1.Detection of breast cancer metastasis-related genes by co-word analysis and network analysis

Objective To provide the reference for early diagnosis and treatment of breast cancer by detecting its metastasis-related genes.Methods Breast cancer metastasis-related genes were searched from PubMed-covered papers with their conception matched according to the MetaMap.A gene-gene matrix was generated using data a-nalysis software.An interaction network of breast cancer metastasis-related genes was established using Ucinet 6 and its related indexes were analyzed.Results tp53, thra, erbb2, esr1, cdh1, egfr, nr4al and cd69 were the core genes for breast cancer metastasis.Conclusion Co-word analysis can show breast cancer metastasis-related genes. However, the role of cd69 in breast cancer metastasis remains unclear and is thus necessary to be further confirmed.
2.Efifcacy observation of intravenous iron and oral iron on anemia patients with chronic hemodialysis and study on the difference of oxidative stress reaction

Objective To explore the safety and efifciency of oral iron therapy and intravenous iron therapy in anemia patients with maintenance hemodialysis,in order to gain experience for clinical treatment of this disease. Methods 88 cases collected in the ifrst people's hospital of Changshu city from July 2010 to March 2013 were divided into research group(n=44) and control group(n=44) by random number table. Control group was given oral iron therapy, and research group was t given intravenous iron. The differences of hemoglobin (Hb), reticulocyte, serum ferritin content, superoxide dismutase (SOD) and serum malondialdehyde (MDA) before and after treatment between two goups were recorded and compared. Results The differences of Hb, serum ferritin and reticulocyte numerical before treatment between two groups were not signiifcant, but signiifcant four weeks after treatment, with higher level in research group(P<0.05).The difference of oxidative stress status before treatment between two groups was not signiifcant, but after treatment, the MDA level in research group was higher than control group(P<0.05),while SOD level was lower than control group(P<0.05). Conclusion Compared with oral iron therapy, intravenous iron has better results in improving anemia, but easy to induce oxidative stress reaction.
3.Establishment of age/gender-related serum cycstatin C reference intervals in children by transmission turbidimetry

ObjectiveTo establish pediatric serum CysC reference intervals based on the children's hospital laboratory data in Zhejiang Province and analyse the effects of CysClevels on age and gender. MethodsCysC was one of tests of a routine biochemical screening panel employed for most outpatients and inpatients in the children's hospital, and 8 127 subjects (4 264 boys and 3 863 girls) were selected from 13 567 subjects from laboratory information system according to the exclusion criterion with seriously systemic diseases, cardiovascular diseases, kidney diseases, elevated values of creatinine, urea or ALT for which 1.5-fold upper limit, serum samples with hemolysis, icterus or lipid turbidity, same patients with non-first-time CysC results and living addresses which were not in Zhejiang. The serum CysC was determined by transmission turbidimetry on Roche DPP modular automatic biochemical analyzer. SPSS 17.0 software and EXCEL 2003 were employed for statistical analysis in this study. Results The serum CysC concentration was a Gaussian distribution after log transformation. The mean Log-transformated CysC lg (CysC) concentration of boys in five age groups ( < 1 month, 1 to 3 months, 4 to 11 months, 1 to 2 years and 2 to 16 years) were 0. 224,0. 170,0. 112,0. 061, -0. 011 (mg/L,lg) respectively, and the mean Log-transformated CysC lg(CysC) concentration of girls in five age groups were 0. 222, 0. 164, 0. 089, 0. 057,-0. 010 ( mmg/L, lg) respectively, and no statistically significant differences between Ig( CysC ) and gender in five age groups were found ( t values were 0. 174, 0. 362, 0. 445,- 1. 464 and - 0. 093, respectively,and corresponding P values all were greater than 0. 05 ). The mean lg ( CysC ) concentrations in five age groups were 0. 222,0. 166,0. 100,0. 059, - 0. 010 ( mmg/L, lg), and significant differences between Lg ( CysC ) and ages by Analysis of Variance were observed ( F = 309. 785 and P = 0 in between-groups totally,P = 0 in any two groups). Serum CysC levels were highest in the age of < 1 month, then declined to the age of 2 years and kept stable in the age of 2 to 16 years. The serum CysC reference intervals for children were as follows:0. 95 -2. 92 mg/L in the age of < 1 month, 0. 81 -2.67 mg/L in the age of 1 to 3 months, 0. 65 -2.45 mg/L in the age of 4 tol2 months, 0. 56 -2. 35 mg/L in the age of 1 year and 0. 45 -2. 13 mg/L in the age of 2 to 16 years. ConclusionsThere is no significant effect on pediatric CysC levels with gender but close correlated with age below 2 years old. It is necessary to establish appropriate age-related reference intervals of serum CysC for efficiently evaluating renal function in local children.
4.Whole body hypoxic preconditioning-mediated multiorgan protection in db/db mice via nitric oxide-BDNF-GSK-3β-Nrf2signaling pathway

