1.Screening of receptor that interacted with outer membrane protein 25 during macrophage infection by Brucella abortus

Objective Receptor was screened to find out the interaction between outer membrane protein 25 of Brucella(OMP25) and Brucella infected macrophages.Methods Recombinant OMP25 protein was expressed and purified.The 7 peptide was screened out with OMP25 protein by phage display technology.OMP25-7 peptide interactions were further confirmed by enzyme linked immunosorbent assay(ELISA) after candidate peptides were predicted with bioinformatics software.Four potential 7 peptides were synthetized.Their functions were estimated after Brucella abortus 2308 infected murine macrophages RAW264.7.Results Forty-two cyclic 7 peptides were screened out based on purified OMP25 by the M13 Phage Library Kit.Four peptides,named as ST-7,MT-7,TF-7 and SN-7,were confirmed with ELISA.Colony forming unit(CFU) result demonstrated ST-7,MT-7,TF-7 and SN-7 showed some function for inhibition of intracellular parasite.ST-7 inhibition ratio was 14.4%-51.8% for Brucella abortus 2308; MT-7 inhibition ratio was 49.6%-69.5% for Brucella abortus 2308; TF-7 inhibition ratio was 43.2%-74.4% for Brucella abortus 2308; SN-7 inhibition ratio was 57.5%-82.2% for 2308.ELISA result showed,that compared to control group,tumor necrosis factor α(TNF-α)increased no statistical significance in each time point of ST-7,MT-7 experiment groups (all P > 0.05).There was no statistical significance in 8 h ago of TF-7,SN-7 experiment groups (all P > 0.05),but there was statistical significance in 12,24 h (all P < 0.05).Conclusions We have obtained four effective OMP25 receptors through phage display technology; TF-7 and SN-7 have inhibited Brucella abortus 2308 invasion and intracellular survival in murine macrophage cells,while ST-7 and MT-7 have only prevented the invasion against Brucella abortus 2308.
2.ImmunohistochemicaI Expression and CIinicaI ImpIication of RASSF1A Gene in LaryngeaI Squamous CeII Carcinoma and Keratosis of the Larynx

Objective To observe the RASSF1A expression in laryngeal keratosis and laryngeal squamous cell carcinoma and investigate the relationship between the gene and the occurrence and development of this disease. Methods The specimens of keratosis of larynx(43 cases)and laryngeal squamous cell carcinoma(31 cases)in Oto-laryngology-Head and Neck surgery of Anhui provincial hospital from Dec 2009 to Dec 2012 were collected.Nor-mal vocal fold mucosa from the cadaveric head as control group(8 cases)were collected.Immunohistochemical meth-ods were used to detect the expression of RASSF1A protein in these tissues.The diffuse distribution of brown gran-ules in the cell cytoplasm yielded positive results and the nucleus was not coloring.The percentage of positive cells of every section,and the average standard deviation( ̄x±s)were calculated.SPSS17.0 statistical analysis software was used to analyze the data,P<0.05 for the difference was statistically significant.ResuIts The RASSF1A expes-sion in laryngeal squamous cell carcinoma,normal laryngeal mucosa,squamous cell carcinoma of the larynx and la-ryngeal keratosis were compared.The differences of RASSF1A expression were statistically significant (P<0.05). The difference of RASSF1A expression in keratosis of larynx and normal laryngeal mucosa was not significant (P>0.05).RASSF1A expression in the clinical stage from I to IV of laryngeal squamous cell carcinoma gradually de-clined.During the clinical stage,the difference had statistics significance (P<0.05 ).In high differentiated squa-mous cell carcinoma,moderately differentiated squamous and poorly differentiated squamous,the differences of RASSF1A expression were insignificant (P>0.05).ConcIusion The reduction of RASSF1A expression correlated with laryn-geal squamous cell carcinoma,which has certain significance for the different ion of carcinoma and laryngeal precan-cerous lesion.
3.Analysis of Non-rational Prescriptions in Pharmacy Intravenous Admixture Service

