1.Clinical efficacy of coronary sirolimus-eluting stenting in patients with type 2 diabetes

Objective To evaluate the clinical outcomes of sirolimus-eluting stenting in patients with coronary artery disease and type 2 diabetes.Methods Among 101 patients with coronary artery disease and type 2 diabetes who underwent percutaneous coronary intervention, 67 received sirolimus-eluting stents (SES group) and 34 were treated only with bare metal stents (BMS group). Baseline clinical characteristics, procedural success rate, occurrence of cardiac events during follow-up were recorded and compared between the two groups. Results The procedural success rate was 100% in both groups. During follow-up(average one year), the SES group patients had significantly lower occurrence of cardiac events than those in the BMS group ( 7.5% vs. 32.4%, P= 0.001).Conclusions Sirolimus-eluting stent implantation for patients with coronary artery disease and type 2 diabetes is safe and can reduce major adverse cardiac events in long-term follow-up.
2.Expressions of IL-10, IL-23 and CD86 in lesions of epidermodysplasia verruciformis

Objective To measure the expressions of IL-10, IL-23 and CD86 in lesions of epidermodysplasia verruciformis (EV), and to explore the relationship between cellular immune abnormality and EV pathogenesis. Methods Immunohistochemistry was used to measure the expressions of IL-10, IL-23 and CD86 in tissue samples from 10 patients with EV and 10 normal human controls. Results Three cytokines were observed in all the samples of EV, with the expression score ranging from 3 to 6 and expression intensity from moderate to high. However, of the control specimens, only 1 was positive for IL-10 with the expression score being 3, and expression intensity being moderate. Conclusion The pathogenesis of epidermodysplasia verruciformis may be correlated with the expression abnormality of some cytokines secreted by keratinocytes.
3.Cyclin E, p27 in the Pathogenesis of External Genital Epidermal Tumors

Objective To investigate the significance of cyclin E and p27 in genital epidermal benign and malignant tumors. Methods HPV DNA was examined and typed by polymerase chain reaction (PCR) and restriction fragment length polymorphism (RFLP) in 99 biopsy specimens taken from skin lesions of condyloma acuminatum, external genital carcinoma in situ and invasive squamous cell carcinoma. The expression of cyclin E and p27 in the lesions was examined by immunohistochemistry technique. Results (1) The cyclin E expression in epidermis was notably higher in the lesions than that in normal skin. In HPV-positive samples, the cyclin E expression was higher than that in HPV-negative ones. The cyclin E expression was strongest in invasive squamous cell carcinoma lesions and lowest in condyloma acuminatum lesions. (2) The p27 expression in condyloma acuminatum was slightly higher than that in normal epidermis. But it was notably lower in the lesions of carcinoma in situ and invasive squamous cell carcinoma than that in the normal skin. The expression was also lower in HPV-positive lesions than that in HPV-negative ones. Comparing the three kinds of HPV positive lesions, the intensity of expression was highest in condyloma acuminatum and lowest in invasive squamous cell carcinoma. (3) The study also showed that cyclin E expression was correlated with p27 expression in the lesions. Conclusions The expression of cyclin E and p27 is associated with HPV infection in the lesions. HPV infection might induce epidermal proliferation and malignant transformation by influencing the expression and interaction of cyclin E and p27.
4.Immune function examination and its clinical significance in children with Kawasaki's disease

