1.Research progress in topical application of anti-vascular endothelial growth factor drugs for uveitis-associated cystoid macular edema and choroidal neovascularization
Chinese Journal of Experimental Ophthalmology 2014;32(7):659-663
Uveitis is a kind of inflammatory disease affected in ocular posterior segment.Uveitis sustains a long duration and causes a significant damage of visual function. It often leads to serious complications, such as cystoid macular edema (CME),cataract, glaucoma, retinal detachment and choroidal neovascularization (CNV).In the pathological process of uveitis, inflammatory factors promote the release of vascular endothelial growth factor (VEGF) through a variety of pathways. It has been verified that VEGF level is elevated in vitreous in the patients with uveiticinduced CME.Intravitreal injection of anti-VEGF drugs, including ranibizumab and bevacizumab, can improve the visual acuity in most patients with noninfectious uveitic-induced CME and CNV.However, intravitreously repeated injections of anti-VEGF drugs may be required in some patients.Intraocular injection of anti-VEGF drugs is an optional and effective way for those with persist macular edema and not being suitable for the use of corticosteroid or immunosuppressive therapy.However,anti-VEGF drugs may affect the efficacy of corticosteroid. Therefore, more clinical and basic researches are still needed. The status and progresses in the use of anti-VEGF drugs for uveitisinduced CME and CNV were reviewed.

Journal of Central South University(Medical Sciences) 2014;(11):1186-1190
Objective: To investigate the effect of chloride concentration on the corrosion of Co-Cr alloy and pure Ti in a simulated oral environment. Methods: The electrochemical corrosion tests of pure Ti and Co-Cr alloy were carried out in neutral artiifcial saliva solutions with different NaCl concentrations (0.9%, 2.0%, and 3.0%). hTe morphologies of corroded surface for pure Ti and Co-Cr alloy were observed by scanning electron microscope (SEM). Results: hTe changes in the self-corrosion potentials (Ecorr) for pure Ti and Co-Cr alloy in three kinds of artificial saliva solutions was not obvious. However, the self-corrosion current densities (Icorr) of pure Ti were much lower than those of Co-Cr. TheIcorr of Co-Cr alloy increased in a concentration-dependent manner of NaCl, whereas the breakdown potential (Eb) of Co-Cr alloy decreased in a concentration-dependent manner. hTe potential ranged for the breakdown of oxide film (Ev) was shortened in a concentration-dependent manner of NaCl. There was no obviousdifference in theIcorr of pure Ti with different concentrations of NaCl. hTe breakdown potential was not seen according to the polarization curves. Conclusion: In a certain range, the increase of the concentration of Cl- leads to accelerate the corrosion behavior of Co-Cr alloy, but it does not affect pure Ti.

BACKGROUND:The differentiation of neural stem cells (NSCs) is a hot research. Whether brain-derived neurotrophic factors (BDNF) can induce the differentiation of NSCs into neurons has not been detailed studied. OBJECTIVE:To investigate the effect of different concentrations of BDNF on the differentiation of NSCs from adult rat hippocampus into neurons in the medium with epidermal growth factor. DESIGN:A controlled study. SETTING:Department of Human Anatomy, China Medical University. MATERIALS:The experiment was performed at the Neurotomia Laboratory of China Medical University in August 2007. NSCs isolated from hippocampus of adult SD rats were inoculated in a serum-free medium. Three clean adult SD rats (200-250 g each) were provided by Laboratory Animal Department of China Medical University. Dispositions to the rats were consistent with ethical standards of animals. Epidermal growth factor (EGF) and BDNF were bought from R&D Company. METHODS:①NSCs obtained from rat hippocampus were cultured in the serum-free medium containing basic fibroblast growth factor (bFGF), EGF and B27. ②Single cell clone was obtained via limited dilution for the fourth passage of cells. Identification of NSCs for the passage of cells from monoclonal spheres was performed by Nestin immunocytochemistry. Neurons and astroglial cells were identified by immunocytochemistry for neuron specific enolase (NSE) and glial fibrillary acidic protein (GFAP) a week after differentiation. ③Differentiation for the monoclonal spheres was done to appraisal the proportion of neurons. NSCs were divided for several groups containing 0 μg/L, 50 μg/L, 100 μg/L, 150 μg/L, and 200 μg/L groups according to the doses of BDNF. EGF was added in the media of each group. Immunocytochemistry for NSE was done a week later to count the positive cells. MAIN OUTCOME MEASURES:Proportion of neurons differentiated from NSCs.RESULTS:①NSCs immunocytochemical staining showed that Nestin was positive in monoclonal spheres, and NSE and GFAP were positive in differentiated cells. ②The proportion of differentiation from NSCs into neurons was significantly higher in groups treated with 50 μg/L and 100 μg/L BDNF compared to other groups(t=2.502-5.025, P < 0.05), but there was no significant difference between 50 μg/L and 100 μg/L BDNF groups(P > 0.05). Moreover, no significant difference was detected among 0 μg/L, 150 μg/L and 200 μg/L BDNF groups (P > 0.05). CONCLUSION:The optimal concentration of BDNF is 50 μg/L for differentiation from NSCs into neurons while the concentration of EGF is 20 μg/L.

World Science and Technology-Modernization of Traditional Chinese Medicine 2013;(3):501-508
This study was aimed to explore the mechanism of Xin-Feng capsule (XFC) on pulmonary function of Sjogren's syndrome (SS) rat model based on the TGF-β1-ERK1 signal pathway. A total of 50 SD rats were ran-domly divided into the normal control (NC) group, model control (MC) group, hydroxychloroquine sulfate (HCQ) group, radix paeoniae alba (total glucosides of paeony capsules, TGP) group, and the XFC treatment group, with 10 rats in each group. Except the NC group, the complete Freund's adjuvant (CFA) and the same rat sub-mandibular gland antigen induced method were applied to each rat after metatarsus injection on two feet and CFA after sufficient emulsification of 0.2 mL submandibular gland protein mixed antigen induced SS. The water drink-ing quantity, body weight change, pulmonary function, ERK1, TGF-β1 protein expression and serum cell factor (IL-17, IL-4) changes were detected in each group. The results showed that compared with the NC group, the body quality and serum IL-4 of MC rats were obviously decreased, and the water drinking quantity, submandibu-lar gland / lung index, submandibular gland pathological score, ERK1, TGF-β1 integral and IL-17 were increased (P < 0.01 or P < 0.05), and the pulmonary function parameter was decreased (P < 0.01 or P < 0.05). Compared with the MC group, the body quality, pulmonary function parameters FEF50 and MMF of the XFC group were in-creased, the IL-4 expression was increased, the water drinking quantity, submandibular gland / lung index, sub-mandibular gland pathological score, serum IL-17 expression, ERK1 and TGF-β1 integral were decreased (P <0.01 or P < 0.05). Compared with the HCQ group, the body quality and IL-17 were significantly decreased in the XFC group (P < 0.01), FEF25, FEF75 and MMF were increased (P < 0.01 or P < 0.05). Compared with the TGP group, the lung index and IL-17 were decreased in the XFC group (P < 0.01 or P < 0.05). It was conclud-ed that the lung function of SS rats was declined, which is closely related to the activation of TGF-β1-ERK1 sig-nal pathway. Traditional Chinese medicine of XFC is able to downregulate the TGF-β1 expression and to restrain epithelium-mesenchymal cell transformation, in order to inhibit ERK1 phosphorylation activation and proliferation, decrease immune inflammation, in order to improve SS and lung function.