OBJECTIVE:To analyze the non-irrational prescriptions in pharmacy intravenous admixture services to ensure patient's medication safety.METHODS:We examined the prescriptions in pharmacy intravenous admixture services from Sep.2006 to Feb.2007 and analyzed the non-irrational ones regarding incompatibility,improper choice of infusion carriers,improper medication frequency,and the incompatibility of Chinese medicines etc.RESULTS:The non-irrational prescriptions totaled 126 cases,of which,46 associated with incompatibility,38 with improper choice of infusion carriers,25 with improper medication frequency and 17 with incompatibility of Chinese medicines.Most of the irrational drug use behaviors were corrected by informing the clinic these medication errors.CONCLUSION:Examining prescriptions in pharmacy intravenous admixture services can improve the safety and effectiveness of patient's use of intravenous drugs.
4.Research on eight-year medical education program publications

Objective In order to get the research focuses and trend of 8-year medical educa-tion program in China, articles on eight-year medical education program published from 2003 to 2014 have been analyzed. Method Using Cochrane Systematic evaluation method, 797 articles have been achieved from CMB, CNKI, WangFang and VIP. These papers were then analyzed with methods of bib-liometrics and content analysis. Result The articles on eight-year medical education program are mainly published on 4 journals. The publications focus on 10 domains. The domain of teaching method-ology is a hot research topic as 399 articles have discussed this domain. Most articles on teaching methodol-ogy focus on PBL, only a few papers discuss other teaching methods. In the 233 study on the curricu-lum design, only 42 papers have carried on the overall design course study and the researchers have the same opinion on the problems existing in eight-year program medical education curriculum. 19 articles have discussed the goal of eight-year medical education program. The goals of different schools' program are diverse. With the development of policy research, the eight-year medical educa-tion program education standards and accreditation are valued. Conclusion In the future, the research priorities should focus on pedagogy theories, the comparison to other nations' medical education system, and ed-ucation standards of eight-year medical education program.
5.Three-dimensional virtual reality imaging combined with two-dimensional ultrasound images for the visual evaluation of hip dysplasia in infants

BACKGROUND:Ultrasound has been widely used in developmental hip dysplasia screening in infants and young children, but there are significant differences among different inspection findings, even in the reexamination by the same examiner. Therefore, the accuracy of ultrasound imaging in the diagnosis is of insufficient evidence.
OBJECTIVE:Through virtual reality three-dimensional reconstruction images showing the structure of acetabulum and femoral head space, to obtain a new parameter in order to enrich measurement standards of Graf ultrasonic diagnosis technology and to further improve the credibility of the diagnosis of developmental dysplasia of the hip.
METHODS:Ten fresh cadaver specimens of Chinese infants were selected, including five males and five females. Two-dimensional ultrasonic method was employed for hip screening, and at the same time, multi-slice spiral CT was used for three-dimensional reconstruction fol owed by volume reconstruction and fitting of surface reconstruction to display the anatomic structure around the hip and to simulate Graf’s two-dimensional ultrasound method to measure a andβangles on three-dimensional virtual reality images.
RESULTS AND CONCLUSION:As confirmed by three-dimensional reconstruction, Graf’s two-dimensional ultrasound could be used to measure a and angles, truly reflecting the development of bony and cartilaginous acetabulum. Graf’s two-dimensional ultrasound contributed to the proper evaluation of beonatal hip dysplasia, which can be used as a routine examination for developmental dysplasia of the hip. These indicate that to master Graf’s ultrasound technology and strictly grasp the evaluation standards for developmental dysplasia of the hip is the important guarantee for diagnosis of developmental dysplasia of the hip.
6.Clinical Observations on Heat-sensitive Point Dazhui (GV14) Moxibustion for the Treatment of Cervical Spondylotic Radiculopathy

