1.Finite element analysis of unilateral and bilateral pedicle screw fixation before and after segment fusion in transforaminal lumbar interbody fusion

Objective To explore the difference between before and afteintervertebral fusion in transforaminal lumbainter-body fusion (TLIF) by unilateral obilateral pedicle screw (PS) fixation .MethodThe L3 -5 Cdatbased on the normal people were used to rebuild the L3 -5 three-dimensional finite elemenmodel(INT) and TLIF operative model .On thibasi,the unilateral Pfixation (M1) and bilateral Pfixation (M2) modelbefore the intervertebral fusion and the unilateral Pfixation (M3) and bi-lateral Pfixation (M4) modelafteintervertebral fusion were buil.Aftesetting the boundary and loading the burden ,the ante-flexion ,retroversion ,lateral bending and rotation of lumbavertebrwere simulated and the L 4 -5 segmenanguladisplacemenand the stresdistribution of implanwere recorded .ResultThe unilateral and bilateral Pfixation model before fusion reduced the range of motion(ROM ) of segmencompared with the Inmodel ,the bilateral fixation wareduced greatethan the unilateral fixa-tion ,buthe difference aftefusion wasignificantly decreased .The peak Von Misestressebefore fusion in the unilateral Pfixa-tion wasignificantly highethan thain the bilateral fixation ,while theipeak Von Misestresseaftefusion trended to be con-sisten.Conclusion The unilateral Pfixation aftefusion can provide the consistensegmenstability same to the bilateral Pfixa-tion .The long-term effecof TLIF with unilateral Pfixation isimilato thawith the bilateral Pfixation .
2.Biological features and ultrastructure of rabbit mesenchymal stem cells in vitro

Objective To isolate and culture the rabbit mesenchymal stem cells,and explore the biological features in vitro.Methods Two milliliter of bone marrow was extracted from the tibia of juvenile rabbit under anesthesia and sterile conditions,and mesenchymal stem cells were obtained by density gradient centrifugation and inoculated to 100ml culture flask.The morphological changes,adherence rate and ultrastructure of the stem cells were observed.The surface markers and growth cycle of the mesenchymal stem cells were detected by flow cytometry,and biological features were observed by inducing the cells differentiate into osteoblasts.Results The adherence of mesenchymal stem cells started 2 hours after culturing,basically finished 20 hours later,and more than 90% of the cells were fused at the 15th day.Observed with transmission electron microscopy,the cultured cells showed typical ultrastructure of stem cells: numbers of microvillus mainly distributed on one side of the cells,irregular nuclei with indentation,and lots of cell organs.Analysis for the cell surface markers indicated positive expression of CD44 and CD90 and negative expression of CD45 in mesenchymal stem cells.Cell growth cycle assay showed that most mesenchymal stem cells(about 97%) stayed in stage G1 and G2.Three weeks after osteoblastic induction,the mesenchymal stem cells gradually produced small calcium salt crystals,the cells were positive in alkaline phosphatase staining,and Von Kossa staining showed many calcium salts in the cell clone as black reaction deposits.Conclusions The rabbit bone marrow mesenchymal stem cells isolated by density gradient centrifugation are of high purity and stable growth rate,and the cells' characters can be still maintained after serial subcultivation.Since easy to operate on isolation and proliferation,the mesenchymal stem cells could be used as the tissue engineering seed cells in repairing of intervertebral disc degeneration.

4.Influence of Sanguisorba Officinalis Tannins Extract on Proliferation of Human Renal Tubular Epithelia Cells Induced by TGF-β1

Objective:To study the preventive effect of Sanguisorba officinalis on renal fibrosis by observing the influence of San-guisorba officinalis tannins extract (STE) on the proliferation of human renal epithelia (HK-2) induced by transforming growth factor-β1 ( TGF-β1 ) . Methods:HK-2 cells were cultured in DMEM medium with high glucose containing 10% fetal bovine serum. The cul-tured cells were divided into 5 groups, including the blank control group, TGF-β1 group (5 ng· ml-1 TGF-β1), intervention group 1 (5 ng·ml-1 TGF-β1 +12. 5μg·ml-1 STE), intervention group 2 (5 ng·ml-1 TGF-β1 +25μg·ml-1 STE) and intervention group 3(5 ng·ml-1 TGF-β1 +50 μg·ml-1 STE). The changes of cell morphology were observed under an inverted microscope and the in-fluence of SET on the cell proliferation was detected by CCK8 assay. Results: TGF-β1 could significantly induce the proliferation of HK-2 and promote the cell fibrosis with significant difference when compared with the control group (P<0. 05). However, after trea-ted with STE, the cell proliferation was inhibited obviously (P<0. 05) and the cells morphology tended to be normal in a dose-depend-ent manner. Conclusion:STE can inhibit the proliferation of HK-2 and prevent renal fibrosis to some extent.
5.Analysis of high risk factors of liver metastasis in patients with postoperative colorectal cancer

Objective To screen the risk factors of liver metastasis in patients with postoperative colorectal cancer and to set up a mathematic model.Methods One hundred and sixty-eight patients received radical surgery were retrospectively analyzed.Logistics model regression was used to identify the risk factors and ROC test to complete a mathematic model.Results The 3 and 5-year incidence rate of liver metastasis was 25.0% and 33.3%,respectively.The risk factors of liver metastasis were lymph node metastasis (LNM),tumor invasive depth (TID),histological grading (G) and preoperative serum tumor markers (PSMs).The mathematic model was:logit (P) =-0.931 + 0.973 × LNM + 0.302 x TID + 0.637 x G + 0.042 x PSMs.Conclusion The risk factors of liver metastasis were LNM,TID,G and PSMs,and the mathematic model could be set up.

Developmental changes in children can affect drug disposition and clinical effects. A physiologically-based pharmacokinetic (PBPK) model is a mathematical model that can be used to predict blood drug concentrations in children and gain insight into age-dependent physiological differences in drug disposition impact. Pediatric PBPK (P-PBPK) models have attracted attention over the past decade. With the concerted efforts of academia, pharmaceutical companies, and regulatory agencies, there are more and more examples of pediatric clinical studies using PBPK models. Nevertheless, the number of P-PBPK models and their predictive performance still lag behind adult models. By referring to the literature, we study the process of children adapting to adult absorption, distribution, metabolism, and excretion (ADME) parameters and analyze the general principles of P-PBPK model establishment. In addition, we summarize the functions and application examples of commonly used P-PBPK modeling software to provide a basis for the rational application of modeling software.
10.Aesthetic repair of medial upper lip defects

Objective To explore the aesthetic results of medial upper Iip repair for skin and soft tissue defects using local flap.Methods According to the location and the size of upper lip defect,the modified rhomboid flap of 60 ° above the defect was designed to repair the wound in philtrum;the O-L flap along philtral column to vermilion border was designed to cover the wound close to the peak and lateral to philtral column.Results Twenty-one postoperative patients that underwent pigmented nevus removal (12 female,9 male;ages 16-33 years) had defects in philtrum for 10 cases and close to the peak and philtral column for 11 cases.The diameter of the defect ranged from 0.5 cm to 1.0 cm.All flaps survived primarily without any complications and follow-up was for 1 to 18 months with excellent outcomes in all cases.All patients were satisfied with contour and functions.No pigmented nevus recurrence occurred.Conclusions The flap designed according to the aesthetic principle could be used to repair medial upper lip defect of medium-sized,which not only obtains functional reconstruction,but also satisfies aesthetic results,with aesthetic subunits saved and scar concealed.
