Aim To investigate the reversal of resistance to doxorubicin by CJZ3,a lomerizine derivative,in doxorubicin-resistant human myelogenous leukemia(K562/DOX)cells.Methods The effect of CJZ3 on P-glycoprotein(P-gp)function and resistance to doxorubicin in K562/DOX cells were observed by using flow cytometry and MTT assay. Results CJZ3 increased intracellular uptake and accumulation of rhodamine123(Rh123)in K562/DOX cells in a concentration-related manner, significantly inhibited the efflux of Rh123 from the cells, and increased the doxorubicin-induced cytotoxicity, apoptosis rate,Caspase-3 activity in doxorubicin-treated cells and the intracellular accumulation of doxorubicin. Conclusions The reversed effect of CJZ3 on resistance to doxorubicin in K562/DOX cells was associated with its inhibitory effect on P-gp function in vitro.

Journal of Environment and Health 2007;0(11):-
Objective To preliminarily investigate the conditions of the performance of chlorine dioxide generator for drinking water disinfection and know the overall situation of the industry. Methods From 2007 to 2008,6 companies'products were chosen for this investigation according to certain rules, and their performance indicators, such as chlorine dioxide output and conversion rate were tested by using the Five-step iodimetry method. Results On-site generating technology was commonly used, and the average conversion rate of the compound type generator was 57.06%,and the pure type generator was 84.78%.Conclusion The conversion rate of the pure type generator is better than the compound type, and the newly invented pure type generator has much better performance and lower disinfection cost.

AIM: To assess the relative agreement of GAT and NCT in IOP measurement by comparing the differences between Goldmann applanation tonometer (GAT) and non-contact tonometer (NCT) in intraocular pressure (IOP) detection.METHODS: IOP of 529 eyes of 265 volunteers were measured with both NCT and GAT, respectively.RESULTS: The measurement results of NCT were lower than that of GAT, there was significant difference between the IOP measured with NCT and GAT (19.13 vs23.43, t=22.644, P<0.05). With the increasing of IOP values, the difference magnitude was greater, especially in IOP group that was more than 30mmHg, but the correlation coefficient became lower.CONCLUSION: The measurement results with NCT are lower than that of GAT. When the IOP with the NCT is in borderline value, it need be corrected with GAT, in order to discover the pathologically elevated IOP and avoid the misdiagnosis and mistreatment of glaucoma.

ObjectiveTo observe the effect of moxibustion with medicine cake on mRNA expression of matrix metalloproteinases-2,9 (MMP-2,MMP-9)in artherosclerosis (AS) plaques of rabbits, and to explore the effect of moxibustion with medicine cake to the stability of artherosclerosis plaques of rabbits.MethodsAS rabbit models were established by feeding high-fat diet. 75 New Zealand big-eared rabbits were divided into 5 groups, blank group (blank control group), model group (AS model group), direct moxibustion group (AS model + moxa cone direct moxibustion), moxibustion on medicine cake group (AS model + moxibustion on medicine cake), western medicine group (AS model + atorvastatin), 15 rabbits in each group. Then the mRNA expression of MMP-2, MMP-9 was detected in the atherosclerotic plaques of rabbits with in situ hybridization.ResultsThe expressions of MMP-2, MMP-9 in the atherosclerotic plaques of rabbits were all be controlled in direct moxibustion group, moxibustion on medicine cake group, western medicine group (P<0.01 or P<0.05). The expressions of MMP-2 and MMP-9 in the moxibustion on medicine cake group and western medicine group were obviously lower than those of the direct moxibustion group.ConclusionThe expression of MMP-2, MMP-9 in the atherosclerotic plaques of rabbits can all be controlled in direct moxibustion group, moxibustion on medicine cake group, and western medicine group; as well as the plaques can all be made stable. The efficacy in the moxibustion on medicine cake group and western medicine group is superior to that of the direct moxibustion group.

Objective To evaluate the stability and biodistribution of a novel SPECT-MRI bi-functional agem 99Tcm-Gd-DTPA-DG in tumor-bearing nude mice. Methods DTPA-DG was synthesized and then conjugated with Gd2O3 to generate Gd-DTPA-DG. The tumor-bearing nude mice were scanned by MRI to evaluate the tumor targeting ability of Gd-DTPA-DG. The orthogonal experiment was applied to optimize pH value of reaction medium and reaction temperature. The radiolabeling efficiency was measured by thin layer chromatography. The distribution of 99Tcm-Gd-DTPA-DG in nude mice was evaluated by scintigrapy in vivo. The % ID/g was measured at different time after intravenous injection of 99Tcm-Gd-DTPA-DG. Results The tumor was significantly enhanced by Gd-DTPA-DG with MRI. The radiochemical purity of 99Tcm-Gd-DTPADG was about 98.5% and remained 96.2% at room temperature for 6 h. The tumor was well visualized by 99TcmGd-DTPA-DG SPECT at 2 h after injection. The tumor uptake was (1.48 ±0.12) %ID/g, and the rumor to muscle radioactivity ratio was 2.91. Conclusions MRI contrast of Gd-DTPA-DG may enhance tumor detection. 99Tcm-labeled Gd-DTPA-DG may be useful for tumor imaging and might have a potential role as a SPECT-MRI bi-functional agent.
