1.Changes of CD8⁺ T cells in dextran sulfate sodium-induced colitis mice pretreated with oral immune regulation.

BACKGROUNDIt has been reported that CD8(+) regulatory cells could be induced upon oral tolerance. The purpose of this study was to investigate the changes of CD8α(+) T cells in dextran sulfate sodium (DSS)-induced colitis mice pretreated by oral immune regulation.
METHODSThe effects of five low oral doses of colitis-extracted proteins (CEP) on colitis were evaluated by clinical manifestation and histological lesions. The percentages of CD8α(+) T cells gating on CD3(+) T cells were evaluated in the gut-associated lymphoid tissues (GALT) and the spleens by flow cytometry. Differences between the two groups were compared by Student's t test or Mann-Whitney U test.
RESULTSCompared to bovine serum albumin (BSA)-fed control mice, administration of CEP resulted in marked alleviation of colitis. The proportion of CD8α(+) T cells, not only in intraepithelial lymphocytes (IELs) and lamina propria lymphocytes (LPLs) of the large intestine (LI) but also in spleen from CEP-fed colitis mice, was significantly higher than that from BSA-fed colitis mice (LI-IELs: (71.5 ± 5.4)% vs. (60.1 ± 4.3)%, P < 0.01; LI-LPLs: (60.7 ± 5.2)% vs. (51.9 ± 4.7)%, P < 0.01; spleen: (24.1 ± 3.6)% vs. (20.3 ± 4.1)%, P < 0.05; n = 8). Mucosal repair in repair-period mice five days after termination of DSS treatment was also accompanied by an increase of CD8α(+) T cells in large intestinal mucosal lymphocytes (LI-IELs: (72.1 ± 3.7)% vs. (61.5 ± 4.5)%, P < 0.01; LI-LPLs: (62.1 ± 5.7)% vs. (52.7 ± 3.6)%, P < 0.01; n = 8). The proportion of CD3(+) T cells increased in Peyer's patches (PPs) and decreased in mesenteric lymph nodes (MLNs) from colitis mice compared to untreated mice, whereas the change pattern of CD3(+) T cells in PPs and MLNs from CEP-fed colitis mice was just on the contrary.
CONCLUSIONImprovement of DSS-induced colitis resulted from oral immune regulation is associated with an increase in CD8α(+) T cells in spleen and large intestinal mucosa.

2.Uncoupling protein and nonalcoholic fatty liver disease.

OBJECTIVETo review the current advances on the role of uncoupling protein (UCP) in the pathogenesis and progress of nonalcoholic fatty liver disease (NAFLD).
DATA SOURCESA comprehensive search of the PubMed literature without restriction on the publication date was carried out using keywords such as UCP and NAFLD.
STUDY SELECTIONArticles containing information related to NAFLD and UCP were selected and carefully analyzed.
RESULTSThe typical concepts, up-to-date findings, and existing controversies of UCP2 in NAFLD were summarized. Besides, the effect of a novel subtype of UCP (hepatocellular down regulated mitochondrial carrier protein, HDMCP) in NAFLD was also analyzed. Finally, the concept that any mitochondrial inner membrane carrier protein may have, more or less, the uncoupling ability was reinforced.
CONCLUSIONSConsidering the importance of NAFLD in clinics and UCP in energy metabolism, we believe that this review may raise research enthusiasm on the effect of UCP in NAFLD and provide a novel mechanism and therapeutic target for NAFLD.

3.Application of magnetic resonance imaging with intraoperative color Doppler ultrasound in the treatment of patients with polyacrylamide hydrogel injected for breast augmentation: a retrospective study of 204 cases for 12 years

