1.Establishment and evaluation of primary allergic contact dermatitis mouse model

Objective To establish and evaluate a new mouse model of allergic contact dermatitis (ACD)-primary ACD mouse model.Methods To establish classical ACD mouse model,mice were sensitized with 1-fluoro-2,4-dinitrobenzene (DNFB)on day 1 and elicited with DNFB on day 6,and primary ACD mouse model was built by painting Of 10 μL 0.2%DNFB once on the ear.Mice were kilied 24 hours after the elicitation in classical model group and 6 days after the sensitization in primary model group;skin specimens were obtained from the right ear of these mice.Ear swelling after painting was used as clinical index.ELISA and real time RT-PCR were performed to measure the expression of IL-2,IFN-γ and IL-4 in these specimens.Local lymph node assay was carried out and flow cytometry was used to detect the proliferation and activation of T lymphocytes in local lymph nodes.Results Ear swelling respouse was observed 6 days after ear challenge with DNFB in primary ACD,which was the same as the time interval from back elicitation to occurrence of ear inflammation in classical ACD.Kinetics of the inflammatory response to DNFB during primary ACD Wag similar to that during classical ACD.In both models,the ear tissues were mainly infiltrated by mononuclear cells;a significant increase was observed in the tissue levels of IL-2 and INF-γ as well as in the proliferation and activation of T lymphocytes in local lymph nodes;while there were no significant changes in the level of IL-4,compared with the normal control mice.Conclusions Similar to classical ACD,Thl type cell-mediated immune responses were reproduccd in primary ACD mousc model,and each model can take the place of the other in comparative research.
2.Values of serum AFP,GGTⅡ and GP73 in diagnosis of primary hepatocellular carcinoma

Objective To explore the early diagnostic values of serum alpha -fetoprotein (AFP),gamma -glutamyltransferase Ⅱ (GGTⅡ),and Golgi protein 73 (GP73)in patients with primary hepatocellular carcinoma (PHC).Methods The serum specimens of 100 pa-tients with liver diseases (50 cases of hepatitis and liver cirrhosis and 50 cases of PHC)and 50 healthy people were collected in our hospital from February 2013 to February 2014.Electrochemical luminescence technique,specific immuno -membrane adsorption assay,and enzyme-linked immunosorbent assay were used to measure the serum levels of AFP,GGTⅡ,and GP73.Comparison of continuous data between multiple groups was made by analysis of variance,and comparison between two groups was made by q test.The receiver operating character-istic (ROC)curves of single or combined test results were made,and the areas under the ROC curves (AUCs)were calculated.The sensi-tivity,specificity,and AUCs of AFP,GGTⅡ,GP73,and the combined test were analyzed and compared.Results The level of serum GGTⅡ in the PHC group was significantly different compared with those in the other two groups (F =16.224,P <0.05),but there was no significant difference between the normal group and the hepatitis and liver cirrhosis group (P >0.05).Significant differences in serum levels of AFP and GP73 were observed by paired comparison between the PHC group,hepatitis and liver cirrhosis group,and normal group (F =193.128,F =20.231,P <0.05 for both).When assayed alone,the specificities of GP73,GGTⅡ,and AFP were 69%,64% and 51%, respectively,and the sensitivities were 92%,84%,and 76%,respectively.In combined test,the specificity was 94.6% and the sensitivi-ty was 98.8%.Conclusion The GP73 test is the best performer in the single assays.Combined test of serum AFP,GGTⅡ,and GP73 shows a good diagnostic value for PHC with greatly improved specificity and sensitivity.
3.Mechanism of T helper cell for demyelinated optic neuritis