ObjectiveThe purpose of the study was to summarize the characteristics of cellular and humoral immunity in children with Kawasaki's disease (KD),and to explore the role of cellular and humoral immunity in the pathogenesis of KD.MethodsOne hundred and ninety-six subjects with KD were diagnosed and observed.The serum IgG,IgM,IgA and C3,C4 levels were detected by velocity scatter turbidimetry.The levels of T lymphocyte subsets,natural killer cells (NK cells),and B cells were analyzed by flow cytometry.Chi-square test,independent t-test and Spearman analysis were adopted to analyze data.ResultsOne hundred and seventy-two cases were diagnosed as typical KD,24 cases were incomplete KD,17 cases were refractory KD,179 cases responded to IVIG therapy,and 34 cases were CAL.The occurrence of coronary artery lesion (CAL) in the refractory KD group was 52.9%,which was evidently higher than those patients responding to IVIG therapy(14.0%,P<0.05).In the group of KD,the level of CD19 absolute value(1165±556)/μl was higher than that in the control group(P<0.05),the ratio of CD4/CD8 (2.19±0.77) was higher than that in the control group (P<0.05),the levels of serum IgM ( 1.41 ±0.45) g/L,IgA (0.81 ±0.35) g/L,C3 ( 1.31 ±0.26) g/L were higher than those in the control group (P<0.05),the levels of CD8 (671±308)/μl,NK (337±233)/μl absolute value were lower than those in thc control group (P<0.05).In refractory KD subgroup,the levels of CD19 absolute value,serum IgM,C3 and the ratio of CD4/CD8 were higher than those in the subgroup responding to IVIG therapy(P<0.05),while the levels of CD8,NK absolute value were lower than those in the latter(P<0.05).In the CAL subgroup,the levels of CD19 absolute value,serum IgM,IgA and the ratio of CD4/CD8 were higher than those in the non-CAL subgroup (P<0.05),while the levels of CD8,NK absolute value were lower than those in the latter(P<0.05).The differences of all data between typical KD subgroup and incomplete KD subgroup were insignificant (P>0.05).The severity grade of CAL was positively correlated with the ratio of CD4/CD8(P<0.05).ConclusionThere are cellular immunity and humoral immunity disturbances in the acute stage of KD,while cellular immunity disturbances are more striking.Cellular immunity and humoral immunity are all involved in the pathogenesis of KD.The immunity dysfunction is more significant in refractory KD and CAL.The occurrence rate of CAL is high in refractory KD.The ratio of CD4/CD8 is relevant to the severity of vascular injury.
5.Therapeutic Effect of the Mixture of Luteolin and Rutin in MPTP Induced Mouse Model of Parkinson′s Disease

Objective To study the therapeutic effect of the complex mixture of luteolin and rutin ( MLR) on 1-methyl-4-phenyl-1,2,3,6-tetrahydropyridin ( MPTP) induced Parkinson’ s disease ( PD) mouse model. Methods Seventy-two C57BL/6 mice were divided into six groups randomly ( n=12 in each group): the normal control , model control , madopar (50 mg·kg-1) group, MLR at low (140 mg·kg-1), middle (280 mg·kg-1) and high (560 mg·kg-1) dose groups. PD mouse models were established by intraperitoneal injection of MPTP ( 30 mg · kg-1 ) . Pole test and traction performance were recorded to access the body coordinate capability and strength. The tyrosine hydroxylase (TH), dopamine transport protein ( DAT) , and glial fibrillary acidic protein ( GFAP ) positive cells were detected by immunohistochemical method. Dopamine ( DA ) , dihydroxyphenylacetic acid ( DOPAC ) , homovanilic acid ( HVA ) , 5-hydroxytryptamine ( 5-HT ) and 5-hydroxyindoleacetic acid (5-HIAA) in striatum were quantified by HPLC-ECD. Results MLR significantly ameliorated mouse motor coordination ability (P<0. 01 or P<0. 05). MLR at 280 and 560 mg·kg-1 could increase TH-positive neurons by 69. 00%and 77. 95% compared with the normal control group (P<0. 01) and DAT-positive neurons by 68. 53% and 70. 40% compared with the normal control group(P<0. 05), and decrease GFAP-postive astrocyte reactivity. The treatment with MLR at three doses attenuated the monoamine neurotransmitter disorder. Conclusion MLR markedly improves MPTP caused movement coordinate ability injury in mice and exerts therapeutic action on PD by regulating neurotransmitters in brain, inhibiting the inflammatory reaction and alleviating the neuron injury.
6.Effect of baicalein on brain injury induced by 6-hydroxydopamine at different sites in rats