Purpose:
Polyacrylamide hydrogel (PAHG), which had been used widely for breast augmentation, has been banned for more than 15 years. Patients who had been injected PAHG for breast augmentation need evacuation surgery to remove as much as possible. To provide a series of diagnosis and treatment process MRI and intraoperative color Doppler ultrasound are combined for maximal removal of PAHG.
Methods:
The patients who received evacuation surgery in Peking University Third Hospital from 2010 to 2022 after PAHG injection for breast augmentation were included in this research. MR scanning was performed preoperatively and postoperatively in some of these patients and color Doppler ultrasound was applied to help evacuate PAHG intraoperatively. The mean clearance rate of PAHG was calculated according to the MRI outcomes.
Results:
Two hundred and 4 patients had received evacuation surgery after PAHG injection for breast augmentation with an average age of 42.8 years and an average body mass index of 21.2 kg/m 2 . The average PAHG retention time was 13.5 years. Among them, 52 patients underwent pre- and postoperative MRI scanning. The mean three-dimensional (3D) volume of PAHG was 684.8 mL (range, 350.0–1,123.9 mL), and the average residual 3D volume of PAHG was 53.7 mL (range, 12.4–98.3 mL). The mean clearance rate was 92.1%.
Conclusion
MRI and intraoperative color Doppler ultrasound can provide effective and precise location information of PAHG for evacuation surgery, which is a reliable method to ensure the maximal removal of PAHG.
4.Clinical Observation of Moxibustion on Acupoint Feishu for Treatment of Acute Asthma in Children (Cold-Asthma)

OBJECTIVE To observe the clinical effect of moxibustion on acupoint Feishu (BL13) in the treatment of acute asthma in children (cold-asthma),and to provide clinical evidence for treatment of asthma with acupuncture and moxibustion.METHODS 87 children with asthma (cold-asthma) who meet the inclusion criteria were selected and randomly divided into 2 groups.The control group was treated with conventional western medicine,and the observation group was treated with moxibustion on bilateral Feihu Points(BL13) on the basis of routine western medicine.CRP,IgE,IL1,IL6,EOS,pulmonary function and symptom score were selected as observation indexes for the clinical effects comparison of patients in two groups.RESULTS Indexes levels such as CRP,IgE,IL6 and EOS in the observation group were significantly lower than that in the control group after treatment (P<0.05~0.01).FEV1 and PEF levels of patients in the observation group were significantly better than that in the control group after treatment (P<0.05).Symptom score in the observation group was significantly lower than that in the control group after treatment (P<0.05).Apparent rate (73.81%) of patients in the observation group was significantly higher than that in the control group (53.33%) (P<0.05).CONCLUSION Moxibustion on acupoint Feishu in the treatment of acute asthma in children (cold-asthma) can markedly decrease the levels of CRP,IgE,IL6 and EOS in patients,improve pulmonary function,and can achieve remarkable clinical curative effect.
5.Study on the availability and safety of exercises quantizing prescription for type 2 diabetes mellitus

Objective To evaluate the availability and security of exercises quantizing prescription for type 2 diabetes mellitus. Methods 1 to 5 grades of prescriptions were designed according to different exercises. 80 patients of type 2 diabetes mellitus were treated by 1 to 5 grades of exercise prescriptions. One hour after dinner, each patient started from a minimum amount of Exercise Prescription 1 for three days. Then the patient increased the grade of exercise prescription every three days, until to the greatest amount of Exercise Prescription 5. Exercise time of each exercise prescription was 60 minutes. Heart rate, blood pressure, and PaO2 were determined before exercise, instantly after exercise, 4 hours after exercise, and 12 hours after exercise in all patients, and the incidence of hypoglycemia was counted, so that the security of exercises quantizing prescription could be evaluated. Changes of blood glucose, glycosylated hemoglobin, blood fat, waistline, hip circumference, and weight 3 months after discharge were compared to evaluate the availability of exercises quantizing prescription. Results The values of heart rate, systolic blood pressure, diastolic blood pressure and PaO2 were in the normal range instantly after exercise, 4 hours after exercise, and 12 hours after exercise. There was no occurrence of hypoglycemia among the patients instantly after exercise. The incidences of hypoglycemia were 1.25% and 2.5% 4 hours and 12 hours after exercise. After 3 months, fasting blood sugar, 2hFBG and glycosylated hemoglobin in each group significantly decreased. Total cholesterol decreased obviously except in B2 group. Waistline, hip circumference and weight of those patients whose BMI were 25 decreased obviously. Conclusions Exercises quantizing prescription of diabetes mellitus is a safe and effective exercise therapy for type 2 diabetes mellitus. 1 to 5 grades of exercise prescriptions provide more choices of individual exercise therapy for type 2 diabetes mellitus patients.
6.Research on the Development Trend of Pharmaceutical Intermediates Application Technology in China from the Perspective of Patent

