1.HPLC fingerprint of Rumex gmelini from different habitats

Objective To compare the HPLC fingerprint of Rumex gmelini from different habitats by RP-HPLC (DAD). Methods In this paper, 12 different samples were studied. Separation was performed on an Planetsil C_ 18 column, with mobile phase consisting of methanol and 0.1% phosphoric acid-water and with gradient elution at the flow rate of 1.0 mL/min. The UV detection wavelength was 254 nm, column temperature was 35 ℃, and the analysis time was 50 min. Results The results showed that this method has a good repeatability and the ratio of common peaksarea of different samples had some difference. Conclusion This method can be used to establish the chromatographic fingerprint of R. gmelini with high specificity.
2.Differentiation of Cortex Fraxini and its three kinds of confusable species

Object To provide a scientific basis for identification and rational use of the herbal medicine by the pharmacognostic study on Cortex Fraxini. Methods Microscopic identification, TLC, HPLC were used. Results There were remarkable differences in the genuine product, confusing product and fakements of Cortex Fraxini in characteristics of microscopic identification, fluorescence, TLC and HPLC. Conclusion The scientific basis has been established and provided for differentiation of confusing product and fakements.
3.Determination of aesculin and aesculetin in periderm and leaves of Cortex Fraxini among different provenances

Objective: To determine the contents of aesculin and aesculetin in periderms and leaves of Cortex Fraxini among differnet provenances. Methods: HPLC was used with acetonitrile-water(15∶85) as the mobile phase and detected wavelength at 348nm. C 18 column was adopted. Results: The calibration curves were linear. The value of correlation coefficient were 0.9992 and 0.9994, respectively. The average recovery of aesculin was 98.2% and aesculetin was 99.2%. RSD were 2.24% and 2.15%, respectively. The quality differences of Cortex Fraxini among different provenances were remarked. The quality of Cortex Fraxini from Shanxi province was the best. Conclusion: The method is applied in determination and analytics of content of Cortex Fraxini and is rapid, simple and easy to carry out. The method is with the feature of accuracy, repetition and stability.
4.Imaging analysis of paraspinal muscles in single segment degenerative lumbar spondylolisthesis and lumbar spinal stenosis

Objective:To explore the difference and clinical significance of paraspinal muscle degeneration between single-segment degenerative lumbar spondylolisthesis and degenerative lumbar spinal stenosis.Methods:From January 2014 to October 2020, a retrospective analysis of 30 patientswere diagnosed with L 4,5 degenerative lumbar spondylolisthesis, aged 61.63±8.42 years old (range 44 to 82 years old), body mass index 24.07±3.17 kg/m 2 and 30 patientswere diagnosed with L 4,5 degenerative lumbar spinal stenosis, aged 59.67±12.89 years old (range 31 to 80 years old), body mass index 25.29±3.48kg/m 2, both of them went on surgery in department of spine surgery, shengjing hospital, China Medical University.30 healthy people were selected from outpatient physical examination in the control group, aged 58.33±7.36 years old (range 52 to 83 years old), body mass index 25.72±2.58 kg/m 2. These three groups were all male. Select all patients with L 3,4, L 4,5 and L 5S 1 disc level axial MRI images, and use the deep learning automatic segmentation measurement system developed by our hospital and Shenyang Institute of Automation Chinese Academy of Sciences to measure multifidus muscle cross sectional area (MMCSA), erector spinae cross sectional area (ESCSA), multifidus muscle fatty infiltration rate (MMFIR) and erector spinae fatty infiltration rate (ESFIR). One-way ANOVA was used to test the imaging parameters of multifidus and erector spinae of the three groups, and LSD- t test was used to compare the imaging parameters in each segment of paraspinal muscles. Results:The gender of three groups were male, there was no significant difference in age ( H=5.303, P>0.05), and there was no significant difference in body mass index ( F=2.267, P>0.05). Multifidus muscle cross-sectional area in L 3,4: degenerative lumbar spondylolisthesis groupincreased 189.11 mm 2 compared with degenerative lumbar spinal stenosis group ( P=0.010). Multifidus muscle cross-sectional area in L 4,5: compared with healthy people group, degenerative lumbar spondylolisthesis group decreased 205.52 mm 2 ( P=0.002), while degenerative lumbar spinal stenosis group decreased 184.14 mm 2 ( P=0.006). Multifidus muscle cross-sectional area in L 5S 1: compared with degenerative lumbar spinal stenosis group, degenerative lumbar spondylolisthesis group decreased 174.93 mm 2 ( P=0.018); compared with healthy people group, degenerative lumbar spondylolisthesis group decreased 406.06 mm 2 ( P<0.001), while degenerative lumbar spinal stenosis group decreased 231.13 mm 2 ( P=0.002). Erector spinae cross sectional area in L 4,5: compared with healthy people group, degenerative lumbar spinal stenosis group decreased 398.70 mm 2 ( P=0.006). Erector spinae cross sectional area in L 5S 1: compared with degenerative lumbar spinal stenosis group, degenerative lumbar spondylolisthesis group decreased 500.02 mm 2 ( P<0.001); compared with healthy people group, degenerative lumbar spinal stenosis group decreased 455.37 mm 2 ( P<0.001). Compared with healthy group, the multifidus muscle fatty infiltration rate of degenerative lumbar spondylolisthesis group in L 3,4 increased 4.96% ( P=0.001). Compared with degenerative lumbar spinal stenosis group, the erector spinae fatty infiltration rate of degenerative lumbar spondylolisthesis group in L 5S 1 decreased 5.41% ( P=0.004). Compared with healthy group, the erector spinae fatty infiltration rate of degenerative lumbarspinal stenosis group in L 5S 1 increased 5.02% ( P=0.008) . Conclusion:Paraspinal muscle cross sectional area of each segment in degenerative lumbar spondylolisthesis group and degenerative lumbar spinal stenosis group decreased in different degrees. In degenerative lumbar spondylolisthesis group, the degree of multifidus muscle fat infiltration was more significant, while indegenerative lumbar spinal stenosis group,the degree of erector spinal fat infiltration was higher.
5.In vitro effect of Banxiaxiexin Tang and different conbinations on hepatic CYP450 in rats.

