BACKGROUND: Many factors can lead to a large area of skin defects, and tissue-engineered artificial skil transplantation composed by seeding cel s and scaffold materials can be used for skin defect repair. OBJECTIVE: To construct the skin implantation scaffold based on human epidermal stem cel s combined with acel ular dermal matrix in nude mice. METHODS: Human epidermal stem cel s from children’s foreskin were isolated and cultured, and the skin of nude mice was obtained to prepare acel ular dermal matrix scaffold. Then, the human epidermal stem cel s were cultured on the acel ular dermal matrix scaffold to construct artificial skin. Ten Sprague-Dawley rats were equivalently randomly divided into two groups: rats treated by human epidermal stem cel s combined with acel ular dermal matrix scaffold as combined group, and those treated by simple acel ular dermal scaffold as acel ular dermal scaffold group after skin defect models were prepared. RESULTS AND CONCLUSION: The artificial skin was milky white, soft and elastic, not easy to break, and has good flexibility, and good plasticity, and could be processed into different shapes. No obvious exudation appeared in the rat wound of two groups after repair. At 2 weeks after modeling, the transplanted skin showed good growth and skin wound healed gradual y in the combined group. In contrast, scar healing and two animals with transplant failure occurred in the acel ular dermal scaffold group. These results show that human epidermal stem cel s combined with acel ular dermal materials in nude mice can construct the tissue-engineered skin, which exerts good repair effects for skin defects in animals.

AIM: To evaluate the quality of Rhizoma gastrodiea processed by different drying methods. METHODS: Vacuum freeze drying and oven drying were adopted. The content of gastrodin was determined by RP-HPLC. RESULTS: The content of gastrodin of Rhizoma Gastrodiae processed by vacuum freeze drying was significantly higher than that by boil with water and oven drying. CONCLUSION: The method of vacuum freeze (drying) can prevent Rhizoma Gastrodiae from degradation of gastrodin, to keep the appearance, to make Rhizoma Gastrodiae convenient for further processing.

Objective To investigate the relationship between the pathogenesis of leukemia and Flt3 gene by detecting the point mutations in juxtamembrane domain coding by Flt3. Methods The exons 14, 15 of Flt3 gene were detected using PCR, PCR/SSCP and gene sequence in 60 leukemia patients with peripheral blood and bone marrow samples. Results We found Flt3-L576P point mutation in 3 cases in juxtamembrane domain in 60 patients, including 1 patient had a deletion of intron 14. Conclusion Flt3-L576 was a new point mutation site, which way be associated with the pathogenesis and development of leukemia.

ObjectiveTo prospectively investigate the feasibility of contrast material-enhanced magnetic resonance angiography(CE-MRA)for visualization of the spinal vasculature.MethodsTen volunteers and fifteen patients with thoracic or lumbar disk herniation underwent contrast-enhanced MR angiography of the thoracoabdominal aorta and posterior branches.Examination Was performed with three-dimensional CE-MRA technique and 0.3 mmol/kg of gadolinium contrast agent Was administrated.ResultsIn all examinations, the Adamkiewicz artery, the anterior spinal artery(ASA),and their connections witll the aorta were identified. The Adamkiewicz artery originated between T8 and L3 in all examinations and derived from the left side of the aorta in 80% of all examinations.ASA was able to be observed from T6 to L3.The diameter of Adamkiewicz artery Was 0.6-1.2 mm and ASA was 0.6-1.0 mm.Conclusion CE-MRA Can be used to lOCalize Adamkiewicz artery and provide accurate information for clinical approach.

Objective To investigate the prevalence of elevated serum alanine aminotransferase ( ALT) and ALT-related dysmetabolism and risk factors. Methods A total of 12 789 adult participated, with no hepatitis B virus infection or alcohol over-intake, received health check-up, including blood pressure measurement, blood glucose and lipid profile test, liver function evaluation and ultrasonography. The subjects were then assigned into two groups according to the serum level of ALT. Metabolic composition was compared between the two groups. Results The prevalence of elevated ALT activity was 8.69%. The waist circumstance, body mass index (BMI) ,systolic blood pressure (SBP) ,diastolic blood pressure (DBP) ,total cholesterol (TC) ,triglyceride (TG) , low-density lipoprotein cholesterol (LDL-C) , urine acid (UA) , fasting blood glucose (FBG) ,HbAlc,total bilirubin (TBIL) ,-y-glutamyltransferase (GGT) ,alkaline phos-phatase ( ALP) of the elevated ALT group were significantly higher than those of the normal ALT group. However, age and high-density lipoprotein cholesterol ( HDL-C) in the elevated ALT group were lower than those in the normal ALT group ( P < 0. 05 ) . The occurrence of overweight, obesity, hyperlipidemia, impaired fasting glucose, diabetes, increased DBP, hyperuricacidemia and fatty liver in the elevated serum ALT group was significantly more than that in the normal ALT group ( P < 0. 05 ) . Multiple logistic regression analysis demonstrated that fatty liver, DBP, BMI.TC, HbAlc were risk factors of elevated serum ALT. Conclusion Non-alcoholic fatty liver disease, lipid metabolic disorders and obesity may be the main risk factors of elevated ALT.

We investigated the correlation between toxin gene exoS,exoU and fluoroquinolone resistance in lower respiratory tract infection with P.aeruginosa so as to provide guidance for reasonable treatment of clinical infections. We collected P.aeruginosa of sputum samples in hospitalized patients from October 2015 to March 2016.The antimicrobial susceptibility was tested by liquid dilution method. The exoS and exoU genes were detected by PCR technique. Results showed that forty-six P.aeruginosa strains were identified from sputum. The exoS and exoU gene positive rate were 86.96 % (40/46) and 69.57 % (32/ 46) respectively, and the highest proportion of genotype was exoS+/exoU+ (60.87%,28/46).Among them,36.96% (17/ 46) were multiple drug-resistant bacteria(MDR).Fluoroquinolone non-sensitive (FQ-NS) strain were 78.95% (15/19) for MDR and 89.47 % (17/19) exoU gene were positive, which was significantly higher than the fluoroquinolone sensitive strains (FQ-S).Compared with the FQ-S strain,FQ-NS strains were serious drug resistance. The drug resistant rate of eefepime and aztreonam were more than 70%,and then meropenem and imipenem were more than 50%.The drugs of lower resistance rate in FQ-NS strain had polymyxin B(10.53%,2/19),amikacin(10.53%,2/19),ceftazidime (15.79%,3/19) and gentamicin (21.05%,4/19).P.aeruginosa of lower respiratory infection carried toxin genes exoS and exoU were higher, the main genetpy was exoS+/exoU+.FQ-NS strains were higher drug resistance rate and a higher proportion of exoU+ strains than FQ-S strains. We should strengthen virulence genes test and drug resistance monitoring in clinical practice.

Objective To determine whether the incidence of pseudolesions of the liver caused by falciform ligament was correlated with liver function. Methods MRI of 150 consecutive cases were evaluated for the presence of pseudolesions caused by ligament compression. All cases were divided into two groups according to the Child-Pugh classification of liver function: Group I-normal liver function and Grade A(98 cases);Group II-Grades B and C(52 cases). Results The incidence of pseudolesions was 31.63% in groups I and no artifacts in Group II.The incidence of pseudolesions was significantly higher in Group I compared with those in Group II.Conclusion The artifacts in the liver caused by falciform compression on MR contrast-enhanced scan images may be seen more frequently in the patients with well liver function than in those with impaired function.