OBJECTIVE:To assist enterprises to understand the technical points a nd development trend of pharmaceutical intermediates industry ,and to provide new development ideas and technical support for pharmaceutical intermediates application technology. METHODS :Taking the core technology involved in pharmaceutical intermediates as the research object and divided into four levels ,the patent data of pharmaceutical intermediates published (announced)before Feb. 2nd,2020 in State Intellectual Property Office (SIPO),INCOPAT and PatSnap patent databases were searched. Patent analysis method was used to analyze the patent application trend and patent applicants of pharmaceutical intermediates ,and the secondary and tertiary technologies of pharmaceutical intermediates were analyzed in detail. Finally ,SPSS 24.0 software was used for curve fitting prediction analysis of the number of patent applications for pharmaceutical intermediates in China. RESULTS :A total of 12 103 related patents were retrieved. Under the secondary technology classification ,there were 331 patents for antibiotic drug intermediates ,474 patents for antipyretic and analgesic drug intermediates ,1 227 patents for cardiovascular drug intermediates ,and 1 550 patents for anticancer drug intermediates. The numbers of patent applications for pharmaceutical intermediates in the world and China were increasing year by year ,and Chinese mainland had the largest number of patent applications. The number of patent applications in Jiangsu , Shanghai and Shandong ranked the top 3. Among the top 10 patent,8 were domestic enterprises or universities ,and they also ranked the top 2 in the composition of pharmaceutical intermediates applicants in China. Among the secondary technologies of pharmaceutical intermediates ,cardiovascular pharmaceutical intermediates accounted for the largest proportion of patent applications (32%),followed by anticancer pharmaceutical intermediates (30%). Among the tertiary technologies ,there were patent applications for the products (35%),methods(35%)and uses (28%)of pharmaceutical intermediates. The curve fitting of the number of patent applications for pharmaceutical intermediates in China showed that the current trend of patent applications was the best fitted with the cubic curve model ,and it was predicted that the number of patent applications in 2020 and 2021 will be 688 and 781 respectively. CONCLUSIONS :The technology of pharmaceutical intermediates has developed rapidly in China ,and universities and enterprises have considerable R&D strength. In addition to expanding in technological innovation ,enterprises can also consider the development idea of school-enterprise cooperation and patent cross licensing among enterprises to help the better deve lopment of China ’s pharmaceutical intermediates industry.
7.The study on exercise safety among elderly diabetes patients

0bjective To observe effects and safety of exercise in elderly patients with diabetes mellitus in order to offer the guideline for the diabetic education about exercise.Methods We designed the five sport prescriptions about the different intensity and evaluated the effect of exercise training on plasma glucose, heart rate.blood pressure and a consumption of the difie,rent exercises.Twentv patients with diabetes mellitus in elderly individuals were evaluated before and after zero hour,4 hours,and 12 hours while exercises were over. And we followed up the effects after 12 weeks.Results The level of glucose in different sport time and in different exercise prescriptions showed significant(P<0.05).The change about the blood pressure in different sport time and in different exercise prescriptions has no statistical significance(P>0.05).After the 1 2 weeks following,the level of plasma glucose,ltbAlc,albumin in urine,triglyeeride,cholesterol,low density lipoprotein and blood pressure decreased(P<0.05).But the change about the weight and high density lipoprotein did not have statistical significance(P>0.05).Conclusions It is effecfive and safe to carry out individual exercise prescriptions among elderly patients with diabetes mellitus.
8.ST-elevated acute myocardial infarction happening 1 month post stent implantation: late thrombosis in-stents or new lesions?