To study the effect of Banxiaxiexin Tang and different conbinations on CYP450 in rat liver microsomes, from the point of liver metabolism, evaluate significance of Banxiaxiexin Tang compatibility. The rats were randomly divided into five groups: Banxiaxiexin Tang group, pungent-swelling group, bitter-descending group, sweet-invigorating group and control group, which were all given decoction by gavage. Using liver microsomes in vitro incubation method, probe substrate were incubated and their metabolites was detected by ultra-high performance liquid chromatography, then was calculated metabolic rate to reflect the drug-treated liver microsomes CYP2C6, CYP2E1, CYP3A1/2 activity. The results showed that comparing with the control group, both Banxiaxiexin Tang group and bitter-descending group showed inhibition on all enzyme subtype (P < 0.01), pungent-swelling group showed significant inhibition on CYP2C6, but no inhibition on CYP2E1 and CYP3A1/2; sweet-invigorating group showed inhibition on CYP2C6 and CYP2E1, but no inhibition on CYP3A1/2. Compared the inhibition on CYP with the three conbinations, bitter-descending group was significant higher than other groups. Banxiaxiexin Tang group showed inhibition on rat liver microsomes CYP450, and the activity maybe come from bitter-descending group.

6.The value of CT radiomics in predicting treatment outcomes in patients with nasal polyps

Objective:To evaluate the predictive efficacy of sinus CT radiomics for treatment outcomes in nasal polyp patients undergoing endoscopic sinus surgery.Methods:A retrospective cohort study was conducted at the First Affiliated Hospital of Sun Yat-sen University, including 194 patients with nasal polyps treated between January 2015 and December 2019. The cohort comprised 132 males and 62 females, aged 16 to 75 years. Patients were divided into a training set ( n=135) and an internal validation set ( n=59). An external validation set ( n=34), consisting of 22 males and 12 females aged 16 to 59 years, was included from January 2020 to December 2021. Disease control was evaluated using the criteria from the European Position Paper on Rhinosinusitis and Nasal Polyps 2020 (EPOS 2020). Radiomic features were extracted from sinus CT images and analyzed using the least absolute shrinkage and selection operator (LASSO) regression. Models combining radiomic and clinical features were developed to predict treatment efficacy. Results:The radiomics and combined models, based on four selected features, outperformed the clinical feature model in the training set, with AUC values of 0.901 and 0.915, versus 0.874, respectively. In the internal validation set, AUCs were 0.839, 0.832, and 0.716. Despite reduced AUCs in the external set, the radiomics model maintained good generalizability (0.748, 0.764, 0.620). Decision curve analysis showed significant clinical benefits in both radiomics and combined models.Conclusion:The CT-based radiomics model demonstrates significant predictive power in identifying refractory nasal polyps, suggesting its potential for clinical application in treatment outcome prediction.
7.Effects of lncRNA SNHG12 on the proliferation,migration and invasion of prostate cancer cells by targeting miR-495-3p/PI3K/Akt signaling pathway

