1.Effect of Abstract of Particulates Sampled From the Air of a Railway Transport Station of Hazard Chemicals on Rat TypeⅡ Pneumocytes and the Antagonism of Antioxidants

Objective To know the effect ofof the particulates sampled from the air of the railway transport station of hazard chemicals on rat typeⅡpneumocytes and the antagonism of antioxidants. Methods The rat typeⅡpneumocytes cultured by routine method were pre-treated with antioxidants N-acetylcysteine (NAC), Vitamin C and Vitamin E, then were exposed to the abstract of particulates. The damages of rat typeⅡpneumocytes and DNA cross-links were detected with MTT assay and ethidium bromide fluorescence assay. Results The abstract of particulates caused obvious damages and DNA cross-links in the rat typeⅡpneumocytes and the significant dose-effect relationship was seen. NAC, Vitamin C and Vitamin E reduced significantly the damages and DNA cross-links. Conclusion Abstract of the particulates sampled from the air of the railway transport station of hazard chemicals may induce the damages and DNA cross-links in rat typeⅡpneumocytes and these adverse effects can be antagonized by antioxidants such as NAC, Vitamin C and Vitamin E.

Objective To investigate the micronucleus frequency in mice peripheral blood reticulocytes induced by air particulate pollutants in a railway transport station of chemical dangerous goods (RTSCDG) Methods Male BALB/C mice were selected and randomly divided into exposure groups and control groups One time exposure experiment was carried out among mice exposed to acetone extracts of air particulate pollutants at doses of 20,40,80,160 mg/kg by intraperitoneal injection Multiple time exposure experiment was carried out among mice exposed to acetone extacts of air particulate pollutants at dose of 10 mg/kg by consecutive intraperitoneal injection one time per day for 5 days The peripheral blood was sampled from mice tails and the micronucleus frequencies in reticulocytes were observed at 24, 48 and 72 h after the exposure Results The frequencies of micronuclei in peripheral blood reticulocytes revealed good time response relationship and reached the highest levels at 48 h after exposure,at the same time the frequencies of micronuclei in dose groups of 20,40,80,160 mg/kg were all higher than those in control group The multiple frequencies exposure experiment with lower dose showed that the frequencies of micronuclei in peripheral blood reticulocytes in exposure group always revealed significantly higher levels compared with those in control group during the 24th~72th hour after the exposure ( P

As a result, the product conformed to British Pharmacopoeia (1968 ) in colour, lustre, purity and heavy matals. The yield was 20%.
4.The effect of trimetazidine on alcoholic cardiomyopathy and plasma brain natriuretic peptide

Objective To observe the effect of conventional therapy plus trimetazidine on alcoholic cardiomyopathy (ACM) and plasma brain natriuretic peptide (BNP).Methods Eighty-six cases of ACM patients were divided into control group and observation group by random digits table method with 43 cases each.Control group was treated with conventional therapy,while observation group was added trimetazidine.Course of treatment was 3 months.The left ventricular ejection fraction (LVEF),left ventricular end diastolic diameter (LVEDD),6 min walk distance,plasma BNP before and after treatment and the efficiency in two groups was observed and compared.Results LVEF,LVEDD,6 min walk distance,BNP before treatment in control group were respectively (37.2 ±7.4)%,(57.6 ±7.4) mm,(312.8 ±21.6) m,(846.2 ±63.7)μg/L,and the indicators in observation group were respectively (38.5 ±8.1)%,(57.1 ±6.8) mm,(316.5 ± 23.9) m,(857.6 ± 61.4) μ g/L.All indicators between two groups was no statistically different (P>0.05).LVEF,LVEDD,6 min walk distance,BNP after treatment in control group were respectively (43.5 ±8.6)%,(54.3 ± 6.4) mm,(511.6 ± 26.7) m,(679.4 ± 51.3) μg/L,and there were significant difference compared with those before treatment (P < 0.05).LVEF,LVEDD,6 min walk distance,BNP after treatment in observation group were respectively (51.6 ± 9.2)%,(51.2 ± 6.3) mm,(579.3 ± 25.1) m,(536.5 ± 50.6)μ g/L,and there were significant difference compared with those before treatment (P < 0.05).There were significant difference in LVEF,LVEDD,6 min walk distance and BNP after treatment between two groups (P < 0.05).The total effective rate in observation group was 90.7% (39/43),which was significantly higher than that in control group [72.1% (31/43)] (x2 =4.914,P < 0.05).Conclusion Adding trimetazidine on the basis of conventional therapy can improve the cardiac function and myocardial remodeling of ACM patients and improve clinical efficacy.
5.Down-regulation of suppressor of cytokine signaling 3 expression in hypothalamus attenuates high-fat dietinduced obesity in rats

ObjectiveTo investigate whether hypothalamic silencing of suppressor of cytokine signaling 3( SOCS3 ) by RNA interference ( RNAi ) attenuates diet-induced obesity and leptin resistance in rats.Methods Hypothalamic SOCS3-deficient rats were established by means of lentiviral vector ( LV ) mediated RNAi technique,and then were fed with a high-fat diet for 8 weeks.After the rats were sacrificed,the serum leptin and insulin concentrations were measuredbyRIA, andthe expressionof SOCS3inhypothalamus was detectedby immunohistochemistry and realtime PCR.ResultsThe immunostaining showed that SOCS3 protein expression was significantly reduced and the expression of SOCS3 mRNA was decreased by 49% in SOCS3 RNAi group compared to the controls( P<0.01 ).The rats with hypothalamic SOCS3 knockdown exhibited a significant hampering in gaining body weight with lowered concentrations of leptin[ ( 8.18±2.10 vs 10.85±2.23 ) ng/ml ],insulin[ ( 18.89±4.88 vs 26.78±6.01 )mU/L],glucose [ (4.89±0.91 vs 6.26± 1.41 )mmol/L] and triglyceride [ (0.47 ±0.10 vs 0.62 ±0.16)mmol/L] when exposed to a high-fat diet(P<0.05 or P<0.01 ).ConclusionThe results provide evidence that rats with hypothalamic SOCS3 silencing by RNAi are resistant to diet-induced obesity,leptin resistance,and metabolic disorder.

