1.In vitro morphological observation on the effect of exogenous collagen on the construction of tissue-engineered tendon

BACKGROUND: Collagen type Ⅰ possesses low antigenicity and provides special functions of scaffold structure for cell migration, proliferation and secretion.OBJECTIVE: To observe the morphological change after combination of transformed human embryonic tendon cells and artificial materials with collagen type Ⅰ, so as to provide the experimental basis for constructing tissue-engineered muscle tendon in vitro.DESIGN: Randomized and controlled observation.SETTING: Department of Orthopaedics, West China Hospital, Sichuan University.MATERIALS: This experiment was carried out at the Department of Transplantation Immunology, West China University of Medical Sciences from July to September 1998. Human hair, carbon fiber and polyglycolic acid were combined with collagen and then set corresponding groups respectively. Simply hair, carbon fiber and polyglycolic acid control groups were set respectively. Artificial materials: Healthy adult hair (30 cm in length, 80 g in weight), carbon fiber knitted strip (Lanzhou carbon factory )and polyglyolic acid fasciculi (Johnson , American) were used in this experiment.METHODS: ①In the group of human hair combined with collagen,nine healthy adult hairs were chosen . Three hairs as one bundle were knitted into tendon with 0.8 cm in length and 0.4 cm in diameter and a knot was tied respectively at the two ends. ②In the carbon fiber combined with collagen group, the purchased carbon fiber knitted strip was separated and made into artificial materials with the same diameter and length as those of human hair tendon. Two ends of the artificial materials were packed with 5/0 silk thread. ③In the polyglycolic acid combined with collagen group, three polyglycolic acid fasciculi were weaved into bundle with the same diameter and length as those of human tendon,and two ends were packed with 5/0 silk thread. ④The process of knitting scaffold materials was the same in control groups as in the corresponding experimental groups. ⑤ Five pieces were prepared in each scaffold material. After sterilized by 60Co, the scaffold materials were put in collagen type Ⅰ solution for 0.5 hour, then taken out and dried at roomtemperature for use in the experimental groups. ⑥transformed human embryonic tendon cell, density for 3×106/mm3, was used for combined culture in each group, and cell number and morphology were observed at different time points after culture under an inverted microscope and scanning electron microscope.MAIN OUTCOME MEASURES: ① Observation of cell growth and adherence under an inverted microscope ② Observation of cell morphology and material adherence under scanning electron microscope RESULTS: ① Observation of cell growth and adherenceunder an inverted microscope :In each experimental group, at two hours after culture, cells gathered around the materials, and some cells adhered to materials presented round; At three days after culture, the number of cells adhered to materials began to increase, morphology changed from round to oval, and the co-existence of round and oval cells was seen;While in each control group, at two and three hours after culture, few cells adhered to the three materials. On day five after culture, in each experimental group, cell morphology turned into fusiform, the number of cells increased further and a great many fusiform cells grew among carbon fiber materials; while in the control group, most cells presented round and ellipse, and the number of cells was less as compared with the corresponding experimental group. ② Cell morphology and material adherence under scanning electron microscope: On day 5 after in vitro culture, most tendon cells on the surface of materials presented spindle-shape in each experimental group. Apophysis stretched out from the tendon cells, grew along longitudinal axon of the materials. Silk and piece-like substance appeared among the apophysis and weaved each other. While there were fewer cells adhered in control group than in corresponding experimental group, cells also presented fusiform or round, but no silk or piece-like substance was found, also no apophysis appeared.CONCLUSION: Human hair, carbon fiber and polyglycolic acid combined with collagen type Ⅰ will facilitate cell adherence. Cells increase around the materials and co-existence of round and oval cells could be seen, and cells turned into fusiform, suggesting that exogenous collagen facilitate the cells to adhere onto the materials and proliferate.
2.Localization of sentinel lymph node with indirect lymphography and the application in head and neck tumor.

Whether the sentinel lymph node (SLN) metastasis has important clinical significance for the therapy and prognosis of tumor. Sentinel lymph node biopsy (SLNB) has became the most accurate clinical method to confirm the status of sentinel lymph node. And the key of the success of SLNB is the localization of SLN. The methods used to locate SLN mainly are blue dye tracer method, radionuclide tracer technique, blue dye tracer method combined with radionuclide tracer technique, indirect lymphography, NIR imaging, and photoacoustic imaging. This article demonstrates the principle, application method and contrast agents of the indirect lymphography and the application in head and neck tumor.

3.Optimization of manganese peroxidase production from Schizophyllum sp. F17 in solid-state fermentation of agro-industrial residues.

