1.STUDIES ON THE CHEMICAL COMPOSITION AND AN-TITUMOR ACTIVITY OF THE GLYCOPROTEIN FROM CHLAMYS (AZUMAPECTEN)FARRERI

Glycoprotein of Chlamys ( Azumapecten ) farreril ( GCFI) was prepared from the skirt of scallop Chlamys (Azumapecten)farreri by extraction with hot water,isolation and purification with column chromatography "on DEAE-cellulose and Sephadex G-100 successively. GCFI was hydrolyzes with enzymes to obtain glycoprotein of Chlamys (Azumapecten)farreri IKGCFID-The chemical composition and antitumor activity of GCFI and GCFII were studied. The result showed that GCFI and GCFII were two glycoproteins in which the composition and content of protein and sugar were different. They could obviously inhibit Sarcoma 180 cells proliferation of mice with the inhibition rate of 47. 29% and 46. 97%,respectively. Pharmacological test indicated that antitumor activity of GCFI and GCFII were associated with the structure of sugar in the molecules.
2.STUDIES ON THE PHARMACODYNAMICS OF ALGAE PREPARATION

The anti tumor and immune activity of Algae Compound Preparation were studied in this paper. The results showed that Algae Compound Preparation could inhibit the growth of S 180 tumor planted to mice with the inhibition rate of 37.98%~50.38%. The life of ascites tumor mice were also prolonged with the prolongation rate of 21%~51.33%. At the same time the activity of apleen cells and the phagocytosis activity of macrophage were also enhanced.
3.EFFECTS OF GLYCOPROTEIN FROM CHLAMYS (AZUMAPECTEN) FARRERI ON TUMOR GROWTH AND IMMUNE FUNCTION IN MICE

Objective: To investigate the antitumor activity and immune enhancing effect of glycoprotein from Chlamys (Azumapecten) farreri (FGP). Methods: S 180 bearing mice were used as animal model. The effects of FGP on tumor weight and immune function were observed in S 180 bearing mice. Results: The results showed that FGP could markedly decrease tumor weight and promote the phagocytic rate of macrophages, NK activity and increase weight of immune organs. Conclusion: FGP can inhibit the growth of S 180 tumor and enhance cell mediated immune function. The antitumor effect of FGP may be related to its immune enhancing activity.
4.Isolation of antarctic microorganisms and screening of antitumor activity

There were some samples collected from the Antarctic soil and South Ocean water for isolation of microorganisms and screening of their antitumor bioactivity by lethality bioassay using brine shrimp and a flow cytometric bioassay.There were 259 stains were isolated from the samples,11% of the Antarctic microorganisms showed strong antitumor activity.This result showed that the Antarctic microorganisms have a good potential in bioactive metabolites researching.
5.Study on the isolation of marine fungi , screening of their antitumor activity and fermentation conditions

Objective To isolate fungi from sea sediment and sea water samples collected from the neritic environment in Qingdao, screen their antitumor activity and study on their fermentation condition. Methods The antitumor activity was assayed by flow cytometry in mouse tsFT210 cells. Results The medium added penicillin and streptomycin possesses good selectivity. 207 strains of fungi were isolated and among them, 19 strains showed antitumor activity, and one fungus(Z 83200)pocesses strong cell apoptosis inducing activity.
6.Familial pedigree analysis of Shen-yin deficiency syndrome in families with type 2 diabetes mellitus GU.

To analyse the familial aggregation and genetic predisposition of Shen-yin deficiency syndrome (SYDS) in families with diabetes mellitus type 2 (DM2). Methods One hundred and forty-one DM2 patients were collected from 32 family lines in Nanjin area, in which the probands were differentiated as DM2 with SYDS. On them, genetic analysis on the characteristics of SYDS was conducted using pedigree analysis, morbidity and heritability of the first-degree relatives of the probands were calculated, and the action of familial SYDS factor on the genesis of the syndrome was assessed by multiple factors regression analysis. Results The morbidity rate of SYDS in the first-degree relatives of the probands was 33.71%, and the heritability, calculated by Falconer formula, was 80.6%. The fitting result of regression analysis showed that familial factor played an important role in SYDS genesis (OR = 5.61, P = 0.001), but DM2 itself is not an independent risk factor for it. Conclusion DM2 with SYDS shows the tendency of familial aggregation and genetic predisposition, genetic factor is associated with the genesis of the syndrome. Pedigree research is a good method for exploring the relationship between syndrome and genetic factor.

7.Effect of Methotrexate on Cell Growth of Human Monocyte-like Cell Line U937 and Its Induction of Apoptosis