Objective To investigate the clinical efficacy of heat-sensitive point Dazhui(GV14) moxibustion in treating cervical spondylotic radiculopathy. Method Ninety-six patients with cervical spondylotic radiculopathy were randomly allocated to a heat-sensitive point Dazhui moxibustion (observation) group of 30 cases, a non-heat-sensitive point Dazhui suspended moxibustion (control) group of 33 cases and a medication group of 33 cases. The observation group received heat-sensitive point Dazhui moxibustion; the control group, non-heat-sensitive point Dazhui suspended moxibustion; the medication group, oral administration of Jingtong granules. The clinical symptoms were assessed and serum hypersensitive C-reactive protein (HsCRP) and interleukin-8 (IL-8) were measured in the three groups before and after 15 days of treatment. Result All the three treatments had a certain effect on cervical spondylotic radiculopathy. The therapeutic effect was best in the observation group (P<0.05). After treatment, serum HsCRP and IL-8 contents decreased in all the three groups compared with before (P<0.05). They were even lower in the observation group than in the control and medication groups (P<0.05) but had no statistically significant differences between the control and medication groups (P>0.05). Conclusion The therapeutic effect of heat-sensitive point Dazhui moxibustion is better than those of non-heat-sensitive point Dazhui suspended moxibustion and oral Jingtong granules. It is a more ideal way to treat cervical spondylotic radiculopathy. The mechanism of its action may be related to post-treatment decrease in inflammatory factors in the lesion.
7.Evaluation of transverse percutaneous fixation with iliosacral screws via the second sacral vertebra for longitudinal sacral fractures

Objective To explore the safe bone channels for transverse placement of sacroiliac screws via the second sacral vertebra in the fixation of longitudinal fracture of the sacrum by digital analysis and clinical observation.Methods Firstly,abdomen CT scanning was performed in 50 healthy adult volunteers.They were 30 males and 20 females,aged from 18 to 56 years (mean,34.6 years).After their CT images were transformed by software Materialise Mimics Innovation Suite 15.0,three-dimensional (3D) models of the pelvis were generated,stored in stereolithography format,and transferred into software Imageware 12.0.In the standard antero-posterior position,the sacrum was segmented and the points of 2D image coordinate geometric boundary were extracted to generate an optimal channel for transverse placement of sacroiliac screws via the second sacral vertebra.Secondly,we admitted 10 patients with longitudinal fracture of the sacrum.They were 7 men and 3 women,aged from 20 to 51 years (average,38.3 years).By Tile classification,4 cases were type B2 and 6 type C1.All the fractures were Denis region Ⅱ ones.Pubic ramus fracture was complicated in 3 cases.After traction reduction of the tibial tubercle was conducted for patients with obvious displacement,the optimal channel was calculated on the basis of the CT data.On the lateral images of the sacrum acquired before operation by C-arm fluoroscopy,the optimal channel for transverse placement of sacroiliac screws via the second sacral vertebra was located.After the skin was marked,2.5 mm Kirschner wire was drilled and the 6.0 mm hollow screws were fixated along the Kirschner wire.Postoperative CT scanning images and 3D reconstructed models were analyzed to validate the accuracy of screw placement.Results The projection of safety bone channel on the sagittal fluoroscopy of S2 vertebral body displayed an irregular water drop shape;the projection area in males (213.9 ± 52.4 mm2) was significantly larger than that in the females (171.6 ±49.6 mm2) (P < 0.05).The average length of the channel in men (135.2 ± 12.9 mm) was significantly longer than that in women (121.1 ± 10.1 mm);the average diameter of the screw in men (10.2 ± 0.9 mm) was also significantly larger than that in women (9.1 ±0.8 mm) (P < 0.05).The postoperative X-ray and CT scanning images showed satisfactory positions and lengths of the screws.The screw lengths averaged 98.2 mm;the operation time averaged 25.6 min.No nerve or vascular injury,loosening or breakage of the sacroiliac screws occurred in the patients.Follow-ups for 12 to 24 months revealed no other complications.Conclusion The safety channel for transverse placement of sacroiliac screws via the second sacral vertebra can be determined using the lateral 3D reconstruction images of the sacrum in preoperative planning,which facilitates the percutaneous transverse fixation of longitudinal fracture of the sacrum.
8.Stability of Cefoperazone Sodium /Tazobactam Sodium for Injections in Fructose-contained Sodium Chloride Injection or Invertose Injection

