1.Effects of cylin-dependent kinase 5 expression inhibition on apoptosis of mouse podocyte

Objective To investigate the expression of Cdk5 in high glucose-induced mouse podocytes and the effects of inhibi-ting Cdk5 expression on podocyte apoptosis .Methods (1)Mouse podocytes cultured in vitro were divided into normal glucose group ,mannitol group ,high glucose group and the high glucose group was further divided in to 0 ,6 ,12 ,24 ,48 h gorup depend on the induce time .Changes of Cdk5 expression in each group were detected by Western blotting .(2)The expression of Cdk5 in podo-cyte was inhibited by Cdk5 miRNA plasmid .The podocytes were divided into 4 groups :NG group(normal glucose) ,HG group(high glucose) ,HG + S group(high glucose + scrambled plasmid) and HG + C group(high glucose + Cdk5 miRNA plasmid) .The podo-cyte apoptosis rates were detected by flow cytometry and terminal deoxynucleotidyl transferase dUTP nick end labeling (TUNEL) , and the expressions of Bcl-2 ,Bax and cleaved caspased-3 were detected by Western blotting respectively after 48 h .Results Com-pared with normal glucose group ,the expression of Cdk5 was significantly increased in podocytes of high glucose group (P< 0 .05) . The expression of Cdk5 in podocytes was inhibited by Cdk5 miRNA plasmid .Compared with HG and HG + S group ,the podocyte apoptosis rate of HG + C group was significantly decreased(P< 0 .05) .The expression of cleaved caspase-3 protein and Bax /Bcl-2 ratio were also decreased (P< 0 .05) .Conclusion High glucose could increased the expression of Cdk5 in podocytes .The downregu-lation of Cdk5 expression could decrease the podocyte apoptosis induced by high glucose .
2.Influence of Electroacupuncture at Acupoints Zusanli(ST 36)and Taichong(LR3) with Different Frequencies on Rats Swimming Endurance

Objective: To observe the influence of electroacupuncture with different frequencies on rats swimming endurance and sift out the optimal frequency parameter.Methods: Wistar rats were randomly allocated to control group, non-acupoint group and electroacupuncture with low frequency group (EA with 2 Hz group), electroacupuncture with medium frequency group (EA with 15 Hz group), and electroacupuncture with high frequency group (EA with 100 Hz group), acupoints Zusanli (ST 36) and Taichong (LR 3) were punctured and rats load swimming time was used as an index, and the change of swimming endurance time before and after acupuncture was observed. Results: After electro-acupuncture, swimming time was increased in 15 Hz and 100 Hz groups respectively, with the significant differences between before and after acupuncture (P<0.05). There was also a significant difference in the percentage of increase in swimming time between 15 Hz group and other groups (P<0.05).Conclusion: It is suggested that electroacupuncture at acupoints Zusanli (ST 36) and Taichong (LR 3) with a frequency of 15 Hz could enhance rats's swimming endurance, and 15 Hz might be an optimal frequency parameter during the electrical stimulation.
3.Effect of Robot-Assisted Training on Lower Extremities of Patients with Stroke: A Systematic Review of Randomized Controlled Trials

Objective To evaluate the effectiveness of robot-assisted training on lower extremities of patients with stroke. Methods PubMed, Medline, EMbase, Cochrane Central Register of Controlled Trials (CENTRAL), ISI Web of Knowledge, Ovid, China National Knowledge Infrastructure (CNKI), Wanfang Data and VIP Database for Chinese Technical Periodicals were searched for the randomized controlled trials (RCTs) about robot-assisted training for lower extremities of patients with stroke from the date of establishment to March 2013. The bibliographies of the retrieved studies were also searched. 2 independent researchers evaluated the included studies using the risk of bias provided by Cochrane Library. The extracted data were analyzed by RevMan 5.1 software. Results A total of 12 trials were discovered.Compared with conventional training, the robot-assisted training increased the score of Fugl-Meyer assessment (lower extremity)(WED=3.97, 95%CI: 2.84~5.10, Z=6.89, P<0.00001) and Berg Balance Scale (WED=3.13, 95%CI: 0.80~5.45, Z=2.63, P=0.008). However,the results of stride frequency (WED=8.40, 95%CI: -0.95~17.75, Z=1.76, P=0.08) and speed (WED=0.03, 95%CI: -0.02~0.07, Z=1.18, P=0.24) were not statistically significant. According to the Jadad Score of included studies, 4 of them were of low quality and 8 of them were of high quality. Conclusion The current evidence shows that to some extent, robot-assisted training could improve the lower extremities function.

5.Progress of Clinical Study on Acupuncture Treatment of Chemotherapy-induced Peripheral Neuropathy in the Recent Decade

Literatures on acupuncture-moxibustion treatment of chemotherapy-induced peripheral neuropathy (CIPN) in the recent decade were searched in the databases of Pubmed, MEDLINE, Biological Abstracts, EMBASE, Sinomed, CNKI, Wanfang, VIP, et al. The general situation was comprehensively analyzed and reviewed from experimental design, treatment method, efficacy evaluation, mechanism research and so on. The results showed that acupuncture has a certain therapeutic effect, but the research is still in the preliminary stage, the relative literatures are insufficient and in a low quality. There is still a controversy on the efficacy of acupuncture in the treatment of CIPN. High quality, large sample and multicenter randomized controlled trials and systematic reviews are needed to verify the efficacy of acupuncture. The mechanism of acupuncture treatment of CIPN is still unclear and needs further research and exploration.
6.Research Progress of the Mechanism of Acupuncture Analgesia Based on Electroacupuncture Frequency

