1.Prediction of the fluctuating intervals of intraocular pressure readings in mild and medium myopia after photorefractive keratectomy

ObjectiveTo predict the fluctuating intervals of intraocular pressure readings in mild and medium myopia after photorefractive keratectomy(PRK).MethodsThe intraocular pressure(IOP)readings of 383 eyes,of which spherical equivalent were less than-6.0 diopters(D)before and over 1 a after PRK,were measured with noncontact tonometer(NCT).The IOP data were analyzed with paired-sample t test,linear correlation and regression.Regression equation was used to predict the fluctuating intervals of IOP readings after PRK.ResultsThe IOP readings after PRK(10.38±2.08)mmHg were lower than those before PRK(15.43±2.74)mmHg.The difference of IOP readings between before and 1 a after PRK was significant (P<0.001).The postoperative IOP readings were related to those of preoperation (r=0.625,P<0.001)and the regression equation was Y=0.474X+3.064[X=IOP reading before PRK(mmHg),Y=IOP reading followed over 1 a after PRK(mmHg)].If IOP reading before PRK was X0,the 95% tolerant interval of IOP readings followed over 1 a after PRK was 0.5X0-0.1,0.5X0+6.3(mmHg).ConclusionIOP readings before PRK can be used to predict the fluctuating intervals of IOP readings in mild and medium myopia after PRK.
2.Influence of different fixation targets in accuracy of Jackson cross cylinder examination

Objective To evaluate the influence of black dots,isolated line above the best correct visual acuity and multiple lines containing the best correct visual acuity in the accuracy of Jackson cross cylinder examination.Methods Forty students were chosen(11 males,29 females,between 24 to 29 years old).Routine examination of phoropter was used to check under the maximum plus to maximum visual acuity(MPMVA) status.The sequence of three fixation targets was chosen randomly.Astigmatism chart was used to judge the best one when the different results were obtained.Results Among the 80 eyes of 40 students,12 eyes were spherical(the cylinder degree below 0.25 DC).The response of three fixation targets was the same in 19 eyes(27.9%),23 eyes(33.8%)were sensitive to the black dots target,14 eyes(20.6%)were sensitive to the isolated line above the best correct visual acuity,12 eyes(17.6%) were sensitive to multiple lines containing the best correct visual acuity.Kruska Wallis Test was used to detect the difference among three groups.No significant difference was found(?2 =4.026,P=0.134).Conclusion According to the efficiency of examination,communication and judgment,black dots target is recommended although no difference is detected among three fixation targets.
3.Monotherapy with levetiracetam for absence epilepsy in children

Objective To assess the efficacy and safety of levetiracetam (LEV) monotherapy in children with absence epilepsy.Methods Fifty five children [ 34 males,21 females,aged (8.1 ± 4.8) y ] with typical absence seizures were treated with LEV ( n =25 ) or valproic acid ( VPA,n =30) from Jan 2009 to Jun 2010.Patients were followed up for 1y; the seizure frequency,electroencephalogram (EEG) and side effect were observed at 6th month and 12th month after the initiation of therapy.Results During the follow-up,none had aggravated symptoms of epilepsy.There were no significant differences in seizure frequency(t =0.76,P =0.450; t =0.67,P =0.504),abnormal EEG ( x2 =0.120,P =0.729 ; x2 =0.043,P =0.836 ) and seizure control ( x2 =0.051,P =0.821 ; x2 =0.078,P =0.780) after 6 and 12 months of treatment between two groups.No death or severe side effects were observed; 2 cases (8.0%) in LEV group had mild adverse effect( drowsiness and restlessness)and 2 cases (6.7% ) in VPA group had weight gain and nausea.Conclusion The results suggest that monotherapy with LEV is effective and safe for treatment of childhood absence epilepsy.
4.Effects of anisodamine on apoptosis in cardiomyocytes and inflammatory response in overtrained rats

