1.The Changes of Circulating Th17 and Tc17 Cells in Patients with Colorectal Polyp,Adenoma and Cancer

Objective To compare ratio of circulating Th17 over Tc17 cells in patients with colorectal polyp (CRP), colorectal adenoma (CRA) and colorectal cancer (CRC),and analyse their relationship. Methods The ratio of circulating Th17 over Tc17 cells in 93 patients(17 CRP, 34 CRA and 42 CRC)and 17 healthy donors were examined by flow cytometry;Serum levels of IL-17A and IL-23 were tested by ELISA. Results The ratio of circulating Th17 over Tc17 cells and the se-rum levels of IL-17A and IL-23 were the lowest in healthy control groups, medium in CRP and CRA groups and the highest in CRC groups (P<0.05). The proportions of Tc17 cells in CRA groups were significantly higher compared to those in CRP groups,but no statistical significance of proportion of circulating Th17 cells as well as serum levels of IL-17A and IL-23 be-tween CRA and CRP groups was observed (P>0.05). Further analysis showed that there was a positive correlation between proportions of circulating Th17 and Tc17 cells (rs=0.594, P<0.001). Conclusion The proportions of circulating Th17 and Tc17 cells were positively related and the ratio of Th17 over Tc17 was markedly increased from CRP group or CRA group to CRC group.
2.Clinical Study on Different Acupuncture and Moxibustion Methods for Prevention and Treatment of Nausea and Vomiting Caused by Chemotherapy

Objective To observe efficacy of different acupuncture and moxibustion methods for treatment of nausea and vomiting caused by chemotherapy. Methods One hundred and sixty-three cases were randomly divided into electroacupuncture group (42 cases), warm acupuncture combined with electroacupuncture group (37 cases), ginger moxibustion group (44 cases) and control group (40 cases). All groups received postoperative adjuvant chemotherapy. Three acupuncture and moxibustion groups received electroacupuncture, warm acupuncture combined with electroacupuncture, ginger moxibustion to stimulate bilateral Zusanli (ST36), Neiguan (PC6), respectively, once a day. The control group was given tropisetron hydrochloride 5 mg+normal saline 100 mL, 30 min before chemotherapy intravenously, once a day to the end of chemotherapy. The number and degree of vomiting and the serum 5-HT content were tested, and the safety test was conducted. Results The complete control rate and effective control rate of acute vomiting of electroacupuncture group and warm acupuncture combined with electroacupuncture group were higher than those of ginger moxibustion group and control group (P<0.05). The complete control rate of delayed vomiting in warm acupuncture combined with electroacupuncture group was significantly higher than that of electroacupuncture group, ginger moxibustion group and control group (P<0.05); The effective control rate was higher than that of ginger moxibustion group and control group (P<0.05); Electroacupuncture group and Ginger moxibustion group were higher than the control group (P<0.05). The scores of acute vomiting in electroacupuncture group and warm acupuncture combined with electroacupuncture group were lower than others (P<0.05). In delayed vomiting, warm acupuncture combined with electroacupuncture group's score was lower than other groups, with statistical significance (P<0.05); Electroacupuncture group and ginger moxibustion group were lower than that incontrol groups, with statistical significance (P<0.05). The levels of serum 5-HT in electroacupuncture group and ginger moxibustion group were higher than warm acupuncture combined with electroacupuncture group (P<0.05), but lower than the control group (P<0.05). No obvious adverse reactions were found in each group. Conclusion Different acupuncture methods had different clinical effects on acute vomiting and delayed vomiting caused by chemotherapy.
3.Changes of circulating Tc17 cells in progression of colorectal cancer

Objective:To explore the changes and possible mechanisms of circulating Tc 17 cells in patients with colorectal cancer ( CRC) following disease progression .Methods:The peripheral blood were collected from 54 patients with colorectal cancer . the proportions of Tc17 cells in peripheral blood mononuclear cells (PBMCs) and cultured PBMCs treated by IL-1β, IL-6 and TGF-βin different concentrations were determined by flow cytometry;the levels of IL-1β, IL-17A, IL-23 and IL-6 in sera were measured by ELISA.Results:The proportions of Tc17 cells and the levels of IL-1β, IL-17A, IL-23 and IL-6 were significantly higher than those in healthy controls (P<0.05).Compared with early CRC, the proportions of Tc17 cells and the levels of IL-1β, IL-17A in advanced CRC were significantly decreased, but the level of IL-6 was significantly increased (P<0.05).The difference of IL-23 level between early and late groups was not significant (P>0.05).In vitro experiments confirmed that IL-1βor high concentrations of IL-6 and TGF-βcould significantly increase the number of Tc 17 cells in PBMCs.Conclusion: The changes of circulating Tc 17 cells in the progression of colorectal cancer are possibly modulated by IL-1β, IL-23, IL-6 and TGF-β.