OBJECTIVE:To study the stability of Cefoperazone Sodium /Tazobactam Sodium in Fructose-contained 0.9% sodium chloride injection or Invertose Injection.METHODS:The contents of Cefoperazone Sodium /Tazobactam Sodium within 6 hours after mixing with Fructose-contained 0.9% sodium chloride injection or Invertose Injection were determined by HPLC,meanwhile the changes in the appearances and the pH values were observed.RESULTS:The contents of Cefoperazone Sodium and Tazobactam Sodium in Fructose-contained 0.9% sodium chloride injection or Invertose Injection were stable within 6 hours,and the mixed solutions were clear and their pH values were stable on the whole.CONCLUSION:Cefoperazone Sodium /Tazobactam Sodium can de mixed with Fructose-contained 0.9% sodium chloride injection or Invertose Injection for clinical use.
9.Genotyping analysis of 151 clinical isolates of Mycobacterium tuberculosis from Uygur in south of Xinjiang

Objective To investigate the application of the multiple locus variable numbers of tandem repeats(MLVA) in genotype Mycobacterium tuberculosis(MTB) strains isolated from Uygur in south of Xinjiang, and to understand the characteristics of genotype and distribution. Methods One hundred and fifty-one Mycobacterium tuberculosis strains were collected from Uygur in south of Xinjiang which contains three regions, Kashgar, Hotan and Kizilsu kirghiz. Twenty-four tandem repeats loci in the total genome of MTB were analyzed by PCR and agarose gel electrophoresis method. The characteristics on polymorphism of DNA fingerprinting of 151 MTB strains were analyzed with BioNumerics 5.0 software. Results Twenty-four MLVA loci of 151 MTB strains were analyzed respectively. The results showed that there were obvious polymorphisms of VNTRs. The clustering of genotype showed that these strains could be categorized into 8 gene groups( Ⅰ , Ⅱ , Ⅲ ,Ⅳ, Ⅴ ,Ⅵ,Ⅶ ,Ⅷ) and 151 genotypes. Sixty-seven isolates(44.4% ) belonged to group Ⅵ. 23.2% were group Ⅷ including 35 genotypes, 20. 5% were group Ⅳ including 31 genotypes. The group Ⅵ prevailed mostly in the Kashgar. The group Ⅲ prevailed mostly in the HOTAN. Conclusion The results showed there were obvious polymorphisms of VNTRs of Mycobacterium tuberculosis clinical strains preliminarily. Group Ⅵ was the predominant prevalent strain in south of Xinjiang.
10.Navigation template for sacral fracture fixation using three-dimensional reconstruction and reverse engineering technique

Objective To provide a new method for sacral fracture fixation by means of 3D recon-struction and reverse engineering technique. Methods Pelvis images of 3D CT scan were obtained from patients with sacral fracture. The digital data were transferred into a computer workstation. 3D models of pelvis were reconstructed using Amira 3.1 software and saved in STL format. The 3D fracture models were imported into Imageware 10.0 software. Different situations of reduction (total reduction, half reduction and no-reduction) were simulated using Imageware 10.0 software. The extract locations and the best directions of inserting iliosacral lag screws were defined using Reverse Engineering according to the 3 situations to before navigation templates were designed according to the posterior anatomic features of the ilium and the insertion channels. Exact navi-gational templates were manufactured by rapid prototyping. Drill guides were sterilized and used intraoperatively to assist surgical navigation and placement of iliosacral lag screws. Results Accurate screw placement was confirmed with postoperative X-ray and CT scanning. The navigation templates were found to be helpful and highly accurate. Conclusion The navigation template may be a useful method for mini-invasive fixation of sacroiliac joint fracture.