Objective To evaluate the effects of anisodamine on apoptosis in cardiomyocytes and inflammatory response in overtrained rats. Methods Twenty-four male Wistar rats, weighing 200-220 g, were randomly divided into 3 groups ( n = 8 each) : control group (group C) , overtraining group (group O) , anisodamine group (group A) . The model of overtraining-induced acute heart injury was established by exhausting swimming. Anisodamine 10 mg/kg was given intraperitoneally 20 min before overtraining in group A. Blood samples were taken at 6 h after overtraining for measurement of serum CK-MB activity. The rats were then sacrificed and myocardial tissues taken for determination of TNF-α content and NF-κB activity (by immunohistochemistry) . The apoptosis rate was detected by flow cytometry. Results The CK-MB activity, apoptosis rate, TNF-α content and NF-κB activity were significantly higher at 6 h after overtraining in groups O and A than in group C, while lower at 6 h after overtraining in group A than in group O ( P < 0.05) . Conclusion Anisodamine can inhibit apoptosis in cardiomyocytes by reducing inflammatory response in overtrained rats.
5.High-mobility group box protein1 increases tumor necrosis factorαexpression in human bronchial epithelial cells

Objective To investigate the effect of high-mobility group box protein1 (HMGB1) on the expression of TNF-αand its mechanism in 16HBE in vitro. Methods groups with different HMGB1 (0, 100, 500, 2 000 ng/mL) concentration was set; RAGE antagonizing groups were as control, HMGB1-2000ng, anti-RAGE and anti-RAGE+HMGB1. The changes of TNF-αmRNA and secretion were determined by quantitative PCR and ELISA. RAGE protein level was measured by western blotting. Results HMGB1 intervention and TNF-α expression of 16HBE presented a positive dose-dependent relationship. Thechanges of RAGE was HMGB1positively concentration dependent. In comparison with HMGB1 2 000 ng/mL group, anti-RAGE+HMGB1showed a remarkable reduction of TNF-α secretion. Conclusion In vitro, HMGB1 increases TNF-α expression in 16HBE with a dose-dependent manner through RAGE.
6.Establishment of a guinea pig model for biological diagnostic reagent of tuberculosis

Objective To establish a guinea pig model for diagnostic reagent of tuberculosis.Methods By single or multiple subcutaneous injection of heat-killed H37 Rv in different doses in the groin of guinea pigs to establish a model of positive response to 0.1 mL (5 IU) standard tuberculin ( TB-PPD) skin test.Results Three doses of heat-killed H37 Rv ( 0.2 mg/mL, 0.3 mg/mL and 0.5 mg/mL ) could be used to generate the model of biological diagnosis of tuberculosis.After 24 and 48 hours, the diameter of red spot by TB-PPD skin test was 15.4 ±2.3 mm when a dose of 0.2 mg/mL heat-killed H37 Rv was administered for immunizing and allergizing the guinea pigs.The biggest red spot was induced at doses of 0.3 mg/mL and 0.5 mg/mL.The test results showed that the immune response induced by multiple njection to immunizing and allergizing guinea pigs was not significantly different than that induced by single immunizing injection, and the first skin test was better than the second, third and fourth skin test (P≤0.05).In addition, the body weight of the guinea pigs was still increasing after infection with heat-killed H37 Rv, and ulcers occurred in the injection sites in some guinea pigs.Conclusions A single subcutaneous injection of 0.2 mg/mL heat-killed H37 Rv in guinea pigs can be used well to establish a reliable model for biological diagnostic reagent of tuberculosis.Increasing the sensitizing dose and multiple sensitization can not increase the intensity of the delayed-type hypersensitivity ( DTH) response.
7.Anti-metastasis effect of thymoquinone on human pancreatic cancer.

Recent studies reported that thymoquinone (TQ), a component derived from the medicinal spice Nigella sativa (also called black cumin), exhibited inhibitory effects on cell proliferation of many cancer cell lines. This study was performed to investigate the anti-metastatic effect of thymoquinone on the pancreatic cancer in vitro and in vivo. The results showed that thymoquinone suppressed the migration and invasion of Panc-1 cells in a does-dependent manner. To investigate the possible mechanisms involved in these events, Western blotting analysis was performed, and found that thymoquinone significantly down-regulates NF-kappaB and MMP-9 in Panc-1 cells. In addition, metastatic model simulating human pancreatic cancer was established by orthotropic implantation of histologically intact pancreatic tumor tissue into the pancreatic wall of nude mice. And administration of thymoquinone significantly reduced tumor metastasis compared to untreated control. Furthermore, the expression of NF-kappaB and MMP-9 in tumor tissues was also suppressed after treatment with thymoquinone. Taken together, the results indicate that thymoquinone exerts anti-metastatic activity on pancreatic cancer both in vitro and in vivo, which may be related to down-regulation of NF-kappaB and its regulated molecules such as MMP-9 protein. Consequently, these results provide important insights into thymoquinone as an antimetastatic agent for the treatment of human pancreatic cancer.
8.Sentinel lymph node detection of rabbit tongue by indirect CT lymphography combined with nanocarbon tracer.