Acute myeloid leukemia (AML) is considered to be a malignancy that is intrinsically resistant to methotrexate (MTX). As compared to acute lymphoblastic leukemia (ALL) blasts, AML blasts, except those of acute monocytic leukemia (AML-M(5)), form fewer amounts of long chain polyglutamate of MTX (MTXPG), when incubated with MTX, thus providing an explanation for their lack of responsiveness to MTX. To explore the novel approach of treatment in patients with AML-M(5), the U937 cell line, which has the monocytic characters, was used. Cell growth inhibition was mearsured by XTT assay after 24 and 48 hours in the continuous presence of various concentrations of MTX ranging from 1 nmol/L to 100 micro mol/L. After 24 hours MTX treatment, the IC(50) value for U937 cells was 0.04 micro mol/L. After 48 hours treatment, the IC(50) was 0.037 micro mol/L and IC(90) was 0.39 micro mol/L. To understand the mechanism of MTX cytotoxicity, the process of cell death was analyzed. A variety of assays, including trypan blue exclusion, flow cytometry, light microscopy (Wright's staining) and DNA fragment electrophoresis, were performed. There were no significant apoptotie changes after shorter exposure of MTX (4 and 6 hours). After 8 hours at various concentrations of MTX treatment ranging from 5 nmol/L to 10 micro mol/L, the percentage of the cells in the pre-G(1) (apoptotic) was 3.2% at 0.1 micro mol/L and it reached a peak of 18.2% at 5.0 micro mol/L. The DNA synthesis in S-phase was inhibited from 41.2% (0.01 micro mol/L) to 19.1% (10 micro mol/L). DNA ladder band, a feature of apoptosis, was observed. The arrest of cell growth and apoptotic properties induced by MTX have lead to its evaluation as a potentially therapeutic agent in the treatment of AML-M(5).
8.Study on the correlation between stressors and mental health status of medical undergraduates of local medical colleges in internship: a case study of Binzhou Medical College

Objective To understand the status quo of stressors and mental health of undergradu-ates of local medical colleges in internship and provide relevant countermeasures so as to provide scientific reference for psychological health counseling for them. Methods The stress source scale and mental health symptom self-rating scale for medical students in internship (SCL-90) coded by Chang Lirong et al was adopted to investigate. The questionnaire survey was conducted on the stress and mental health status of 390 interns in Binzhou Medical College, and 384 valid questionnaires were collected, with the effective rate of recovery as 98.46%. The questionnaire results were analyzed by sample t test, variance analysis and Pearson correlation analysis. Results A total of 24.1% of students in local medical schools were under moderate pressure (1.17±0.63). The highest pressure source was examination and employment (1.58±0.74), and the other followed were knowledge and skills (1.43 ±0.68), interpersonal relationships (1.11 ±0.54), patients (1.06±0.53), medical conditions (1.00±0.53) and work load (0.95±0.52) respectively. The scores of various stressors were significantly and positively correlated with most of the mental health factors. Conclusion The undergraduates of local medical colleges and universities are generally under stress in internship, which would affect their mental health. It is suggested that the interns should be guided from the aspects of employment, study and psychology so as to help them relieve their stress and develop good psychological qualities.
9. KRAS mutations analysis in mucinous epithelial lesions of the endometrium

Objective:
To investigate the frequency of KRAS mutation in mucinous epithelial lesions of the endometrium, and analyze the correlation between KRAS mutation and the clinicopathologic features.
Methods:
The cohort included forty-three cases of mucinous epithelial lesions of the endometrium selected from July 2015 to October 2017 from Beijing Obstetrics and Gynecology Hospital, and 22 control cases. Genomic DNA was extracted from formalin-fixed paraffin-embedded tissue sections. Polymerase chain reaction amplification for KRAS exons 2 and 3 was performed, followed by sequencing using capillary electrophoresis. The Fisher exact test was used to compare the prevalence of KRAS mutation among the different groups.
Results:
The patients′age ranged from 33 to 77 years [mean (55.12±9.34) years, median 55 years]. None of the eight cases of endometrial hyperplasia with mucinous differentiation without atypia showed KRAS mutation. The frequency of KRAS mutations was 1/10 in endometrial atypical hyperplasia, 1/12 in endometrioid carcinoma, 4/11 in endometrial atypical hyperplasia with mucinous differentiation (EAHMD), 6/15 in endometrioid carcinoma with mucinous differentiation (ECMD) and 8/9 in mucinous carcinoma (MC), respectively. 10.Research on the relationship between smartphone addiction and mental health of medical freshmen

Objective:To understand the current situation of smartphone addiction and mental health of medical freshmen, and conduct research on the relationship between smartphone addiction and mental health, so as to provide evidence for the prevention and intervention of smartphone addiction among medical freshmen.Methods:A stratified random sampling method was used to select 279 freshmen in the medical profession of Binzhou Medical University as the research objects, and the revised smartphone addiction scale (SAS-C) and mental health symptom self-rating checklist scale (SCL-90) were selected for testing. SPSS 19.0 was used for t test. Results:A percentage of 29.71 medical freshmen had smartphone addiction problems, which was generally above average. The SCL-90 total scores and the scores of somatization, interpersonal sensitivity, depression, anxiety, hostility, psychosis and other factors in the smartphone addiction group were significantly higher than those in non-smartphone addiction group. The total score of smartphone addiction in medical professional freshmen was significantly correlated with the total score of SCL-90. In addition to the interference factors of daily life, other factors had positive correlation with various factors of SCL-90. Interpersonal sensitivity and obsessive-compulsive symptoms had a significant predictive effect on smartphone addiction (β=0.24, 0.26, P < 0.01), which were important variables affecting mobile phone addiction of medical freshmen. Conclusion:Many medical freshmen have the problem of smartphone addiction, which may cause a series of psychological problems, especially obsessive-compulsive symptoms, interpersonal sensitivity, anxiety and psychosis.
