1.Detection of chromosome 8 abnormalities in carcinoma of endometrium (endometrial carcinoma of uterus) by fluorescence in situ hybridization( FISH)

Purpose: To study the relationship between chromosome abnormalities and the pathogenesis, development and prognosis of endometrial carcinoma of uterus. Methods: FISH was used to detect endometrial carcinoma, endometrial hyperplasia and normal endometrium , 10 cases in each group, respectively. Results: All of the 10 carcinoma cases had chromosome 8 mutation, including trisomy, monosomy, tetraploid and in 10 cases of endometrial hyperplasia with trisomy of chromosome 8 were found. But there were no mutations of chromosome 8 in the normal group. There was significant difference in the three groups. Conclusions: Abnormalities of chromosome 8 are significantly associated with the process of pathogenesis, development of endometrial carcinoma. The abnormalities may occur in the early stage of the carcinoma, and may have the significant association with clinical stage and pathological differentiation among the patients.
2.Effects ofpneumoperitoneum on the airway pressure and pulmonary infection of patients with different body mass index in laparoscope

Objective To make clear the patients in minimally invasive surgery through explo-ring effects of pneumoperitoneum and pulmonary infection on the airway pressure of patients with dif-ferent body mass index (BMI)under general anesthesia.Methods A total of 849 cases of patients with trachea cannula in pneumoperitoneum surgery inr general anesthesia under different position were studied.To observe the changes of airway pressure and the ratio of pulmonary infection within one week of patients in general anesthesia.Propofol,Remifentanil and Atracrium were used in general an-esthesia.BIS and TOF were used to detect the degree of muscle relaxation.Results The airway pres-sure was increased obviously in patients of Head-down Tilt under same pneumoperitoneum pressure(P<0.05);the airway pressure was increased in patients with BMI>30 kg/m2 (P <0.05).There was no obvious increase in pulmonary infection rate in all patients.Conclusion Pneumoperitoneum could increase airway pressure and pulmonary infection of patients with low head position and BMI > 30 kg/m2 under same pneumoperitoneum pressure.Therefore,shorten the operation time and decreased pneumoperitoneum pressure could be used to prevent pulmonary infection.

Antigen Rv2628 of Mycobacterium tuberculosis is associated with latent tuberculosis infection. In this study, Rv2628 was prokaryotic expressed and purified, its immunological characteristics was evaluated with macrophage cell line RAW264.7 and BALB/c mice. The results show that Rv2628 was mainly expressed in form of inclusion body confirmed by SDS-PAGE, and could react with rabbit anti-H37Rv polyclonal antibody detected by Western blotting assay, indicating that the protein had an effective immunoreactivity. The interactions between Rv2628 and macrophage cell line RAW264.7 confirmed that it could effectively induce cells to produce pro-inflammatory cytokines, the relative expression level of IL-6 mRNA was higher than the control group in 1-12 h. BALB/c mice were subcutaneously immunized with Rv2628 protein, the production of IFN-gamma and IL-4 in the spleen cells was determined by Sandwich ELISA, in the Rv2628 immunized group, the level of IFN-gamma was significantly higher than that of IL-4 (P < 0.000 1). It indicated the protein induced Th1-tendency immune responses. At the same time, Rv2628(11-30) peptide used as coating antigen, the murine serum antibody titer detected by indirect-ELISA was 1:1 600, which demonstrated that Rv2628 could also induce humoral immune responses. In summary, Rv2628 could induce specific pro-inflammatory cytokines, affectively induce strongly Th1-tendency immune response and humoral response, it could be a potential target for developing subunit vaccine against TB. In addition, it laid foundation for probing the cross-talk between M. tb and host.