Aim To explore the therapeutical effect and mechanism of baicalein on two 6-hydroxydopamine (6-OHDA ) induced Parkinson′s disease (PD ) rat models,which received unilateral lesions of the left medial forebrain bundle (MFB ) or caudate putamen (CPu ) made by stereotaxic injection of 6-OHDA (MFB-M,CPu-M).Methods PD rat models were established by microinjection of 6-OHDA into MFB or CPu.The anti-tremor effect of baicalein on PD rat models was examined.Spontaneous activity was recor-ded. Dopamine (DA ), dihydroxyphenylacetic acid (DOPAC)and homovanilic acid (HVA)in striatum were quantified by HPLC-ECD.The tyrosine hydroxy-lase (TH)and OX-42 positive cells were detected by immunohistochemical method.The morphological vari-ation of the neurons was confirmed by analysis at an ul-trastructural level.Results Baicalein significantly in-creased the spontaneous activity in CPu-M.The elec-tromyography (EMG ) recordings revealed that com-pared with 6-OHDA group,the tremor frequency in ba-icalein group was decreased by 55% in MFB-M,and by 60% in CPu-M.6-OHDA treatment decreased DA levels in the striatum,while treatment with baicalein attenuated the DA decreases in CPu-M.Moreover,ba-icalein treatment could increase TH-positive neurons and decrease OX-42-postive microglia compared with 6-OHDA group in both MFB-M and CPu-M.Conclu-sions In the present study,it is illustrated that ①microinjection of 6-OHDA into the MFB and the CPu could cause different pathological changes of PD, which is important for efficacy evaluation;②baicalein showed the ability to alleviate the behavior symptoms in PD-rats at different stages by improving motor function and attenuating muscle tremor;③therapeutic effect of baicalein was produced by inhibiting the inflammatory medium production and release,anti-apoptosis,chan-ging dopamine catabolism, and inhibiting dopamine turnover.
7.Detecting and Typing of HPV DNA in Tissues of Some External Genital Proliferative Diseases

Objective To study the possible relationship between human papillomaviruse (HPV) infection and external genital proliferative diseases. Methods HPV DNA was detected using polymerase chain reaction (PCR) with a consensus primer and typed by using restriction fragment length polymorphism (RFLP) method in 151 paraffin-embedded biopsy specimens from seven kinds of external genital epidermal proliferative lesions. Results In 30 cases of condyloma acuminatum, HPV DNA was positive for all cases, among which HPV6 and HPV11 accounted for 60% and 26.7%, respectively. In 40 cases of bowenoid papulosis, 5 cases of genital Bowen′s disease and 6 cases of erythroplasia of Queyrat, the positive rates of HPV DNA were 55%, 100% and 33.3%, respectively, and the predominant type was HPV16. In 18 cases of external genital invasive squamous cell carcinoma HPV DNA positive rate was 27.8%, and HPV16 was detected in all of the 5 HPV-positive cases. HPV DNA was negative in 32 cases of leukoplakia and 20 cases of extramammary Paget′s disease. Conclusions Condyloma acuminatum is mainly caused by HPV6 infection, followed by HPV11; HPV16 infection is closely associated with the pathogenesis of bowenoid papulosis and external genital squmous cell carcinoma including Bowen′s disease and erythroplasia of Queyrat. In external genital invasive squamous cell carcinoma, HPV16 infection may be one of the multiple carcinogenic factors. No clear relationship is found between HPV infection and leukoplakia or extramammary Paget′s disease.
8.Clinical study of CD64 combined with chemokine in neonatal sepsis

