1.Bioequivalence evaluation of secnidazole tablets in healthy male volunteers

AIM: To compare the bioavailability of the test and reference formulation of secnidazole (2 g) tablets under fasting conditions. METHODS: This bioequivalence study was carried out in 20 healthy male Chinese volunteers according to a single dose, two-sequence, crossover randomized design. Fifteen blood samples per period were collected over 96 h, and plasma secnidazole concentrations were determined by locally validated high performance liquid chromatography (HPLC) assay and pharmacokinetic parameters were analyzed by the non-compartmental and compartmental methods. RESULTS: Plasma concentration-time profiles were adequately described by a one-compartment open model with first-order absorption. The main pharmacokinetic parameters of secnidazole test and reference tablets were as follows: tmax were (2.30±1.06) and (2.28±1.10) h, Cmax were (49.63±6.35) and (46.17±4.24) mg/L, t1/2 were (28.84±3.41) and (29.05±4.01) h, AUC0-96 were (1832.06±180.15) and (1847.14±204.14) mg·h-1·L-1, respectively. The relative bioavailability of test tablets was (99.99±11.92)%. CONCLUSION: The results indicate that the two formulations of secnidazole tablets are bioequivalent in the rate and extent of absorption.
2.The effects of fluor-hydroxyapatite coating on the osseointegration and peri-implantitis of orthodontic micro-implants

Objective:To study the effects of fluor-hydroxyapatite (FHA) coating titanium alloy on the osseointegration and peri-implantitis of orthodontic micro-implant.Methods:Titanium of FHA alloy (FHA group) and titanium alloy(control group) orthodontic micro-implants were respectively planted into buccal alveolar bone in mandibular premolar area of rabbits.Scanning electron microscope was used to observe the osseointegration around the micro-implants.ELISA was employed to detect TNF-α in the gingival crevicular fluid around the implants.Results:The FHA-coating titanium alloy orthodontic micro-implants led to higher bone density,smaller marrow cavity,and lower TNF-α level and shorter lasting period of TNF-α over-expression than the controls (P < 0.05).Conclusion:The FHA-coating titanium alloy orthodontic micro-implant has better histocompatibility and may inhibit peri-implantitis.

Objective To evaluate changes in retinal expression of three heat shock proteins(HSPs)HSP27,HSP70 and HSP90 in normal and experimental glaucomatous rats's retina,and to explore the potential relationship between HSPs with glaucoma optic neuropathy.Design Experimental study.Participants 60 Wistar rats.Methods Rats were randomly divided into ocular hypertension group(n=30)and sham control group(n=30).The right eye was designed as the experimental eye,and the left as the control eyes.Intraocular pressure(IOP)was elevated by using underwater bipolar electro coagulation condensate 3 episcleral and limbal veins on the right eye of rats to establish of animal models of glaucoma.IOP were monitored twice weekly with a Tonopen.In treated eyes,IOP ranged from 27 to 35 mmHg throughout the study.Rats were killed with euthanasia at 10,20,or 60 days following the occurrence of ocular hypertension,with retina dissected free and protein isolated.Protein was used for Western blot analysis and probed with specific HSP antibodies,and normalized to ?-actin levels.Main Outcome Measures IOP,Western bolt.Results Western blot analysis demonstrated that HSP27 protein levels in the retinas were elevated as much as 197% at 10,20 and 60 days following the induction of ocular hypertension.No changes in protein levels were observed for HSP70 or HSP90 in retina from ocular hypertensive eyes.Conclusions The stress protein HSP27 is upregulated in retinas from ocular hypertensive rats.No changes in HSP70 or HSP90 were observed.The upregulation in HSP27 appears to be a gene specific event associated with elevated IOP,its expression may be a potential relationship associated with optic neuropathy in glaucoma.
5.Comparison of the Effect of Different Adjustment Methods (Occlusion) in the Restoration of Posterior Teeth

Objective To investigate the effect of different adjustment methods (occlusion) in dental implant repair. Methods 96 cases of patients with dental implant repair treated in our hospital from June 2015 to February 2017 were selected as the research object and randomly divided into the control group and the observation group with 48 cases in each group. The biting surfaces of the patients were reduced in the observation group, but the patients in the control group were treated with the original adjustment methods (occlusion). The bite balance and masticatory function of the two groups were evaluated at 3 months and 6 months after repair respectively. The occlusal balance was set up by computer to set the X and Y axes, and the X-axis and Y-axis values of the two groups were monitored. And measured the molars asymmetry index(AMOF), to calculate the ratio of each tooth occlusal force, and the larger the AMOF value, the more asymmetrical the bite force is. Chewing function score of 10 points, the higher the score that chewing better. Results Compared with the control group, the indexes of occlusal balance in the observation group at 3 months after repair were significantly increased (P<0. 05), and at 6 months after repair, the AMOF (P<0. 05). Compared with the control group, the scores of chewing function in the observation group at 3 months and 6 months after repair were significantly increased, there was statistical difference between the two groups Significance (P<0. 05). Conclusion The effect of biting surface reduced (occlusion) in dental implan.
6.The effects of β tricalcium phosphate/collagen scaffold loaded with human bone morphogenetic protein 2 plasmid on the osteogenesis ability of MC3T3-E1 cells

Objective:To study the effects of beta tricalcium phosphate(β-TCP)/collagen scaffold loaded with human bone morphogenetic protein 2 (hBMP2) plasmid on the osteogenesis ability of MC3T3-E1 cells.Methods:hBMP2 DNA plasmid-modified β-TCP/collagen scaffold and the naked plasmid(control) were constructed.MC3T3-E1 cells were respectively in vitro cultured onto the β-TCP/collagen scaffold with hBMP2(Z) and with control plasmid(Z0),on peace dish with the saffold and hBMP2(M) and with the control plasmid(M0).The surface morphology of the samples was observed by SEM.Osteogenesis of the cells was examined by alkaline phosphatase activity(ALP) test,real-time fluorescent quantitative PCR for the detection of Runx2,OCN,ALP and OPN mRNA expression.Data were statistically analyzed.Results:The composite sample surface of plasmid DNA containing hBMP2 modified β-TCP/collagen was porous;group Z and M showed highter ALP activity and higher mRNA expression of Runx2,OCN,ALP and OPN than group Z0 and M0;so did group Z than group M.Conclusion:Porous β-TCP/collagen scaffold loaded with BMP2 DNA is potential for osteoinduction.
