1.Correlation between liver injury and levels of interleukin-18,transforming growth factor-β1,and HBVDNA In patients with chronic hepatitis B

Objective To explore the correlation between liver injury,fibrosis and levels of serum interleukin(IL)-18,transforming growth factor(TGF)-β1,and HBV DNA in patients with chronic hepatitis B(CHB).Methods Sixty-seven CHB cases were collected as experimental group and 20 healthy controls were enrolled as healthy control group.Serum levels of IL-18 and TGF-β1 were analyzed using enzyme linked immunosorbent assay(ELISA),and serum HBV DNA level was measured by fluorescent quantitative polymerase chain reaction(PCR).Liver biopsy was performed in 48 cases.Data analysis was done by variance analysis and data with heterogeneity of variance were analyzed by rank sum test.Partial correlation was employed to analyze the correlations between different data.Results The serum levels of IL-18,TGF-β1,alanine aminotransferase(ALT)and total bilirubin(TBil)were all higher in patients with hepatitis B and cirrhosis than those in controls(P<0.01).With the progression of liver injury,the levels of IL-18 and TGF-β1 were correspondingly increased,and the differences of TGF-β1 level between different groups were statistically significant(P<0.01),with the highest level in the patients with liver cirrhosis.There was no significant difference of the levels of ALT and TBil between the high and low viral load groups,but compared with control group,the differences were both significant(F=10.970,F=7.528;F=14.698,F=13.395;all P<0.05).TGF-β1 level gradually increased with fibrosis stage of CHB increasing from SO to S4,and there were significant differences between two stages except S3 and S4(P<0.01 or P<0.05).The difference of IL-18 level was only significant between SO stage and other stages.There was no significant difference of HBV DNA level among all stages. Partial correlation analysis indicated that IL-18 positively correlated with ALT and TBil(r=0.4806 and r=0.5047,respectively,both P<0.01).HBV DNA had no correlation with IL-18,TGF-β1,ALT and TBil.Conclusions Serum IL-18 and TGF-β1 levels play important roles in the progression of liver injury.Serum TGF-β1 level is correlated closely with post-hepatitis cirrhosis. Serum HBV DNA level is not significantly correlated with liver injury and fibrosis.

Objective To explore the infiltration and apoptosis of eosinophilia(EOS)and the influence of death receptor(Fas) and B cell lymphoma-2 (Bcl-2) expression after Belamcanda and Ephedra Decoction on asthma rats.Methods40 healthy rats of Holland were selected as the research subjects,they were divided into four groups: control group,asthma model group, dexamethasone group,Belamcanda and Ephedra Decoction group with 10 rats in each group.Rat asthma model was prepared by repeated stimulation of egg protein,the symptoms of asthma in each group were evaluated;the number of EOS infiltration in lung tissue was detected by staining method;EOS apoptosis was detected by methyl thiazolyl tetrazolium(MTT) method;Blot Western was used to detect the expression of Fas and Bcl-2 protein;the flow cytometry was used to detect the apoptosis of EOS in rat.ResultsCompared with the rats in the model group, dexamethasone group, Belamcanda and Ephedra Decoction group rats asthma symptoms were significantly reduced, scores were significantly lower (P< 0.05),there was no significant difference between Dexamethasone group and sheganmahuang Decoction group.In the asthma model group,the number of EOS infiltration and EOS% in venous blood and lung tissue were significantly higher than those in control group,EOS apoptosis rate was significantly lower than that of the control group (P< 0.05).In Dexamethasone group,Belamcanda and Ephedra Decoction group infiltration number of EOS and EOS% were significantly lower than that of model group but higher than the control group, the apoptosis rate of EOS protein was significantly higher than the model group and the control group (P< 0.05),there was no significant difference between Dexamethasone group and sheganmahuang Decoction group.The expression of Fas in dexamethasone group,Belamcanda and Ephedra Decoction group was significantly higher than the control group and model group (P< 0.05),Bcl-2 protein was significantly lower than the control group and model group (P< 0.05),there was no significant difference between Dexamethasone group and sheganmahuang Decoction group.Cell flow cytometry showed that the control group had no cell apoptosis,a large number of apoptotic cells were appeared after Belamcanda and Ephedra Decoction treat with.ConclusionBelamcanda and Ephedra decoction can induce apoptosis of EOS expression,it may be associated with the upregulation of Fas, downregulation of Bcl-2 expression.
3.Inhibitory effects of recombinant human Mullerian inhibiting substance on cell proliferation in two human ovarian carcinoma cell lines

