2.Application Value of 18F-FDG PET/CT in Ovarian Cancer

Objective To evaluate the value of PET/CT in preoperative assessment and postoperative monitoring of ovarian cancer. Methods A retrospective analysis was conducted in 45 patients of ovarian neoplasm with clinical records underwent 18F-FDG PET/CT, including 10 patients underwent PET/CT before surgery and 35 patients after surgery. The clinical follow-up time was 6 months at least. The diagnosis based on pathology and clinical follow-up data. Results (1) The sensitivity, specificity and accuracy of PET/CT in detecting ovarian cancer were 94.6%,75.0%and 91.1%, respectively. (2) Ten patients before surgery were all detected tumor by PET/CT, but 2 of them were false positive based on pathologic results. (3) Two patients with non-standard surgery were detected tumor by PET/CT. In 33 patients after standard surgery, 6 patients were no tumor detected by PET/CT. In addition,4 patients with normal CA125 and no signs of recurrence and metastasis were detected tumor by PET/CT. The pathology and clinical follow-up data supported the results. 23 patients with higher CA125 were diagnosed recurrence and metastasis based on pathology and clinical follow-up data, 21 of them were detected tumor by PET/CT. Conclusion 18F-FDG PET/CT plays an important role in preoperative assessment, early diagnosis and accurate positioning of recurrent and metastasis of ovarian cancer. It can be used to guide the clinical treatment.
3.Clinical effects of gelatin sponge infiltrated by mouse nerve growth factor in local treatment of peripheral nerve injury

BACKGROUND:Several studies have attempted to apply mouse nerve growth factor to local lesions of peripheral nerve and found that local injection of mouse nerve growth factor can promote nerve recovery, which is superior to systematic application. OBJECTIVE:To evaluate the clinical effects of gelatin sponge infiltrated by mouse nerve growth factor in the repair of peripheral nerve injury. METHODS:Thirty-six patients with single peripheral nerve injury, including 16 males and 20 females, aged 18-48 years, were randomly divided into two groups: 18 patients in case group underwent debridement and neuroanastomosis, and then the injured nerve was wrapped by gelatin sponge which was infiltrated by mouse nerve growth factor and folowed by plaster fixation, anti-inflammatory therapy, neurotrophy and circulation improvement therapy; the other 18 patients in control group were treated only with debridement and neuroanastomosis and other conventional therapies. At 4 weeks after treatment, electrophysiological examination was performed. In addition, sensory and motor function of the distal end of injured nerve was evaluated at 6 months after treatment. RESULTS AND CONCLUSION: Sensory evoked potential and motor evoked potential showed that the recovery rate was 78% (n=14) and 83% (n=15) respectively in the case group, while 57% (n=10) and 66% (n=12) in the control group. There was significant difference between the two groups (P < 0.05). The total effective rate was 94.4% (n=17) in the case group and 83.3% (n=15) in the control group, which were statisticaly better in the case group than the control group (P < 0.05). These findings indicate that it is significantly effective to treat peripheral nerve injury by gelatin sponge infiltrated by mouse nerve growth factor that has good biocompatibility.
4.The Metabolomic Study of Learning and Memory Function of APP/PS1 Double Transgenic Mice Improved by Gouteng San

Objective:The non targeted high-throughput urine metabolomics technology was used to study the pathogenesis of APP/PS 1 double transgenic mice and the mechanism of action of Gouteng san.Methods:5-month-old APP/PS 1 double transgenic mice were test with Morris water maze for spatial learning ability.Then we employed the non targeted high-throughput urine metabolomics technology to study the pathogenesis of APP/PS1 double transgenic mice based on the metabolic network.The focus investigation of the key pathways and the observation of the treatment by Morris water maze and metabolic level have been used after spatial learning ability damaged confirmed.Results:The comparison between APP/PS1 double transgenic mice and normal mice suggested that a significant longer was existed in former,which was call-back by Gouteng san.With the non targeted high-throughput urine metabolomics analysis and pathway focused analysis,we found certain signals from metabolic profiling,which was identified to be 6 biomarkers associated with learning and memory function by mass spectrometry analysis or authoritative database.Respectively,they were taurine,pteroylglutamic acid,neopterin,glutaurine,2-oxoglutarate and dihydroneopterin.They were mainly related to taurine metabolism and folate metabolism and represented an effective callback.Conclusion:Gouteng san possess a favorable effect on learning and memory ability of APP/PS1 double transgenic mice,6 biomarkers may be a potential target for the pathogenesis of APP/PSI double transgenic mice and provide experimental basis for the study of Gouteng san.
5.Relationship between the serum chemokine IP-10 and RANTES levels and Interferon therapeutic early response in patients with chronic hepatitis B

Objective To investigate the relationship between the serum chemokine IP-10 and RANTES levels and Interferon therapeutic early response in patients with chronic hepatitis B(CHB).Methods 50 patients with chronic hepatitis B were chosen into interferon therapy.After 12 weeks,they were devided into three groups:complete response A、partial response B、non response C group.HBV-DNA was detected by PCR;The serum chemokines( IP-10 and RANTES) were measured by Luminex Liquichip technology.Results The base HBV DNA and RANTES levels of three groups weren't significantly different (P <0.05) ;The base ALT and IP-10 levels of A group were significantly higher than that in B and C group( P < 0.05).The IP-10、RANTES contents of A group in therapeutic 4th week were significantly lower than that before interferon therapy(P < 0.05 );There were no significant differences in B、C group (P >0.05) ;The levels changes of IP-10、RANTES、HBV DNA and ALT in therapeutic 12th week were significantly different between the three groups ( P < 0.05 ),The level of ALT in 50 patients has positive correlation with IP-10 level (P < 0.05) ;The level of HBV DNA in 50 patients had positive correlation with RANTES level( P < 0.05 ) ;The base level of IP-10 had positive correlation with the change of HBV-DNA contents in therapeutic 12th week( P <0.05 ) ;The change of ALT level in reponse patients in therapeutic 12th week had positive correlation with the change of IP-10 、RANTES levels( P < 0.05 ).Conclusion The decrease of IP-10,RANTES level in CHB patients received 12weeks interferon-α therapy could lead to reduce liver inflammation;The base IP-10 level probably was relevant to the early response in CHB patients received interferon-α therapy.
6.Evaluation of vortex within left ventricle in patients with dilated cardiomyopathy by vector flow mapping

