1.The in vitro dissolution of total composition of the tablet of rhizomes of Ligusticum chuanxiong components and in vitro-in vivo correlation by the method of area under the absorbance-wavelength curve.

To discuss the availability of evaluation on the dissolution studies of the multicomponents in traditional Chinese medicine, the in vitro dissolution of total composition of the tablet of rhizomes of Ligusticum chuanxiong components and its correlation with the in vivo were studied by the method of area under the absorbance-wavelength curve (AUAWC). Taken the tablet of rhizomes of Ligusticum chuanxiong components which is composed of sodium ferulate and ligustrazine hydrochloride as subject model, the dissolution tests were carried out with basket method. The plasma concentrations of tablets in different rats were determined by AUAWC at different interval times. The in vivo absorption percentage was calculated by Wagner-Nelson equation to evaluate the in vitro and in vivo correlation. According to the results, the cumulative dissolution in vitro of total composition of tablets of rhizomes of Ligusticum chuanxiong components at 60 min was 90.65% in water by AUAWC. The in vivo pharmacokinetics is fitted with an one-compartment model. The linear equation based on the cumulative dissolution rate (fr) and absorption percentage (fa) at 5, 10, 20, 30 and 60 min was fa = 0.819 7 fr+0.183 and the correlation coefficient was 0.959 5, which showed a good correlation between the in vitro dissolution and the in vivo absorption percentage. The method of AUAWC can be used accurately, feasibly and conveniently to evaluate the in vitro and in vivo correlation of total composition of tablets of rhizomes of Ligusticum chuanxiong components, which will provide better guidance to study the in vitro and in vivo correlation of sustained release preparation etc under complex system of traditional Chinese medicine in the future.
2.Effect of Acupoint External Application of Herbal Drugs on Pathological Changes of Nasal Mucosa of Allergic Rhinitis Mice

【Objective】To observe the effect of acupoint external application of herbal drugs on pathological changes of nasal mucosa of allergic rhinitis mice. 【Methods】 Sixty BALB/c mice were divided into 5 groups: A(normal),B(model),C(treated with external application of herbal drugs on acupoints),D(treated with dexamethasone) and E(treated with PBS).Except group A,the mice in other groups were sensitized by ovalbumin(OVA).On the 15~(th) of the first sensitization,group C received external application of medicinal disc(mainly composed of Semen Sinapis, Rhizoma Corydalis,Herba Asari,Radix Kansui,Fructus Xanthii,etc.) on Dazhui (GV14)point,group D received intraperitoneal injection of 0.1?g/L dexamethasone(0.5?mL for each mouse) one hour before nasal dripping of OVA solution,and group E received external application of paper disc on Dazhui point and intraperitoneal injection of(0.1?g/L) PBS(0.5 mL for each mouse) one hour before nasal dripping of OVA solution,qd,for 4 weeks.After treatment,the mice were executed and the nasal mucosa was fixed in 4% paraformaldehyde.Routine paraffin slices were prepared and stained by haematoxylin and eosin.The pathological features of nasal mucosa were observed under light microscope,and eosinophil(EOS),neutrophil and lymphocyte count was performed in different groups.【Results】Compared to group A,the infiltration of EOS and neutrophil in nasal mucosa was obvious in group B(P0.05).【Conclusion】The infiltration of EOS in nasal mucosa plays an important role in the pathogenesis of allergic rhinitis and the decrease of EOS infiltration may be one of the therapeutic mechanism of external application of herbal drugs on acupoints for allergic rhinitis.
3.Determination and Analysis of Cordyceps Militaris(Fr) Link's Amylose Content

Objective: To compare amylose content of five varieties of Cordyceps Militaris sample to supply a basis for devoloping and urilizing amylose of Cordyceps Militaris. Methods: Phenol sulfuric acid procedure was used.Results: There is great difference in amylose in the case of cross comparision but not obvious in sequence reference.. Conclusion: Amylose chiefly exists in mycelium.
4.The Community Succession of Sarcosaphagous Insects on Pig Carcasses in Summer Indoor and Outdoor Environment in Shenzhen Area

