1.Effects and mechanism of penhyclidine hydrochloride on myocardial ischemia reperfusion injury in rat

Objective To explore effects and mechanism of penhyclidine hydrochloride (PHC) on myocardial ischemia reperfusion injury in rat.Methods Sixty Sprague Dawley (SD) adult male rats were randomly divided into sham operation group(SHAM group),myocardial ischemia reperfusion injury group(MIRI group) and penhyclidine hydrochloride treatment (PHC group),and 20 rats in every group.Compared left ventricular function of 3 groups after 24 h of ischemia reperfusion.Selected 10 rats of every group and compared the myocardial infarction size after ischemia reperfusion,and the less 10 rats' the level of tumor necrosis factor-α (TNF-αt) and nuclear factor-kB(NF-kB) in myocardial tissue were measured.Results After 24 h of ischemia reperfusion,there was no significant myocardial infarction size in SHAM group,while that in MIRI group were (39.4± 3.7)%,higher than that in PHC group (20.5 ± 2.3)%,and the difference was significant (P < 0.01).The level of left ventricular end-systolic volume (LVESV) of rats in SHAM,MIRI and PHC groups were (0.138 ± 0.024),(0.457 ± 0.032) and (0.280 ± 0.027) ml respectively,left ventricular end-diastolic volume(LVEDV) were (1.012 ± 0.065),(1.395 ± 0.076) and (1.183 ± 0.102) ml respectively,and left ventricular ejection fraction (LVEF) were (85.6 ± 7.3) %,(43.2 ± 4.5) % and (64.8 ± 5.1) % respectively,and there were statistically significant differences between groups (F =54.12,32.70,35.46,P < 0.001).The level of TNF-αtand NF-kB in rats of SHAM,MIRI and PHC group were (5.4 s0.7),(34.2 ±2.9) and (21.6 ± 1.3) pg/mg respectively and (87.6 ± 7.3),(134.2 ± 9.6) and (116.3 ± 8.7) respectively,and there were statistically significant differences between groups (F =59.40,17.97,P < 0.001).There were statistically significant differences between MIRI and SHAM groups,PHC and SHAM groups in terms of above factors (P <0.05).Conclusion PHC can decrease the inflammatory reaction and protect the myocardial ischemia reperfusion injury in rat.
3.Determination of hupzine A in Huperzia serrata by HPLC

Object To compare the hupzine A (Hup A) in Huperzia serrata (Thunb.) Trev. obtained by different extracting methods and investigate the amount of alkaloids and the content of Hup A from different parts of the plants and from different places. Methods Using HPLC for the determination of Hup A. Results The content of Hup A in the stem and leaf is richer than that in the root. The content of Hup A from Guizhou, Guangdong and Anhui Provinces is 0.018%, 0.021% and 0.020% repectively; The difference of extract method of Hup A is no prominence. Conclusion The content of Hup A in the ground is richer than that of underground, and there are some difference in the content of Hup A obtained from different places.
4.Clinical therapeutis effect of ginsenoside Rg3 plus chemotherapy on serum vascular endothelial growth factor in aged gastric cancer patients

Objective To evaluate the effects of ginsenoside Rg3 combined with XELDX regimen on advanced carcinoma of stomach.Methods Totally 93 postoperative patients with advanced gastric cancer were randomly divided into two groups:control group(n = 53) who received only XELDX regimen,and study group (n= 40) who were treated with ginsenoside Rg3 + XELDX.The objective response rate,KPS score and serum VEGF levels in the two groups were detected.Results (1) Before treatment,treatment group and control group showed that serum VEGF levels were (387.63±159.74) μg/L and (378.56± 169.36) μg/L,respectively (P<0.05).After 8 weeks of treatment,serum VEGF of the treatment group decreased to (207.74 ± 115.71) μg/L,the level of the control group VEGF decreased to (246.53 ±107.63) μg/L(P<0.05).(2) The efficacy rate was 57.5% (23/40) in treatment group and 45.3%(24/53) in the control group (P>0.05).(3) KPS score increasing rate was higher in treatment group (65.0%) than in control group (39.6%)(x2=5.87,P<0.05).Conclusions Ginsenoside Rg3 combined with chemotherapy in advanced gastric cancer patients can reduce serum VEGF levels and improve the quality of life of patients.
5.Screening of binding proteins interact with phosphotyrosine-interacting domain of DOC-2 by yeast two hybrid system