Objective To evaluate left ventricular function in the way of intracavitary vortex by vector flow mapping(VFM),and to demonstrate the formation and variation of the vortex. Methods Twenty-six patients with dilated cardiomyopathy(DCM) and 44 healthy controls were involved. The velocity distribution imagings on the section plane of the flow in the left ventricle were obtained by VFM, the location of the vortex and its variation in every phase of cardiac cycle were focused,and series parameters were measured,such as the diameter of the vortex (transverse diameter, vertical diameter), number of the vertex. Results relaxation(e) period. Vortices were observed mostly during diastolic period, nearby the anterior leaflet of mitral valve,middle upper 1/3 of left ventricle,and their shapes changed 2 - 3 times. The diameters of the vortex (transverse diameter,vertical diameter) , number of the vertices in one cardiac cycle during 7 phases mostly nearby the cardiac apex, middle lower 2/3 of left ventricle, and their shapes changed 4 - 5 times.Compared with control group,the diameters of the vortex (transverse diameter, vertical diameter) during 7 phases in DCM group were bigger,and number of the vertices were more ( P <0.01 ). The diameters of the vortex (transverse diameter, vertical diameter), number of the vertices within DCM group in one cardiac cycle during 7 phases were significantly different ( P < 0.01 ). Conclusions VFM can demonstrate the variation of intracavitary vortex. Vortex in DCM group were significantly bigger than those of control group.
7.Effects of local hyperthermia on phosphatidylinositol 3-kinase gene expression in Langerhans cells from human papillomavirus(HPV)-infected skin

Objective To investigate the effect of local hyperthermia on the mRNA expression of phosphatidylinositol 3-kinase(P13-K)in Langerhans cells(LCs)transmigrating from HPV-infected skin.Methods Tissue samples were collected from 5 female patients with condyloma acuminatum(CA)and 5 normal human controls.then equally divided into three parts to receive local hyoerthermia treatment at 37℃,42 ℃ and 45℃,respectively,for 30 minutes.After another 12-hour incubation in RPMI 1640 culture medium,transmigrating cells were collected and LCs were purified.Real-time quantitative reverse transcription-PCR was performed to detect the mRNA expressions of P13-K p85a and p110α in purified CD1a~+ LCs from these tissue samples.Results In CD1a~+ LCs transmigrating from normal skin and lesions of CA.the expressions of P13-K p85a and P110α mRNA decreased with the increase of temperature.Atier local hyperthermia treatment of normal skin at 37℃,42℃,45℃,the relative mRNA expression level was 1.00±0.00,0.78±0.13,0.54±0.17,respectively.for P13-K p85α in transmigrating LCs,1.00±0.00,0.80±0.11,0.61±0.12,respectively.for P13-K P110α;there was a significant difierence among the three temperatures in bOth parameters(both P<0.01).In the case of lesions of CA.the relative mRNA expression level of P13-K p85αand p110α was 1.00±0.00 and 1.00±0.00 under treatment at 37℃,0.20±0.11 and 0.49±0.21 at 42℃.0.1±0.08 and 0.09±0.03 at 45℃.respectively;significant difierence was also noted among the three treatment temperatures(both P<0.01).The decrease in P13-K P110α mRNA expressions under hyperthermia treatment at 42 ℃ and 45℃ compared with those at 37℃was greater in LCs from CA lesions than that from normal skin.Conclusions Local hyoerthermia could remarkably inhibit the expression of P13-K genes.which may enhance immunity and favor elimination of HPV.
8.Improved PCR-based subtractive hybridization, a new strategy on cloning differential expression genes in apoptotic MCF-7 cells

Aim To clone apoptosis-related genes from human MCF-7 breast cancer cells and to analyze the character of the method used in the process. Methods A poptotic cell model of MCF-7 cells was established with the apoptotic tumor cells induced by the all-trans-retinoic acid. The apoptotic gene was cloned from the model by improved PCR-based subtractive hybridization. Results 5 clones were identified to be related to apoptosis by reverse dot blot, 4 of them were known genes, and 3 were related to apoptosis. A novel gene, named apmcf-1, coded for 47 amino acid was identified. This gene was accepted by Genbank, the accession number was AF141882. Conclusion This improved PCR-based subtractive hybridization may be an efficient way in cloning differential expression gene.
9.Resection of giant neurofibroma in a case of neurofibromatosis type 1 followed by replantation of the skin from tumor surface

Neurofibromatosis type 1 combined with giant neurofibroma is very rare, and treatment is quite difficult. October 2019, a 23 year-old female patient of neurofibromatosis type 1 combined with giant neurofibroma at the shoulder and back was treated at the Department of Burn and Plastic surgery, the First People’s Hospital of Bijie Municipal. The giant neurofibroma was resected and the skin from the tumor surface was replanted to cover the wound. During the following period of 3 months, satisfactory results were achieved for both donor site and recipient site.
