1.Study on the intervention effect of continued nursing on the quality of life of patients with chronic obstructive pulmonary disease

Objective To explore the effect of continued nursing intervention on quality of life of patients with chronic obstructive pulmonary disease. Methods The study took use of randomized controltest. All the 130 patients who were on admission because of acute exacerbation of chronic obstructive pulmonary disease were randomly divided into the intervention group (62 cases) and the control group (68 cases). The intervention group received telephone and home follow-up for one year, and the control group was provided conventional care. Lung function, SGRQ and dyspnea degree were collected on one-month,three-month, six-month, twelve-month after intervention. Results Lung function and activity scores of SGRQ, dyspnea degree and six minutes walk test between two groups had statistical significance after intervention. Conclusions Continued nursing intervention can postpone the rate of FEV1% declining, reduce dysnea degree, decrease the frequency of acute exacerbation, increase the quality of life of COPD patients.
2.MicroRNA expression profiles and miR-10a target in anti-benzoa pyrene-7, 8-diol-9, 10-epoxide-transformed human 16HBE cells.

OBJECTIVETo screen miRNA profiles of malignantly transformed human bronchial epithelial cells, 16HBE-T, induced by anti-benzo[a]pyrene-trans-7,8-diol-9,10-epoxide (anti-BPDE), and to analyze putative miR-10a targets in 16HBE-T.
METHODSA novel microarray platform was employed to screen miRNA profiles of 16HBE-T cells transformed by anti-BPDE. Microarray data for miR-10a and miR-320 were validated using quantitative real time polymerase chain reaction (QRT-PCR). The expression of a putative target for miR-10a, HOXA1, was analyzed by reverse transcription polymerase chain reaction (RT-PCR) and QRT-PCR.
RESULTSIn comparison with the vehicle-treated cells (16HBE-N), 16HBE-T exhibited differential expression of 54 miRNAs, in which, 45 were over-expressed and 9 were down-regulated. The five most highly expressed miRNAs were miR-494, miR-320, miR-498, miR-129, and miR-106a. The lowest expressed miRNAs were miR-10a, miR-493-5p, and miR-363*. Three members of miR-17-92 cluster, miR-17-5p, miR-20a, and miR-92, showed significantly higher abundance in 16BHE-T as miR-21, miR-141, miR-27a, miR-27b, miR-16 and miRNAs of the let-7 family. The putative target for miR-10a, HOXA1 mRNA was up-regulated 3-9-fold in 16HBE-T, as compared with 16HBE-N.
CONCLUSIONThe findings of the study provide information on differentially expressed miRNA in malignant 16HBE-T, and also suggest a potential role of these miRNAs in cell transformation induced by anti-BPDE. HOXA1 is similarly up-regulated, suggesting that miR-10a is associated with the process of HOXA 1-mediated transformation.

3.MicroRNA profiles of malignantly transformed cells induced by anti-benzo-a-pyrene-trans-7, 8-dihydrodiol-9, 10-epoxide.

OBJECTIVETo screen microRNA (miRNA) profiles of malignantly transformed cells induced by anti-benzo-a-pyrene-trans-7, 8-dihydrodiol-9, 10-epoxide (BPDE) and to look for miRNAs which is expressed differently between malignantly transformed cells and normal human bronchial epithelial cells 16HBE.
METHODSExperimental group was the malignantly transformed 16HBE which was induced by cultured with final concentration 2.0 micromol/L of BPDE which was dissolved in dimethyl sulphoxide. The control group was 16HBE that was cultured with minimal essential medium including dimethyl sulphoxide. 327 miR-NAs were tested be-tween those two groups with miRNA microarray analysis. MiR-10a that was down expressed and miR-320 that was overexpressed were selected to be validated by miRNA specific quantitative real-time reverse transcriptase chain reaction (miR qRT-PCR).
RESULTS327 human miRNAs were tested with miRNA microarray analysis. 55 miRNAs were found expressing differently between those two groups and of which 46 were overexpressed and 9 were down expressed. Some data were validated by quantitative RT-PCR.
CONCLUSIONmiRNAs expressed significantly between malignantly transformed 16HBE and normal cells and this helps us look for unique miRNAs of malignantly transformed cells induced by BPDE, but there should have more sufficient evidences to prove their functions in malignant cells.

