1.Effect of isoflurane on expression of p-GSK-3β and β-catenin in neural stem cells in hippocampus of developing rats

Objective To evaluate the effect of isoflurane on the expression of phosphorylated glycogen synthase kinase-3 beta (p-GSK-3β) and β-catenin in neural stem cells (NSCs) in the hippocampus of developing rats.Methods Twenty-four 7-day-old Sprague-Dawley rats,weighing 15-20 g,were divided into 2 groups (n =12 each) using a random number table:control group (group C) and 2% isoflurane group (group Ⅰ).Group C inhaled 30% oxygen for 4 h.Group Ⅰ inhaled 2% isoflurane in 30% oxygen for 4 h.Six rats were randomly selected from each group,and 5-bromodeoxyuridine (BrdU) 200 mg/kg was intraperitoneally injected immediately before anesthesia to assess the proliferation of NSCs in the hippocampal dentate gyrus.The rats were sacrificed at 6 h after the end of anesthesia,and hippocampi were isolated for determination of the number of BrdU positive cells in the dentate gyrus (by immunohistochemistry) and expression of p-GSK-3β and β-catenin in hippocampal tissues (by Western blot analysis).Results Compared with group C,the number of BrdU positive cells was significantly decreased,and the expression of p-GSK-3β and β-catenin was down-regulated in group Ⅰ (P<0.05).Conclusion Isoflurane can inhibit the proliferation of NSCs in the hippocampal dentate gyrus of developing rats,and the mechanism may be related to down-regulation of the expression of p-GSK-3β and β-catenin.
2.Clinical Observation of Acupuncture for Idiopathic Oligoasthenozoospermia

Objective To observe the clinical efficacy of acupuncture in treating male idiopathic oligoasthenozoospermia. Method A hundred patients with oligoasthenozoospermia were recruited and randomized into a treatment group and a control group, 50 cases in each group. The treatment group was intervened by acupuncture, while the control group was by placebo acupuncture at non-meridian points. The clinical efficacies and changes of the relevant parameters of sperms after treatment were observed. Result The clinical efficacy of the treatment group was significantly better than that of the control group (P<0.05); in the treatment group, the relevant parameters of sperms were significantly improved after intervention (P<0.05); after intervention, the changes of the parameters in the control group were statistically insignificant (P>0.05); after intervention, the improvements of the relevant parameters of sperms in the treatment group were more significant than that in the control group (P<0.05). Conclusion Acupuncture can effectively improve the sperm density, sperm motility, and sperm viability, and enhance the recovery rate in treating infertility, and it’s easy-to-operate, without significant adverse reactions, safe, and reliable.

4.Influence of seawater immersion of open abdominal wound on function and pathological changes in liver in dog

Objective To investigate the influence of seawater immersion of open abdominal wound on function and pathological changes in liver in dog.Methods Twenty healthy dogs were divided randomly into two groups.A perforating abdominal wound was was made in each animal of both groups.Animals in control group(n=10)only abdominal wound was made,while abdominal wound was followed by seawater immersion in the animals of seawater immersion group(n=10).Blood samples were taken before the trauma(0 hour)and at 4,8,12,16 and 20 hours after the trauma and water immersion for both groups.Total bilirubin(TB),alanine transaminase(ALT),aspartate transaminase(AST),lactate dehydrogenase(LDH),blood ammonia(NH3),prothrombin time(PT),tumor necrosis factor-?(TNF-?),interleukin-6(IL-6)and endotoxin(ETX)were assayed.Experimental animals were sacrificed 24 hours after trauma,and the pathological changes in hepatic tissue were studied.Results TB,ALT,AST and PT were higher in seawater immersion group,than those in control group,with a marked elevation at 16 hours.In seawater immersion group,LDH was significantly higher than those in control group at 4 and 12 hours,and NH3 was increased significantly at 4 hours in experimental group(P

Objective To evaluate the effect of isoflurane anesthesia on Wnt3a expression in the hippocampus of developing rats.Methods Twenty-four 7-day-old male Sprague-Dawley rats,weighing 125-155 g,were randomly divided into 3 groups (n=8 each) using a random number table:control group (group C),4 h inhalation of 2% isoflurane group (group I4),and 6 h inhalation of 2% isoflurane group (group I6).The rats were sacrificed at 6 h after the end of treatment in each group,and the hippocampi were removed for determination of Wnt3a mRNA expression (by real-time reverse transcriptase polymerase chain reaction) and Wnt3a expression (by Western blot).Results Compared with group C,the expression of Wnt3a and Wnt3a mRNA in hippocampi was significantly up-regulated in I4 and I6 groups (P<0.05).Compared with group I4,the expression of Wnt3a in hippocampi was significantly up-regulated (P<0.05),and no significant change was found in the expression of Wnt3a mRNA in hippocampi in group I6 (P>0.05).Conclusion The mechanism of isoflurane-induced neurotoxicity is probably related to upregulation of Wnt3a expression in the hippocampal tissues of developing rats.
7.Construction of core Competence Index System for nursing emergency personnel in infectious disease emergencies——thinking on nursing emergency rescue based on COVID-19 epidemic