Background Optic neuritis is one of the common clinical neuro-ophthalmic diseases.Recurrent optic neuritis can cause irreversible axonal damage and visual impairment,and no prevention method is effective up to now.Its pathogenesis is considered to be associated with the body's immune imbalance.Objective This proposal tried to establish experimental autoimmune encephalomyelitis (EAE) and explore the immunomodulatory effects of T helper (Treg),Th17 cell in the model of optic neuritis.Make the immune mechanism of demyelinating optic neuritis clear.Methods EAE model was established to observe the clinical symptoms and visual electrophysiological changes of optical neuritis in mice;hematoxylin-eosin staining to observe the histological changes in the mouse models of EAE; immunohistochemical staining to detect optic nerve axon damage markers β-amyloid peptide precurso (β-APP) protein expression quantity changes;reverse transcription PCR (RT-PCR) to detect the dynamic expression of forkhead/winged helix transcription factor p3 (Foxp3),transforming growth factor-β (TGF-β),interleukin-1β (IL-1β),IL-17,IL-10 in optic nerve tissue.Results After immunization 11 days,neurological symptoms of EAE mice started to appear,hematoxylin-eosin staining showed optic nerve tissue infiltration of inflammatory cytokines; immunohistochemical staining showed enhancement of axonal injury markers β-APP positive staining; flash visual evoked potential(F-VEP) test showed 7,11,14,19,23,28 days,compared with the normal mice,P1 prepatents of model group were prolonged after modeling,the differences were statistically significant (t =4.487,15.203,16.364,11.540,11.959,16.163,all at P<0.05).When 7 days after modeling,the difference of N1-P1 amplitude between normal group and model group was no statistical significance (t =-0.992,P =0.378); while 11,14,19,23,28 days after modeling,compared with normal mice,N1-P1 amplitudes of model mice were significantly lower,the differences were statistically significant(all at P< 0.05).The expressions of TGF-β,IL-1β,IL-17,Foxp3,IL-10 mRNA among different time points had stastistie significant differences (F =12.721,15.015,14.343,69.374,68.290,all at P =0.000),compared with the normal group,when 11 days,14 days after modeling,TGF-β mRNA levels were significantly increased ; 19 days,23 days after modeling,IL-1β mRNA levels were significantly increased;7 days,11 days after modeling,IL-17 mRNA levels were significantly increased ;7,11,14,19,23,28 days after modeling,Foxp3 mRNA levels were significantly lower; 19,23,28 days after modeling,IL-10 mRNA levels were significantly lower,the difference were statistically significant (all at P< 0.05).Conclusions Th17 cells launch demyelinating optic neuritis immune injury,and Th1 subgroup performs the function of maintaining inflammatory lesions,Treg cell subgroup is abnormal much earlier than that of the Th2 subgroup.The imbalance of Th17/Treg cell may be involved in the pathogenesis of demyelinating optic neuritis.
4.Validity and reliability of the simplified Chinese version of modified Barthel index for Chinese stroke patients

Objective To study the validity,reliability and sensitivity of the simplified Chinese version of modified Barthel index (MBI) in evaluation of activities of daily living(ADL) of Chinese stroke patients. Methods Forty Chinese stroke patients (26 males and 14 females,aged 59.65±11.68 years) participated in the study.The duration of the disease averaged 21.20±19.44 days.All the patients were evaluated for three times with MBI.Barthel Index (BI) and the motor component of Functional Independence Measure (M-FIM)independently by twophysiatrists.Spearman correlation coefficients between MBI and Barthel index (BI),between MBI and M-FIM were used to test the validity of MBI.Intraclass correlation coefficients (ICC) were used to test both interrater and intra rater reliability of MBI.Sensitivity of MBl was analyzed with the Wilcoxon signed rank test and paired t-test.Results MBI was highly correlated with BI in each item and total score(r=0.816~1.000,P<0.01),and with M-FIM in total score(r=0.935~0.981,P<0.01).There were high reliability with MBI in the results collected by same assessor and different assessors(ICC=0.866~0.997).Furthermore,MBI was more sensitive than BI in majorities of its items and its total scores as well.Conclusions MBI has good validity and reliability as well as sensitivity in assessment of ADL of Chinese stroke patients.

Endoplasmic reticulum is an important organelle in eu-karyotic cells.Endoplasmic reticulum stress (ERS)is usually triggered under cell hypoxia,carbohydrate undersupply or medi-cal treatment.Now,present studies show that ERS could activate several cell signal pathways and the UPR pathway is most widely researched.When UPR activates cell signal pathway,it can reg-ulate cancer drug resistance by involving with DNA damage re-pair,apoptotic suppression and autophagy.
6.PREDICTIVE FACTORS OF LIFE QUALITY OF ADULT PATIENTS WITH CHRONIC TONSIL-LITIS AFTER TONSILLECTOMY