Objective To find out the endoscopic characteristics of peptic ulcer disease in Monglian area.Methods 15826 cases in hospital within past 17 years were retrospectively analyzed.Results In all endoscopic cases,2298 cases of peptic ulcer were found,the detecting rate was 14.52%.Among them,58.98%(1376 cases) were of duodenal ulcer,32.16%(739 cases) were of gastric ulcer,7.96%(183 cases) were of complex ulcer.The detecting rates of male and female patients were 18.53% and 8.06% respectively,Monglian and Han nationalities were 18.43% and 13.15% respectively.Conclusions The detecting rate of peptic ulcer in Monglian nationality is higher than in Han nationality at Monglian area,has singnificant difference(P

Objective To understand the relationship between GATA -4,-5,-6 gene mutations and con-genital heart disease(CHD),and to provide grounds for early prevention and genetic counseling of children with CHD. Methods GATA -4,-5,-6 coding regions exons and the flanking intron sequences in 1 98 CHD patients were screened,including 66 cases of the ventricular septal defects,84 cases of the atrial septal defects,and 48 cases of the nonsyndromic conotruncal heart defects patients.A total of 300 healthy subjects were selected as controls.The acquired sequences were aligned with which those publicized in GenBank by the aid of program BLAST.All exons and bilateral partial intron -exon boundaries of GATA -4,-5,-6 genes were amplified by the polymerase chain reaction (PCR). The PCR products were purified and directly sequenced by automatic DNA sequence equipment.The acquired GATA -4,-5,-6 gene sequences were compared with GenBank standard gene sequences with the aid of program BLAST. Results A heterozygous missense mutation in the GATA -4 gene was identified in a ventricular septal defect patient and a persistent truncus arteriosus patient.The mutation was located in c.799G >A(p.V267M)in exon 4 of GATA -4. Multiple aligenment of GATA -4 proteins across species demonstrated that altered amino acid was highly conserved. Transcription factor GATA -5,-6 screening showed no mutations in children with CHD in this study.Conclusions Transcription factor GATA -4 gene mutation may be associated with the occurrence of CHD.Transcription factor GATA -4 gene may be susceptible gene in human CHD.
9.Relationship between the polymorphism of the AT2R gene 1675A/G and the effect of indapamide sustained release tablets in female hypertension patients

Objective To investigate the relationship of the1675A/G polymorphism of AT2 gene with the therapeutic effect of indapamide sustained release tablets in female patients with primary hypertension.Methods Two hundred and twenty female patients with primary hypertension were treated with Indapamide Sustained Release Tablets ( 1.5 mg · qd) for 8 weeks.The blood samples from the patients were collect to determine AT2 gene polymorphism by PCR combined with HRM and sequencing.Results Two hundred and five patients completed the test.In female patients,the therapeutic efficacy of indapamide sustained telease tablets among different AT2R genotypes( AA:70.6%,AG:71.6%,GG:71.4% ) showed no significant difference ( x2=2.53,P=0.49 ),neither do the decline of BP after therapy ( F=0.39 and 0.19 respecrively,P > 0.05).Conclusion The AT2 genotype was assumed to be not correlated to the blood pressure lowering response to Indapamide Sustained Release Tablets in female primary hypertension patients.
10.The effect of triptolide on HIF1αand VEGF expressions under hypoxia in human umbilical vein endothelial cells

Objective To investigate the effect of triptolide (TP) on the expression of hypoxia inducible factor 1 alpha (HIF1α) and vascular endothelial growth factor (VEGF) in the human umbilical vein endothelial cells (HUVECs) under hypoxia. Methods (1) HUVECs were treated with 0, 40, 80, 160 and 320 nmol/L TP (named with hypoxia group, TP40 group, TP80 group, TP160 group and TP320 group, respectively) under the hypoxic condition (37℃, 5%CO2, 1%O2, 94%N2) for culturing 12 hours. Meanwhile, cells cultured under normoxia condition (without TP added) were set as the normoxia group. Western blot assay was used to detect the expression of HIF1αin each group. (2) The cells were divided into normal control group, hypoxia group and TP80 group. The immunofluorescence method was performed to detect the localization of HIF1α in cells. (3) Expressions of VEGF were detected by Western blot assay in TP80 group and hypoxia group. (4) The cells were divided into hypoxia group, TP80 group, TP80+KF20 group (80 nmol/L TP and 20μmol/L KC7F2), and TP80+KF30 group (80 nmol/L TP and 30 μmol/L KC7F2). After 12-hour culturing, Western blot assay was used to detect the expressions of HIF1α and VEGF in each group. Results (1) Under the normoxia condition, no HIF1α was detected in HUVECs. The expression level of HIF1αwas significantly increased in TP80 group than that in hypoxia group (P<0.05), while there was no significant change in expression of hypoxia HIF1αin TP160 group and TP320 group compared with that of hypoxic group. (2) The immunofluorescence result showed that HIF1α was mainly expressed in the nucleus. (3) The expression of VEGF was significantly increased in TP80 group than that in hypoxia group (P < 0.05). (4) After the intervention of KC7F2, HIF1αand VEGF expression levels were significantly decreased in the TP80+KF20 group and the TP80+KF30 group than those in the TP80 group (P<0.05). Conclusion TP can improve the expression of HIF1αand VEGF to accelerate the proliferation of endothelial cells under hypoxia condition.