Manganese peroxidase (MnP), a crucial enzyme in lignin degradation, has wide potential applications in environmental protection. However, large-scale industrial application of this enzyme is limited due to several factors primarily related to cost and availability. Special attention has been paid to the production of MnP from inexpensive sources, such as lignocellulosic residues, using solid-state fermentation (SSF) systems. In the present study, a suitable SSF medium for the production of MnP by Schizophyllum sp. F17 from agro-industrial residues has been optimized. The mixed solid medium, comprising pine sawdust, rice straw, and soybean powder at a ratio of 0.52:0.15:0.33, conferred a maximum enzyme activity of 11.18 U/g on the sixth day of SSF. The results show that the use of wastes such as pine sawdust and rice straw makes the enzyme production more economical as well as helps solve environmental problems.

4.Application of DWI with various b values in diagnosis of clear cell renal cell carcinoma Fuhrman grades SHEN

Background and purpose:Renal cell carcinoma is the most common type of adult renal cancer, and the Fuhrman grading system is the most widely accepted independent indicator for the prognosis of kidney tumors. This study aimed to explore the correlation between the conventional diffusion weighted imaging (DWI) with various b values and Fuhrman grade of clear cell renal cancer, and assess the diagnostic efifciency of ADC values at different b values in differentiating Fuhrman low- and high-grade tumor. Methods:Thirty-three patients with pathologically proved clear cell renal cell cancer (CCRCC) and qualiifed images were included for the research. The diagnostic efif-ciency of ADC values at different b values for differentiating Fuhrman low-(1 and 2) and high-grade (3 and 4) tumor were also assessed and compared by receiver operator characteristic curve (ROC), and the optimum sensitivity, speciifc-ity and accuracy were selected using Youden index. Results:The 33 patients consist one tumor of Fuhrman grade 1, 14 of grade 2, 16 of grade 3 and 2 of grade 4. A negative correlation was found between the ADC0-800, ADC0-400-800, ADC0-600-1 200, ADC0-400-800-1 200 and ADCtotal with Fuhrman grade (r:-0.553,-0.511,-0.603,-0.645,-0.610, respectively), among these the ADC0-400-800-1200 showed the strongest correlation. The AUCs [area under the (receiver operator characteristic curve) ROC curve] of those parameters for differentiating the low-and high-grade tumor were 0.789, 0.757, 0.813, 0.844 and 0.835, among which the ADC0-400-800-1 200 reveal the best result, but the difference was not statistically signif-icant (P>0.05). The sensitivity of the ADC values were 86.7%, 73.3%, 60.0%, 86.7%and 86.7%;The speciifcity were 66.7%, 77.8, 72.2%, 77.8%and 72.2%. Conclusion:ADC0-800, ADC0-400-800, ADC0-600-1 200, ADC0-400-800-1 200 and ADCtotal re-vealed signiifcant correlation with the Fuhrman grade of clear cell renal cancer, and providing high diagnostic efifciency in differentiating Fuhrman low-and high-grade tumor.
5.Effect of SIRT6/NF-kappa B signal axis during hematopoietic stem and progenitor cell senescence induced by tert-butylhydroperoxide in vitro

BACKGROUND:SIRT6/NF-κB is the important signal axis to cellsenescence, but the effect of SIRT6/NF-κB signal axis to hematopoietic stem and progenitor cell(HSC/HPC) senescence is unclear. OBJECTIVE:To induce (t-BHP) in vitro and to investigate the role of SIRT6/NF-κB signal axis in Sca-1+HSC/HPC senescence induced by tert-butylhydroperoxide in vitro. METHODS:Sca-1+HSC/HPC was isolated and purified by magnetic activated cellsorting. Sca-1+HSC/HPC senescence was induced by 100μmol/L tert-butylhydroperoxide in vitro. The senescence-associatedβ-Galactosidase staining, cellcycle analysis and culture of mixed hematopoietic progenitor cellwere used to investigate the biological effects of tert-butylhydroperoxide on Sca-1+HSC/HPC senescence. The expression of senescence associated SIRT6, NF-κB mRNA and protein was examined by real-time fluorescence quantitative PCR and western blot assay. RESULTS AND CONCLUSION:Compared with control group, the percentage of positive cells expressing SA-β-Gal and cells in G0/G1 phase increased and the number of forming colony of mixed hematopoietic progenitor decreased in the aging group. It showed lower expression of SIRT6 and higher expression of NF-κB in the aging group. The SIRT6/NF-κB signal axis may play a key role in the Sca-1+ HSC /HPC senescence inducted by tert-butylhydroperoxide.
6.Adult stem cells and possible mechanisms of its differentiation--editorial.

Adult stem cells are the multi-potential cells, which exist in fetal and adult tissues. It can reproduce itself (undergo self-renewal) or give rise to more specialized (differentiated) cells. Under certain inducing conditions, adult stem cells can acquire the ability to differentiate into different tissue cells. Multipotent adult progenitor cells (MAPC), an alternative name of adult stem cell given by Catherine Verfaillie, existing in bone marrow, can differentiate into cells with characteristics of mesodermal, neuroectodermal, and endodermal lineages in vitro at the single-cell level. MAPC can also contribute to most cell types when injected into the blastocyst. Adult stem cell differentiation implies that different cell lineages are derived from a single initial cell; all differentiated cell types are functional in vitro and in vivo; and engraftment is robust and persistent in the physiological and pathological situations. The possible mechanisms may underlie the differentiation: various tissue-specific stem cells are present in different organs; adult stem cells would be reprogrammed when removed from their usual microenvironment and introduced into a different niche that imparts signals to activate a novel genetic program needed for the new cell fate. And true multi-potential stem cells persist in postnatal life. In the future, multi-potent adult stem cells might then be used for therapies of degenerative or genetic disorders of multiple different organs.