Objective To explore the predictive factors of life quality of adult patients with chronic tonsillitis after tonsillectomy .Methods 68 adult patients with chronic tonsillitis treated with tonsillectomy admitted to our hos-pital from January, 2013 to June, 2014 were divided into the observation group (SF-36 score<50 points, n=38) and the control group (SF-36 score>50 points, n=30) according to SF -36 scale scores 1 week after operation. Life quality ( SF-36) score of the 2 groups before and after treatment were statistically analyzed .The predictive fac-tors of life quality of adult patients with chronic tonsillitis after tonsillectomy were analyzed by single factor analysis and logistic factors analysis .Results Life quality scores of preoperative eight dimensions in the 2 groups including physiological functions, physiological roles, physical pain had no statistically significant difference (p>0.05).Life quality scores of the two groups at weeks 1, 2 and 4 after surgery were higher than those before surgery .Life quality scores of the observation group after treatment were lower than those of the control group in the same period ( p<0.05).The results of single factor analysis showed that relative factors of life quality of adult chronic tonsillitis after tonsillectomy were gender, age, Hamilton depression rating scale (HAMD) scores and Hamilton anxiety rating scale ( HAMA) scores, etc..The results of logistic multifactor analysis showed that age , HAMD scores and HAMA scores were independent predictors of life quality of adult chronic tonsillitis after tonsillectomy .Conclusion Independent predictive factors of life quality of adult chronic tonsillitis after tonsillectomy includw age , HAMD scores and HAMA scores.Therefore, diets and sports can improve disease resistance ability of aged patients .And psychological condi-tion regulation can also effectively improve patient's life quality .
7.Validity and reliability of Wolf motor function test on assessing the upper extremities motor function of Chinese stroke patients in acute stage

Objective: To study validity and reliability of Wolf motor function test (WMFT) on assessing the upper extremities motor function of acute stroke patients so as to guide clinical application. Method: Twenty-three stroke patients in acute stage participated in the study. Patients were assessed twice with WMFT and the upper extremities motor function test of Fugl-Meyer movement assessment (U-FMA) within one week. The correlation between the assessment results of WMFT and U-FMA were analyzed to study the validity and reliability of WMFT. Result: WMFT was highly correlated with U-FMA in total scores (r =0.922, 0.929, P<0.001). There were high correlation in all items of WMFT between two assessment sessions. The intraclass correlation coefficients (ICC) were 0.989, 0.997 for intrarater and 0.980, 0.991 for interrater. Conclusion: The WMFT is valid and reliable on assessing upper extremities motor function of stroke patients in acute stage.
8.Diagnosis and treatment of inherited platelet function disorders

Inherited platelet function disorders(IPFD)is a rare hereditary disease characterized by various degrees of bleeding tendency with or without thrombocytopenia.Due to the lack of standardized evaluation system in laboratory test, the diagnosis and classification of IPFD are difficult.Thus, the incidence of IPFD may be underestimated.At present, combined with symptoms and laboratory tests, the second generation sequencing technology is conductive to the fast and accurate diagnosis of complex hemorrhagic diseases of this type and can prompt assessment and treatment.However, there still exists no effective targeted treatment for IPFD.This paper was aimed at introducing the diagnosis and treatment of IPFD.
9.Preliminary Studies on Factors Affecting Content of Magnolol in Magnolia Bark Three Agents Decoction

Objective: To study the factors affecting the content of magnolol which is one of the main active components in Magnolia Bark Three Agents Decoction. Methods: The contents of magnolol in extracts of Magnalia Bark Three Agents Decoction (medicinal materids were decocted alone, together or in different compatibleness, and residuals) were determined by HPLC. The effects of use amount of Radix et Rhizoma Rhei and Fructus Aurantii Immaturus, decoction time, and extraction times on magnolol content were studied by orthogonal design.Results: The content of magnolol in Magnolia Bark Three Agents Decoction of which medicinal materials were decocted together was over double that of which they were decocted alone. The dissolution sate of magnolol increased with the increasing of the use amount of Fructus Aurantii Immaturus and extraction times. Conclusion: Fructus Aurantii Immaturuns and extraction times are the main factors affecting magnolol content in Magnolia Bark Three Agents Decoction.
10.Effect of poly (ADP-ribose) polymerase inhibitor on liver metastasis of colorectal carcinoma CT26 cell line in mice

Objective To investigate the effect of poly (ADP-ribose)polymerase (PARP) expression inhibited by 5-AIQ on the liver metastasis in colorectal carcinoma CT26 cell lines. Methods Liver metastasis model of colorectal carcinoma was established by intrasplenic inoculation of colorectal carcinoma CT26 cell in BALB/c mice. Treated by 5-AIQ (100 or 500 ?mol/L),CT26 cells were inoculated to the spleen of the mice,or the rats were first inoculated of CT26 cells,then treated by 5-AIQ (3 or 10 mg/kg) ip. The changes spleen primary carcinoma and liver metastasis carcinoma were observed. The PARP expression in spleen primary carcinoma was detected by Western blotting. Results Significant difference was found in the splenic carcinoma size,the number of liver metastatic nodules,the grading of liver metastatic nodules in the 5-AIQ treated group and control group(P0.05).