OBJECTIVE:
To investigate the value of indirect CT lymphography (CT-LG) combined with nanocarbon tracer suspension staining in the sentinel lymph node (SLN) detection of rabbit tongue (cancer).
METHOD:
Eight rabbits were divided into two groups, i. e. normal control group (6 rabbits) and VX2 tongue carcinoma group (2 rabbits). Pre-contrast CT images were taken for all rabbits. The mixture of iohexol and nanocarbon tracer suspension (iohexol 0.25 ml and nanocarbon tracer 0.25 ml, respectively, volume ratio = 1:1) was used as contrast agent of indirect CT-LG. Indirect CT-LG was performed as the contrast agent was injection into right ventrolateral submucosa of the tongues. CT images were acquired at 1, 5, 10 and 15 min after injection.
RESULT:
There were no cervical lymph nodes clearly identified in any group on the pre-contrast CT images. After the contrast agent injection, indirect CT-LG showed an ipsilateral SLN with rapid enhancement which was the only enhanced SLN, located lateral to the larynx-trachea region in all rabbits of VX2 control group and carcinoma group. The CT attenuation values of SLN were (58.80 ± 5.45) HU, (134.80 ± 28.92) HU, (91.00 ± 13.08) HU and (63.80 ± 4.32) HU at 1, 5, 10 and 15 minutes after indirect CT-LG. After the cervical anatomy guided by CT scan, all of the right deep SLN showed a distinct enrichment of black dye highly consistent with the locations on the indirect CT-LG images.
CONCLUSION
It is more accurate for sentinel lymph node detection by indirect CT lymphography combined with nanocarbon tracer staining.

9.Investigation of Hand Germ-Carrying status of Healthcare Workers in Burn Wards

OBJECTIVE To investigate hand hygiene of healthcare workers in burn ward in our hospital and provide scientific basis for the effective prevention and control of hospital infection in burn ward.METHODS Selected randomly 30 healthcare workers in burn ward and sampled from hands for bacterial culture.RESULTS The average colony of hand pathogens of the 30 healthcare workers was 105.42 CFU/cm2,the maximam was 1093 CFU/cm2.Among the hand pathogens,40.91% were Bacillus subtilis,36.36% were micrococcus and 13.64% were Staphylococcus aureus.CONCLUSIONS Hand hygiene of healthcare workers are not optimistic in burn ward.Strengthening the education of hand hygiene and raising the executable rate of hand hygiene are important measures to prevent and control the hospital infection in burn ward.
10.Effects of intracranial co-infusion of Aβ_(1-42) and thiorphan on Macaca Rhesus hippocampal formation

Aim To observe the change of amyloid, acetylcholine transferase and glial fibrillary acidic protein expression in Macaca Rhesus hippocampal after infused the Aβ_(1-42) and thiorphan and explore the possibility of the establishment of Macaca Rhsus AD model in brain.Method The Rhesus monkeys were anesthetized (im), the skull was exposed by a midline scalp incision, and oriented craniotomy was performed on left side by dental drill.First, neprilysin in cerebral cortex and basal nucleus was consumed by infusion thiorphan. Then cerebral cortex and basal nucleus were slowly infused with fibrilla Aβ_(1-42). Finally, the cannula for thiorphan infusion was implanted into the basal nucleus.Miniosmotic pump (Alzet MODEL 2ML4,) was subcutaneously fixed by bio gel 454 on the calvaria (Loctite Co. Ltd,USA) according to the manufacturer's instructions.After 50 days' survival, animals were deep anesthetized with ketamine and sacrificed. The pathological changes were observed by HE staining and immunostaining in monkey brains.Result Neuronal loss and a proliferation of microglia were detected in hippocampal formation by HE staining.Immuno-staining showed Aβ_(1-42),ChAT and GFAP positive cells density were 0.59±0.05,0.21±0.04 and 0.19±0.04 separately.Compared with control group, the density in experimental groups showed distinct difference in statistic analysis (P<0.01).Conclusion The same pathological change was detected in the thioaphan and Aβ_(1-42) infusion in Macaca Rhesus hippocampal formation as what was found in AD patients.
