Wilson’s disease is an autosomal recessive disorder of copper metabolism. The resultant accumulation
of copper primarily damages the liver and brain, resulting in hepatic, neurological and psychiatric
symptoms. There have been many recent studies advancing the understanding of Wilson’s disease in
Asia. There are indications that the incidence of Wilson’s disease in parts of Asia may be relatively
high. Many genetic studies have identifi ed various hot spots in theATP7B gene in a variety of the Asian
populations. Screening of these hotspot mutations may thus be useful in confi rming the diagnosis.
Despite the advances in treatment, lack of familiarity by the health care profession resulting in late
diagnosis, and poor access to treatment particularly among those from the developing economies
remain areas of major concern.
3.Efficacy of phacoemulsification auxiliary hookinterred the anterior chamber treated failure of glaucoma combined with cataract surgery

AIM: To explorethe therapeutic efficacy of treating cataract phacoemulsification remove combined with trabeculectomy early postoperative failure with open sclera flap recanalization by phacoemulsification auxiliary hook interred the anterior chamber.
METHODS:Twenty-seven cases ( 27 eyes ) of glaucoma combined cataract surgery(3mo) in the early failure were treated with that the phacoemulsification auxiliary hook into anterior chamber under the original sclera flap, open from the inside have adhesion of the sclera flap. Postoperative follow-up 12mo, the intraocular pressure and best corrected visual acuity were observed.
RESULTS: The postoperative follow- up was 12mo. There were functional filtering bleb in 14(52%) eyes; and the IOP was below 21mmHg in 17 eyes(63%) without any anti- glaucoma drug, and in 7 eyes ( 26%) with 1 - 2 categories of lower intraocular pressure( IOP) drugs, and 3 eyes(11%), failure and the success rate was 89% after 12mo. Average intraocular pressure after 1wk, 1mo, 3mo, 6mo and 12mo were(7. 1±4. 3) mmHg,(10. 5±5. 1) mmHg,(15. 1±4. 8 ) mmHg, ( 16. 8±5. 2 ) mmHg, ( 17. 3±5.1 ) mmHg, significantly lower than the IOP preoperatively(30. 2±6. 8) mmHg (P<0. 01). Average visual acuity ( all of the following were best corrected vision) after 1wk, 1mo, 3mo, 6mo and 12mo was(0. 32±0. 52),(0. 52±0. 42),(0. 55±0. 39),(0. 53±0. 47),(0. 54±0. 42 ). Postoperative 1wk visual acuity deeper than preoperative( 0. 46 ± 0. 44 ) ( P < 0. 05 ), more than all improved compared with preoperative ( P < 0. 05 ). Postoperative anterior chamber bleeding in 5(19%) eyes, all absorbed itself in 3 to 5 days; ciliochoroidal detachment in 7 ( 26%) eyes, except one case need operate, others were conservative treatment to heal. Postoperative continuous low intraocular pressure concurrent macular edema was 1 eyes(4%), 4mo healed after treatment.
CONCLUSION: It is a safe and effective method thattreating phacoemulsification cataract remove combined with trabeculectomy early postoperative failure with open sclera flap recanalization by phacoemulsification auxiliary hook interred the anterior chamber.
4.Synthesis of seven sinapine analogs and their effects on the tension of smooth muscle of intestines isolated from rabbit.

Seven sinapine analogs (6a-6g) were synthesized using cinnamon acid or benzoic acid and their derivatives as starting materials, which obtained from substituted benzaldehyde and malonate. The structures of target compounds were characterized by IR, 1H NMR and elemental analysis. The effects of compounds 6a-6g on the smooth muscle of intestine isolated from rabbit were studied, and the experimental results showed that compounds 6a, 6d and 6g had diastolic action, while 6f had contractile action.
5.Preoperative imaging evaluation of hilar cholangiocarcinoma

Recently, the incidence of hilar cholangiocarcinoma (HC) has gone up. Preoperative imaging evaluation plays an important role in guiding clinician to make reasonable operating program. MR cholangiopancreatography (MRCP) can make accurate preoperative classification of HC, and,combined with conventional MR scans, is able to perform an all-in-one examination, making accurate overall preoperative evaluation of HC.

7.Effect of CsA on mitochondria stress after microperfusing glutamate and calcium into tree shrews' hippocampus

AIM: To observe the changes of glutamate and calcium within the hippocampal microenvironment in mitochondrial stress.METHODS: A lateral hippocampus was microperfused with glutamate and calcium chloride solution by a kind of single-pumped push-pull perfusion system in Tree Shrews.At 24 h,the expression of cytochrome C(Cyt C)was observed by immunochemistry.Also,the hippocampus was removed,then mitochondria and cytoplasmic fragment were divided by low temperature centrifugation and the distribution of cytochrome C was assessed through Western blotting.The relative amounts of caspase-3 and caspase-9 mRNA were evaluated by real time fluorescence polymerase chain reaction.In the treated group,cyclosporin A(CsA,40 mg/kg) was intravascularly injected at 6 h after perfusion of glutamate-calcium chloride solutions into the hippocampus and inspected the above-mentioned items at 24 h.RESULTS: In the glutamate-calcium group,compared with the control group,cytochrome C immunoreactivity increased and the content of hippocampal mitochondrial cytochrome C decreased.Also,the cytochrome C was detected in cytosol.Cyclosporin A treatment at 6 h after microperfusion,the cytochrome C expression weakened and no Cyt C in cytosol fraction was observed.By real time PCR,in relation to the control group,the caspase-3 and caspase-9 mRNA was higher in the glutamate-calcium group.Cyclosporin A treatment cut down both caspase-3 and caspase-9 mRNA contents.CONCLUSION: The accumulation of glutamate and calcium may promote Cyt C release,caspase cascade activation and the mitochondrial stress.The neuroprotection of CsA may results from uniquely inhibiting the mitochondrial permeability transition pore,and preventing Cyt C release and caspase activation.
8.The opening of neuronal mitochondria permeability transition pore affects respiratory function during thrombotic cerebral ischemia in tree shrews