Objective: To screen for proteins which can interact with phosphotyrosine-interacting domain (PID) of differentially expressed gene in human ovarian cancer cell line DOC-2 by yeast-two hybrid technique, so as to provide evidence for the signal pathway of DOC-2. Methods: The cDNA sequence of human DOC-2 gene was amplified and its PID domain (nDOC-2) was subcloned into the bait vector pGBKT7 of yeast two-hybrid system; the product was then used to screen an embryo brain cDNA library and the proteins interacting with nDOC-2 were identified. Quadrople dropout(QDO) medium and X-?-gal were used for selecting the positive clones. PCA was used to analyze the amplified sequence. After elimination of the false positive clones, the positive clones were sequenced and analyzed by bioinformatic methods. Results:Twenty-one candidate positive clones were obtained and 3 of them were plasmids encoding Homo sapiens partial mRNA for betaglycan (TBR III gene), Homo sapiens protocadherin gamma subfamily C 3 (PCDHGC3), and APLP1(amyloid beta precursor-like protein 1).Conclusion: The proteins obtained in this study may play important roles in the signal pathway of DOC-2, which provides a new orientation for DOC-2 gene therapy of ovarian cancers
6.Relationship of angiogenesis and clinical characteristics of GA-secreting pituitary adenomas

Objective To investigate MVD, TSP-1, TGF-?1 expression in GH-Secreting Pituitary Adenomas by immunohistochemistry, and to correlate data with clinical characteristics.Methods The protein expression of TSP-1, TGF-?1 in 48 surgical specimens (21 invasive cases; 27 non-invasive cases) of pituitary adenomas was measured using immunohistochemical method. The relationship between the expression and clinical properties was examined. MVD was measured by detecting CD34.Results Compared with the noninvasive group, no difference of expression of CD34(t=2.257; P=0.083) was observed. The expression of TSP-1 in invasive group was low. The expression of TGF-?1 was higher in invasive cases than that in noninvasive ones. The expression of TGF-?1 had positive correlations with MVD; but there was no correlation between the expression of CD34 and the invasion of pituitary adenomas. In addition, MVD count was not associated with the expression of TSP-1. Size, sex or rate of recurrence did not influence MVD and TSP-1 expression. Conclusion MVD values do not necessarily represent angiogenesis in pituitary adenomas. TGF-?1 may increase MVD, and TSP-1 does not affect MVD in pituitary adenomas and angiogenesis may be regulated by other pathway.
7.Outcomes of primary total hip replacement in patients with ankylosing spondylitis

Objective To review the characteristics and outcomes of primary total hip replacement(pTHR) in patients with ankylosing spondylitis(AS). Methods Eighteen patients (26 hips) with AS underwent pTHR from January 1991 to September 2003 were reviewed retrospectively. Sixteen patients (24 hips) were followed-up. The mean duration of follow-up was 85 months (ranging from 23 to 169 months). All patients were evaluated using Harris score standardization. Results There were 15 hips with hip ankylosis( 57.7%). The average time of the operation was 167.4 minutes (ranging from 90 to 350 minutes),the average volume of bleeding in the operation was 773.9 ml (ranging from 300 to 2000 ml),the postoperative average drainage was 488.6 ml(ranging from 50 to 1130 ml). The rate of excellent and good was 87.5 percent. Conclusion The pTHR is a very effective treatment for patients with AS. The perfect rehabilitation care is one of the main factors influencing the outcomes of pTHR for patients with AS.
8.Diagnosis and surgical treatment of a special type of Maisonneuve injury

