1. Rapid determination of 17 anti-inflammatory and analgesic chemicals illegally added into Chinese patent medicine and health foods by UPLC-MS/MS

Objective: To estabilsh a rapid and accurate method for the determination of 17 anti-inflammatory and analgesic chemicals (paracetamol, aspirin, phenacetin, chlorpheniramine maleate, rofecoxib, piroxicam, lornoxicam, meloxicam, prednisone acetate, sulindac, naproxen, dexamethasone acetate, phenylbutazone, oxaprozin, celecoxib, diclofenac sodium, and indomethacin) which were illegally added into the Chinese patent medicines (CPM) and the health foods. Methods: The UPLC-MS/MS method was adopted. The samples were extracted with methanol by ultrasonic processing and separated on a Waters Acquity BEH C18 column (100 mm × 2.1 mm, 1.7 μm) with 0.1% formic acid methanol (A) and 0.1% formic acid water (B) as the mobile phase by gradient elution (0-4 min, 40% A; 4-5 min, 40%-50% A; 5-6 min, 50%-60% A; 6-12 min, 60%-80% A; 12-13 min, 80% A; 13-14 min, 80%-40% A) at a flow rate of 0.2 mL/min, and the column temperature was 40℃. A positive-ion (ESI+) source and a MRM mode were used to separately and quantitatively determinate the 17 anti-inflammation and analgesia chemicals. The obtained molecular ions, fragment ions, and retention time for MRM channels were used to identify the 17 chemicals by comparison with those of reference substances. The obtained peak areas were used to determinate the accurate content of chemicals in commonly used drugs in clinic. Results: A good resolution of the 17 chemicals, including paracetamol, aspirin, phenacetin, chlorpheniramine maleate, rofecoxib, piroxicam, lornoxicam, meloxicam, prednisone acetate, sulindac, naproxen, dexamethasone acetate, phenylbutazone, oxaprozin, celecoxib, diclofenac sodium, and indomethacin was obtained under this UPLC and MS/MS condition. The limits of qualitation and quantitation were in the range of 0.3-5.0 and 0.9-15.0 ng/g. The standard addition recoveries were in the range of 90.5%-113.8%. The six components in samples (paracetamol, prednisone acetate, diclofenac sodium, indometacin, chlorpheniramine maleate, and naproxen) were detected. Conclusion: The method is simple, accurate, and has high sensitivity, which can be used for the qualitative and quantitative determination of illegally added chemicals in CPM and health foods.
2.Clinical Significance of Measuring Interleukin-13 in Serum and Pulmonary Function in Infants with Bronchiolitis

Objective To evaluate the clinical significance of measuring IL-13 in serum and dynamics of pulmonary function in infants with bronchiolitis.Methods A total of 50 infants with bronchiolitis(30 in acute phase,20 in recovery phase) and 20 infants in healthy controls were recruited into this study.IL-13 level in serum were detected with Sandwich enzyme linked immunosorbent assay(ELISA),at the same time the object′s respiratory function were tested,then the above indexes were compared.Results The level of serum IL-13 in recovery phase of bronchiolitis was significantly lower than that in acute phase of bronchiolitis and those of both 2 groups were significantly higher than that in control group(all P0.05).Conclusions IL-13 plays a role in pathogenesis in infants with bronchiolitis.The reduction of level of IL-13 will contribute to recovery of infants with bronchiolitis.During the acute phase of bronchiolitis higher resistance can be seen.
3.Experimental study of the anti-melanoma effect of dentritic cells pulsed with RPL8 peptide

Background and purpose:Studies have shown that ribosomal protein L8 (RPL8) is shared by melanomas, gliomas and ovarian carcinomas. A peptide of RPL8 signiifcantly stimulated proliferation and cytokine expression of the hepler T cell clone and lymphocytes in melanoma patients. RPL8 may stimulate anti-tumor immunity, making RPL8 an attractive candidate for therapeutic intervention. In this study, we prepared DC pulsed by RPL8 (RPL8-DC) and investigate the anti-tumor effect of RPL8-DC on melanoma in mice.Methods: The recombinant protein was achieved through IPTG induction in E. coli and identiifed with Western blot. Bone marrow-derived DC was loaded with RPL8 protein. RPL8 and CD11c, CD80, MHC-Ⅰ, MHC-Ⅱmolecules on dentritic cells were monitored by lfuorescence microscope and FACS analysis, respectively. The anti-tumor effect of T cells in vitro was detected by MTT assay. Subcutaneous tumors were induced in C57BL/6 mice using B16 cells. The tumor volumes were measured after injection with RPL8-DC. Results:The puriifed protein was combined with speciifc antibodies. DCs pulsed by RPL8 were visualized under lfuorescent microscopy. CD11c, CD80, MHC-Ⅰ, MHC-Ⅱmolecules on DCs were up-regulated after stimulation with RPL8 and LPS. B16 cells were inhibited by T cells stimulated with RPL8-DC. The inhibition rate of tumor cells was 70%in RPL8-DC group when effector-to-target ratio was 30∶1, which was higher than PBS and DC groups. Inhibition of growth could be observed more signiifcantly in mice after the treatment with RPL8-DC. The mice receiving the therapy of RPL8-DC were able to survive much longer than the mice receiving control therapy. Conclusion:The DC pulsed by RPL8 protein can inhibit the growth of melanoma.
4.Clinical studies on different acupuncture time for primary dysmenorrhea

