5.A study on DNA damage induced by the treatment with the mycotoxins of Alternaria alternata in vitro and the inhibitory effects of R Ⅲ on the DNA damage

The alkaline elution and fluorometrie DNA assay were adapted to the evaluation of DNA single-strand breaks in the cultured human fetal lung 2BS cell. the DNA damage was induced in vitro by the treatment with Alternariol monomethyl ether (AME) and Alternariol (AOH) which were the main mycotoxins of Alternariol alternata. The results showed that both AME and AOH could induce the DNA single-strand breaks in the cultured human fetal lung 2BS cells and there existed the dose-response association betweenthe fractions of DNA remaining on the filters and the doses. The fractions of DNA remaining on the filters were 0.57?0.04, 0.45?0.02, 0.30?0.02, 0.18?0.01 respetively under the AME concentration of 10, 25, 50, 100?g/ml and were 0.68?0.3, 0.54?0.01, 0.47?0.03, 0.34?0.01 respetively under the AOH concentration of 0.1, 1, 5, 10?g/ml when DNA was eluted for 6hr. Objective:To promote the application of barcode technology and establish comprehensive barcode management platform for medical equipment in order to achieve precise management of medical equipment just like ID.Methods: Combined with information management of barcode was effectively applied to the medical equipment asset; analyzed the underlying application of barcode technology in the field of equipment asset management; finally, to clarify the application effect of barcode technology in precise equipment asset management.Results: The application of barcode technology has realized dynamic management for the whole life cycle of medical equipment, and effectively improved management method and efficiency of hospital medical equipment asset.Conclusion:Through informatization to support precise management for medical equipment, and this method can form more scientific and technological barcode technology management mode for medical equipment asset. Objective To understand how resistance and whether its compensatory hyperinsulinemia will influence the ovulatory function and sex hormone level,especially in the testosterone level and blood lipid level and to observe the effects of metformin on the polycystic ovary syndrome with pseudoacanthosis nigricans and the insulin resistance.Methods The method of the research is to carry out a three-month tretment for 30 patients who were suffering from the polycystic ovary syndrome with the pseudoacanthosis nigricans and the insulin resistance.During the treatment much attention was drawn to the following aspects of the patients.Such as the body mass index,the waist hip ratio,the fasting blood glucose,the fasting insulin,two hours after postcibal blood glucose and postcibal insulin,the insulin sensitive index,the change of blood lipid and sex hormone level,and the skin lesion change.Results After the treatment,all the indexes fell except that the insulin sensitive index rose.There was significancely difference(P

Objective To study the two-phase flow dynamics distribution and red blood cell distribution under the fluid-solid coupling interaction in left coronary artery at the typical time point within one cardiac cycle,and to investigate the formation and development mechanisms of left coronary artery atherosclerotic plaque Methods The blood was regarded as a two-phase fluid.Based on fluid-solid interaction between blood and vascular wall,the computational fluid dynamics method was used to make the transient numerical simulation of two-phase flow in the left coronary artery under fluid-solid interaction;the distribution of blood flow in the left coronary artery at the typical time point within one cardiac cycle was studied,the relationship between hemodynamic parameters and the formation of atherosclerotic plaque was analyzed.Results A lowspeed eddy zone existed in an area between the distal segment of circumferential branch and the proximal outside of blunt-edge branch of the left coronary artery,where both internal wall shear stress and red blood cell volume fraction were very small and the blood flow pattern was very complicated.Conclusion At the lowspeed eddy zone that carries small wall shear stress,the lipid concentration polarization and macromolecular material deposition are easy to be produced.The area that has less red blood cells is liable to develop hypoxia,resulting in increased vascular wall permeability and intimal injury,which will activate the immune system,causing lipid accumulation in vascular wall and intimal hyperplasia and,thus,to induce the formation of atherosclerotic plaque.(J Intervent Radiol,2017,26:253-257)
9.Screening of binding proteins interact with phosphotyrosine-interacting domain of DOC-2 by yeast two hybrid system

Objective: To screen for proteins which can interact with phosphotyrosine-interacting domain (PID) of differentially expressed gene in human ovarian cancer cell line DOC-2 by yeast-two hybrid technique, so as to provide evidence for the signal pathway of DOC-2. Methods: The cDNA sequence of human DOC-2 gene was amplified and its PID domain (nDOC-2) was subcloned into the bait vector pGBKT7 of yeast two-hybrid system; the product was then used to screen an embryo brain cDNA library and the proteins interacting with nDOC-2 were identified. Quadrople dropout(QDO) medium and X-?-gal were used for selecting the positive clones. PCA was used to analyze the amplified sequence. After elimination of the false positive clones, the positive clones were sequenced and analyzed by bioinformatic methods. Results:Twenty-one candidate positive clones were obtained and 3 of them were plasmids encoding Homo sapiens partial mRNA for betaglycan (TBR III gene), Homo sapiens protocadherin gamma subfamily C 3 (PCDHGC3), and APLP1(amyloid beta precursor-like protein 1).Conclusion: The proteins obtained in this study may play important roles in the signal pathway of DOC-2, which provides a new orientation for DOC-2 gene therapy of ovarian cancers
