1.Simultaneous Determination of Vinyl Chloride and Vinylidene Chloride in Drug Packaging Materials Con-taining Polyvinyl Chloride/Polyvinylidene Chloride by HS-GC

Objective:To develop a method for the simultaneous determination of vinyl chloride and vinylidenechloride in drug packaging materials containing polyvinyl chloride /polyvinylidene chloride by HS-GC.Methods: A Stabilwax (30.0 m ×0.25 mm, 0.25 μm) capillary column and an FID detector were adopted .The headspace equivalent temperature was 80℃for 30 min and the col-umn temperature was 40℃.The inlet temperature was 190℃and the detector temperature was 210℃.N2 was used as the carrier gas . The flow rate was 1.0 ml · min-1 .Results: Vinyl chloride had a good linear relationship within the range of 0.5-2.5 μg ( r =0.996 7), and the average recovery was 90.4%(RSD=0.9%, n=9).Vinylidenechloride had a good linear relationship within the range of 1.0-5.0 μg (r=0.999 0), and the average recovery was 90.0%(RSD=0.6%, n=9).Conclusion: The method is fast and accurate in the simultaneous determination of vinyl chloride and vinylidenechloride in drug packaging materials containing polyvinyl chloride/polyvinylidene chloride .
2.Studies on the distribution of interstitial Cajal-like cells in ampulla region and the myoelectric activities of the sphincter of Oddi of guinea-pigs

Objective To investigate the distribution of interstitial Cajal-like cells(ICLC) in ampulla and the myoelectric activities of the sphincter of Oddi(SO) of adult guinea-pigs.Methods C-Kit immunohistochemical staining was carried out on whole-mount preparations of guinea-pigs' ampulla region.The myoelectric activities of SO were recorded by using the contact electrode on mucosa.Result C-Kit-positive ICLC were observed in both exterior and inner wall of ampulla.Some of these cells were distributed in the circular and longitudinal smooth muscle cell(SMC) layers assuming a spindle-like shape.Another sub-group of ICLC,which were located between the longitudinal and circular SMC layers,were shown to have oval or triangle cell bodies with projection of more than three long processes.These cells interlaced into a cellular network,closely resembling the interstitial cells of Cajal(ICCs),around the myenteric nervous plexus(ICC-MY) in the duodenum.At the inner side of circular SMC layer of the exterior wall,ICLC were similar to ICCs associated with the deep muscular plexus(ICC-DMP) of small intestine.In the inner wall of the ampulla,a large amount of ICLC were found in the SMC layers or bundles,running parallel with them.At the opening of the ampulla,a number of ICLC surrounded the opening to form a ring.The spontaneous rhythmic myoelectric activities could be recorded in the SO.Conclusion It is suggested that the ICLC might contribute to the regulation of the spontaneous rhythmic contraction,and it might be related to development of motility disorders of the SO.
3.Villi Chromosome High-throughput Gene Sequencing of 52 Cases of Missed Abortion

Objective To understand the relationship between embryonic chromosomal abnormalities and missed abortion by high-throughput gene sequencing, so as to provide a basis for guiding the next pregnancy.Meth-ods By chromosome detection on villi chromosome of 52 cases of missed abortion by high-throughput gene sequen-cing technique, the types of chromosome abnormalities and the proportion were investigated.Results Among villi samples from the 52 cases of missed abortion, chromosomal abnormalities were founded in 28 cases, including 24 ca-ses of number abnormality (17 cases of trisomy, 6 cases of triploid and a case of monomer) and 4 cases of structural abnormalities.Conclusion Embryonic chromosomal abnormality is the main cause of missed abortion in early preg-nancy.High-throughput gene sequencing technology used to detect the villi chromosome is comprehensive, fast and accurate, helping to clarify the cause of missed abortion and guide the next pregnancy reasonably.
4.Cloning of ribosomal protein L27 cDNA from Jilin Shuangyang sika deer

