1.Effect of mandibular protrusion on expression of type X collagen in condylar cartilage of young adult rats

Objective:To examine the relation between the alteration of condylar endochondral in response to mandibular protrusion and the action time length by establishing a model of mandibular protrusion on young adult rats. Methods: 75 female Sprague-Dawley rats aged 9 weeks were divided into two experimental groups and one control group. In two experimental groups, bite jumping appliances created forward positioning of the condyle for twelve hours and whole day, respectively. The experimental rats, together with the control rats, were sacrificed on days 3, 7, 14, 21 and 30, respectively. Tissue sections were cut in the sagittal plane through the mandibular condyle and were processed for in situ hybridization and immunostaining of type X collagen. Results: The peak of type X collagen protein expression in 24-hour experimental groups appeared on day 21 which was on the top of all experimental groups, while in 12-hour experimental groups it was found on day 30. The results of in situ hybridization were basically in agreement with the results of immunostaining. Conclusion: Both intermittent and continuous mandibular advancement can provoke endochondral ossification in young adult rat condylar, while 24-hour ones can produce more obvious and quicker effect.
2.Establishment and Application of Hyperinsulinemic-Euglycemic Clamp Technique in Conscious Rats

Objective To establish the hyperinsulinemic-euglycemic clamp(HEC) technique in conscious rats, and to explore the effect of acute infusion of lipid on glucose infusion rate (GIR) in rats. Methods Ten SD rats were random-ly divided into two groups, 5 rats for each group. The right jugular vein and left carotid artery were catheterized and under-went a HEC with infusion of lipid (intralipid group) for 6 hours, and with continuing infusion of 5%glucose (control group). The plasma levels of free fatty acid(FFA) and GIR were measured by HEC method. Results The level of FFA concentration increased by 17.6-fold, and GIR was reduced by 27%in the intralipid group compared to those of control group (P<0.001). Conclusion The rat model of HEC has been successfully established by intravenous intralipid infusion, which can be con-firmed by HEC technique.
3.Intraductal radiofrequency ablation under direct vision of choledochoscopy for the treatment of malignant common hepatic duct stricture

Novel intraductal radiofrequency ablation for malignant biliary stricture indicated good therapeutic prospect.In traditional intraductal radiofrequency ablation,the radiofrequency catheter was placed through endoscopic retrograde cholangiography or percutaneous transhepatic cholangiogram under the supervision of X-ray.In February 2014,a patient with intrahepatic adenocarcinoma of the bile duct and malignant common hepatic duct stricture was admitted to the Shenzhen People's Hospital.The patient received cholecystectomy + choledocholithotomy + T tube drainage 3 months prior to the admission.The results of histopathological examination indicated reactive hyperplasia.The patient recovered uneventfully after the operation.One week before the admission,the patient had jaundice and umber urine and received choledochoscopy + common bile duct dilatation + histopathological examination + biliary stent placement.The results of pathological examination confirmed that the patient had intrahepatic adenocarcinoma of the bile duct.The patient received intraductal radiofrequency ablation through T-tube fistula under direct vision of choledochoscopy,and recovered well after the operation.Intraductal radiofrequency ablation under direct vision of choledochoscopy not only has the advantages of accurate positioning and real-time monitoring,but also facilitate cyclical repeat treatment.
4.A comparative analysis of surgical treatment to primary tumor in patients with metastatic breast cancer

Objective:Until recently, no consensus has been reached about the treatment of primary tumor in patients with meta-static breast cancer, and whether or not to excise it has not yet reached agreement. This study aimed to evaluate the value of surgical and nonsurgical treatment of primary tumor by analyzing the clinical data of patients with metastatic breast cancer. Methods:This re-view includes the data of 120 metastatic breast cancer patients. Their clinical data in Xiangyang Central Hospital (Hubei province) from January 2005 to December 2012 were collected. All cases were divided into surgical and nonsurgical groups, and the overall survival and symptomatic local progression rates were analyzed. Results:The 120 patients had a median follow-up of 52 months (range=10-92 months). A total of 55 cases were in the surgical group, 30 of whom had surgery before the metastatic diagnosis, and 65 cases were in the nonsurgical group. No significant differences were observed regarding the tumor classification, lymph-node classification, and meta-static site of the tumor in the two groups. Patients in the surgical group experienced longer overall survival (49 months vs. 33 months, P=0.016) and lower rates of symptomatic local progression (14.5%vs. 46.2%, P<0.001). Conclusion:This study demonstrated that the overall survival and symptomatic local control in the surgical group were better than those in the nonsurgical group. However, this hy-pothesis remains to be proved by multicenter clinical trials.