Objective To investigate the effects of bone marrow mesenchymal stem cells administration on VEGF,SDF-1 and TGF-?1 expressions and heart function in myocardial infarcts rats.Methods MSCs were harvested from SD rats,cultured and passaged in vitro,then the cells were purified by density gradient centrifugation and adhesive-screening method.Rats were randomly divided into 2 groups.In the control group(n=8) the animals were treated with saline of the same volume as placebo.Four weeks after the injection,the VEGF,SDF-1 and TGF-?1 expressions and heart function were examined.Results Four weeks after the transplantation,Immunohistochemistry showed expression of VEGF and SDF-1 increased markedly in experiment group,and expression of TGF-?1 decreased.Heart function was improved in the experiment group(P
5.Determination of Particle Size of Gefitinib by Laser Scattering Method

Objective:To establish and validate a method for the determination of particle size and size distribution of gefitinib. Methods:A Malvern Mastersizer 3000 laser size analyzer and a Hydro LV wet autosampler were used,a light scattering method for the determination of particle size and size distribution was adopted to analyze the particle size of gefitinib,and then the methodological study was carried out. The pump speed was 2 000 r·min -1 ,the obscuration was 8% -20% ,the measurement time of background and sample was 10 s,the sample RI was 1. 500,and the sample absorbance was 0. 00. Results:The D10 of 3 batches of gefitinib was less than 7 μm,the D50 was less than 15 μm and the D90 was less than 25 μm,which all met the preparation requirements of final products. Conclusion:The method is accurate,simple and repeatable,and suitable for the particle size analysis of gefitinib.

Objective To investigate the effects of DanzhiⅠ on rats acute skin trauma model.Method Perforate back of the rats was made with special puncher to establish acute trauma model.Wound surface was applied with Danzhi Ⅰ and the healing process was observed.Result Danzhi Ⅰ significantly shrinked the rim of raw surface and shorten the time of healing.The number of completely healed rats increased obviously.Conclusion Danzhi Ⅰ can significantly accelerate the healing of acute skin trauma of rats.
7.miR-34a partially reverses inhibition of CEES-exposed keratinocytes migration via ERK1/2 pathway

Objective To explore the effect of 2-chloroethyl ethyl sulfide(CEES) poisoning on keratinocyte migration and the regulatory role of microRNA(miR)-34a.Methods MTS was used to detect the viability of cells exposed to CEES in order to select an appropriate dose of CEES exposure in this in vitro model.The protein level of keratin 5 and keratin 10 was detected to assess cell differentiation status .Scratch assay was applied to evaluate cell migration ,and miR-34a silencing in keratinocytes was achieved by transfecting chemically synthesized miR-34a specific miRNA inhibitor.t-ERK1/2 and p-ERK1/2 levels closely related to cell migration were detected using Western blotting .Results An in vitro CEES exposure model of keratinocytes was established at the optimal concentration of 0.5 mmol/L CEES in the viability test , and this dose was chosen to evaluate cell migration changes .The migration of cells was significantly inhibited 24 h after CEES exposure , accompanied by no changes in morphology and keratin 5/10 levels.Silencing of miR-34a significantly increased the migration of cells exposed to CEES , which could be blocked by adding 5 μmol/L U0126 , an ERK1/2 phosphorylation selective inhibitor.Conclusion Silencing of miR-34a can significantly increase keratinocyte migration and partially reverse the inhibition of CEES-caused migration , which could be mediated by ERK 1/2 pathway activation .