Objective:To report the diagnosis and surgical treatment of a special type of Maisonneuve injury.Methods:A total of 4 patients were treated at Department of Orthopaedics, Beijing Chao Yang Hospital from January 2015 to July 2019 for Maisonneuve injury. They were 3 males and one female, aged from 34 to 61 years (average, 45.3 years). All injuries were closed, initially manifested as posterior dislocation of the ankle on X-ray films and X-ray and CT re-exams after manual reduction showed fine reduction with no obvious fracture of the ankle joint. Consequently the diagnosis of their Maisonneuve injury was missed in emergency visits, but re-exams in outpatient visits showed separation of distal tibiofibular syndesmosis. All the patients were treated by restoration of the fibular length, fixation of the distal tibiofibular syndesmosis and repair of the triangular ligament. American Orthopaedic Foot and Ankle Society (AOFAS) ankle-hindfoot score was used to evaluate functional recovery of the ankle joint at 12 months after operation.Results:All the 4 patients were followed up for 12 to 14 months (average, 12.7 months). The fractures united after 110 to 185 days (average, 149.3 days). No post-operative complications like infection, delayed union or nonunion were observed. The AOFAS score at 12 months ranged from 82 to 96 points (average, 90.5 points), giving 3 excellent and one good cases.Conclusions:The Maisonneuve injury which is clinically manifested as posterior dislocation of the ankle with no obvious fracture of the ankle joint is likely to be missed in clinical diagnosis. Therefore, X-ray exam of the full length tibia and fibula should be taken in physical examination after reduction for the patients with simple posterior dislocation of the ankle. Once the special type of Maisonneuve injury is diagnosed, surgical treatment is indicated. Satisfactory treatment efficacy can be achieved by fixation of the distal tibiofibular syndesmosis and repair of the triangular ligament.
9.Effect of radiation on collagen type I mRNA expressions in mouse osteoblast in vitro

Objective Cancer radio-therapy may induce bone damage of the patients.collagen type I gene expressions in osteoblast after radiation indicates the influence of radiation on the function of early and late osteoblast.Methods Bone marrow stromal cells were differentiated into osteoblasts in vitro.and the characteristics was indentified.The collagen type I expressions in early and late stage osteoblasts exposed to 1~4Gy radiation were examined by RT-PCR.Results Compared to control group,collagen type I gene expressions increased in early osteoblast after 1~3 Gy radiation (P<0.05),while the gene expressions in late osteoblast that cultured 10 days decreased.Collagen type I gene expression in late stage ostoblast after 4 Gy irradiation was greatly higher than that in early stage osteoblast (P<0.01).Conclusion After 1~3 Gy irradiation,the collagen type I expression in early osteoblast was enhanced,indicating the increased ability of bone formation.The exposure to 1~3 Gy decreased collagen type I expression in late osteoblast and weakened the ability of bone formation.The result of high expression of collagen type I in late osteoblast after 4 Gy irradiation may be the manifestation of compensatory function.
10.Effects of pulsed electromagnetic fields on the precursor cells of osteoclasts and osteoblasts

Objective To study the in vitro and in vivo effects of pulsed electromagnetic fields (PEMFs) on osteoclast and osteoblast precursor cells.MethodsTo observe the in vitro effect of PEMFs,femur bone marrow cells of 8 week old female SD rats were collected.According to different treating doses,rats were divided into four treatment groups and one control group.After the treatment,the clones of granuloeyte/maerophage colony forming unit(CFU-GM) and fibroblast colony forming units(CFU-F) were measured respectively.To observe the combined in vitro and in vivo effect of PEMFs,8 week old female SD rats were randomly divided into three groups: 2-70 group,ovariectomization (OVX) group and SHAM group.Rats in the 2-70 group and OVX group were bilateral ovariectomized,while rats in the SHAM group were sham-ovariectomized.12 weeks after ovariectomization,the 2-70 group was exposed to PEMFs while the other groups were left untreated.Then,femur bone marrow cells of the rats were collected.According to the way whether the groups were treated with PEMFs,the cells were divided into six groups: 2-70 with/without treatment,OVX with/without treatment,SHAM with/without treatment.After the treatments,the clones of CFU-GM and CFU-F were measured respectively.Resultsin vitro effect of PEMFs: Compared with the control group,the CFU-GMin the treated groups reduced while the CFU-F increased.PEMFs effect in vitro and in vivo: The CFU-F in treated groupsincreased,whileno.significantdifferencesofCFU-GMwerefoundamongthegroups.Conclusion PEMFs has inhibitory effect on osteoelast precursor cells and enhances the proliferation of osteoblast precursor cells when simply applied in vitro.When PEMFs was applied in combined manner of in vitro and in vivo,it shows that PEMFs enhance the proliferation of osteoblast precursor cells but has no inhibitory effects on osteoelast precursor cells.
