1.Relationship between RIG-Ⅰ and IFNs signaling pathway

Objective To study the regulatory effect of interferons(IFNs) on retinoic acid-inducible gene-Ⅰ(RIG-Ⅰ) and the roles of RIG-Ⅰ in IFNs signaling pathway. Methods RIG-Ⅰ expression before and after IFNs treatment in mouse embryonic fibroblasts(MEFs) were analyzed with Northern blotting and semi-quantitative RT-PCR assay.MEFs isolated from wild-type and RIG-Ⅰ-/-mice were used to test growth inhibition and antiviral activity of IFNs with MTT assay and cytopathic effect inhibition assay. Results Both IFN-? and IFN-? could induce RIG-Ⅰ expression in MEFs.Treated with 100 U/mL IFN-?,growth inhibition and antiviral activity of MEFs from wild-type mice were more significant than those from RIG-Ⅰ-/-mice.With the absence of RIG-Ⅰ,the antiviral protective role IFN-? plays was significantly weaker than the wild type. Conclusion RIG-Ⅰ gene is a novel mediator of interferon effects on cells.It may participate in the inflammation responses mediated by IFNs through modulating cytokines production.
2.Investigation of psychological process of children with leukemia

Objective To explore the psychological demand of leukemia in different periods,understand the psychological characteristics of children and to provide psychological nursing basis. Methods The real mental feeling in diagnosis, treatment and rehabilitation of leukemia patients was surveyed through interview and questionnaire. Results The psychology of leukemia was influenced easily by external factors without regularity. The psychological characteristics included: fear, dependence, calm, confident, loneliness,disappointment, and accept ion. Conclusions Psychological change of leukemia patients has no obvious regularity. Nursing staff shall provide proper psychological nursing according to the different characteristics of psychological.
3.Influence of different incision protection styles on incision bacterial growth in cesarean section

Objective To analyze the protective effects of different incision protection styles on incision bacterial growth of cesarean.Methods Totals of 300 patients of cesarean section were divided into three groups randomly with 100 patients in each group:group one which operation film was pasted after operation sheet draped,the operation film was pasted before operation sheet draped in group two,and traditional skin towel was applied in group three.Subcutaneous tissue fluid was dipped after skin incision and before skin suture in each patient.The tissue fluid was bacterial cultured,and the influence of different incision protection styles on incision infection in cesarean was judged according to the bacterial growth.Results The preoperational tissue fluid had no bacterial growth in group one,group two and group three (x2 =1.31,P >0.05).The postoperative tissue fluid had zero bacterial growth in group one,and one in group two,and seven in group three.The bacterial growth rate in group three was higher than that in group one and group two (11.67% vs 0% and 1.67%) (x2 =4.69,P < 0.05),but there was no significant difference between group one and group two (P >0.05).Conclusions The operation film pasted after operation sheet drape may be more effective on prevention of incision bacterial growth in cesarean section.
4.Effects of glycine on apoptosis in murine cardiomyocyte suffering from ischemia and hypoxia.

OBJECTIVETo investigate the effects of glycine on apoptosis in murine cardiomyocyte suffering from ischemia and hypoxia.
METHODSThe primary passage of cultured cardiomyocytes from neonatal rats were subjected to ischemia and hypoxia, and the cells were divided into IH (without other treatment), and G (with treatment of 5 mmol/L glycine) groups. Normal murine cardiomyocytes served as control (C group). Cardiomyocytes were cultured for 6 hours in vitro. Apoptosis, mitochondrial membrane potential and its distribution, the condition of mitochondria permeability transition pore (mPTP) were observed with expression of fluorescence intensity. The activity of caspase-3 was observed by Laser Scanning staining.
RESULTS(1) Apoptosis: the fluorescence intensity in IH group was obviously higher than that in G and C groups (P < 0.01). (2) Mitochondrial membrane potential: the fluorescence intensity in IH group was 32 +/- 7, which was obviously lower than that in G and C groups (52 +/- 4, 73 +/- 4, respectively, P < 0.01). (3) The condition of mPTP: the intensity in IH group was 27 +/- 4, which was obviously lower than that in G and C groups (62 +/- 8, 90 +/- 7, respectively, P < 0.01). (4) The activity of caspase-3: the activity of caspase-3 in IH group was obviously higher than that in G and C groups (P < 0.01).
CONCLUSIONGlycine can inhibit apoptosis in cardiomyocytes subjected to ischemia and hypoxia,and the effect may be attributable to changes in mitochondrial membrane potential, lessening opening of mPTP, alleviation of calcium overload , and decrease in activity of caspase-3.

5.Study on metronidazole resistance to Helicobacter pylori from three populations with different ethnics in Yunnan.