Nerve agent not only inhibit acetylcholinesterase ( AChE) at an early stage, but also induce prolonged and progressive neuroinflammation and delayed neurodegeneration.Recently, the US National Institute of Health ( NIH) has sponsored some major programs of toxic mechanisms and treatment of nerve agents, which aims at the development of quick and effective treatment to acute intoxication and delayed effect.The experimentally effective new antidotes mainly include AChE-targeting drugs, broad-spectrum reactivators and scavengers, antiinflamatory and nerve protection drugs.
9.Effects of cognitive behavioral therapy on psychological and physiological status of hypertension patients in community

Objective To investigate the influence of psychological intervention in emotional disorder on blood pressure,lipids and glucose in hypertensive patients in community.Methods 599 hypertensive in community were divided to intervention group (n =302) and control group (n =297) by randomization.The control group received routine medication.Additionally the intervention group got another psychological intervention which lasted 6 months.An SCL-90 scale was adopted to evaluate patients'psychological change of mental health,blood sugar,low density lipoprotein cholesterol,total cholesterol,systolic pressure and diastolic blood pressure before and after treatment.Results Somatization,compulsion,interpersonal relationship,depression,anxiety,hostility and terror in intervention group((1.44±0.29),(1.78 ±0.39),(1.66±0.38),(1.63 ±0.48),(1.43 ±0.35),(1.37 ± 0.52) and (1.28 ± 0.41) respectively) had a lower score than control group ((2.01 ± 0.59),(2.04±0.68),(1.88 ±0.38),(1.84±0.59),(1.92±0.54),(1.61 ±0.53)and(1.60±0.50)).The differences were significant(P< 0.01).After intervention,blood glueose,low density lipoprotein,total cholesterol,systolic pressure,and diastolic pressure were ((4.65 ± 0.811) mmol/L,(2.97 ± 0.702) mmol/L,(4.86 ± 0.884)mmol/L,(134.11 ± 13.39)mm Hg(1 mm Hg =0.133 kPa) and (93.73 ±7.61)mm Hg respectively) and before intervention((5.09 ± 0.710) mmol/L,(3.23 ± 0.584) mmol/L,(5.36 ± 1.138) mmol/L,(150.13 ± 12.23) mm Hg and (89.64 ± 8.35) mm Hg respectively).The differences were significant (P< 0.05).Conclusion Cognitive behavioral therapy can improve community hypertension patients' physiology and psychology,which is good for blood pressure,lipids,glucose control.
10.Effect of glucose concentrations on energy metabolism in 16 HBE cells exposed to CEES

Objective To compare the changes in energy metabolism in 2-chloroethyl ethryl sulfide(CEES)-poisoned bronchial epithelial cell 16HBE cultured in media at different glucose concentrations .Methods Bronchial epithelial cell 16HBE was cultured in high (4.5 mg/ml) or low (1.1 mg/ml) glucose medium and exposed to a sulfur mustard simulant CEES of 0.2, 0.5, 1.0 mmol/L.Cell growth and cytotoxicity were tested using MTS .ATP, ADP and AMP were detected by HPLC and the value of ATP/ADP, total adenine nucleotides ( TAN) and energy charge ( EC) was subsequently calculat-ed.Mitochondrial oxidative phosphorylation-related proteins, COX-10 and ISCU, were detected using Western blotting . Rhodamine 123 was applied to detect the mitochondrial membrane potential using flow cytometry .Results Low glucose accelerated the growth and energy metabolism of 16HBE cells in regular culture , and the contens of ADP , TAN, COX-10 and ISCU in low glucose group were significantly higher than those in high glucose group .CEES exposure (≥0.5 mmol/L) significantly affected cell viability in both high and low glucose groups , with significant difference between the two groups exposed to 1.0 mmol/L CEES.In high glucose group, 24 h after 0.5 or 1.0 mmol/L CEES exposure, the contents of ATP, ADP and TAN were significantly increased , while ATP/ADP and EC decreased .In low glucose group , ADP, AMP and TAN significantly decreased, while ATP/ADP and EC increased 24 h after 1.0 mmol/L CEES exposure.The mi-tochondrial membrane potential (MMP) also changed differently after 0.5 mmol/L CEES exposure.MMP in high glucose group marginally increased at 3 h, and significantly increased at 8-12 h (P<0.05), and returned to normal at 24 h. MMP in low glucose group showed a transient decrease at 5 h (P<0.01), and back to normal at 8 h.The protein levels of COX-10 and ISCU were significantly increased in high glucose group 24 h after 0.5-1.0 mmol/L CEES exposure , but sig-nificantly decreased in low one 24 h after 1.0 mmol/L CEES exposure .Conclusion When 16HBE is cultured at a high or low glucose concentration , the cell growth, stress responses and energy metabolism including MMP , COX-10, ISCU and ATP production are in different status before or after CEES exposure .High glucose could protect against CEES exposure .