6.Rapid determination of phosphodiesterase 5 inhibitors in invigorative health food by paper spray ionization mass spectrometry method

Objective A rapid method was constructed for detection of 3 kinds of phosphodiesterase 5 (PDE5) inhibitors (sidenafil,vardalafil,tadalafil) in invigorative health food by paper spray ionization mass spectrometry (PSI-MS method).Methods The characteristic ions of the PDE5 inhibitors could be used for preliminary identification and semiquantitative analysis with internal standard method using PSI-MS method.Results The 24 kinds of commercially available health-care products includes capsule,tablet,pill,powder,wine,syrup and liquid were tested.Results from PSI-MS were consistent with the HPLC-UV date.The calibration curves of PSI-MS has a good linearity in a given range.The linear coefficients of analytes were higher than 0.99.The LODs of 3 kinds of PDE5 inhibitors were lower than 1.0 mg/L.The RSDs of this method ranged from 20% to 24%.Conclusion The PSI-MS method was rapid,accurate and targeted,which is compliant for quickly screening the PDE5 inhibitors in large complex matrix samples.

8.Effects of extraction orthodontic treatment on the upper airway dimensions in young adult patients with malocclusion

Objective:To investigate the effects of extraction orthodontic treatment on the upper airway dimensions in young adult patients with malocclusion,and to evaluate the relationship between changes in the craniofacial morphology,position of incisor and upper airway dimensions.Methods:26 patients with upper and lower first premolar extraction and 21 patients without extraction were recruited.Cephalometry and dental cast analysis were performed pretreatment and posttreatment.Results:No statistical difference was found in the parameters of the upper airway dimensions in both non-extraction group and extraction group after treatment.In linear correlation analysis,SPAS was positively correlated with cephalometric variables U1-Y and L1-Y.MAS and IAS was positively correlated with cephalometric variables SNB,U1-Y and L1-Y.The multiple regression analysis revealed that the change of U1-Y had a significant positive effect on SPAS and IAS,and the change of L1-Y also had a significant positive effect on SPAS and MAS.Conclusion:The upper airway dimensions can not be altered during extraction orthodontic treatment,while the changes of incisors are significant associated with the change of the upper airway dimensions.
9.STUDY ON CULTURE CONDITION OF MARINE VITAMIN B_(12)-PRODUCING BACTERIA

The vitamin B 12 Productivity of 14 marine bacterial strains isolated from the China Sea was determined microbiologically using Escherichia coli 113 3. Tests were made on the culturing media and culture conditions for Vitamin B 12 production. The results showed that the culturing medium in which the extract from soybean cake with seawater and some nutriments were added was suitable to recognize further some marine Vitamin B 12 producing bacterial strains. The medium with appropriate cobalt chloride and some salts added may be useful for Vitamin B 12 biosynthesis. The PH value of the fermentation for the testing strains was raised with the lapse of time for fermentation. A better Vitamin B 12 production may be obtained by keeping the fermentation liquid slightly basic till about 96h. No.2627 strain in all the strains tested showed well in the productivity. The mixed cultivation of some strains (including No.2627 strain) tested often has potentiality and superiority for Vitamin B 12 production.
10.Study on the Preparation Process and Quality Control of Naringin Liposome Gel

OBJECTIVE:To optimize the preparation process of Naringin liposome gel,and to establish the quality control method of the gel. METHODS:The preparation method of Naringin liposome was investigated by single factor test with encapsula-tion percentage as index. The phosphatide concentration,the proportion of phosphatide to cholesterol and the proportion of phospha-tide to drug in the liposomes were optimized by orthogonal design. Using formability,spread performance and stability as compre-hensive evaluation indicator,the dosage of carbopol and triethanolamine and drug-loading amount in the gels were optimized by or-thogonal design. The quality control method of the gel was established preliminarily. RESULTS:Naringin liposomes were prepared by the method of ethanol injection;the optimal formulation of the liposomes was as follows as phosphatide 30 mg/ml,the propor-tion of phosphatide to cholesterol 3∶1,the proportion of phosphatide to drug 10∶1;that of the gels was as follows as carbopol 0.30 g,triethanolamine 1.0 g,drug-loading amount 1.0 g/20 g. Average encapsulation efficiency of validation test was 40.19% for Lipo-some(RSD=0.10%,n=3);comprehensive score was 9.8,average content of naringin was 0.58%(accounting for 96.67% of la-bel amount)for gels. The quality control method of the preparation was established,i.g. identification,content determination. CON-CLUSIONS:The optimal preparation formulation is feasible,and the preparation is controllable in quality.