5.Combined Action of ACE Gene I/D and GNB3 Gene C825T Polymorphisms on Essential Hypertension in Northern Han Chinese

Essential hypertension (EH), a complex polygenic disease, is considered to the result of the genetic interaction of multiple gene alterations in concert with environmental factors. Evidences showed that angiotensin-converting enzyme (ACE) gene and G protein beta3 subunit (GNB3) gene are both important susceptibility genes for EH, and that there exists putative biological connection between the two genes in developing hypertension. To investigate whether hypertension was affected by gene-gene interaction between the two genes in the northern Chinese Han population, a case-control association study including 502 hypertensive cases and 490healthy controls was conducted, selecting the ACE gene I/D polymorpinsm and the GNB3 gene C825T polymorphism. Linkage disequilibrium analysis revealed a significant nonrandom distribution only in male hypertensives, indicating that interaction between ACE gene and GNB3 gene may predispose males to the occurrence of hypertension. Multivariate stepwise logistic regression in single locus analysis, with adjustment for common risk factors for hypertension, demonstrated that the OR for DD/ID versus Ⅱ for hypertension among men was significant (OR 1.57; 95% CI, 1.09 ~2.27; P = 0.016) in dominant genetic model. In combination analysis stratified with respect to gender, slightly significant ORs were found after adjustment in males: OR for TT vs CC, 0.11; 95%CI, 0.01 ~0.99; P = 0.049 within ACE DD genotype; OR for DD/ID vs Ⅱ, 1.52; 95% CI, 1.01 ~2.29; P = 0.047 within GNB3 CC+CT genotype. The results suggest that ACE, or a nearby gene, is a male-specific susceptible gene for hypertension, and that there may exist epistatic gene-gene interaction between ACE D allele and GNB3 825C allele.
6.Analysis of the Trend of Emergency Drugs in the Inpatients of Our Hospital during 2012-2016

OBJECTIVE:To provide reference for the supply of emergency drugs. METHODS:The data of emergency drugs in the inpatients of our hospital were collected from hospital information system during 2012-2016,including the proportion and age of emergency patients,drug,clinical departments and disease types. The trend of emergency drugs was analyzed by using SPSS 16.0 software. RESULTS:The proportion of emergency patients and discharged patients were increasing year by year. The number of patients in <10 year-old and 10-19 year-old showed a declining trend year by year. The number of patients in 30-39 year-old,50-59 year-old and >60 year-old showed an increasing trend year by year. Among emergency drugs,the use frequency of Heparin sodium injection,Dexamethasone sodium phosphate injection and Flumethasone injection showed a declining trend year by year;the use fre-quency of Hemagglutination for injection,Penehyclidine ether injection,Atracurium besylate injection and Propofol injection showed an increasing trend year by year. Among clinical department,the number of pediatric patients showed a declining trend year by year;the number of patients in obstetrics department,emergency department,cardiology department,micro hand surgery department and pediatric surgery department showed an increasing trend year by year. Among emergency disease types,the number of patients with re-spiratory system disease,infectious and parasitic diseases showed a declining trend year by year;the number of patients with circula-tion system disease,injury and poisoning,certain other consequences of external factors,digestive system diseases,pregnancy,child-birth and puerperium disease as well as some cases originated in the perinatal period showed an increasing trend year by year. CON-CLUSIONS:With the change of the proportion,age and disease type of emergency patients,the type and utilization rate of emergen-cy drugs have changed. It is necessary to adopt effective measures to guarantee the supply of emergency drugs.
7.Effect of electroacupuncture at different segments of heart meridian on the electric discharge activities of the superior sympathetic plexus in rabbits

