1.Effect of 8-nitrochyrsin on the proliferation and apoptosis of HL-60 cells

Objective To investigate the effect of 8-Nitrochyrsin on cell proliferation and cell apoptosis of HL-60 cells in vitro.Methods 8-Nitrochyrsin on proliferation of HL-60 cells was detected by MTT assay;The trypan blue exclusion method was used to count the number of viable and dead cells,then the survival rate and growth curve were achieved;DNA ladder bands were observed by DNA agarose gel electrophoresis;HL-60 cell apoptotic percentage was detected by flow cytometry with PI staining.Results 8-Nitrochrysin inhibited proliferation of HL-60 cells in a concentration-dependent manner;8-Nitrochyrsin possess the inhibitory effect on the proliferation and growth of HL-60 cells;8-Nitrochrysin possess the significantly effect of inducing apoptosis of HL-60 cells.Conclusion 8-Nitrochyrsin possess the inhibitory effect of the proliferation and growth of HL-60 cells and significantly induces apoptosis of HL-60 cells.

Objective To investigate the dose distribution and radiation risk of Varian thoracic cone beam computed tomography (CBCT) with default parameters with reference to Monte Carlo simulation and International Commission on Radiological Protection (ICRP) report 110.Methods EGSnrc/BEAMnrc code was used to simulate the material, thickness, and geometry of the kV CBCT source (kVS) to establish the kVS model.A benchmarked MCSIM code was applied to calculate the dose distribution in the ICRP phantom after the scan with the standard thoracic parameters (110 kV, 20 mA, and 262 mAs), and the conversion coefficient of absolute dose was obtained in a spherical phantom following the TG-61 protocol.The results of Monte Carlo simulation were validated by PDD and Profile in a water phantom and the measurement of the absolute dose in the computed tomography dose index (CTDI) phantom and Alderson phantom.The models including BEIR VⅡ were used to evaluate the radiation risks.Results With reference to the criterion of 3%/1 mm, the uncertainties of PDD and Profile were less than 2%.The difference between the measured and calculated values was<2.9% in the CTDI phantom and ≤0.05 cGy in the Alderson phantom.In the ICRP110 phantom, the doses to the left lung, right lung, left breast, right breast, heart, thyroid, trachea, cancellous bone, and cortical bone were 1.28, 1.39, 1.74, 1.80, 1.46, 0.48, 0.88, 0.85, and 1.84 cGy, respectively.The relative risks of ischemic heart disease, breast cancer, lung cancer, thyroid cancer, and tracheal cancer in a standard scan were1.001 , 1.009, 1.019, 1.000, and 1.008, respectively. Conclusions The accumulated dose and long-term risks of CBCT during image-guided thoracic radiotherapy cannot be neglected and should be effectively controlled.
4.Analysis of ecological environment factors of Himalayan marmot based on global positioning system, remote sensing and geographical information system in Ynshu City, Qinghai Province

Objective To analyze the ecological environmental differences of hibernation hole and temporary hole of Himalayan marmot based on global positioning system (GPS),remote sensing (RS) and geographical information system (GIS) in Yushu City,Qinghai Province.Methods Hibernation holes and temporary holes were located based on GPS.Ecological environment factors related with Himalayan marmot,such as land surface temperature (LST),digital elevation model (DEM),normalized different vegetation index (NDVI),were extracted based on RS and GIS.The ecological environment differences between hibernation hole and temporary hole were explored based on epidemiological statistical method.Results Monthly LST (degree Kelvin,K) of hibernation hole was slightly higher than that of temporary hole in the same period.Yearly day LST of hibernation hole was 1.1 K higher than that of temporary hole in the same period.Yearly night LST of hibernation hole was 0.3 K higher than that of temporary hole in the same period.Monthly NDVI of temporary hole was slightly higher than that of hibernation hole in the same period.Yearly NDVI of temporary hole was 0.001 higher than that of hibernation hole in the same period.DEM of temporary hole was about 40 m higher than that of hibernation hole.The top two ratios of soil type factors were both alpine meadow soil and dark felty soil for hibernation hole and temporary hole,which were 62.1% and 29.3%,64.4% and 25.6%.The top two ratios of land cover factors were both rainfed cropland and mosaic cropland for hibernation hole and temporary hole,which were 43.3% and 25.8%,40.0% and 26.2%.Conclusions Soil type factors and land cover factors for hibernation hole and temporary hole are similar.Lower DEM and higher LST are chose for hibernation hole,and better NDVI is chose for temporary hole,which is good for food intake.
5.Protective Effect of Sulfated Gastrodia Elata Polysaccharides on the Apoptosis of PC12 Cells Induced by Corticosterone

Objective: To investigate the protective effect of sulfated Gastrodia elata polysaccharides (GEPS) on the apoptosis of PC12 cells induced by corticosterone (CORT).Methods: After modifying GEP chemically, GEPS was obtained.PC12 cells were pretreated with GEPS (0, 250, 500 and 1 000 μg·ml-1) for 30 min before cultivating with CORT for 48 h.The cellular viability, lactate dehydrogenase (LDH) release and morphological observation were determined respectively by CCK-8 assay, LDH assay, inverted microscope and DAPI fluorescein stain method.Results: With the pretreatment of GEPS, the survival rate of PC12 cells increased significantly (P<0.05) in a dose-dependent manner.Compared with CORT injured group, GEPS attenuated the release of LDH with statistical significance (P<0.05), and LDH leakage decreased with the concentration of increase GEPS.Under an inverted microscope, PC12 cells incubated with CORT became shrinkage, and aggregated with decreased diopter and bad adhesion,and lots of cells floated.However, with the pretreatment of GEPS, the living status of PC12 cells improved markedly.Compared with CORT injured group, GEPS obviously reduced the apoptosis of PC12 cells.Conclusion: GEPS exhibits protective effects on the apoptosis of PC12 cells induced by CORT, while when compared with GEP, GEPS shows the similar effects.The other bioactivities of GEPS need further studies.
6.Quick cognitive screening scale for the elder:development, reliability and validity