Objective: To seek the optimal acupuncture time for primary dysmenorrhea and provide clinical basis for optimal acupuncture treatment protocol. Methods:A total of 90 eligible cases were randomly allocated into three groups, 30 cases in each group. Points Guanyuan (CV 4), bilateral Zusanli (ST 36) and Sanyinjiao (SP 6) were selected for patients in all three groups, with a different treatment duration: 15 min in group A, 30 min in group B and 45 min in group C. Then the clinical efficacy in each group was evaluated by pain symptom scoring. Results:As for the pain symptom scores, there were statistically significant intra-group differences between before and after treatment in three groups (allP<0.05); coupled with statistically significant inter-group differences between group B and the other two groups (bothP<0.05). As for clinical efficacy, there were statistical differences between group B and the other two groups (bothP<0.05), indicating that 30 min of acupuncture is the optimal duration in the treatment of dysmenorrhea. Conclusion:With the same needling manipulation, 30 min of acupuncture treatment achieves a better efficacy for primary dysmenorrhea.
6.Follow-Up on Puberty Development and Final Height in 41 Children with Simple Virilizing Form of 21-Hydroxylase Deficiency

Objective To explore the puberty and height development in childern with congenital adrenal hyperplasia(CAH) due to 21-hydroxylase deficiency(21-OHD) with the simple virilizing(SV) form.Methods Patients of SV 21-OHD,diagnosed and treated in Tianjin Children's Hospital from Jan.1970 to Jan.2008,were treated with cortisone acetate or hydrocortisone after diagnosed,and blood investigations were performed as part of monitoring,and dosages were adjusted accordingly to obtain normal growth velocity and slow bone age(BA) development.Gonadotropin-releasing hormone agonist(Gn-RHa) was given after the appearance of central precocious puberty(CPP).Forty-one patients(18 females,23 males) had achieved final height before Dec.31st 2008,and their puberty and height development were analyzed in this longitudinal study by contrast the ages and statures of obtaining final height(FH),chronological age(CA) and BA of G2/B2 stage,duration from G2/B2 stage to obtain FH and stature development of different gender during this period and the influences of compliancy on the ages and statures of obtaining FH were contrasted.Results With regarding to the age of testicular volume ≥4 mL,78.26% boys were before 9 years old.As far as the age of breast development was concerned,38.89% girls were before 8 year old and 66.67% menarche occurred before 10 year old.The differences between the duration and stature development of the period from the state of G2/B2 to skeletal maturation of different gender were significant (Pa

8.Study on the difference of A-scan and IOL Master in measuring intraocular lens power

AIM: To compare the difference of A-scan and lOL Master in intraocular lens power measurement.
●METHODS:Two hundred and twenty-six patients (230 eyes) with age-related cataract were included in the study. Before surgery, axial length was measured by A-scan and lOL Master respectively and corneal curvature was measured by auto refractometer. lntraocular lens power was calculated according to the SRK-T formula. Corneal curvature was measured by auto refractometer and the refractive outcome was performed by phoropter three months after cataract surgery.
●RESULTS:The mean axial length was (23. 48 ± 1. 94) mm measured by A-scan and (23. 75±1. 96) mm measured by lOL Master. There was significant difference between them ( P < 0. 05 ). After random grouping, the preoperative and postoperative mean corneal curvature in A-scan group was (43. 94±1. 81) D and (43. 98±1. 87) D respectively. There was no statistically significant difference between them (P>0. 05). And the results were (44. 10 ± 1. 57 ) D and ( 44. 11 ± 1. 58 ) D in lOL Master group. There was no significant difference between them (P>0. 05); The mean absolute refractive error (MAE) in A-scan group was ( 0. 47 ± 0. 27 ) D and in lOL Master group (0. 41±0. 19) D. The difference was significant (P<0. 05).
●CONCLUSION: lOL Master is proved to be slightly more accurate than A-scan for lOL power calculation.
9.Influence of Mild Hypothermia on Serum ?-Endorphin,Neuron-Specific Enolase in Children with Severe Viral Encephalitis

Objective To explore the effect of mild hypothermia on serum ?-endorphin(?-EP),neuron-specific enolase(NSE) and its protection to encephalon in children with severe viral encephalitis.Methods Fifty-six cases with severe viral encephalitis were divided into mild hypothermia group(30 cases) and regular treatment group(26 cases).Regular treatment group was given combined therapy such as decreasing intracranial pressure,antiviral therapy,supportive and symptomatic treatment.While mild hypothermia group was given mild hypothermia treatment besides regular treatment,and the change of serum ?-EP,NSE before treatment and the first day,5~th and 10~th day after treatment in the two groups were determined.Results There were no significant difference of serum ?-EP,NSE before treatment and in the first day after treatment,while it decreased in 5~th and 10~th day after treatment.Serum ?-EP,NSE in mild hypothermia group were significantly lower than those of regular treatment group(P

Objective To understand the current cognition status of stroke rehabilitation nursing knowledge in young nurses of the department of neurology.Methods Self-designed questionnaires were used for investigating 273 young nurses between 20 and 30 years old of the department of neurology of 4 tertiary hospitals in Zhengzhou.Results The current status of stroke rehabilitation nursing knowledge of the nurses surveyed was generally at a low level.There were no significant differences among nurses with different education degrees,professional titles and whether their relatives suffered from stroke.However,there were significant differences among nurses with different professional lives and whether the department carried out rehabilitation care.Conclusions Nursing educators and managers need to find a new way to improve the level of stroke rehabilitation nursing knowledge of the young nurses.
