The treatment plan of facial neuritis in Evidence-based Guidelines of Clinical Practice in Acupuncture and Moxibustion (2011 edition) is discussed, and case information of facial neuritis during the recent five years in department of acupuncture and moxibustion, PLA General Hospital, is retrospectively analyzed. In accordance with anatomy of the facial nerve to form the acupuncture prescription, the detailed diagnosis and treatment method for facial neuritis are introduced. The advantages of the diagnosis and treatment method for facial neuritis are summarized, hoping to establish a more comprehensive, standardized and unified treatment plan.

A promoter fragment (7S?P ) of ? subunit gene was isolated by PCR from genomic DNA in various soybean accessions, including cultivars Nannong99-10,N2899,Nannong 88-1, and wild soybeans Jiangpu -1 and ZYD4174.The sequences of 7S?P fragment from these five soybean accessions shared 99% homology, this indicated that the promoter regions of ? subunit gene were conserved. Meanwhile , sequencing analysis showed that the 7S?P fragment contained several seed-specific motifs, such as RY motif, AGCCCA motif, ACGT motif and A/ T rich motif. The expression vector pBI121-7S?P was constructed with the 7S?P fragment (from Nannong99-10) and the GFP reporter gene for functional analysis. Arabidopsis plants were transformed by Agrobacterium mediated method. Southern blot results showed that the 7S?P had been integrated into the genome of Arabidopsis. Assay of GFP expression in the seeds of transgenic Arabidopsis was determined to identify the function of 7S?P promoter. The results showed that 7S?P was a seed-specific promoter.

BACKGROUND:With the development of genetic engineering and tumor molecular biology, gene therapy for tumors has become a new treatment modality.
OBJECTIVE:To explore the effect of the KDR gene silencing on the proliferation and invasion capacity of breast cancer MCF-7 cel s.
METHODS:Interfering RNA (siRNA) sequences for smal molecule KDR gene was designed and transferred into human breast cancer MCF-7 cel s. Then, RT-PCR and western blot assay were used to detect the KDR mRNA and protein expression. Flow cytometry, cel counting kit-8 test and Transwel test were employed to detect the cel cycle, proliferative capacity and invasion capacity of breast cancer MCF-7 cel s after the KDR gene silencing.
RESULTS AND CONCLUSION:After 48 hours of KDR silencing, the mRNA and protein expressions of KDR in MCF-7 cel s were decreased obviously;MCF-7 cel s arrested at G0/G1 stage and the number of cel s at S stage was reduced. Cel proliferation was inhibited significantly. The amount of cel s passing through the filtering membrane became less. After KDR gene silencing, the proliferation and invasion of breast cancer MCF-7 cancer stem cel s were inhibited remarkably, indicating that KDR gene silencing may be a new target for the effective treatment of breast cancer.

Objective Toinvestigatetherelationshipbetweendifferenttypesofinternalwatershed infarctionandtandemstenosesofinternalcarotidartery(ICA).Methods Atotalof55patientswith internal watershed infarction confirmed by head MRI and diffusion-weighted imaging (DWI )examination were enrolled. They all underwent the extracranial internal carotid artery (ICA ) ultrasonography and intracranial cerebral artery MR angiography (MRA)examinations. According to the findings of imaging,the 55 patients with internal watershed infarction were divided into a simple internal watershed infarction (IWSI)group and an internal watershed infarction accompanied with ipsilateral cortical watershed infarction (C-IWSI)group. The relationship between the two types of internal watershed infarction and tandem stenoses of ICA was analyzed. Results (1 ) Of the 55 patients with internal watershed infarction,24 cases (43. 6%)were in the internal watershed infarction group and 31 cases (56. 4%)were in the C-IWSI group. The ipsilateral vascular stenosis were ICA 20 cases (36. 4%,including extracranial segment 11 cases and intracranial segment 17 cases),middle cerebral artery (MCA)44 cases (80. 0%), and tandem stenoses of ICA 15 cases (27. 3%). (2)Ipsilateral tandem stenoses of ICA:2 cases were in the IWSI group (intracranial ICA+MCA 2 cases);13 cases were in the C-IWSI group (extracranial ICA+intracranial ICA +MCA 4 cases,extracranial ICA + intracranial ICA 1 case,extracranial ICA + MCA 2 cases,and intracranial ICA+MCA 6 cases). (3)Compared with the IWSI group,the incidences of ipsilateral ICA stenosis and tandem stenoses of ICA in patients of the C-IWSI group were higher (54. 8%[n=17]vs. 12. 5%[n=3],41. 9%[n=13]vs. 8. 3%[n =2]),and there were significant differences(P =0. 001, 0.006]). The incidences of extracranial and intercranial ICA stenosis were higher than those of the IWSI group (35. 5%[n=11]vs. 0,45. 2%[n=14]vs. 12. 5%[n=3]),and there were significant differences (P=0.003,0.009).Conclusion Inthedifferenttypesofinternalwatershedinfarction,theincidenceof tandem stenoses of ICA is different. The IWSI patients with ipsilateral cortical watershed infarction often accompany by tandem stenoses of ICA.