By organizing literature, summarizing and analyzing the relevant experimental and clinical studies, this article analyzed the study status of the analgesic action of electroacupuncture with different frequencies from the chemical mechanism, neuro- physical mechanism, molecular mechanism, and action mechanism. It brought up that electroacupuncture of different frequencies can activate different regions in the central system and generate different central opioid peptides, excite different receptors and afferent neurons, and different acupuncture manipulations can induce different neural electrical information encoding sequences. This article has provided theoretical evidences for future researches on the mechanism of acupuncture analgesia.
7.Protection of vitamin C on the cardiac injury induced by nano-titanium dioxide in mice

OBJECTIVE To observe the protection of vitamin C on the cardiac injury induced by 50 nm titanium dioxide inmice.METHODS Kunming mice were ad mistered by ig of vitamin C 100,200 and 400 mg·kg -1 for 2 d.And then the mice were ad mistered by ig of nano-TiO2 2 g·kg -1 and vitamin C (100.0,200.0 and 400.0 mg·kg -1 )for 3 d,the interval of treatment with nano-TiO2 and vitamin C was 4 h.The mice were scarified 24 h later after the last ad ministration.Electrocardiogra m (ECG)was determinated by physiological recorder.The myocardial enzy mes activities in serum and superoxide dismutase (SOD)and glutathione peroxidase(GSH-Px)activities in serum and myocardial tissue were determinated by bioche mical method.Cometassay was used to detect the DNA da mage of the heart. Heart tissue was used for histopathological exa mination by HE staining.RESULTS Co mpared with the control,ECG showed higher S-T and T-wave a mplitude of nano-TiO2 2 g·kg -1 (P<0.05).The myocar-dial enzy mes activities significantly increased and activities of SOD and GSH-Px significantly decreased in nano-TiO2 group,compared with the control group(P <0.05).Cometassay showed that olive tail mo ment (OTM)was significantly increased after nano-TiO2 2 g·kg -1 ,compared with the control group (P<0.05).The histopathology showed ede ma of myocardial cells,myofibril disorders and increasing infla mmatory cells.Vita min C 100,200 and 400 mg·kg -1 can decrease S-T in ECG,OTM,myocardial enzy mes activities,increase the SOD and GSH-Px activities in serum and myocardial tissue;reduce myocardial hypertrophy and infla mmatory cells.CONCLUSION nano-TiO2 can induce myocardial injury inmice and vitamin C can alleviate the da mage.The mechanism may be associated with the antioxidant ability of vitamin C inmyocardial tissue.
8.Effect of Pomegranate Tannins on Endogenous Metabolites in Rat Urine

Objective:To study the influence of pomegranate tannins on the endogenous metabolites in the urine of normal rats by metabonomics method. Methods:The rats were divided into two groups, the blank control group and tannins group. The rats in tan-nins group were orally administered 78. 5 mg·ml-1 pomegranate tannins for 4 weeks. The urine samples were collected and used in the metabonomics study. The urine data were collected by HPLC-MS and comparatively analyzed using PCA and PLS-DA method in SIM-CA-P software. Results:The score points of tannins group displayed obvious distance compared with those of the blank control group, and the changes were the most obvious on the 22nd administration day. Conclusion:Endogenous metabolites in rat urine are obviously influenced by pomegranate tannins, and the research provides the basis for the further study of pomegranate tannins.
9.Effects of Pneumoperitoneum with Carbon Dioxide on Implantation and Growth of Tumor Cells

Objective To study whether carbon dioxide used to establish pneumoperitoneum has an influence on port-site and intraperitoneal implantation and metastasis of tumor cells. Methods R 15 hepatic cancer cells were injected into 30 Wistar rats’ peritoneal cavities 1 hour before operation, then the 30 Wistar rats were randomly divided into 3 groups: gasless group, helium group and carbon dioxide group. The suspension was exposed to the gas environment for 2 hours, all animals were killed after 28 days and the port-site and intraperitoneal implantation and metastasis of tumor cells were examined.Results On port-site, intestinal serous coat, mesentery, greater omentum and diaphragm, the weights of tumor cells, in carbon dioxide group were (326.7?230.3) mg, (626.2?215.9) mg, (476.2?204.8) mg,(2 536.5?906.7) mg and (384.5?149.9) mg respectively; in helium group were (235.6?107.3) mg, (414.2?148.4) mg, (261.8?92.6) mg, (1 633.4?247.3) mg and(220.0?57.9) mg; in gasless group were (145.0?42.4) mg, (221.5?108.2) mg, (212.5?109.6) mg, (797.5?335.9) mg and 113.0 mg.The weights of carbon dioxide group showed a significant increase, compared with helium group and gasless group (P 0.05). Conclusion The insufflation of carbon dioxide promotes intraperitoneal tumor implantation and growth compared with helium and gaslessness in a rat model.
10.Purification and Characterization of Laccase from Monodictys asperospera (Cooke & Massee) Ellis

A new wood-degrading fungus Monodictys asperospera(Cooke & Massee) Ellis with a high level of laccase production was chosen to study.This laccase was purified by ammonium sulfate precipitation,DEAE-cellulose and sephacryl S-300.Purification of about 8.1 fold was achieved with an overall yield of 5.7%.Its molecular weight was estimated to be about 77 kD.The optimum temperature and pH of the lac-case activity were 55?C and 6.0,respectively.Kinetic studies of the laccase showed that the Km and the Vmax for using syringaldazine as substrate was 0.163 mmol/L and 0.194 mmol/(L.min),respectively.The carbo-hydrate content was 18.14%.In addition,it was found that laccase activity was significantly inhibited by Cu2+.
