1.Effect of long intergenic noncoding RNA-p21 knockdown on radiosensitivity of hypoxic tumor cells

Objective To investigate the effect and mechanisms of lincRNA-p21 knockdown on radiosensitivity of hypoxic tumor cells.Methods The expression of lincRNA-p21 in SMMC7721 and U251 MG cells after hypoxia treatment were detected by QRT-PCR.Lentivirus vector carrying lincRNA-p21 siRNA or scrambled oligos were constructed.SMMC7721 and U251MG cells with stable integration of lincRNA-p21 siRNA or scrambled sequence were generated through lentiviral-mediated gene transfer.Cell cycle and apoptosis of hypoxic tumor cells were analyzed by flow cytometry.Radiosensitivity of hypoxic tumor cells were measured by cloning formation assay.The expressions of HIF-1α and GLUT1 protein were detected by Western blot.Results Under hypoxia condition (1% O2) for 24 and 48 h, lincRNA-p21 strongly increased (t =5.13, 7.49, 8.90, 10.11, P < 0.05) in SMMC7721 and U251MG cells in a timedependent manner, but this increase was suppressed in the SMMC7721 and U251MG cells with stable integration of lincRNA-p21 siRNA (t =144.81, 334.32, P < 0.05).When SMMC7721 and U251MG cells were cultured under hypoxia condition (1% O2) for 48 h, knockdown of lincRNA-p21 induced G2/M phase arrest (t =7.05, 10.18, P < 0.05) and apoptosis (t =42.27, 24.79, P < 0.05), reduced the expressions of HIF-1α and GLUT1 protein, and enhanced radiosensitivity with a radiosensitization ratio of 1.23 and 1.31, respectively.Conclusions Hypoxia treatment elevate the expression of lincRNA-p21.Knockdown of lincRNA-p21 enhance radiosensitivity of hypoxic tumor cells accompanied with G2/M phase arrest, apoptosis induction and the decrease of HIF-1α protein expression.

Mammal multidrug and toxin extrusion proteins (MATEs) play an important role in the transport of organic cations in the body. MATEs mediate the final excretion step for multiple organic cation drug used clinically and important endogenous substances. This article reviews the discovery, type, gene coding and polymorphism, body distribution, classification of substrates and inhibitors and their research method of MATEs. The article also discusses the major research significance of MATEs with examples.
3.Construction and identification of small interfering RNA expression vector targeting EOLA1 gene

Objective To construct small interfering RNA(siRNA) expression vectors targeting EOLA1 (Endothelial-overexpressed lipopolysaccharide-associated factor 1) gene, and to assay their effects on the expression of EOLA1. Methods GFP-EOLA1 fusion protein expressive vector pEGFP-N2/EOLA1 was constructed and transfected into ECV304 cells. The transfected cells were cultured in M199 containing G418 (400?g/ml) for 5 weeks to screen the cell line stably expressing GFP-EOLA1 fusion protein. For interfering EOLA1 target gene, complementary oligonucleotides were desiqned and synthesized at different sites. The oligonucleotides were inserted into pSinencer3.1/H1 plasmid. Then, the vector was transfected into the ECV304 cells stably expressing GFP-EOLA1 fusion protein and the inhibiting affection to target gene EOLA1 was identitied by the observation of the green fluorescence in transfected cells with inverted fluorescent microscope and Western immunoblot assay. Results The ECV304 cell line stably expressing GFP-EOLA1 fusion protein was constructed, and the siRNA vector of EOLA1 can knockdown EOLA1 gene expression specifically. Conclusion The siRNA vectors specifically interfering EOLA1 expression were constructed successfully, which would be useful in the study of function of EOLA1 gene.
4.Anti-inflammatory cytokines and type 2 diabetes