Objective To explore the growing development and community succession of main sarcos-aphagous insects on pig carcasses in summer indoor and outdoor environment in Shenzhen area and to estimate the postmortem interval (PMI). Methods From early May to August in 2013, in Forensic Med-ical Examination Center of Shenzhen Public Security Bureau, the main insect species and the decomposi-tion process were observed in two adult pig carcasses of simulative indoor and outdoor environment. The different decomposition stages and the community succession of insects were recorded. Results The indoor and outdoor pig carcasses showed skeleton 412.5 and 325 hours after death, respectively. The main species of flies on pig carcasses were Chrysomya megacephala, Chrysomya rufifacies and Chrysomya chani. The main species of beetles were Crecphilus maxillosus, Necrobia ruficollis, Saprinus splendens and Dermestes maculatu. The dominant species of flies in the outdoor pig carcasses obviously produced the second generations due to the effect of mass rainfall, nor in the indoor pig carcasses. Conclusion There are regular patterns on the community succession of insects on pig carcasses in summer indoor and out-door environment in Shenzhen area. The activity patterns of seven typical insects and their larva show important value for estimating PMI.
5.GROWTH INHIBITION OF LUNG CANCER CELL LINE in vitro BY TELOMERASE ANTI-SENSE OLIGONUCLEATIDES

Objective To study the inhibition of lung cancer cell line by telomerase anti\|sense DNA,and discuss the possibility of using it in clinical treatment. Methods A phosphorothioate oligonucleotide(PS\|ODN) with sequence identical to the repeat sequence of the mammalian telomere 5′\|d(TTAGGG)\|3′ and a control scrambled sequence 5′\|d(TGTGAG)\|3′ were incubated with a lung cancer cell line.The effects of PS\|ODN on cell line growth,colony\|forming and growth shape were detected.The in vivo efficacy of this PS\|ODN was evaluated in a 801\|D nude mouse model.Once tumors were established these animals were administered PS\|ODN or saline for 15 days. Results Telomease anti\|sense DNA inhibit telomerase activation of cell line 801\|D growth and colony\|forming.The activity of the 6\|mer telomere mimic demonstrated a dose dependency.No activity was observed with the scrambled controls.A significant decrease in tumor weight was observed in animals given PS\|ODN, but not followig saline\|treated animale.Conclusion\ These results demonstrated that short hexameric oligonucleotide telomere exerts the growth inhibitory effect on lung cancer cell in vitro and in vivo, and suggest the potential utility of telomerase anti\|sense DNA as cancer cell inhibitors.\;[
6.The extraction of sperm DNA from mixed stain using DNase-Ⅰ purification combined with alkaline lysis method

Objective To establish a method of sperm DNA extraction in mixed stain by using DNase-Ⅰ purificationcombined with alkaline lysis method in forensic science.Methods 79 mixed stain samples of criminal cases were collected.Sperm DNA was extracted using the purification of DNase-Ⅰ binding alkaline lysis method.16 STR loci were genotyped with fluorescent multiplex amplification system.The typing results were compared with that of extracted using two-step differential extraction procedure.Results Of all 79 mixed stain samples,64 samples were genotyped successfully by using DNase-Ⅰ purification combined with alkaline lysis method while 57 samples were genotyped successfully with two-step differential extraction procedure.There was significant difference between two methods(P=0.039).The purification of DNase-Ⅰ binding alkaline lysis method had a higher success rate and lower cost than that of two-step differential extraction procedure.Conclusion Purification of DNase-Ⅰ binding alkaline lysis method can increase the typing success rate of the mixed stain samples.The method is simple,rapid and easy to be automated,and suitable for forensic identification test.