OBJECTIVETo evaluate the prevalence of Helicobacter pylori (H.pylori) resistance to metronidazole among three populations in Yunnan.
METHODSSusceptibilities to metronidazole among 109 H. pylori strains (33 H. pylori strains from Han, 31 H. pylori strains from Bai and 45 H. pylori strains from Naxi ethnic populations) were tested by Epsilometer test (E-test).
RESULTSIn 109 H. pylori strains, the overall metronidazole resistance rate was 67.89%. There were no significant difference in the metronidazole resistant rates of H. pylori among Han, Bai, Naxi populations Yunnan in terms of the distribution on age and upper gastroduodenal diseases. In the facet of gender, metronidazole resistant rate of H. pylori was significantly lower in Han males than in females (chi2=5.304, P=0.027), but not seen in the Bai or Naxi peoples.
CONCLUSIONMetronidazole resistance rate of H. pyloriin Yunnan was high, but no significant difference was found among Han, Bai, Naxi peoples in the province.

6.Identification of metastasis-related microRNAs of hepatocellular carcinoma in hepatocellular carcinoma cell lines by quantitative real time PCR.

OBJECTIVETo identify the metastasis-related miRNAs in hepatocellular carcinoma (HCC) cell lines.
METHODSA qRT-PCR method was established and optimized.
RESULTSAll candidate metastasis associated miRNAs except miR-124a were expressed in high metastasis cell line MHCC97H and low metastasis cell line MHCC97L, while some miRNAs were differentially expressed between liver cancer cell line (HepG2) and hepatic cell line (L02) (P less than 0.05), these miRNAs include: miR-148b (1.96+/-0.51 vs 3.76+/-0.28), miR-9 (-4.38+/-0.86 vs -1.10+/-0.53), miR-30c (8.41+/-0.40 vs 6.82+/-0.29), miR-338 (3.14+/-0.29 vs -2.36+/-0.32), miR-34a (0.71+/-0.40 vs -2.95+/-0.26), Let-7g (-4.07+/-0.55 vs -6.98+/-0.56). miR-148b expression was about 4 times higher than miR-148a [5.46 (IQR 4.25-6.67) vs 1.29 (IQR 0.94-1.64)] in all cell line tested (Z=-5.097, P=3x10(-7)).
CONCLUSIONThis study may help to understand the biological significance of miRNAs in HCC metastasis.

7.Research on related factors for hypoglycemic episodes in patients with type 2 diabetes mellitus based on continuous glucose monitoring system

Objective To identify related factors for hypoglycemic episodes in patients with type 2 diabetes mellitus(T2DM)through continuous glucose monitoring(CGM). Methods The included 147 patients with T2DM were those who had undergone CGM for 5 days in our ward of Department of Endocrinology and Metabolism,Huashan Hospital from Dec 2018 to Oct 2019. The general information, laboratory parameters and CGM parameters of the patients were collected. According to whether there wasan episode of hypoglycemia during the monitoring period,the patients were divided into non-hypoglycemia group and hypoglycemic group. A single hypoglycemia episode was defined as a sensor monitoring blood glucose of less than 3.9 mmol/L and lasting for more than 15 minutes.CGM parameters included the mean blood glucose(MBG),standard deviation(SD),coefficient of variation(CV),the differences between maximum and minimum blood glucose (BG) levels (ΔBG),mean amplitude of glycemic excursions (MAGE)and the percentage of time in range(%TIR)of BG at <3.9 mmol/L,3.9-7.8 mmol/L,>7.8 mmol/L,3.9-10.0 mmol/L,and >10.0 mmol/L. Results Logistic regression analysis showed that lower estimated glomerular filtration rate(eGFR)levels,increased use of insulin and its analogs and lower MBG levels were associated with hypoglycemic episodes. Spearman correlation analysis showed that the MBG level and the %TIR of BG>7.8 mmol/L and BG>10.0 mmol/L were negatively associated while glycemic variability(GV)levels(SD,CV,ΔBG,MAGE)and % TIR of BG at 3.9-7.8 mmol/L were positively associated with hypoglycemic episodes. Pearson correlation analysis showed that the duration of hypoglycemic episodes was positively correlated with the use of sulfonylureas and CV levels. Conclusion Lower eGFR levels,increased treatment with insulin and its analogs and lower MBG levels were related factors for hypoglycemic episodes in patients with T2DM.
8.Relationship between work-family balance and depression among employees in organization: A cross-lagged regression analysis

Objective:To investigate the relationship between employees'work-family balance (WFB) and depression.Methods:Totally 259 employees from 4 departments in a petrochemical enterprise were assessed with the Work-Family Balance Scale (WFBS) and Patient Health Questionnaires-9 (PHQ-9) and were conducted a 4-months follow-up study via cross-lagged regression.Results:There were significantly negative correlation between the scores of WFBS and PHQ-9 (r =-0.12--0.22,Ps < 0.05).Cross-lagged regression revealed that WFBS score at time 1 was not a predictor of PHQ-9 at time 2 (β =0.04,P > 0.05),and PHQ-9 score at time 1 was a negative predictor of WFBS score at time 2 (β =-0.13,P < 0.05).Conclusion:There may be a close relationship between work-family balance and depression,and depression could predict work-family balance among employees in organization.
9.Value of plasma NT-proBNP for diagnosing heart failure in patients with previous myocardial infarction