Compared with proinflammatory processes in the development of type 2 diabetes,our knowledge on anti-inflammatory cytokines is rather limited.Anti-inflammatory cytokines such as adiponectin,interleukin 1 receptor antagonist(IL-1RA),transforming growth factor-β1 (TGF-β1),growth differentiation factor-15 (GDF-15),omentin,and secreted frizzled-related protein 5 (SFRP5) are strongly associated with increased risk of type 2 diabetes:Adiponectin and omentin levels decreased before type 2 diabetes,in contrast,concentrations of IL-1RA,TGF-β1,GDF-15 are increased.Change of SFRP5 levels in impaired glucose tolerance and type 2 diabetes remains controversial.This paper will summarize and recommend studies that investigated associations between circulating levels of anti-inflammatory cytokines and type 2 diabetes.
5.Analysis of factors associated with incision infection in elderly general surgery patients

Objective To explore factors associated with incision infection in elderly patients undergoing general surgery and thus provide a basis for its clinical prevention.Methods Retrospective clinical data of 2985 elderly patients who underwent routine surgical operation after admission to the Department of General Surgery from Feb.2009 to Dec.2013 were analyzed.Patients with incision infection following surgery were assigned into the infection group,while patients without incision infection served as the control group.Single factor analysis and multivariate Logistic regression analysis were conducted to identify factors responsible for incision infection.Results There were 91 cases with incision infection,constituting a rate of 3.0%.Compared with the control group,patients in the infection group showed an older average age [(67.3±5.8) years vs.(63.0± 5.6) years,t=1.906,P<0.05],a longer average operating time [(2.13±0.26) hours vs.(1.87±0.24) hours,t=10.149,P<0.001],and a longer average incision length [(8.14±2.62) cm vs.(7.59±2.14) cm,t=2.396,P<0.01].In addition,compared with the control group,more patients in the infection group received prophylactic antibiotics (74.7% vs.83.3%,x2 =4.819,P=0.032),exhibited malnutrition (20.9% vs.10.0%,x2 =11.418,P=0.001),and had diabetes mellitus (11.0% vs.5.1%,x2 =5.968,P=0.015).Therefore,independent risk factors for incision infection included age (OR=1.89),operating time (OR=3.32),nutritional status (OR=2.08) and diabetes mellitus (OR=3.18).Conclusions A few risk factors may contribute to incision infection.Clinicians should take necessary preventive measures accordingly to minimize its occurrence.
6.Antibiotic Resistance of Acinetobacter baumannii Causing Lung Infection in Elderly Patients and Its Clinical Analysis

OBJECTIVE To explore the clinical features of Acinetobacter baumannii lung infection and drug resistance situation.METHODS An analysis was conducted on clinical features and risk factors of 79 cases of A.baumannii lung infection in elderly patients from Jan 2005 to Jun 2008.Drug resistance of A.baumannii to 12 kinds of antimicrobial drugs such as ciprofloxacin,ceftazidime,ceftriaxone,imipenem and amikacin was tested.RESULTS An onset of A.baumannii infections in the lungs of elderly patients usually appeared during the critical period of some underlying diseases.More clinical manifestations as fever,lung and wet La-hypoxemia existed.Accepting antibiotic therapy,pulmonary diseases,mechanical ventilation,ICU wards,etc were the risk factors.There was a high mortality rate with multi-drug resistant(MDR) A.baumannii.Bacterial resistance to cefoperazone/sulbactam,piperacillin/tazobactam was lower while its resistance to other antibiotics was higher.The detection rate of MDR was 37.97%.CONCLUSIONS Elderly patients with A.baumannii infection have a high incidence of lung infection,but lacking characteristic clinical signs and symptoms.There is an increasing trend in the detection rate of MDR.It is of great significance to reduce the incidence rate of A.baumannii lung infection and prevent the spread of multi-drug resistant strains by strengthening the monitoring of pathogens and their resistance and choosing antibiotics according to susceptibility testing.
7.Analysis on Pathogens and Their Drug Resistance of Tuberculosis Patients with Lower Respiratory Tract Infection