Objective To explore the effects of long non-coding RNA(lncRNA)small nucleolar molecule RNA host gene 12(SNHG12)targeting inhibition of miR-495-3p/phospholipinositol-3-kinase(PI3K)/protein kinase B(Akt)signaling pathway on the proliferation,migration and invasion of prostate cancer cells.Methods The expressions of SNHG12 and miR-495-3p in prostate cancer tissues and cells(LNCaP,C4-2,DU145)were detected with real-time fluorescence quantitative PCR(qRT-PCR).After DU145 cells were divided into si-NC,si-SNHG12,si-SNHG12+anti-miR-NC and si-SNHG12+anti-miR-495-3p groups,the expressions of SNHG12 and miR-495-3p were detected with qRT-PCR;the targeting relationship between SNHG12 and miR-495-3p was determined with dual luciferase assay;cell proliferation was assessed with MTT assay;cell migration and invasion were evaluated with Transwell assay;the protein expressions of proliferating cell nuclear antigen(PCNA),N-cadherin,and E-cadherin were detected with Western blot.Results The expressions of SNHG12 were significantly increased,while the expression of miR-495-3P was significantly decreased in prostate cancer tissues and cells(LNCaP,C4-2,DU145)(P<0.05).Knockdown of SNHG12 decreased DU145 cell activity,lowered the protein expressions of PCNA and N-cadherin,reduced the number of migrating and invading cells,but increased the protein expression of E-cadherin(P<0.05).SNHG12 targeted and negatively regulated miR-495-3p,and down-regulation of miR-495-3p reversed the effects of SNHG12 knockdown on the proliferation,migration and invasion of prostate cancer cells.Compared with the si-NC group,the si-SNHG12 group had significantly decreased expressions of p-PI3K and p-Akt(P<0.05).Compared with the si-SNHG12+anti-miR-NC group,the si-SNHG12+anti-miR-495-3p group had significantly increased protein expressions of p-PI3K and p-Akt(P<0.05).Conclusion lncRNA SNHG12 can promote the proliferation,migration and invasion of prostate cancer cells through targeted inhibition of miR-495-3p/PI3K/Akt signaling pathway.
8.Predictive efficacy of low grip strength and grip asymmetry on postoperative outcomes in elderly patients with colorectal cancer

Objective To observe the efficacy of low grip strength and grip asymmetry in predicting multiple postoperative outcomes in elderly patients with colorectal cancer. Methods A total of 257 elderly colorectal cancer patients were selected as study objects. Data on general information, grip strength indicators and adverse postoperative outcomes were collected from all patients. Associations of low grip strength, grip asymmetry with adverse postoperative outcomes were explored by Logistic regression. Receiver operating characteristic curves were plotted to further analyze the efficacy of low grip strength and grip asymmetry alone and their combination in predicting adverse outcomes of patients. Results Grip strength, grip strength ratio, and the combination of grip strength and grip strength ratio could effectively predict the occurrence of complications. The areas under the curve (AUCs) and 95% confidence interval (95%CI) were 0.649 (0.587 to 0.707), 0.627 (0.565 to 0.686), and 0.672 (0.611 to 0.729) respectively. Grip strength ratio and the combination of grip strength and grip strength ratio could effectively predict prolonged hospital stays, with AUC(95%CI) of 0.671 (0.610 to 0.728) and 0.577 (0.514 to 0.638), respectively. Grip strength and the combination of grip strength and grip strength ratio could effectively predict the occurrence of frailty, with AUC (95%CI) of 0.585 (0.522 to 0.646) and 0.586 (0.523 to 0.647), respectively. Conclusion This study innovatively reveals the predictive efficacy of multiple grip strength indicators for postoperative outcomes in colorectal cancer patients. Low grip strength and grip asymmetry are effective in predicting the occurrence of complications in patients, grip asymmetry is effective in predicting the occurrence of prolonged hospitalization, and low grip strength is effective in predicting the occurrence of frailty in elderly patients with colorectal cancer.
9.Clinical value of combined detection of ApoA1 and AFP in young and middle-aged patients with pulmonary tuberculosis