Object To develop the gene resources of Jilin Shuangyang sika deer,a rare animal in China and reveal the pharmacological functions of deer medicinal material on the molecular level. Methods A cDNA plasmid library of spleen cells derived from Shuangyang sika deer was set up. Some housekeeping genes were cloned and analyzed. Results One of the cDNA sequences and it's deduced amino acid sequence in deer share high homology with a group of cDNA sequences encoding ribosomal protein L27 in human,rat,Canis and mouse and their corresponding amino acid sequence respectively. It has the completed open reading frame as well. Conclusion A novel full-length cDNA encoding Shuangyang sika deer ribosomal protein L27 is discovered. This sequence has been deposited in Genbank under accession number AF373231.
5.Liquid chromatography-mass spectrometry for measurement of serum 25 ( OH ) D level

Many diseases are associated with vitamin D deficiency.Serum 25 (OH)D levels can serve as objective indicators of the nutritional status of vitamin D.Liquid chromatography-mass spectrometry (LC-MS) method may be applied for routine measurement of 25 ( OH ) D levels in clinical practice.This article reviewed the significance of detecting 25 (OH) D2 and 25 (OH) D3,compared the advantages and disadvantages of different 25 (OH) D measurement methods,and introduced the application of LC-MS method for routine detection of serum 25 (OH) D2 and 25 (OH) D3 levels following the standard by National Institute of Standards and Technology of America.

Objective To study the influence of unilateral and bilateral amplification on the effect of hearing aid evaluation .Methods Using the subjective method that International Outcome Inventory for Hearing Aids (IOI-HA) and objective method that medium acoustic intensity (65 dB SPL) word recognition score(WRS) to evaluate the effect of unilateral and bilateral hearing aid fitting of middle -aged severe sensorneural hearing loss .Results Hearing aid were used for severe sensorneural hearing loss and the improvement of monosyllables and sentences in quiet and noise test of unilateral were 35 .73% ,43 .15% ,43 .23% ;the improvement of monosyllables and sentences in quiet and noise test of bilateral were 37 .90% ,51 .33% ,54 .86% .The WRS of bilateral was higher than unilater-al .The score of IOI-HA was 15~37 ,meaning patients with severe sensorneural hearing loss were satisfied with hearing aid ,and there was no statistical significance between unilateral and bilateral fitting .Conclusion The bilat-eral hearing aid fitting was better than unilateral .Binaural hearing loss are recommended to fit bilaleral hearing aids .
7.Clinical significance of microrna-625 expression in esophageal squamous cell carcino-ma

Objective: To analyze the correlation of miR-625 expression with clinicopathological characteristics in esophageal squa-mous cell carcinoma (ESCC) and to explore the effect of miR-625 on the migration and proliferation of ESCC cells. Methods:The expres-sion level of miR-625 was determined through real-time PCR in 86 paired human ESCC tissue specimens and tumor-adjacent normal esophageal tissue specimens, ESCC cell lines, and esophageal epithelial cell line. The associations of miR-625 expression with clinico-pathological characteristics and survival in ESCC patients were analyzed. Transwell and CCK-8 assays were performed to examine the effect of miR-625 expression on migration and proliferation of ESCC cells. Results:Compared with tumor-adjacent normal specimens, miR-625 was significantly downregulated in ESCC tissue specimens (P<0.05). MiR-625 expression was decreased in ESCC cell lines com-pared with human esophageal epithelial cell lines (P<0.05). Lower miR-625 expression was associated with poorer prognosis and sur-vival. The migration and proliferation abilities of ESCC cells were inhibited by miR-625 overexpression (P<0.05). Conclusion:MiR-625 acts as a tumor suppressor gene in the development and progression of ESCC, suggesting that miR-625 may serve as an efficient prog-nosis biomarker and a potential therapeutic target for ESCC.
8.Role of tyrosine kinase receptor RON in tumor growth and metastasis:research progress

Recepteur d′origine Nantais(RON),a tyrosine kinase receptor ,is a growth factor receptor belonging to the proto-oncogene met family and has been proved to display abnormal expres?sion in many types of tumors. The RON receptor is activated by binding to the ligand macrophage stim?ulating protein,overexpression of the receptor,variants of the proto-oncogene and by point mutations of the kinase region. The downstream transduction of RON by mitogen-activated protein kinases and phosphoinositide3-kinase signaling pathways can help regulate the proliferation,apoptosis,migration and invasion of tumor cells. A better understanding of the mechanisms and related signaling pathways of RON activation in tumor progress and development will provide more information for the RON-based target therapy.
9.Freiberg's disease treated with dorsal closed Ⅴ-shaped osteotomy and Charlotte breakage screw fixation