BACKGROUNDST-elevated acute myocardial infarction (STEAMI) happening in the first month post percutaneous coronary intervention (PCI) is almost related to acute thrombosis or subacute thrombosis in-stents. This study aimed to investigate the possible causes of myocardial infarction one month later.
METHODSPatients who had a history of successful PCI, and received coronary angiography or re-PCI due to STEAMI were included in this study. The AMI-related lesions and previous angiographic findings such as the number of lesions, the degree of the stenosis, the type of stents and acute results of last PCI were recorded. If the AMI-related lesion was localized in-stents or at the edge of stents (distance apart from the edge < or = 5 mm), it was defined to be late thrombosis; otherwise as a new-lesion induced AMI.
RESULTSOne hundred and ninety-two patients aged 40 - 79 years were included in this study. New lesions, as the cause of STEAMI, were found in 144 patients (Group A, 75%), and late thrombosis in 48 patients (Group B, 25%). Almost all newly built thromboses were found at the sites of previous insignificant lesions (diameter stenosis < 50%). There was a significant difference in the average time from previous PCI to AMI ((30.1 +/- 12.4) vs (20.3 +/- 11.9) months) between the two groups. Diabetes mellitus (DM) and drug-eluting stent (DES) utilization were associated with markedly higher morbidity of late thrombosis in adjusted Logistic regression (hazard ratio (HR) 3.4, 95% confidence interval (CI) 1.1 - 10.9 and 5.3, 95% CI 1.1 - 26.5).
CONCLUSIONSSTEAMIs happening 1 month after PCI are more likely to develop from previous insignificant lesion rupture than from late thrombosis in-stents. Moreover, DM and DES are associated with the high incidence of late thrombosis, which may indicate that intensive antiplatelet therapy should be considered in patients with diabetes.

9.Mechanical characteristics of the spirochete flagella in 2D micro-periplasmic space

Objective To investigate mechanical characteristics of the spirochete flagella with tight-fitting ribbon configuration in micro-periplasmic space. Methods The 2D model of two parallel plates was used to simplify the periplasmic space, and the effects of flagellum spacing and eccentricity on force and torque acted on the spirochete flagella, and wall shear stress acted on the spirochete protoplasmic cylinder were studied by using numerical simulation method. Results (1) The relationship between the flagellum horizontal force and eccentricity was presented as a parabolic curve, and the peak value of the flagellum horizontal force was mainly caused by the gradual increase of pressure difference at two sides of the cylinder and the resistance viscous force as well. Flagellum spacing had no significant influence on flagellum horizontal force. (2) The relationship between the flagellum torque and eccentricity was presented as an exponential curve, and smaller flagella spacing would cause bigger flagella torque. (3) Flagellum spacing had no significant effect on wall shear stress of the protoplasmic cylinder, but it would be increased with the number of flagella and the eccentricity increasing. Conclusions Numerical simulation results in this study can qualitatively reflect mechanical characteristics of the spirochete flagella, and also provide references for further understanding the morphology of spirochete as well as its kinematic mechanism and pathogenic characteristics.
10.Test-retest reliability of Mandarin monosyllable lists:a multi-center study in Chinese dialectal regions

Objective To develope 22 Chinese Mandarin monosyllable lists with good psychometrical equivalence.This study was to evaluate the test-retest reliability of these lists when it was used in speech recognition test in normal hearing dialectal speakers.Methods Seven cities including Dalian,Shahghai,Hangzhou,Wuhan,Guangzhou,Fuzhou and Xiamen were selected as testing centers which contain 6 typical Chinese dialectal regions including north of China,East of China,north of Fujian,south of Fujian,Guangdong and mid-south of China.At each center,22 local normal hearing people were selected to ioin this study.Every participant was tested by each recognition test of all 22 lists twice in twosessions and same test order respectively.The second run of testing was carried out within 10 days-1 month since first run of testing.Results There was a significant correlations between scores obtained at the two sessions(r=0.682.P<0.01).Paired student-t test had shown that a gross seore of all dialectal participants was significantly higher than that of initial test to retest(P<0.01).The mean increment of score was(2.7±10.1)%.A significant difierence of test-retest score in 7 sites was 19.8% and it was equal to 5 test items.A one way ANOVA analysis had indicated that there were statistically significant difference between the score improvement of 7 test sites(P<0.01).Another analysis had shown that there was no significant correlation between test-retest score improvement and intra-session intervals(P=0.947).Conclusions Mandarin monosyllabic recognition test seems to be more stable,and the present study has indicated a systematic differences in Chinese Mandarin monosyllable recognition scores between test and retest.Monosyllable recognition test iS not susceptible to memorv effect.Pearson's correction analysis is not suitable to evaluation for test-retest reliability.