AIM:To investigate the inhibitory effects of recombinant human Mullerian inhibiting substance on cell proliferation in human ovarian carcinoma cells (OVCAR8 and SKOV3 cell lines). METHODS:The expression of MISIIR protein and the localization of MISIIR protein were analyzed by Western blotting and confocal spectral microscopy,respectively. Cell apoptosis and cell cycle were detected by flow cytometry (FCM). Cell viability was determined via MTT method. Clone formation test was used to detect oncogenicity in vitro.RESULTS:The MISIIR protein expression in OVCAR8 cells but not in SKOV3 cells was observed. MISIIR expression was seen on the OVCAR8 cell surface and in the cytoplasm with both antibodies. After treated with rhMIS for 48 h,the cell viability was significantly decreased in OVCAR8 cells. rhMIS inhibited the oncogenicity of OVCAR8 cells greatly. The cell apoptosis of OVCAR8 cell exposed to 10 mg/L rhMIS was (31.3±2.1)%,and OVCAR8 cells in the G_1 phase were increased by (70.4±3.0)%. Compared to SKOV3 cells the differences were significant (P<0.01). CONCLUSION:Recombinant human Mullerian inhibiting substance suppresses the growth of MISIIR-positive ovarian cancer cells by inducing apoptosis and cell cycle arrest. We predict that rhMIS might be a new target to treat human ovarian malignancies.

5.Effect of signal transduction inhibitors on human endometrial carcinoma cells with differential PTEN gene expression

Objective To investigate the apoptotic and proliferation effects of signal transduction inhibitors on human endometrial carcinoma cells with different PTEN gene expression. Methods FTEN antisense oligonucleotide and pcDNA3.1/PTEN vector contained PTEN gene were transfected into endometrial carcinoma cells (HEC-1A and Ishikawa). The expression of PTEN protein was detected by confocal spectral microscopy. The endometrial carcinoma cells (HEC-1A, HEC-1A-PTEN-null, Ishikawa, Ishikawa-PTEN) were treated with signal transduction inhibitors, RG-14620, SB203580 (SB) and rapamycin, respectively. Cell apoptosis morphology, cell apoptosis and cell cycle were detected by Hoechst 33258 staining and flow cytometry. Cell viability was determined by methyl thiazolyl tetrazolium assay. Results The PTEN protein expression in two endometrial carcinoma cells (Ishikawa, HEC-1A) was exchanged by PTEN antisense oligonucleotide blocked and pcDNA3. 1/PTEN stable transfected. After treated with RG-14620, SB and rapamycin, marked morphological changes of apoptosis were observed in HEC-1A-PTEN-null and Ishikawa cells. The cell apoptosis of HEC-1A-PTEN-null and Ishikawa cells exposed to SB were significantly increase [(31.6±0.8)% and (37.8±0.8)%, respectively], the G1 phase cells were increased to (84.1±3.2)% and (87.5±1.9)%. While cell viability was significantly decreased in HEC-1A-PTEN-null and Ishikawa cells, the cell viability of HEC-1A-PTEN-null and Ishikawa cells exposed to SB were (54.0±2.1) % and (49.0±1.7) %. Conclusion Loss of PTEN in endometrial carcinoma cells may improve the G_1 phase cells and apoptotic effects, inhibit the cell proliferation, which due to the sensitivity of cells to related signal transduction inhibitors.
6.Effect of Pneumatically Ballistic Extracorporeal Shockwave on Chronic Nonspecific Low Back Pain

Objective To observe the clinical effect of pneumatically ballistic extracorporeal shockwave on chronic nonspecific low back pain. Methods 66 cases with chronic nonspecific low back pain were randomly divided into treatment group (n=33) and control group (n=33). There were 1 case drop-out in the treatment group and 2 in the control group. The treatment group accepted pneumatically ballistic extracorporeal shockwave, and the control group accepted hot magnet for 2 weeks. They were assessed with Visual Analogue Scale (VAS) and Finger-floor Distance (FFD) before and after treatment. Results The scores of VAS and FFD improved in both groups after treatment (P<0.05), and the difference before and after treatment were more in the treatment group than in the control group (P<0.05). Conclusion The pneumatically ballistic extracorporeal shockwave is more effective on chronic nonspecific low back pain than hot magnet.
7.Comparative analysis of CT and MRI in evaluating therapeutic efficacy of primary liver cancer after transcatheter arterial chemoembolization with lipiodol