Objective To investigate the value of neutrophil CD64 combined with monocyte chemotactic protein 1 (MCP-1),interleukin-8 (IL-8) and interferon-inducible protein-10 (IP-10) in the clinical diagnosis of neonatal sepsis.Method Cases of neonatal sepsis from March 2015 to June 2016 in the department were chosen as sepsis group.35 neonates with non-infection diseases were selected as noninfection group and 40 healthy newborn infants were assigned as control group.The level of CD64 in blood were detected by flow cytometry,while the level of MCP-1,IL-8 and IP-10 of the serum in the three groups were detected by automatic biochemical analyzer.The differences between groups were compared by single factor ANOVA.The ROC curves of sepsis diagnosed by whole blood CD64,serum MCP-1,IL-8 and IP-10 were drew.Result The level of CD64,IL-8 and IP-10 of the neonatal blood in the sepsis group were significantly higher than that in the non-infection group (P < 0.05) and control group (P < 0.05).There were no significant difference between the sepsis group and non-infection group in MCP-1 (P > 0.05),but significantly higher than that in the control group (P < 0.05).The levels of serum MCP-1 and IP-10 in the non-infection group were significantly higher than those in the control group (P < 0.05),but there was no significant difference between the non-infection group and control group in CD64 and IL-8 (P >0.05).The optimal thresholds of blood CD64,MCP-1,IL-8 and IP-10 in the diagnosis of sepsis were 35.0 MFI,58.6 ng/L,60.3 ng/L,0.46 μg/L.The sensitivity and specificity of the diagnosis of sepsis were 92.8% and 90.6% in CD64,70.0% and 42.6% in MCP-1,78.5% and 68.0% in IL-8,72.8% and 54.6% in IP-10,97.1% and 94.6% when combined.Conclusion The combination test of CD64,MCP-1,IL-8 and IP-10 can improve the sensitivity and specificity of the diagnosis of sepsis.
9.Relationship between expression of PIK3CA protein and the clinicopathology of esophageal squamous cancer

Objective To analyze the expression and clinical significance of PIK 3CA protein in squamous cell carcinoma of the esophagus .Methods Forty -five samples of surgical resection of esophageal squamous cell carcinoma were collected as the observation group ,and 45 normal tissues adjacent to the carcinoma were selected as the control group.The expression of PIK3CA protein in the tissues of esophageal squamous cell carcinoma and the corresponding para cancerous tissue was detected by immunohistochemistry ( IHC).Results The results of IHC showed that the positive expression rate of PIK 3CA protein in esophageal squamous cell carcinoma was 53.33%, which in adjacent normal tissues was 0.00%,the difference was statistically significant (χ2 =32.727,P<0.01).The positive expression rate of PIK3CA protein was correlated with lymph node metastasis ,differentiation and clinical stage of esophageal squamous cell carcinoma (χ2 =5.180,8.201,13.566,P=0.023,0.017,0.001),and was not correlated with the degree of tumor invasion (χ2 =0.952,P =0.329).Conclusion The expression of PIK3CA protein is abnormal in esophageal squamous cell carcinoma ,and the expression of PIK3CA protein plays a role in the occurrence and development of esophageal squamous cell carcinoma .
10.Effects of problem-solving therapy on breastfeeding self-efficacy and psychological status among primiparas

Objective:To explore the effects of problem-solving therapy on breastfeeding self-efficacy and psychological status among primiparas, to provide basis for improving the breastfeeding of primipara.Methods:A total of 102 primiparas were divided into experimental group and control group by random number table method, each group contained 51 cases. Finally, 47 cases in the experimental group and 49 cases in the control group completed the study. The control group received routine nursing, while primiparas in the experimental group carried out problem-solving therapy. Before and after 6-week intervention, the effects was assessed by Breastfeeding Self-Efficacy Scale (BSES), Edinburgh Postnatal Depression Scale (EPDS) and General Well-being Schedule (GWB), respectively.Results:After intervention, the scores of primapara feedig cognition, feeding feeling, feeding skills and BSES total scores were (21.13±2.97) points, (65.47±6.63) points, (31.94±2.59) points, (118.53±8.47) points in the experimental group, significantly higher than (19.43±3.28) points, (61.76±5.20) points, (30.53±2.01) points, (111.71±6.11) points in the control group ( t values were 2.658-4.507, P<0.01); the scores of EPDS were (6.04±1.49) points in the experimental group, significantly lower than (6.92±2.08) points in the control group; the scores of life satisfaction and interest, depression and pleasure, energy, relaxation and tension in General Well-Being (GWB) and total scores were (7.51±1.71) points, (20.19±2.47) points, (21.68±2.32) points, (17.06±2.74) points, (90.45±4.96) points in the experimental group, significanlty higher than (6.41±1.82) points, (18.71±2.98) points, (20.57±1.87) points, (15.78±1.79) points, (84.61±5.26) points in the control group, the difference was statistically significant ( t values were 2.380-5.587, P<0.05 or 0.01). Conclusion:Problem-solving therapy can improve breastfeeding self-efficacy and alleviate postnatal depression as well as enhance subjective well-being of primipara women.