The beneficial effects of hypoxic preconditioning are abolished in the diabetes. The present study was designed to investigate the protective effects and mechanisms of repeated episodes of whole body hypoxic preconditioning (WBHP) in db/db mice. The protective effects of preconditioning were explored on diabetesinduced vascular dysfunction, cognitive impairment and ischemia-reperfusion (IR)-induced increase in myocardial injury. Sixteen-week old db/db (diabetic) and C57BL/6 (non-diabetic) mice were employed. There was a significant impairment in cognitive function (Morris Water Maze test), endothelial function (acetylcholineinduced relaxation in aortic rings) and a significant increase in IR-induced heart injury (Langendorff apparatus) in db/db mice. WBHP stimulus was given by exposing mice to four alternate cycles of low (8%) and normal air O2 for 10 min each. A single episode of WBHP failed to produce protection; however, two and three episodes of WBHP significantly produced beneficial effects on the heart, brain and blood vessels. There was a significant increase in the levels of brain-derived neurotrophic factor (BDNF) and nitric oxide (NO) in response to 3 episodes of WBHP. Moreover, pretreatment with the BDNF receptor, TrkB antagonist (ANA-12) and NO synthase inhibitor (LNAME) attenuated the protective effects imparted by three episodes of WBHP. These pharmacological agents abolished WBHP-induced restoration of p-GSK-3β/GSK-3β ratio and Nrf2 levels in IR-subjected hearts. It is concluded that repeated episodes of WHBP attenuate cognitive impairment, vascular dysfunction and enhancement in IRinduced myocardial injury in diabetic mice be due to increase in NO and BDNF levels that may eventually activate GSK-3β and Nrf2 signaling pathway to confer protection.

6.Effect of group self-affirmation training on stigma in rectal cancer patients with colostomy

Objective? To explore the intervention effects of the group self-affirmation training on the stigma in the rectal cancer patients with colostomy. Methods? By convenience sampling, 89 rectal cancer patients with colostomy in Yancheng Third People's Hospital from April 2017 to April 2018 were selected and divided into the experimental group (n=45) and control group based on the random number table(n=44).The experimental group received group self-affirmation training on the basis of the conventional nursing care while the control group received the conventional nursing care. The self-designed General Information Questionnaire, Social Impact Scale(SIS), Self-Esteem Scale(SES) were used to evaluate the stigma and level of self-esteem before and after intervention. Results? Before intervention, the two groups had no statistical difference in the scores of stigma (t=-0.323, P=0.747) and level of self-esteem(t=-0.270,P=0.788); after intervention the experimental group had lower scores than the control group in stigma (t=-3.008, P<0.01) and higher scores than the control group in self-esteem(t=4.033, P<0.01) with statistical significance. Conclusions? Group self-affirmation training can reduce the stigma of rectal cancer patients with colostomy and improve their self-esteem.
7.Analysis of genetic variant in a fetus featuring pontocerebellar hypoplasia type 6.

OBJECTIVE:
To explore the genetic basis for a fetus with cerebellar dysplasia and widened lateral ventricles.
METHODS:
The couple have elected induced abortion after careful counseling. Skin tissue sample from the abortus and peripheral venous blood samples from both parents were collected for the extraction of genomic DNA, which was then subjected to whole exome sequencing. Candidate variant was verified by Sanger sequencing.
RESULTS:
Prenatal ultrasonography showed increased nuchal translucency (0.4 cm) and widened lateral ventricles. Magnetic resonance imaging revealed infratentorial brain dysplasia. By DNA sequencing, the fetus was found to carry compound heterozygous variants c.1A>G and c.1564G>A of the RARS2 gene, which were inherited from its father and mother, respectively. Among these, c.1A>G was known to be pathogenic, but the pathogenicity of c.1564G>A was unreported previously. Based on the American College of Medical Genetics and Genomics guidelines, the c.1564G>A variant of RARS2 gene was predicted to be likely pathogenic(PM2+PM3+PP3+PP4).
CONCLUSION
The compound heterozygous variants c.1A>G and c.1564G>A of RARS2 gene contributed to the fetus suffering from pontocerebellar hypoplasia type 6, which expanded variant spectrum of RARS2 gene.