9.Expression of cathepsin B in acutely photodamaged fibroblasts and its significance

Objective To investigate the changes in cathepsin B (CatB) expression in acutely photodamaged human dermal fibroblasts (HDFs) and their significance.Methods HDFs were isolated from the foreskin of children,and subjected to primary culture and subculture.The fourth-to eighth-passage HDFs were used in the following experiment.HDFs were divided into two groups to receive irradiation with different doses of ultraviolet A (UVA) for different durations (acutely photodamaged group) or remain unirradiated (control group).Cell counting kit-8 (CCK8) assay was conducted to evaluate the proliferative activity of HDFs after irradiation with UVA at 5,10,15,20 and 25 J/cm2 respectively.Western blot and quantitative real-time reverse transcription PCR were performed to measure the protein and mRNA expressions of CatB respectively in HDFs at 24,48 and 72 hours after exposure to UVA at 10 J/cm2,and at 48 hours after exposure to UVA at 10,15,20 and 25 J/cm2.Statistical analysis was carried out by analysis of variance and least significant difference (LSD) test using the SPSS 13.0 software.Results UVA radiation induced a decrease in the proliferative activity of HDFs.When the dose of UVA was ≤ 10 J/cm2,the survival rate of HDFs maintained higher than 85%,and significant differences were observed in cell survival rate between unirradiated and irradiated HDFs at 24,48 and 72 hours (all P < 0.05).Western blot showed that the gray value of CatB protein in the acutely photodamaged group irradiated with 10 J/cm2 UVA was significantly higher than that in the control group at 24 hours (0.76 ± 0.14 vs.0.35 ± 0.01,P < 0.05),48 hours (1.34 ± 0.38 vs.0.45 ± 0.12,P< 0.05) and 72 hours (0.82 ± 0.09 vs.0.61 ± 0.06,P< 0.05).Increased mRNA expressions of CatB were also observed in the acutely photodamaged group compared with the control group at 24 hours (0.149 ± 0.009 vs.0.089 ± 0.015,P < 0.05),48 hous (0.173 ± 0.009 vs.0.091 ± 0.010,P < 0.05) and 72 hours (0.185 ± 0.158 vs.0.111 ± 0.017,P < 0.05) after UVA radiation at 10 J/cm2.The gray value of CatB protein was 0.99 ± 0.07,1.49 ± 0.14,1.89 ± 0.08,2.07 ± 0.06 in HDFs at 48 hours after exposure to UVA of 10,15,20 and 25 J/cm2,respectively,significantly higher than that in the control group (0.60 ± 0.05,all P < 0.05).Similarly,the mRNA expression of CatB was up-regulated in HDFs at 48 hours after UVA radiation at 10,15,20 and 25 J/cm2 compared with the unirradiated HDFs.Conclusion The protein and mRNA expressions of CatB are up-regulated in acutely photodamaged HDFs induced by UVA radiation.
10.Clinical features and risk factors of invasive fungal infections in children with acute leukemia

Objective To analyze the clinical features and risk factors of invasive fungal infections (IFI) in children with acute leukemia.Methods Ninety-six acute leukemia children complicated with IFI admitted in Guangzhou Women and Children's Medical Center during January 2005 and February 2017 were retrospectively reviewed, and 96 cases of acute leukemia without IFI admitted at the same period were randomly selected as control group.The clinical manifestations of IFI were analyzed, multivariate Logistic regression was used to study risk factors of the complication of IFI in pediatric acute leukemia.Results Among 96 children complicated with IFI, fungus were detected in samples from sputum, bronchoalveolar lavage fluid, or blood in 78 cases, in which 42 cases (43.75%) were oral infection, 36 cases (37.50%) were pulmonary infection.Candida albicans (33.33%, 26/78) was the most commonly isolated pathogen, followed by Candida parapsilosis (20.51%, 16/78) and Candida tropicalis (20.51%, 16/78).Univariate analysis revealed hormone-containing chemotherapy, neutropenia (< 0.5 × 109/L), time duration of neutropenia ≥ 10 days, usage of carbapenem antibiotics and combined drug administration ≥2 types were associated with fungal infection (P < 0.05 or <0.01).Multivariate Logistic regression showed that the time duration of neutropenia ≥ 10 days (OR =11.390, 95% CI 4.145-55.263, P < 0.01),usage of carbapenem antibiotics (OR =4.825, 95% CI 1.681-13.842, P < 0.01) and hormone-containing chemotherapy (OR =2.220, 95% CI 1.542-8.246, P < 0.05) were the independent risk factors of IFI.Conclusion Rational usage of antibiotics and effective measures taken to restore the granulocytes can help to reduce the incidence of IFI in children with acute leukemia.