Objective To explore the clinical application value of combined detection of apolipoprotein A1(ApoA1)and alpha-fetoprotein(AFP)in young and middle-aged patients with pulmonary tuberculosis.Meth-ods A total of 82 young and middle-aged patients with pulmonary tuberculosis admitted to the hospital from December 2020 to December 2021 were selected as the observation group,and 85 healthy people who under-went physical examination in the hospital during the same period were selected as the control group.The clini-cal,lipid,inflammation and iron metabolism related indexes were compared between the two groups.Multiva-riate Logistic regression was used to analyze the influencing factors of pulmonary tuberculosis in young and middle-aged patients.The area under the receiver operating characteristic(ROC)curve(AUC)was used to e-valuate the application value of ApoA1,AFP and their combination in the diagnosis of pulmonary tuberculosis in young and middle-aged patients.Results Compared with the control group,the observation group had sig-nificantly higher levels of direct bilirubin,creatinine,AFP and high-sensitivity C-reactive protein(P<0.05),and significantly lower levels of triglyceride,total cholesterol,high-density lipoprotein cholesterol and ApoA1(P<0.05).Multivariate Logistic regression analysis showed that ApoA1 was an independent protective factor(OR=0.119,95%CI 0.019-0.754,P=0.024)and AFP was an independent risk factor(OR=1.554,95%CI 1.138-2.122,P=0.006)for pulmonary tuberculosis in young and middle-aged patients.ApoA1,AFP and the combined prediction of young and middle-aged tuberculosis AUC were 0.644(P=0.006),0.262(P<0.001),0.678(P=0.001),respectively.Conclusion The low level of ApoA1 is an independent protec-tive factor for pulmonary tuberculosis in young and middle-aged patients,and the high level of AFP is an inde-pendent risk factor for pulmonary tuberculosis in young and middle-aged patients.The combined detection of ApoA1 and AFP has good value in predicting pulmonary tuberculosis in young and middle-aged patients.
10.The effect of estimated glomerular filtration rate on outcome of patients with acute ischemic stroke after intravenous thrombolysis with recombinant tissue plasminogen activator

Objective To explore the association between estimated glomerular filtration rate (eGFR) and prognosis of acute ischemic stroke (AIS) patients who were treated by intravenous thrombolysis with recombinant tissue plasminogen activator (rt-PA).Methods We consecutively screened AIS patients who were treated by intravenous thrombolysis with rt-PA from January 2006 to September 2016 in Tangshan Gongren Hospital.According to eGFR value of patients at admission,the eligible patients were divided into two groups:normal eGFR group (eGFR ≥ 90 ml ? min-1 ? 1.73 m-2) and decreased eGFR group (eGFR < 90 ml? min-1 ? 1.73 m-2).The incidence of symptomatic intracerebral hemorrhage (SICH),early neurological deterioration (END) at 24 hours and seven days after thrombolysis,mortality within seven days and 90 days,and excellent recovery at 90 days were compared between the two groups.The OR with 95% CI and the adjusted OR with 95% CI were analyzed by univariate and multivariate Logistic regression models.Results A total of 258 patients were enrolled,including 182 cases in the normal eGFR group and 76 cases in the decreased eGFR group.After adjusting for the potential confounders,multivariate Logistic regression analysis showed that the rates of SICH (13.2% (10/76) vs 3.3% (6/182),OR =3.859,95% CI 1.313-11.341),END at 24 hours (21.1% (16/76) vs 8.2% (15/182),OR =2.958,95% CI 1.347-6.495) and seven days (32.9% (25/76) vs 12.6% (23/182),OR =3.129,95% CI 1.555-6.293),mortality within seven days (22.4% (17/76) vs 6.0% (11/182),OR =4.079,95% CI 1.588-10.477) and 90 days (23.7% (18/76) vs 9.9% (18/182),OR =2.457,95% CI 1.050-5.749) were higher in the decreased eGFR group than in the normal eGFR group.On the other hand,the chance of excellent recovery at 90 days (22.4% (17/76) vs 43.4% (79/182),OR =0.435,95% CI 0.229-0.824) was less in the decreased eGFR group than in the normal eGFR group.Conclusion Decreased eGFR may not only increase the risks of SICH,END and death,but also reduce the chance of 90-day excellent recovery in AIS patients after intravenous thrombolysis with rt-PA.
