (TCM), which states that "reinforcing mother viscera for treating deficiency of child viscera". Based on the principle of inter-generating relationship in the five elements that earth generates metal, by strengthening the functions of the internal organs belonging to "earth" (spleen and stomach) to indirectly nourish the internal organs belonging to "metal" (lungs and large intestine). Using reinforcing earth for generating metal in the treatment of lung cancer, according to the principle of "reinforcing mother viscera for treating deficiency of child viscera", by strengthening the function of "earth" viscera, the treatment effect of "metal" viscera can be achieved. The metabolic disorder, inflammatory response, and immune regulation caused by the changes in the composition and structure of intestinal microbiota are involved in the occurrence and development of lung cancer, which is consistent with the theory of “the external and internal relationship between the lung and the large intestine” in TCM. From the perspective of intestinal microbiota microecology, the paper discusses the pathogenesis of "earth disease leads to metal disorders" that the gut microbiota dysbiosis causes lung cancer and the treatment method of "reinforcing earth for generating metal" by regulating the microecological balance of gut microbiota in lung cancer, suggesting the great significance of "reinforcing earth for generating metal" therapy aiming at microecological balance of gut microbiota in the treatment of lung cancer, in order to provide a different perspective for the treatment of lung cancer with TCM.
10.Prokineticin 2 mediates peripheral and central sensitization of somatic pain

Prokineticin 2 (PK2) is a newly discovered chemokine, which participates in various physiological functions of the body by binding to receptors PKR1 and PKR2. PK signaling pathway is a newly discovered important regulatory pathway for the occurrence and maintenance of pain after tissue injury and nerve injury in recent years. It plays a key role in regulating injury-related nociceptive events and is a potential therapeutic target for many diseases. The activation of PKRs can induce pain sensation and participate in the sensitivity of pain receptors to different stimuli. The PK system (PKs and PKRs) is an important link involved in inflammation and pain transmission in immune cells. PK2 is involved in the regulation of pain perception by activating PKR1 and PKR2 on primary sensory neurons. In rat primary sensory neurons, PK2 also enhances gated ion channel current through the PKC signaling pathway, inhibits GABA-activated currents, and sensitizes purine nucleotide P2 receptor (P2X). This paper reviews the research progress of PK2 in physical pain. We hope to find new drugs for the treatment of inflammatory pain that target the PKs signaling pathway in future studies.
