1.Cyclooxygenase-2 inhibitors enhance the antileukemia activity of STI571

AIM: To investigate whether celecoxib,a cyclooxygenase-2(COX-2) inhibitor,potentiates the anti-leukemia activity of STI571 in K562 cells.METHODS: K562 cells were treated with STI571,celecoxib or combination of both at different concentrations in suspension culture.Cell proliferation was documented by MTT assay,and cell apoptosis was determined by flow cytometry and morphology.Meanwhile,RT-PCR was applied to analyze the probable mechanism underlying the effects of the drugs.RESULTS: The combination of STI571 and celecoxib dramatically suppressed the proliferation of K562 cells,in which 0.25 ?mol/L STI571 and 40.0 ?mol/L celecoxib enhanced the inhibiting rate to 76.1%?1.6%.Furthermore,the combining administration of drugs significantly promoted the apoptosis induced by STI571,which showed characteristic changes of morphologic features and increase in sub-G_1 cells.By using RT-PCR technique,the expression of COX-2 had no decline by single administration of celecoxib or STI571.However,a progressive down-regulation was caused by coadministration of two drugs.In contrast with COX-2,the expression of VEGF had no changes at any time.CONCLUSION: The administration of celecoxib alone only inhibits the proliferation of K562 cells.Combination treatment with STI571 and celecoxib promotes the apoptosis induced by STI571.
3.Effect of lifestyle quantilization based-weight management on overweight or obesity occupational groups

Objective To study the effects of lifestyle quantilization based-weight management on overweight or obesity police officers.Methods One hundred and seven overweight or obesity police officers received lifestyle quantilization based-weight management (i.e.Jinbi weight management) and were then assigned to the excellent performance group (group A,n =50),good performance group (group B,n =42)and loss to follow-up group (group C,n =15).Dietary habits,body weight,height,waist circumference (WC),blood pressure (BP),total cholesterol (TC),triglyceride (TG),low-density lipoprotein cholesterol (LDL-C),high-density lipoprotein cholesterol (HDL-C) and fasting plasma glucose (FPG) were measured before and after the intervention.After 8 weeks' intervention,the changes of above parameters were recored.Student's t test was used for data analysis.Results Of group A and B,83 reported weight loss (group A t =13.31,group B t =5.04 ; both P < 0.05).In group A,body weight,body mass index (BMI),WC,body fat and visceral fat index were significantly decreased,in contrast to body water (t values were 13.31,13.72,10.8,8.59,6.83 and-6.62,respectively; all P < 0.05).However,there were no significant changes of BP,FPG,TC,TG,LDL-C and HDL-C in group A.Following intervention,daily dietary energy intake of group A was reduced by 74.1 k J,fat intake was decreased by 11.6 g,energy ratio of dietary fat was decreased by 1.8%,energy ratio of cereal was increased by 4.2%,and sodium chloride and cooking oil was decreased by 1.3 g and 10 g,respectively.Conclusion Lifestyle quantilization based-weight management shows effectiveness among overweight or obese police officers,and thus may be recommended for other functional communities.
4.Analysis and future prospect of State Science and Technology Awards in the medical fields from 2000 to 2020

Objective:By analyzing State Science and Technology Awards in the medical fields from 2000 to 2020, this paper aims to provide possible references for the development, transformation and application of scientific and technological achievements under the context of new evaluation mechanism, to further improve the effectiveness of scientific research management.Methods:Accessing in to the website of National Office For Science & Technology Awards to obtain information of Science & Technology Awards in the field of Medicine, analyze and characterize the different types of awarded scientific and technological achievements.Results:The awarded medical achievements in the past 21 years were mainly aimed at the common and frequently encountered diseases in China, with the most achievements related to cancer and tumor disease, followed by cardiovascular disease and infectious disease. Universities accounted the highest proportion of awards thus becoming the main force of medical scientific and technological innovation, while more balance was needed in regional and organizational distribution. The first prizes of State Natural Science Award and State Technological Invention Award are visibly vacant.Conclusions:Priorities should be given to foster original scientific and technological achievements, to train elite medical talent, to solve the regional imbalance of medical innovation strength and to develop high-tech medical enterprises with international competitiveness, thus solving major medical challenges.
5.Effect of congrong powder preparation on erythrocyte parameter and index of blood rheology in rats.

OBJECTIVETo observe the effect of Congrong powder preparation on erythrocyte parameter and the index of blood rheology in healthy rats.
METHODThe 50 rats were randomly divided into 5 groups ( physiological saline group, testosterone propionate group, large-dose administered group, moderate-dose administered group, small-dose administered group). Blood were taken to determine erythrocyte parameter and the index of blood rheology.
RESULTCongrong powder preparation had no effect on erythrocyte parameter. There was an increase in whole blood viscosity, whole blood reduction viscosity, and in the erythrocyte rigidity index in the large-dose administered group.
CONCLUSIONCongrong powder can influence deformability and some related characters of erythrocytes.

6.Progress of N-myristoylation in oncology treatment

N-myristoylation is a type of protein modification and one of the most common types of protein lipidation. N-myristoylation affects cell signal transduction and metabolic pathway reprogramming by changing protein conformation, stability and localization. It is involved in various physiological and pathological processes. Studies have shown that the protein N-myristoyltransferase (NMT) is abnormally expressed in many types of malignancies and involves in development and progression of tumors. Inhibitors targeting NMT have good anticancer effects and provide potential targets for cancer recurrence and metastasis therapy. This article mainly introduces the protein N-myristoylation system and reviews its role and mechanism in the development and progression of tumors, summarizes the latest research progress of targeting protein N-myristoylation for the treatment of cancer in the last 5 years.

The incidence of brain metastases in patients with non-small cell lung cancer (NSCLC) has increased as a result of improved local control rate and survival rate. Prophylactic cranial irradiation (PCI) has been proven to reduce the incidence of brain metastases and improve survival rate in patients with NSCLC. However, the value of PCI for NSCLC is still controversial. This paper reviews the progress of the efficacy and adverse reactions after PCI treatment for patients with NSCLC.
8.Long-term outcomes and influencing factors of idiopathic nephrotic syndrome in children

Pediatric idiopathic nephrotic syndrome (INS) is characterized by massive albuminuria, hypoproteinemia, edema and hyperlipidemia, with a long course and high probability of relapse and prolongation. Long-term complications caused by long-term usage of hormones and immunosuppressants in children with INS seriously affect their physical and mental health and quality of life. Most children with steroid-sensitive nephrotic syndrome can be cured before adulthood, while some of them relapse in adulthood. Long-term prognosis of children with steroid-resistant nephrotic syndrome is poor. There have been few studies in China followed the long-term outcomes and its related factors of children with INS over 10 years. The paper reviewed the literatures on the long-term outcomes of children with INS, including renal survival, growth, mental health, learning and work, marriage and fertility, disease recurrence and long-term related complications, to explore the factors related to the poor long-term outcomes of children with INS and to assist in clinical decision-making and follow-up management.