Objective:To study the distribution pattern of CIK cells re-infused by different manner.Methods:Isotope 32P-? dATP and fluorescence dye CM-DiI were used individually to label CIK cells. CIK cells labeled by the two methods in vitro were inoculated to nude mice by intraperitoneal injection or tail vein injection. Radioactivity quantitative measurement and fluorescence microscopy were used to analysis dynamic distribution of CIK cells among organs of mice.Results:The CIK cells were quickly distributed to organs such as liver, spleen, kidney, lung, stomach and intestine after inoculation into nude mice. Among those organs, the liver, spleen and kidney showed highest distribution concentration of CIK cells. Early stage after infusion, concentration of CIK cells in lung above all reached peak via tail vein, and by means of intraperitoneal injection, distribution of CIK cells in intraperitoneal organs firstly got to max. CIK cells remained alive in liver and spleen for more than 2 weeks.Conclusion:The extensive distribution pattern of CIK cells among organs shows that CIK cells can be used as drugs against various malignant tumors in organism. Infusion of CIK cells via blood vessel maybe suit for tumor of organs with rich blood supply, and application by means of body-cavity way should suit for malignant effusions and limited lesion in it.

Objective To study the effect of varicocele(VC) on the expressions of hypoxia-inducible factor-1?(HIF-1?) and apoptosis-associated gene Bax in germ cells of adolescent rats,and investigate the mechanism of the infertility resulting from varicocele. Methods The varicocele model was established by partial ligation of the left renal vein in adolescent male Wistar rats. A total of 25 rats were randomly divided into 2 groups,namely varicocele group(n=15) and sham operation group (SOG,n=10). All the animals were killed 30 d after operation. The left testis was collected and the expression of HIF-1? and Bax was detected by immunohistochemical method. Objective To explore the proliferative and apoptotic effect of 3 different siRNAs targeting survivin in human bladder cancer cell line EJ28. Methods Three siRNAs targeting survivin and 1 fluorescence-labeled siRNA as a negative control were chemically synthesized. Transfetion efficiency was observed under fluorescence microscope after transfecting the fluorescence-labeled siRNAs. EJ28 cells were divided into 6 groups: siRNA164 group, siRNA167 group, siRNA389 group, and negative control group, lipofectin group and cell control group. The proliferation of EJ28 cell was detected by MTT assay at 24, 48 and 72 h after the transfection. Apoptotic rate was detected by Annexin V-FITC-FCM assay at 48 h.Results All the sequence-specific siRNAs targeting survivin efficiently suppressed the proliferation of EJ28 cells. siRNA 164 had the most powerful effect because of its highest inhibitory rate of (41.32?2.54)% and the highest apoptotic rate of (13.20?0.25)% at 48 h after transfection. Conclusion RNAi targeting survivin has a potential value in the gene therapy of bladder cancer.

Objective To observe the influence of reperfusion after adnexal torsion (AT) on malondialdehyde (MDA), glutathione peroxidase catalase (GSH-Px) and catalase (CAT) contents of ovarian tissue in rabbits. Methods Forty female Japanese long-eared white rabbits were randomly divided into study group (n=32) and control group (n=8). The left adnexa of rabbits in the study group was clockwise twisted three laps, and then fixed on the left abdominal walls. Adnexal detorsion was then done 24 hours after adnexal torsion in the study group, and then the rabbits were divided into 4 groups (8 each). Both ovaries of each group were removed 24h, 48h, 72h and 96h, respectively, after reperfusion. The rabbits in the control group received sham-operation and both ovaries were removed 96h later. The removed left ovaries were used for biochemical detection of GSH-Px, CAT and MDA. All the right ovaries were used as experimental internal-control. Results The activity of GSH-Px declined significantly 24h to 72h after adnexal detorsion (P<0.01), and no significant difference was found when compared with that of control group 96h later (P>0.05). The activity of CAT declined significantly 24h and 48h after adnexal detorsion (P<0.01), then it began to increase after 72h, and it became significantly higher in 96h group as compared with that in control group (P<0.05). MDA content in study groups increased obviously 24h and 48h after adnexal detorsion (P<0.01), and it declined 72h later albeit still significantly higher than that of control group (P<0.01), and then it continuously declined to reach the level of control group 96h later (P>0.05). Conclusions Detorsion after adnexal torsion can affect the degree of oxidative stress injury in rabbits' ovaries as shown by the changes in the activities of GSH-Px, CAT and MDA. With the elongation of detorsion time, ovarian injury will be gradually alleviated.