Objective To evaluate the clinical outcomes of treating Freiberg's disease with dorsal wedge osteotomy and Charlotte breakage screw fixation.Methods From June 2010 to June 2014,11 patients with Freiberg's disease were treated at our department.They were 5 males and 6 females at an average age of 32 years(range,from 15 to 52 years).X-ray revealed osteosclerosis and collapse of the metatarsal head.According to Smillie classification system,there were 5 cases of stage Ⅱ and 6 ones of stage Ⅲ.The duration of symptoms was from 7 to 48 months (average,23 months).The metatarsal heads were rotated to reconstruct the joint surface.After dorsal closed V-shaped osteotomy,Charlotte breakage screws were implanted.Results All wounds healed with no complications in the early postoperative period.The 11 patients were followed up for 14 to 48 months (average,19 months),showing obvious pain relief.X-ray showed that the osteotomy sites got solid union in all patients at an average time of 10 weeks (range,from 8 to 13 weeks) after operation.Average shortening of the metatarsal was 1.5 mm (range,from 1.1 to 1.8 mm).The American Orthopaedic Foot and Ankle Society(AOFAS) score,Lesser Metatarsophalangeal-Interphalangeal Scale(LMIS) score,and range of motion at one day,one month and one year after operation were significantly improved compared with the preoperative ones (P < 0.05).Significant differences were also observed between all postoperative time points (P < 0.05).No such complications happened as referred pain from the adjacent metatarsophalangeal joint,displacement,nonunion,necrosis or infection.All patients returned to sports and recreational activities at 6 months after operation,except one case of stage Ⅲ who had constant swelling in the metatarsophalangeal joint but eventually recovered at 10 months after operation.According to patients' satisfaction at the last follow-ups,including 7 cases were excellent,3 good and one poor.Conclusion Dorsal closed V-shaped osteotomy with Charlotte breakage screw fixation is an effective procedure for Freiberg' s disease of stages Ⅱ and Ⅲ,because it leads to effective reconstruction of the metatarsophalangeal joint,allows early joint motion and avoids a second operation.
10.Effect of insulin on small intestinal smooth muscle in diabetic mice

Objective To investigate the effects of regular insulin (RI)on duodenal smooth muscle in diabetic mice. Methods Diabetes mellitus (DM) model was established by intraperitoneal injection of 150 mg/kg streptozotocin (STZ) in male BALB/c mice. The model mice were divided into DM group and DM treated with RI group with 6 each. Meanwhile, 6 normal mice were served as controls. The mice in treatment group were intraperitoneally injected with 40 U/kg of RI daily.Whereas the mice in DM and control groups were intraperitoneally injected with phosphate buffer solution (pH = 7. 40). After 6 weeks, the small intestinal transit rate of mice was determined by lavage of Indian ink. Interstitial cells of cajal (ICC) in duodenal myenteric plexus were counted using immunohistochemical staining. Slow waves of duodenal smooth muscle cells were recorded with intracellular recordings. Data were analysed by SPSS 17.0 software, and comparisons among three groups were done using LSD test. Results After intervention for 6 months, the clinical presentations,such as more water and food intake and polyuria, were improved in treatment group. The body weight was increased in treatment group [(23.33±3.13) g] compared with DM group [(15.42±1.40) g,P<0.01] ,but dereased compared with control group [(26.78 ± 2.09) g, P<0.05]. The level of blood glucose in DM group was significantly higher than that in control and treatment groups(P<0.01). Small intestine transmission rate was significantly reduced in DM group than that in control and treatment groups (P<0.01), but it was slower in treatment group than that in control group (P< 0. 01 ). Immunohistochemical study showed that the number of c-kit positive cells reduced obviously in DM group than that in control group and treatment group (P<0.05), whereas it was lower in treatment group than that in control group (P < 0.05). The slow wave frequency and amplitude of duodenal smooth muscle cells in DM group were reduced when compared with control and treatment groups (P<0.01) and both were lower in treatment group than that in control group (P<0. 01 ). Conclusion The findings indicate that DM mice have gastrointestinal dysmotility and exogenous insulin may improve small intestinal dysmotility in DM mice.