Objective To develop a Quick Cognitive Screening Scale for Elder(QCSS-E) and examine its reliability and validity.Methods In the light of rating model of MMSE and MoCA,a screening scale was developed to assess cognitive function,which contained 47 items across twelve domains.Five hundred community subjects were interviewed with the QCSS-E,then 150 subjects were retested after two weeks,and 75 subjects were assessed with MMSE and CCAS.The test-retest reliability,internal consistency reliability,empirical validity and constmct validity of QCSS-E were calculated.Results The test-retest reliability and Cronbach's α coefficient were 0.972 and 0.814 for QCSS-E total score,0.656-0.911 and 0.679-0.746 for domain scores.There were low to moderate correlation of 0.133-0.612 among domain scores,correlation domain scores with total score ranged from 0.303-0.664.QCSS-E scores decreased with increasing agc,increased with the level of education increasing,and decreased with the aggravation of brain damage.The QCSS-E was significantly correlated with MMSE(r=0.886) and CCAS (r=0.899).Factor analysis got two factors accounting for 49.4% of variance,which were named vulnerability factor and resistance to damage factor.Conclusion The stability,internal consistency,and validity of the QCSS-E are good and meet with psychometric standard.
7.Folliculotropic mycosis fungoides complicated by eosinophilia: a case report

A 69-year-old man presented with a 3-year history of scattered erythematous patches, perifollicular papules, acneiform lesions(such as milia, cysts) on the head, trunk and limbs as well as alopecia and peripheral eosinophilia. Histopathological examination revealed chronic focal dermal and perifollicular inflammatory infiltration with vascular proliferation and presence of a small number of eosinophils. He was initially diagnosed with folliculitis, and treated with antihistamines and antibiotics. Thereafter, lesional inflammation and pruritus were attenuated. However, plaques and alopecia developed in the occipital area 3 months later. The second histopathology of biopsy specimens revealed a dense dermal infiltrate of lymphoid cells and eosinophils, perifollicular infiltrate with numerous lymphoid cells, eosinophils and atypical lymphocytes migrating into hair follicles. Alcian blue stain showed epidermal mucinosis in follicles. Immunohistochemical examination showed positive staining of atypical cells for CD3, CD4, CD5, CD2, CD43 and ubiquitin carboxyl-terminal esterase L1 (UCHL1), but negative staining for CD20, CD79a, Epstein-barr virus, CD56, phosphoglucomutase-1, myeloperoxidase, CD7, or AE1 and AE3 monoclonal anti-keratin antibodies. T-cell receptor gene rearrangement was undetected. He was diagnosed with folliculotropic mycosis fungoides. Novel skin lesions still emerged after treatment with photochemotherapy (PUVA) plus acitretin. Follow up of the patient still continued.
9.Expression and location of YB-1 protein in hepatocellular carcinoma and their clinical significance

ObjectiveTo study the expression levels of YB-1 protein in hepatocellular carcinoma (HCC) tissues,tissues adjacent to tumour and normal liver tissues,and to explore the clinical significance.MethodsTo study the expression levels of YB-1 protein,immunochemistry was carried out on 58 HCC tissues and their corresponding adjacent tissues to tumour and on 20 normal liver tissues.The YB-1 protein with nuclear translocation was detected by Western blot in HCC tissues,tissues adjacent to tumour,normal liver tissues,HCC cell lines including QGY-7701 and SMMC-7721,and a normal hepatic cell line LO2.ResultsPositive signals of YB-1 protein were detected at a high level in HCC tissues (72.4%,42/58) when compared to tissues adjacent to tumour (41.4%,24/58) and to normal liver tissues (35 %,7/20) (P>0.05).The nuclear expression of YB-1 (31%,18/58) was significantly correlated with the pathologicalgrade,tumour size and portal venous invasion (P<0.05).Using Western blot,the YB-1 protein with nuclear translocation was expressed at a higher level in HCC tissues (0.474±0.107) than in tissues adjacent to tumour and in normal liver tissues (P<0.05).Similar results were obtained in HCC cell lines and the normal hepatic cell line (P<0.05).ConclusionThe high expression of YB-1 in HCC and its nuclear translocation may be involved in human HCC progression.YB-1 may be a potential target for HCC treatment in the future.
10.Expression of Spa-1 and Ki-67 in tissues of extramammary Paget's disease (EMPD)

Objective To investigate the potential role of Spa-1 in the development and metastasis of EMPD.Methods Tissue specimens were resected from 17 patients with primary EMPD,12 patients with invasive EMPD and 9 normal human controls.Immunohistochemistry was performed to measure the expression of spa-1 and Ki-67.Results Positive staining was observed for Spa-1 in cytoplasm,and for Ki-67 in cell nuclei.Spa-1 and Ki-67 were weakly expressed in the basal layer of normal skin.The expression of Spa-1 and Ki-67 in EMPD tissue were statistically higher than those in the normal control tissue (both P<0.01).Increased expression of Spa-1 was noted in invasive EMPD tissue compared with in situ EMPD tissue.The expression of Spa-1 was positively correlated with that of Ki-67 in the tissue of EMPD.Conclusion Spa-1 may play a certain role in the initiation and progression of EMPD.