4.Investigation on awareness of perimenopausal hormone replacement therapy among a part of the medical care personnel in Guiyang

Objective To study the awareness of perimenopausal hormone replacement therapy (HRT) among a part of the medical care personnel in Guiyang . Methods A survey was conducted among 500 medical staff members in 4 hospitals of Guiyang by cluster random sampling using questionnaire about HRT. Results The survey showed that 74.6% (373/500) medical staff thought that the hormone replacement therapy was necessary to perimenopausal women; 96.7% (87/90) of obstetrics and gynecology doctors believed that it was necessary for perimenopausal women to use HRT,which was significantly higher than the doctors of other specialties 68.6% (166/242) and the nurses group 71.4% (120/168) (χ2=28.509, 23.537, P<0.01). Only 5.8%(29/500) of the medical personnel were willing to recommend HRT. In light of the attitude for recommending HRT, the obstetricians and gynecologists group was more significantly higher than the other specialties doctors group (χ2=86.781, P<0.01). Conclusion The knowledge of hormone replacement therapy in part of Guiyang medical personnel is not sufficient;the recommending rate of HRT was low;the side effects of HRT was still a concern. There are differences between obstetrics and gynecology doctors and doctors other specialties and nurses in HRT knowledge.
5.Evaluation of effects of open visiting in very-low-birth-weight infants

Objective To explore the feasibility of open visiting among mothers' of very-low-birth-weight(VLBW) infants and observe effects.Methods Totally 74 eligible VLBW infants were recruited,those who were visited and cared daily by mothers were assigned into the experimental group(n=36) and those who were not visited by mothers were assigned into the control group(n=38).The experimental group received open visiting as well as individualized developmental nursing care,and the control group only received individualized developmental nursing care.Relevant regulations and procedures were designed,and clinical pathways of mothers' involvement in nursing care were developed and implemented.Incidence of infection,time to reach full oral feeding,weight gaining during hospitalization,rate of breast-feeding during hospitalization and 30 days after discharge,hospitalization time and rate of readmission 30 days after discharge were recorded and compared.Results There was no significant difference in incidence of infection between two groups (P>0.05).Time to reach full oral feeding in the experimental group was earlier than that of the control group,weight gaining during hospitalization in the experimental group was faster than that of the control group,rate of breast-feeding 30 days after discharge was higher in the experimental group,and the differences were significant (P<0.05).Hospitalization time in the experimental group was shorter than that of the control group,rate of readmission 30 days after discharge in the experimental group was lower,and the differences were significant (P<0.05).Conclusion Mother's open visiting for VLBW infants in NICU does not increase the incidence of infection,but can shorten time to reach full oral feeding,promote weight gaining,increase rate of breast-feeding 30 days after discharge,reduce hospitalization time,and decrease rate of readmission 30 days after discharge.
6.Effects of Different TCM Exercises for Preventive Treatment of Diseases on Sleep Quality, Depression and Anxiety of College Students

Objective To observe the effects of different TCM exercises for preventive treatment of diseases on sleep quality and multidimensional mental health level of college students.Methods According to the willing of the research objects, 132 college students were divided into Yijinjing group, Baduanjin group, Wuqinxi group, Tai Chi group, and jogging group. They exercised 45 min each time, 5–6 times per week, received central guidance every two weeks, and exercise intensity was timely adjusted based on master of physical activity and reaction to the situation of college students, for 2 months. PSQI, BFS, SDS and SAS before and after intervention in each group were observed. Results After intervention, five kinds of exercises could improve sleep situation of college students, and had their own focus in sleep quality, falling-asleep time, sleep time, sleep efficiency, sleep disorders, sleep medication, daytime dysfunction. There was statistical significance in BFS activity score, thought score, anger score, excitation score, and depression score after intervention in Yijinjing group compared with before intervention (P<0.05); There was statistical significance in activity score, total joy score, calm total score after intervention in Wuqinxi group compared with before intervention (P<0.05). There was statistical significance in SAS total scores of college students in Yijinjing group, Baduanjin group, and jogging group after intervention compared with before intervention (P<0.05). There was statistical significance in SAS standard scores of college students in Yijinjing group, Baduanjin group, and jogging group after intervention compared with before intervention (P<0.05). There was no statistical difference in SDS scores in the five groups before and after intervention (P>0.05).Conclusion 5 kinds of exercises can improve the sleep quality from different aspects. Yijinjing group and Wuqinxi group can improve the mental health of college students, of which Yijinjing group can bi-directionally regulate the positive and negative psychological indicators of college students.
7.The integrated teaching design of the core course of Chinese medicine specialty " Traditional Chinese Medicine Analytics" based on virtual simulation

The course of " Traditional Chinese Medicine Analytics" of Shanghai University of Traditional Chinese Medicine was listed as the Shanghai Key construction course in 2017, was selected as the Shanghai first-class course in 2021, and was selected as the national first-class course in 2023. This article reviews the knowledge points of identification analysis, quantitative analysis, inspection, fingerprint, and specific chromatogram in the course of " Traditional Chinese Medicine Analytics" which are integrated to the virtual simulation experimental projects to construct a knowledge map for this course. With gas chromatography as an example, this article introduces the ideas for constructing virtual simulation experimental projects of gas chromatography analytical techniques in traditional Chinese medicine and evaluates the teaching effect from the dimensions of the degree of student completion, learning performance, and error rate, so as to lay a solid foundation for promoting the integration of virtual simulation technology into the construction of core courses, continuously improve the quality of classroom teaching, and achieve the goal of winning in the classroom.
8.Application of ricin-immunotoxin mediated T cell depletion to allogeneic hematopoietic stem cell transplantation.