7.Investigation about the postoperative rehabilitation and the health knowledge of patients with arteriosclerosis obliterans in lower extremity

Objective To investigate the condition of exercise, medicine compliance and the rehabilitation of patients with arteriosclerosis obliterans (ASO) in lower extremity after the surgery, and also to study the influence of self-efficiency and social support to their rehabilitation. Methods The questionnaire survey was performed in 102 patients who had surgical or interventional therapy of arteriosclerosis obliterans in the lower extremity in our hospital, investigated the general information of the patients, their knowledge of postoperative rehabilitation, medicine compliance, exercise condition, general self-efficiency and social support. Results The postoperative time for the patients with arteriosclerosis obliterans was 2 to 22months, the knowledge rate for postoperative rehabilitation was 7.8%-46.1%.The Morisky score for the patients of taking medicine for the treatment of vascular disease was 2.0,and there was only 6 patients(5.9%)with good medicine compliance. There were 22 patients(21.0%) had completed the ambulation training according to the guidelines, and there were 25 patients(24.5%) had completed the Buerger training. The self-efficiency score of the patients was 2.78 ± 0.76, and the gross score of social support was 32.38 ± 7.89. Spearman correlation analysis shows that the Morisky score for the patients of taking medicine for the treatment of vascular disease was in positive correlation with self-efficiency (r=0.289, P=0.003), but in negative correlation with social support (r=–0.286, P=0.006). Conclusions Most of the patients with arteriosclerosis obliterans have poor medicine compliance, and do not do exercise as required by the guidelines. Patients with high self-efficiency demonstrates medicine noncompliance, however the enhancement of social support will help patients improve their medicine compliance.

Objective To investigate the effects of fluvastatin (Flu) on the levels of estradiol(E2 ) ,tumor necrosis factor-α(TNF-α) ,insulin-like growth factor-1(IGF-1) and bone glaprotein(BGP) in experimental osteoporosis rats .Methods The model of osteo-porosis was established ,all rats were ovariectomized .The rats were given Flu(20 ,10 ,5 mg/kg) through intragastric administration (ig) .After treating for 12 weeks ,weighed all rats ,then sacrificed all rats ,removed the uterus and weighed its wet weight ,and calcu-lated uterus index ;the levels of E2 ,TNF-α,IGF-1 and BGP in serum were detected .Results Flu did not affect the uterus weight in experimental osteoporosis rats ;20 mg/kg Flu could increased the osteoporosis rat′s serum levels of E2 and IGF-1(P<0 .05) ,de-creased the levels of TNF-αand BGP(P<0 .01);10 mg/kg Flu could decreased the osteoporosis rat′s serum levels of BGP .Conclu-sion Flu could increased the levels of E2 ,IGF-1 ,and decreased the levels of TNF-α,BGP in experimental osteoporosis rats .
10.Evaluation of the effects caused by different doses of N-nitrosodiethylamine on establishing multistep carcinogenesis model of cirrhotic liver in rats

ObjectiveTo evaluate the effects caused by different doses of N-nitrosodiethylamine (DENA) on establishment of a multistep carcinogenesis model of liver cirrhosis in SD rats.MethodsFifty male SD rats were randomly divided into control group (n =5 ) and experimental group (n =45 ).Experimental group consisted of 3subgroups(high,median and low dosegroup ),which were intragastrically induced DENA once a week for 10 weeks,by using 70,50,35 mg/kg DENA solution,respectively.MRI was performed for inspecting pathological changes of rat livers in the following weeks.The mortality rates of different groups were calculated.The differences of the incidence of dysplastic nodule (DN)found in three groups were analyzed with Pearson Chi-square test.ResultsThe multistep carcinogenesis development from regenerative nodule (RN) to DN to hepatocellular carcinoma (HCC) was showed in the SD rat model.Two rats in median dose group and 4 rats in high dose group died.The incidence of DNs found in 3 different groups was 20.7% ( 6/29 ),30.6% ( 19/62 ),14.3% (9/63),respectively,which showed no significant difference among three groups(x2 =4.901,P > 0.05).However,significant difference existed between median and high dose group( x2 =4.811,P < 0.05 ).Combined with the MRI presentation,the intragastric dose of 50 mg/kg could be more appropriate among three different doses.ConclusionsThe median dose DENA could induce an ideal multistep carcinogenesis model of HCC which is suitable for investigating DN on MRI.