Objective The study aimed to evaluate the value of plasma NT-proBNP in diagnosing heart failure in patients with previous myocardial infarction.Methods Plasma concentration of NT-proBNP was measured in patients with previous myocardial infarction by ELISA method at admission.Patients were divided into non heart failure group(NYHA class I)and heart failure group(NYHA class Ⅱ-Ⅳ).The NT-proBNP levels were compared between NYHA class Ⅰ,Ⅱ,Ⅲ and Ⅳ,and between heart failure group and non heart failure grou.ROC analyses were performed to evaluate the diagnosing value of plasma NT-proBNP for heart failure and to identify the optimal cut-off point for diagnosing heart failure patients.Results Total 586 patients[aged from 25-83(58 ± 11)years,80% male]with previous myocardial inffarction were enrolled in his study(n =374 of NYHA class Ⅰ,n =99 of NYHA classes Ⅱ,n =82 of NYHA class]Ⅲ,n =31 of NYHA class Ⅳ).Plasma NT-proBNP levels in these four NYHA classes were 484.7(381.6,647.8)pmol/L,907.6(516.6,1290.3)pmol/L,1420.2(879.5,2336.2)pmol/L,2442.6(1695.4,3670.7)pmol/L,respectively(P <0.01).The plasma NT-proBNP level in heart failure group wassignificantly higher than in non heart failure group[1148.2(707.9,2145.3)pmol/L vs.484.7(381.6,647.8)pmol/L,P <0.01].Plasma NT-proBNP level in patients with age≥60 years was significantly higher than patients with age < 60 years[702.3(472.4,1208.5)pmol/L vs.526.6(392.1,855.6)pmol/L,P<0.01].ROC analysis showed that the area under the curve(AUC)for diagnosing heart failure was 0.844(95% CI: 0.809-0.880,P < 0.0l),the optimal plasma NT-proBNP cut-off point for diagnosing heart failure was 700 pmol/L with a sensitivity of 75.9%,a specificity of 79.9%,an accuracy of 78.3%,a positive predictive value of 67.9% and a negative predictive value of 85.3%.The optimal plasma NT proBNP cut-off point was 600 pmol/L for patients ≥60 years old and 800 pmol/L for patients < 60 years old.Conclusion Plasma NT-proBNP level is a valuable parameter for diagnosing hemt failure in patients with previous myocardial infarction.

Objective:To investigate the association between body mass index (BMI) trajectories in children and adolescents and subclinical renal damage (SRD) in adulthood.Methods:4 623 participants aged 6-18 years old were recruited from the ongoing cohort of Hanzhong adolescent hypertension study in 1987, and the subjects were followed up in 1989, 1992, 1995, 2005, 2013 and 2017, respectively. Group-based trajectory modeling was used to identify distinct BMI trajectories in longitudinal analysis. Generalized linear model was applied to examine the association between different BMI trajectories and SRD incidence in adulthood.Results:A total of 2 678 subjects from childhood to adulthood were enrolled in this study. All subjects were divided into three groups according to three distinct BMI trajectories: low-increasing BMI group ( n=1 017), moderate-increasing BMI group ( n=1 353), and high-increasing BMI group ( n=308). Over follow up for 30 years, a total of 248 participants (9.3%) developed SRD. Urinary albumin-to-creatinine ratio (uACR) in low to high-increasing BMI group was 0.9(0.6, 1.4), 1.0(0.7, 1.7), 1.6(0.8, 3.2), respectively ( P trend<0.001), and estimated glomerular filtration rate was 98.5(87.6, 111.6) , 96.2(86.4, 109.7), 95.3 (87.5, 125.0) ml·min -1·(1.73 m 2) -1, respectively ( P trend=0.025). The generalized linear model analysis showed that uACR was increased linearly from low to high-increasing BMI group [ β=3.16(95% CI 1.02-5.31), Ptrend=0.004]. There was no correlation or linear trend between BMI trajectory and estimated glomerular filtration rate [ β=-2.30(95% CI-5.18-0.57), Ptrend=0.117]. Compared with the low-increasing BMI group, the high-increasing BMI group had greater odds of experiencing SRD in adulthood after adjusting for multiple confounders such as age, gender, medical history and lifestyle ( OR=2.83, 95% CI 1.84-4.36, Ptrend<0.001). Conclusions:Higher BMI trajectorie is correlated with higher level of uACR and risk of SRD in middle age. Identifying long-term BMI trajectorie from early age may assist in predicting individuals′ renal function in later life.