This study was aimed to investigate the clinical outcome of ricin-immunotoxin mediated T cell partially depleted HLA/MLC mismatched allogeneic hematopoietic stem cell transplantation. 13 patients with hematological malignancies were treated by ricin-immunotoxin mediated T cell partially depleted allogeneic hematopoietic stem cell transplantations from HLA/MLC mismatched donors, including 6 cases of CML in CP(1), 1 case of ALL in CR(1), 1 case of ALL in CR(2), 1 case of ALL in relapse, 2 cases of AML in CR(1), 1 case of AML in CR(2), 1 case of MDS-RAEBT-AML (M(4)) in CR(1). The results showed that 8 cases were engrafted successfully, 2 cases of them developed grade II acute GVHD and 2 cases developed grade III-IV acute GVHD. Within following-up of 8 - 90 months, 2 patients who experienced grade III-IV acute GVHD died early after transplantation; 1 patient died of late onset of infection; the other 5 patients survived free from diseases. After failure at first infusion, 4 patients were given reinfusion of peripheral blood hematopoietic stem cells from the same donor. 3 out of 4 cases failed to engraft and only one patient got engraftment but died of related complications of transplantation. One patient was performed a second transplantation from a syngeneic donor and survive free of disease until now. In conclusion, T cell partially depleted HLA/MLC mismatched allogeneic hematopoietic stem cell transplantation by ricin-immunotoxin decreases the occurrence of severe acute GVHD but with high risk of rejection, which clinical outcome still needs further evaluation.

9.Effect of protein kinase C and calcium on the thrombin receptors activation.

OBJECTIVETo compare the effects of protein kinase C (PKC) and calcium (Ca2+) on platelet aggregation and platelet membrane surface GP I b expression in thrombin receptors activation, and investigate the role of Gq signal transduction pathway in such activation.
METHODSPeptide SFLLRN (PARI-AP) and AYPGKF (PAR4-AP) were used to stimulate platelet, and the effects of Ro-31-2220 (inhibitor of PKC) and BAPTA/AM (calcium chelator) on the platelet aggregation and GP I b were analyzed.
RESULTSEither 25 micromol/L PAR1 or 250 micromol/L PAR4 peptide could induce absolute platelet aggregation with a reversible internalization of GP I b. Platelet aggregation was inhibited by Ro-31-2220 or BAPTA while the morphological change curve still occurred upon PARs activation. In addition, Ro-31-2220 decreased GP I b centralization upon PAR1 stimulation [(87.00 +/- 0.04)% and (73.00 +/- 0.08)%, respectively at 1, 2 min, P<0.05], albeit it blocked the internalization of GP I b in PAR4 activation [(44.00 +/- 0.01)% and (46.00 +/- 0.05)%, respectively at 10, 30 min, P <0.05]. Meanwhile, GP I b internalization was blocked by BAPTA in both peptides [(94.00 +/- 0.08)% and (95.00 +/- 0.00)% at 1 min, (92.00 +/- 0.02)% and (94.00 +/- 0.01)% at 2 min, (91.00 +/- 0.02)% and (91.00 +/- 0.02)% at 5 min, (90.00 +/- 0.04)% and (87.00 +/- 0.03)% at 10 min, respectively, P <0.05].
CONCLUSIONPKC and calcium play an important role in thrombin receptor activation. Calcium is closely correlated with such activation, being similar in the two PARs signal pathways. PK C promotes GP I b centralization in PAR1 pathway and accelerates GP I b return to membrane surface in PAR4 pathway.

10.Effect of anti-Helicobacter pylori ureB monoclonal antibody on platelet aggregation and activation, and its mechanism study.

OBJECTIVESTo study the effect of monoclonal antibody (McAb) against helicobacter pylori (Hp) ureB, 1F11 on platelet aggregation and activation, and its mechanism.
METHODSThe relativity between human platelet glycoproteins (GPs) and Hp ureB was identified by Western blot and FCM. Platelet aggregation was measured by turbidimetry, and P-selectin and TXB2 assay by ELISA.
RESULTS1F11 could bind to platelet GPIIIa, and ADP-induced platelet aggregation was inhibited by 1F11 in a dose-dependent manner. However, 1F11 had no effect on plasma P-selectin and TXB2 induced by ADP. The FCM results show that the positive rates of platelet binding to FITC-SZ21 was decreased from 99.5% to 77.4% after addition of 1F11.
CONCLUSIONMcAb against Hp ureB 1F11 inhibits platelet aggregation through binding to platelet GPIIIa but does not block platelet activation. There might be crossed-epitopes on Hp ureB and platelet GPIIIa, and Hp infection might be involved in ITP immunopathology.