OBJECTIVE To understand the pathogens spectrum of tuberculosis patients with lower respiratory tract infection in hospital and the drug resistance,so as to provide guidance of rational drug use.METHODS 576 pathogens isolated from the sputum samples of TB patients with lower respiratory tract infection in hospital from 2005 to 2007 and the drug resistance were analyzed.ESBLs test was conducted on the Gram-negative bacill.RESULTS Gram-negative bacilli accounted for 73.6 percent.The first six ones were: Pseudomonas aeruginosa(19.4%),Acinetobacter baumannii(12.0%),Klebsiella pneumoniae(11.1%),E.coli(9.9%),Staphylococcus aureus(9.2%),Candida albicans(8.2%).They were highly sensitive to impinem,Cefoperazone-sulbactam.The drug resistance rates of ESBLs produceing P.aeruginosa and A.baumannii were respectively 2.7%-30.3% and 8.7%-30.4%.The resistance of MRSA to B-lactamase,Fluoroquinolone and Aminoglycoside was 64.7%-100.0%.ESBLs produceing E.coli and K.pneumoniae strains accounted for 52.6% and 45.3%.CONCLUSIONS Lower respiratory tract infection of tuberculosis patients was mainly caused by Gram-negative bacilli,most of which was portunistic pathogens.The pathogens resistance to antibiotics are increasing E.coli and K.pneumoniae produce a higher proportion of EBSLs.To delay the formation of pathogen resistance and control the hospital infection,it is important to enhance drug monitoring and rational use of antibiotics.
8.A Study on Stimulation Effect of Human Interleukin-2 (IL-2) with Carboxymethyl-Pachymaran (CMP) in the Experimental Reaearch and the Clinical Application

The titer of Interleukin-2 (IL-2), prepared from the healthy human peripheral blood lymphoytes which were stimulating-induced by carboxymethy-parchymaran (CMP), was very high and reached clinical drug level after special process. The IL-2 was not only available for intramuscular injection. but also for intravenous in jection or even for intraarterial, injection 44 partients with advanced malignant tumor treated with the combination of IL-2 / LAK and chemotherapy showed an overall objective response rate to 86. 4% (CR + PR: 13, 29.3%); (CR + PR + MR: 22, 50%); (CR + PR + MR + SR : 38, 86. 4%) . 37 Patients with various malignant tumors were treated by low-dose interleukin-2 stinulated with CMP and irradiation with 60Co. The percentages of CR, PR and SR were 51.4%, 46.8%and 2.7%, respectively. Overall effectiveness rate was 97.3%. 1 year survival rate was 86.2%. 16 Patients with various advanced cancers were treated by low-dose interleukin-2 stimulated with CMP and low - dose radiotherapy or chemotherapy. The percentage of CR, PR and SR was 37. 5%, 56. 3%and 6 . 3%, respectively. Overall effective rate was 93. 8%. 1 year survival rate was 87. 5%. The current results indicate that the possibility of combined therapy of the low - dose IL-2/LAK adoptive chemoimmunotherapy and radiotherapy or chemotherapy may be effective in treatment of advanced cancer.

Objective:The glucemic index(GI)and insulin index(II)of wheat flour, oat fiber and resistant starch (RS) were studied.Method:Ten healthy subjects consumed 4 kinds of carbohydrate foods: glucose powder,wheat flour food, oat fiber food and RS food.Up to 120 min after consumption of test materials, blood samples were taken for glucose and insulin analyses.Results:The GI value of wheat flour food, oat fiber food, RS food was 88.24?20.84,60.16?14.16,47.05?10.22 respectively,regarding glucose powder as 100 . According to the ultrasonic manifestations of the gall bladder before laparoscopic cholecystectomy(LC),including size,morphology,wall thickness,presence or absence of gallstone,congenital anomaly of biliary duct,surrounding adhesion and state of common bile duct,tbe patients Werc dividcd into 4 groups:A.Most suitable for LC,B.Sitable for LC,C.Relatively suitable for LC. D.Not suitable for LC.This provided clinicians the basis for patient selection.Comparing with the sur.gical findings,the coincidence rate of B-mode ultrasonography was 97.8%.Preoperative B-mode ul-trasonography of gall bladder has significant clinical value to relieve the patients from pain,decrease the operative risk and conversion rate of LC.