Objective To compare the value of computed tomography(CT)and magnetic resonance im-aging(MRI)in assessing the curative effect of primary liver cancer after transcatheter arterial chemoembolization (TACE). Methods 45 patients with primary hepatocellular carcinoma treated with TACE for 3 ~ 5 times were included in the research. CT ,MRI and digital subtraction angiography (DSA) were performed after treatment. DSA was used as a standard to analyze the lesions ,residual or lesion recurrence and the detection of tumor cap-sule in CT and MRI DSA. Results The scanning accuracy and sensitivity of residual lesions or lesion recurrence after TACE in CT was much lower than MRI ,and there is a significant difference between the two groups (P <0.05). In terms of tumor capsule detection accuracy after TACE ,CT was much lower than MRI(P < 0.05). In detection rate of residual or lesion recurrence in different types of lipiodol deposition after TACE ,CT was much lower than MRI (P < 0.05). Conclusions Compared with the CT test ,MRI detection has a higher accuracy and sensitivity of residual lesions or recurrence after TACE. MRI can not only effectively show the tumor cap-sule ,but also detect residual lesion or lesion recurrence of different types of lipiodol deposition. MRI is superior to CT in the evaluating the curative effect of primary liver cancer after TACE.
8.Value and assessment of P1 component in children with CIs

OBJECTIVE Aim to explore the value and the selection of observation target of P1 component in children with cochlear implant(CI). METHODS 13 children(4.37±0.73 years old) with right side CI and one year of regular post-CI rehabilitation were recruited as CI group. 15 gender and age (4.25±0.84)years matched children with left side external and middle ear malformation were selected as control group. We collected their AEP which showed their reaction to 1000 Hz pure tone and analyzed the P1 component. RESULTS The cut off value between CI group and control groupwas 10.4mV for P1 Amplitude(P1A) and 110.5 mV for P1 Latency(P1L). More precisely, the values of CI group were above the cut off value while the values of control group were below it. Whether choosing P1A or P1L as dividing standard, the AUC were between 0.5 and 0.9(AUC: P1A0.733, P1L0.800), which showed medium distinguishing significance. P1L component showed greater You-den index(0.590>0.471) and sensitivity(0.923> 0.538) while P1A showed greater specificity(0.933>0.667). CONCLUSION P1L shows greater ability in distinguishing the difference between CI group and control group while P1A has advantage in determining their common feature. Generally, P1L shows higher value in studying CI children. We need to make choice between P1A and P1L in different situation and use P1A and P1L standard in series or parallel.
9.Clinical analysis of cryptococcosis neoformans infection in patients with connective tissue disease

Objective:To improve the level of clinical diagnosis and treatment by analyzing the clinical features and relevant factors of cryptococcosis neoformans in patients with connective tissue disease(CTD).Methods:Twelve patients with CTD and cryptococcosis neoformans infection in Peking University People's Hospital from January 2010 to April 2021 were retrospectively enrolled. Clinical and laboratory data, treatment and outcome were collected and analyzed. Independent sample t-test or Rank-sum test was used. Results:The age of the patients ranged from 18 to 85 years old(mean 51 years old), all of whom were female. None of them were exposed to pigeons and their feces. Of the 12 patients, 3 patients suffered from rheumatoid arthritis, 7 patients had systemic lupus erythematosus, 1 patient was diagnosed with primary Sj?gren 's syndrome, and 1 patient was diagnosed as undifferentiated connective tissue disease. Four cases were cryptococcal meningitis, 8 were pulmonary cryptococcosis. None of the 12 patients had immunodeficiency virus infection. All 12 patients were given glucocorticoid alone or combined with immunosuppressive or biological agents. All were detected with positive cryptococcus neoformans antigen in serum; 6 got lumbar puncture, 2 cases were positive for ink stain, cerebrospinal fluid (CSF) culture were positive in 2, in whom 3 had high intracranial pressure, in which the highest one was more than 600 mmH 2O (1 mmH 2O=0.009 8 kPa); 7 cases underwent lung biopsy. Among these patients, all were positive for cryptococcosis neoformans in lung tissue pathological examination; 6 had the number of peripheral lymphocytes less than 1.0×10 9/L, and 2 were detected for the number of CD4 + T cell, which was significantly decreased. As for the initial anti-fungal drug therapy, all cases were treated with fluconazole intravenously; 2 were treated with combined amphotericin, 1 was treated with combined fluorocytosine, 1 was treated with amphotericin and fluorocytosine. Then oral flu-conazole was prescribed as sequential therapy. The whole treatmentcourse ranged from 4 to 21 months. Eleven patients were cured, and 1 was relieved. Conclusion:Patients with connective tissue disease complicated with cryptococcus neoformans infection have atypical clinical symptoms. Treatment with immunosuppressive drugs and glucocorticoids are related causes. Patients with decreased peripheral blood lymphocytes, especially CD4 + T cell, are more susceptible to infection. Early diagnosis and timely treatment are the key to improve the prognosis and cure of the disease.

The review provides an overview on the connotation, extension, and adverse effects, as well as potential underlying mechanisms of fear of missing out in mobile social media among college students, which aims to highlight future attention, as well as prevention and intervention reference on fear of missing out in college students.
