1.Significance of Bcl-2 family in tumor progression and therapy

Bcl-2 family have dual-regulating effects on cell apoptosis mediated by mitoehondrion. The ratio of pro-apoptosis members and anti-apoptosis members closely correlates with tumorigenesis, drug-resist-ance and prognosis. Therefore,Bcl-2 family become important targets in tumor biotherapy. Many strategies have been applied to tumors treatment targeting Bcl-2 family,such as some biological treatment,short peptides and organic small molecules.
2.Determination of Paeoniflorin in Gujing Pill by HPLC

Objective To establish a method for the content determination of paeoniflorin in Gujing Pill. Methods Orthogonal test was used to optimize the condition of sample extraction and paeoniflorin content was detected by HPLC. HPLC was performed with Merck- LiChrospher RP- C18 column(4.6? 250 mm,5 ? m); methanol- water (25 ∶ 75) served as mobile phase and the detection wavelength was at 230 nm, the flow rate being 0.8 mL/min. Results Paeoniflorin has a good linearity within 0.27~ 1.35? g(r=0.9998) and the average recovery was 97.08 % with RSD 2.02 % (n=10). Conclusion This method is simple, sensitive, specific and can be used for the quality control of Gujing Pill.
3.The clinical application of modified preflush in the treatment of CRRT without heparin for patients received dialysis with complication of cerebral hemorrhage

Objective: To explore the clinical effect of modified preflush on the treatment of CRRT without heparin in patients received dialysis with complication of cerebral hemorrhage. Methods: 44 patients received dialysis with complication of cerebral hemorrhage were divided into control group (28 cases) and observation group (16 cases) according to the implemented time of modified preflush. A retrospective analysis was adopted to compare the effect of filter in blood coagulation grade and service life between conventional preflush (control group)and modified preflush (observation group). Results: The differences of I, II, III grade of blood coagulation between the two methods were significant (x2=12.020, P<0.05), and the differences of average service time of filter, transmembrane pressure(TMP), pressure of filter and value of pressure drawdown (ΔP) between the two methods also were significant (t=3.535, t=4.024, t=6.071, t=3.524, P<0.01). Besides, the difference of venous pressure (PV) between them was significant (t=2.166, P<0.05), while the differences of BFR, UFR and Pa between the two methods were not significant (t=0.263, t=1.162, t=0.258, P>0.05). Conclusion: Modified preflush method can make the fiber of filter to obtain more full stretch and soak, and it also can prolong the service of time, and decrease incidence of blood coagulation, and ensure curative effect and smooth process.
4.Construction of lentiviral vector encoding CLEC4M and overexpression of CLEC4M in K-562 cells

Objective To construct the lentiviral vector encoding CLEC4M and prepare K -562 cells with stable overexpression of CLEC4M.Methods The gene sequence of normal CLEC4Mwas cloned by reverse transcription PCR and then inserted into GV166 vector to construct GV166-CLEC4Mlentiviral expression vector,and then lentiviral packaging was performed by transfection of293T cells.The ob-tained lentiviral liquid was used to infect human leukemia cell line K-562.Real-time PCR and Western blot were used to detect the over-expression of CLEC4M in K-562 cells.Results Sequencing showed that the recombinant lentiviral expression plasmid GV166-CLEC4M was successfully constructed.Lentiviruses could efficiently infect K-562 cells,according to real-time PCR.CLEC4Mwas successfully o-ver-expressed in K-562 cells at mRNA and protein levels.Conclusion The construction of lentiviral vector encoding CLEC4M lays a foundation for further study of CLEC4M gene involved in HCV entry into host cells.
5.Values of serum AFP,GGTⅡ and GP73 in diagnosis of primary hepatocellular carcinoma

Objective To explore the early diagnostic values of serum alpha -fetoprotein (AFP),gamma -glutamyltransferase Ⅱ (GGTⅡ),and Golgi protein 73 (GP73)in patients with primary hepatocellular carcinoma (PHC).Methods The serum specimens of 100 pa-tients with liver diseases (50 cases of hepatitis and liver cirrhosis and 50 cases of PHC)and 50 healthy people were collected in our hospital from February 2013 to February 2014.Electrochemical luminescence technique,specific immuno -membrane adsorption assay,and enzyme-linked immunosorbent assay were used to measure the serum levels of AFP,GGTⅡ,and GP73.Comparison of continuous data between multiple groups was made by analysis of variance,and comparison between two groups was made by q test.The receiver operating character-istic (ROC)curves of single or combined test results were made,and the areas under the ROC curves (AUCs)were calculated.The sensi-tivity,specificity,and AUCs of AFP,GGTⅡ,GP73,and the combined test were analyzed and compared.Results The level of serum GGTⅡ in the PHC group was significantly different compared with those in the other two groups (F =16.224,P <0.05),but there was no significant difference between the normal group and the hepatitis and liver cirrhosis group (P >0.05).Significant differences in serum levels of AFP and GP73 were observed by paired comparison between the PHC group,hepatitis and liver cirrhosis group,and normal group (F =193.128,F =20.231,P <0.05 for both).When assayed alone,the specificities of GP73,GGTⅡ,and AFP were 69%,64% and 51%, respectively,and the sensitivities were 92%,84%,and 76%,respectively.In combined test,the specificity was 94.6% and the sensitivi-ty was 98.8%.Conclusion The GP73 test is the best performer in the single assays.Combined test of serum AFP,GGTⅡ,and GP73 shows a good diagnostic value for PHC with greatly improved specificity and sensitivity.
6.Effect of sleeping time on psychological state and quality of life in patients with type 2 diabetes