Objective:To construct the index system of the core competence of nursing emergency personnel in the case of infectious disease outbreak, and to provide reference basis for the assessment and training of the core competence of nursing emergency personnel.Methods:Based on the core competence indicators of nurses defined by the International Nurses' Association and the action framework of WHO's outbreak of infectious diseases as the theoretical framework, the item pool was summarized through semi-structured interviews and research group meetings, the Delphi expert letter consultation method was used to construct the indicator system, and the analytic hierarchy process and the average distribution method were used to determine the weight of indicators at all levels.Results:Two rounds of Delphi expert letter consultation, 36 questionnaires were issued in each round, and the effective recovery rate was 86% (31) and 92% (33), respectively.Finally, it was determined that the core competence index system of nursing emergency personnel for infectious disease emergencies included 5 first-level indicators, 13 second-level indicators and 53 third-level indicators. The coefficient of variation ranged from 0 to 0.173, the weight ranged from 0.074 to 0.525, and the balanced combination weight ranged from 0.165 to 0.266.Conclusion:The index system of the core competence of nursing emergency personnel in infectious disease emergencies is comprehensive and scientific, which can provide theoretical basis for evaluating and training the core competence of nursing emergency personnel in infectious disease emergencies.
8.Comprehensive analysis of insulin products complex disulfide bonds structure by high resolution mass spectrum

The correct pairing of disulfide bonds maintains the correct folding mode and high-level structure formation of peptides and protein drugs, which is crucial for the quality control of products. In order to ensure that the disulfide bonds are correctly paired, disulfide bond analysis is an essential part of peptides and protein drug characterization. Mass spectrometry can be used to analyze disulfide bonds. However, insulin and its analogues have two pairs of disulfide bonds without restriction enzyme cutting site. Conventional collision-induced dissociation (CID) and high-energy induced cleavage (HCD) cannot accurately locate the complex disulfide bond. In our study, three methods were used to localize the complex disulfide, including enzyme digestion combined with key peptide fragment in source decay (ISD) fragmentation method, enzyme digestion combined with partial reduction alkylation method, intact protein source ISD and electron transfer dissociation (ETD) cleavage method, The applicability of insulin aspart, insulin lispro and insulin glargine were also investigated. This study provides a new way for the quality control of disulfide bonding mode of insulin and its analogues, and also provides a reference for the disulfide bond localization of peptides or proteins containing this complex disulfide bond.
9.Three new belamcandaquinones from Ardisia punctata

Aim To study the chemical constituents of Ardisia punctata. Methods Compounds were separated with a combination of multi-chromatography. Their chemical structures were determined on the basis of spectral analysis and single crystal X-ray diffraction. Results Three compounds were isolated from chloroform extract of the roots of Ardisia punctata. Their structures were elucidated as 2-tridecyl-3-[ (2-tridecyl-3-acetoxy-4-methoxy-6-hydroxy) -phenyl ] -6-methoxy-1,4-benzoquinone ( 1 ), 2-tridecyl-3-[ ( 2 -tridecyl-4,6-dihydroxy ) -phenyl ] -6 -methoxy-1,4-benzoquinone ( 2 ) and 2 -tridecyl-3 - [ ( 2 -pentadecyl-4,6-dihydroxyl)-phenyl]-6-methoxy-1,4-benzoquinone (3). Conclusion The three compounds are new1,4-benzoquinone derivatives.
10.Investigation and analysis of the gastrointestinal symptoms in patients with chronic obstructive pulmonary disease

Objective To investigate the gastrointestinal symptoms in patients with chronic obstructive pulmonary disease (COPD) and explore its possible reasons.Methods Thirty-six patients with stable COPD were divided into mild group [21 cases,forced expiratory volume in first second (FEV1) measured values/predictive values ≥50%] and severe group (15 cases,FEV1 measured values/predictive values < 50%) according to the pulmonary function.Replacement of 19 healthy volunteers as control group.The patients and volunteers completed the Gastrointestinal Symptom Rating Scales (GSRS) scores and received electrogastrography,the plasma vasoactive intestinal peptide (VIP) and intestinal fatty acid binding protein (Ⅰ-FABP) were detected.Results The GSRS scores in severe group and mild group was higher than that in control group[(17.73 ± 4.64),(14.29 ± 5.44) scores vs.(8.00 ± 2.29) scores],and severe group was higher than that in mild group,there was significant difference (P < 0.05).The percentage of normal slow waves of fasting and postprandial,the percentage of bradygastria in severe group and mild group were lower than those in control group,there were significant differences(P < 0.05).The level of VIP in severe group was lower than that in mild group and control group [(36.07 ± 9.22) μ g/L vs.(44.16 ± 7.88),(46.53 ± 4.54) μ g/L],the level of I-FABP was higher than that in mild group and control group [(35.80 ± 11.69) μ g/L vs.(27.40 ±9.53),(23.58 ±8.18) μg/L],there was significant difference (P <0.05),but there was no significant difference between mild group and control group (P > 0.05).Conclusion The patients with COPD are accompanied with serious gastrointestinal symptoms and gastric dysrhythmia,severe COPD can be associated with VIP decrease and Ⅰ-FABP rise in peripheral blood.