AIM: The present study explored those changes in brain mitochondrial respiration in different times after thrombotic cerebral ischemia induced by photochemical reaction in primate's animal tree shrew and o bserved the effects of platelet-activating factor (PAF) receptor antagonist gin kgolide B (GB) and immunosuppressor cyclosporin A (CsA) on neuronal mitochondria l respiration and mitochondrial permeability transition pore(MPT) in twenty-four hours after occlusion and showed their neuronprotective mechanism and expatiate the relationship between mitochondrial respiration and MPT. METHODS: The focal thrombotic cerebral ischemia was formed by photochemistry-induce d technology in tree shrews. At 4,24,72 h after focal cerebral ischemia, the n euronal mitochondria were centrifuged. Clark oxygen electrode was used to measur e the changes in neuronal mitochondrial respiration.Pretreatment for experiment al animals with GB and CsA at 6h after occlusion, we centrifuged the mitochondri a and measured the changes in neuronal mitochondrial respiration at 24 h after o cclusion. In addition, experiments were performed in a flurometer by measuring CaCl 2 100 ?mol/L induced centrifuged mitochondrial swelling and GB or CsA were added at the same time. 9.The morphologic changes of dorsal root ganglion after lumbar nerve root being under chronic compression and inflammatory stimulation in rabbits

Objective To observe the morphologic changes of dorsal root ganglion in the lumbar region of rabbits after the nerve root was under chronic compression and inflammatory stimulation. Methods Twenty New Zealand rabbits were recruited for this study, of which 5 served as the control (control group), and the rest were randomized into 3 experimental subgroups: 10d group, 30d group, 90d group, respectively. The autologous nucleus pulposus from the tails (about 5mg) was put into the silastic tube (inner meter of 1.5mm, external diameter 2.5mm and length 12mm), which was inserted into the left L 7 intervertebral foramen to compress the lumbar nerve root. Sham operation was performed with the rabbits in the control group. The nerve root and the dorsal root ganglia were harvested and processed and observed with light microscope and electron microscope after 10d, 30d, 90d, respectively. Results In the 10d group, obvious hyperemia, edema and infiltration of inflammatory cells in the interspace of the intima of the dorsal root ganglia (DRG) could be observed. Pyknosis, degeneration and necrosis were also found in some of the nerve cells. Electron microscopic observation showed that the number of the rough endoplasmic reticulum and mitochondrion decreased, ribosome exfoliated, mitochondrion swelled. In 30d group, typical degeneration and necrosis became more obvious. Electron microscope showed that the number of lysosome and smooth endoplasmic reticulum increased, mitochondrion swelled and its cristae disappeared, nuclei concentrated and deviated. In 90d group, significant proliferation of fibrocyte could be observed. At the same time, dura mater and arachnoid of spinal cord around the nerve root were notably thickened, and became fibrogenesis. Electron microscope also showed the increment of the lysosome and smooth endoplasmic reticulum, the swelling of mitochondrion, the loss of its cristae and the concentration of the nucleolus in the central part of the nuclei. No significant changes were found in the control group. Conclusion Pathological changes of neural degeneration such as edema, inflammatory infiltration could be observed in dorsal root ganglion after the nerve root was under chronic compression and stimulation by autologous nucleus pulposus.
10.Effects of activin A and follistatin on type Ⅰ collagen of cultured rat renal interstitial fibroblasts

Objective To investigate the stimulating effects of activin A(ACT-A) and follistatin(FS) on the secretion of type I collagen(Col I) and the expression of Col I mRNA of cultured rat renal interstitial fibroblasts in vitro.Methods Rat renal interstitial fibroblasts isolated from normal SD rat renal medulla were cultured in vitro.The cells were divided into three groups:different concentrations of ACT-A(group A),different concentrations of FS(group F),and different concentrations of FS plus 30ng/ml ACT-A(group A+F).The expression of Col I mRNA in cultured rat renal interstitial fibroblasts was detected by RT-PCR,and the expression of Col I protein in cultured rat renal interstitial fibroblasts was examined by immunocytochemistry.Results Renal interstitial fibroblasts were successfully cultured in vitro.In present study,the expression of Col I mRNA increased significantly in cultured rat renal interstitial fibroblasts after stimulated by ACT-A in dose-dependent manner(P0.05).FS could inhibit the effects of ACT-A(30ng/ml) on Col I mRNA and Col I protein of the cultured renal interstitial fibroblasts in dose-dependent manner in A+F group(P