BACKGROUND: Correlation between channel and viscera is a core of athemry on acupuncture and meridian, and is also a key entrance for mech anism of acupuncture. This study was designed to discuss theory onacupuncture and meridian through correlation between heart meridian andelectric discharge activities of sympathetic nerve. OBJECTIVE: To investigate the effect of electroacupuncture at differentsegments of heart meridian on the electric discharge activities of the supe rior sympathetic plexus in rabbits. DESIGN: Randomized controlled animal study. SETTING: Central Laboratory of Channel and Viscera Research, AnhuiCollege of Traditional Chinese Medicine. MATERIALS: The experiment was carried out at the Central Laboratoryof Channel and Viscera Research, Anhui College of Traditional ChineseMedicine in March 2004. A total of 36 healthy chinchilla rabbits weighing(2.8±0.3) kg were randomly divided into three groups with 12 in eachgroup, including multi-electro-acupuncturing the wrist (Shenmen region), elbow (Shaohai region) or axils (Jiquan region) sections of heart meridian. METHODS: One rabbit of the same sex whose body mass was less than10% was selected from each group. Superior sympathetic plexus was sepa rated under Olympus anatomic microscope (Japan), and multiple double electrodes materialized as stainless steel sil were embedded. Then, afterfixing with special gelatum (USA), thoracic cavity was closed, temperaturein anus was controlled at (39±0.5) ℃, and 50 mL/L glucose was drippedinto vein for 2 hours before acupuncture. The changes of heart rhythm vari ability (HRV) and electric discharge activities of superior sympatheticplexus were recorded and analyzed. MAIN OUTCOME MEASURES: HRV, ratio between high frequency(HF) and low frequency (LF) of power spectrum. RESULTS: All rabbits were involved in the final analysis. After elec troacupuncture, the total HRV values and electroactivitic frequencies of thesuperior sympathetic plexus were increased, but LF/HF of power spectrumwas decreased. The decrease at the wrist section was superior to others; theaxils section was inferior to elbow section. CONCLUSION: Electroacupuncture at different segments of heart meridi an can improve the electric discharge activities of the superior sympatheticplexus and the leaf-cardiac function. Among the three groups, the relation ship of functions was the closest between the wrist stem of heart meridianand left heart.
8.Cartilage surface of early osteoarthritis in rats induced by papain under scanning electron microscope

BACKGROUND:Osteoarthritis animal models induced by papain are commonly used for studying osteoarthritis.
OBJECTIVE:To observe the morphology of articular cartilage surface during early osteoarthritis process in rats induced by papain and L-cysteine under scanning electron microscope.
METHODS:The right knees of Sprague-Dawley rats were given 0.15 mL mixed solution of 2%papain and 0.03 mol/L L-cysteine in the ratio of 2:1. The left knees were given injections of 0.15 mL 0.9%NaCl, as the control group. Another two rats (four knees) maintained untreated, as the normal control group. Al the rats were observed with scanning electron microscope at 2, 4, 6 weeks after injection, to explore the surface morphology of medial condyle of femur.
RESULTS AND CONCLUSION:The condyles of normal and control groups revealed numerous shal ow pits on the surface of articular cartilage. At 2 weeks after injection, the surfaces of condyles were unsmooth, presenting the shrinkage and distortion. The thinner articular cartilage and local tiny cracks were seen at 4 weeks. The deep and great cracks and cartilage defect were observed on the surface of cartilage at 6 weeks. The time of 4-6 weeks is a reference point for establishing early osteoarthritis model in rats through injections of 2%papain and 0.03 mol/L L-cysteine.

10.Application of laparoscopic training in the standardized residency training of pediatric general surgery

Objective:To explore the application of laparoscopic training in the standardized residency training of pediatric general surgery.Methods:A total of 36 rotating residents who received the standardized residency training in the department of pediatric general surgery of Shenzhen Children's Hospital from January 2017 to December 2019 were selected for laparoscopic training. The training content includes the study and training of laparoscopic theory knowledge, laparoscopic surgery video, skills operation of simulated operating system. Assessment was conducted before and after the training, and statistical analysis was performed to compare the difference of scores before and after the training. The self-evaluation of resident's learning efficiency and the satisfaction with teachers were investigated by questionnaires. SPSS 20.0 was used for paired t-test. Results:After receiving the standardized residency training of pediatric general surgery, the theoretical knowledge and cognition of the application on pediatric general laparoscopic surgery were significantly improved among the 36 residents. The time that the skills spent in vitro simulation box during the simulated operation training was significantly reduced after training, with a statistically significant difference ( P < 0.05). According to the questionnaire survey, resident's satisfaction with self-evaluation of learning efficiency was 97.22% (35/36), their satisfaction with teachers was 94.44% (34/36), and their satisfaction with teaching curriculum was 100.00% (36/36). Conclusion:Laparoscopic training can effectively improve the clinical practice ability of pediatric general surgery residents, which is worthy of promotion.