8.Zhuangtongyin Modulates Ferroptosis via the Nrf2-SCL7A11/xCT-Gpx4 Pathway to Im-prove Cerebral Ischemia-reperfusion Injury

[Objective]To investigate the protective effect of Zhuangtongyin on the Middle Cerebral Artery Occlusion(MCAO)model by modulating ferroptosis through the Nrf2-SCL7A11/xCT-Gpx4 pathway and its underlying mechanism.[Methods]C57BL/6J mice were randomly divided into Sham operation group(Sham),model group(MCAO),low-dose Zhuangtongyin group(ZTY-L),high-dose Zhuangtongyin group(ZTY-H),with 5 mice in each group.The MCAO group was modelled by silica gel embolization,the middle cerebral artery of mice was embolized for 1h,then the silica gel was pulled out and reperfusion was performed after 72 h;and the other operations in the Sham group were the same as those in the MCAO group except that the thread plug was not inserted.The neural function of mice was evaluated by Zea-Longa method.TTC staining was used to evaluate the volume of cerebral infarction.The level of brain injury was evaluated by HE staining and Nissl staining.Prussian blue staining and the expression of iron transport-related carrier receptors TfR1 and DMT1 on mRNA level was detected by qPCR to evaluate the iron ion deposition level in mice brain.The expression of lipid peroxidation-related gene ACSL4 on mRNA level was detected by qPCR,and the content of 4-HNE was detected by ELISA kit to evaluate the lipid peroxidation level of mice brain.The expressions of ferroptosis marker PTGS2 mRNA level was detected by qPCR.The expressions of Nrf2,SCL7A11/xCT,Gpx4 in mice brain tissue were detected by Western-blot and immunofluorescence.[Results]Zhuangtongyin improved the nerve function of mice after MCAO(P<0.05)and the cerebral infarction volume of mice(P<0.05)and alleviate the pathological injury of cerebral cortex cells after MCAO operation.Zhuangtongyin attenuated the accumulation of trivalent iron ions in the brain tissue of mice following MCAO.Additionally,Zhuangtongyin downregulated the expression of TfR1 and DMT1 mRNA(P<0.001),a transporter associated with cellular iron ion uptake,in the brains of post-MCAO mice.Furthermore,Zhuangtongyin reduced levels of lipid peroxidation product 4-HNE(P<0.001)and suppressed ACSL4 mRNA expression in brain tissue post-MCAO(P<0.001).Besides,Zhuangtongyin downregulated the expression of PTGS2 mRNA(P<0.001),in the brains of post-MCAO mice.Zhuangtongyin increased the expression of nrf2 into the nucleus(P<0.001),and increased the expression of xCT and Gpx4 in neurons after MCAO(P<0.001).[Conclusion]Zhuangtongyin can enhance the nerve function and reduce cerebral infarction volume in MCAO/R mice,alleviate the pathological damage of cerebral cortex cells,and modulate the expression of key signaling molecules in the Nrf2-SCL7A11/xCT-Gpx4 pathway.Therefore,it is suggested that the mechanism by which Zhuangtongyin improves MCAO/R injury in mice may involve regulating ferroptosis through the Nrf2-SCL7A11/xCT-GPX4 pathway.

Objective To establish a Nomogram model for predicting the occurrence of stigma in elderly patients with colostomy after colorectal cancer(CRC)surgery.Methods A total of 123 eld-erly patients with colostomy after CRC surgery in the Northern Jiangsu People's Hospital Affiliated to Yangzhou University from June 2022 to December 2023 were selected as research objects,and they were divided into stigma group(n=62)and non-stigma group(n=61)according to the occurrence of stigma.Logistic regression analysis was used to screen risk factors,and a Nomogram was drawn and evaluated for its discrimination and consistency.Results Score of the Social Impact Scale(SIS)in 123 elderly patients with colostomy after CRC surgery ranged from 34 to 90,with an average score of(60.14±12.08)and a median score of 48.Female(95%CI,2.467 to 20.978,P<0.001),presence of stoma complications(95%CI,1.766 to 10.093,P=0.001),moderate to low level of perceived social support(95%CI,1.654 to 12.710,P=0.003),and moderate to low level of psychosocial adaptation(95%CI,1.568 to 10.869,P=0.004)were identified as independent risk factors for stigma in elderly patients with colostomy after CRC surgery.The area under the receiver operating characteristic curve was 0.816(95%CI,0.744 to 0.889);the calibration curve had a slope close to 1,and the Hosmer-Lemeshow goodness-of-fit test showed better results(x2=7.722,P=0.461).Conclusion Female,presence of stoma complications,moderate to low level of per-ceived social support,and moderate to low level of psychosocial adaptation are independent risk fac-tors for stigma in elderly patients with colostomy after CRC surgery.Nomogram model established based on these factors can help identify high-risk populations for stigma,so as to obtain individual-ized precise intervention and reduce the incidence of stigma.