Objective To observe the sleeping time and analyze its impact on the psychological state and quality of life of patients with type 2 diabetes .Methods Convenience sampling was used to recruit 365 patients with type 2 diabetes in Jiangsu Province Hospital on Integration of Chinese and Western Medicine Affiliated to Nanjing University of Chinese Medicine between April 2014 and April 2015 .All the patients were divided into two groups according to the six-hour cut-off point of sleeping time .They were investigated by means of World Health Organization Quality of Life-BREF ( WHOQOL-BREF), Diabetes Distress Scale ( DDS) and General Self-Efficacy Scale ( GSES).Data were analyzed by t-test, chi-square test, Mann-Whitney U test and multivariate regression analysis .Results The average sleeping time was 7.03 h, and the sleeping time of 109 patients was less than 6 h.Patients with less sleeping time had lower quality of life and self-efficacy scores as well as higher diabetes distress scores than those with sleeping time more than 6 h.The differences were statistically significant (P<0.05).Regression analysis showed that sleeping time was the factor of quality of life (β=0.117, P=0.047), self-efficacy (β=0.136, P=0.024) and diabetes distress(β=-0.118, P=0.046).Conclusion Sleeping time affects the psychological state and quality of life of patients with type 2 diabetes.The medical should pay more attention to the quality of sleep of such patients .

Objective To explore the antagonistic effect of Ziyin Xiehuo Recipe(ZYXHR,Recipe for nourishing kidney yin and purging ministerial fire) on estrogen-like activity of environmental endocrine disruptors,nonylphenol(NP) and mixture of NP and bisphenol A(BPA). Methods Totally 30 three-week-old female SD rats were randomly divided into control group(fed with corn oil),exposed group A (feeding NP100 mg / kg ),exposed group B (feeding NP 50 mg/kg+ BPA 200 mg/kg ),treatment group A (NP 100 mg/kg + ZYXHR),treatment group B (NP 50 mg/kg+ BPA 200 mg/kg + ZYXHR). 8.Adverse reactions of lansoprazole combined with Flupentixol and melitracen in the treatment of functional dyspepsia patients

Objective To analyze the curative effect and adverse reaction of lansoprazole combined with droperidol thiadiazole melitracen treatment in patients with functional dyspepsia.Methods From December 2013 to December 2015 in our hospital and accepted treatment of 90 patients with functional digestive adverse as the research object,according to the random number table were divided into the control group and the observation group,45 cases in each group.The control group by oral administration of lansoprazole enteric coated capsules,the observation group taking oral lansoprazole and droperidol thiadiazole melitracen,course lasted for 4 weeks,efficacy and adverse occurrence was compared between two groups of patients.Results After 4 weeks of treatment,the observation group with symptoms score GSRS(postprandial fullness discomfort,early satiety,abdominal pain,epigastric burning sensation were lower than the control group with postprandial fullness discomfort early satiety,abdominal pain,epigastric burning sensation,and the difference was statistically significant(P<0.05); the patients in the observation group after 4 weeks of treatment,the total effective rate(86.7%)was higher than the control group(66.7%)significantly,and difference was statistically significant(P<0.05); symptoms of adverse reactions of two groups of patients in the process of drug use were mild and did not give special treatment,in a few days after the basic relief.Conclusion Lansoprazole combined with fluorine Pai Sai tons of Murray Song Xin can effectively eliminate or reduce the symptoms of functional dyspepsia patients clinical and no serious adverse reactions,it is safe in clinical application.
9.Effects of Smoking on Arsenic Metabolism, Methylation:a Meta-analysis

Objective To investigate whether smoking affects metabolism and methylation of arsenic. Methods Six papers about the relation between smoking and arsenic metabolism, methylation were collected until December,2008, and the data were quantitatively analyzed with random and fixed effect models. Results In the group of smoking, the summary weighted mean difference of percent of inorganic in urinary was 0.59 (95% CI:-0.01-1.18). The summary weighted mean difference of percent of MMA in urinary was 2.44 (95%CI:1.95-2.94). The summary weighted mean difference of percent of DMA in urinary was -3.04 (95%CI:-4.01- -2.07). Current evidence suggested that percent of MMA was higher, percent of DMA was lower in smoking or ever smoking group compared with nonsmoking group. Conclusion Smoking may be considered a risk factor for metabolism and methylation of arsenic on human.
10.Reversal of multidrug resistance by CPUE1 in K562/A02 cells

AIM: To study the effect of CPUE1 on the reversal of multidrug resistance (MDR) in K562/A02 cells and its mechanism. METHODS: MTT assay was used to determine the influence of CPUE1 on the cytotoxicity of vincrisine (VCR) in K562/A02 cells. The effect of CPUE1 on the apoptosis induced by VCR in K562/A02 cells was detected using DNA analysis and Annexin-Ⅴ/PI double stain assay. Flow cytometry was used to investigate the efflux of rhodamine123 (Rh123) by P-glycoprotein (P-gp) in K562/A02 cells. RESULTS: CPUE1 increased the cytotoxicity and apoptosis induced by VCR in K562/A02 cells. At the concentration of 10 ?mol?L -1 ,CPUE1 reduced the IC_ 50 value of VCR from 60.54 to 4.17 ?mol?L -1 . CPUE1 inhibited the efflux of Rh123 and resulted in the increase of intracellular accumulation of Rh123 in K562/A02 cells. CONCLUSION: CPUE1 has strong reversal effect on MDR in K562/A02 cells by inhibiting P-gp activity.
