1.Effect of Yun-Pi Prescription in Different Dosage on Small Intestinal Function of Splenic Asthenia Rats

Objective To observe the effect of Yun-Pi Prescription in different dose on small intestinal function of splenic asthenia rats, so as to explore mechanisms of Yun-pi Prescription in treatment of children apositia. Methods Experimental rats were randomly divided in to six groups:the control group, the model group, the positive control group (Xiao shi Jian er Syrup group), Yun-Pi Prescription in high, middle and low dose group. Measure the changes of body weight and food intake, observe excretory rate of D-xylose of rats. Results Yun-Pi Prescription could increase body weight and food intake, improve excretory rate of D-xylose in splenic asthenia rats. Conclusion Yun-Pi Prescription could improve the small intestinal absorptive function of splenic asthenia rats with dose-effect relationship.

[Objective] To observe the effects of Yun-Pi Prescription of different doss on endurance of rats with splenic asthenia and to explore the mechanism of Yun-pi Prescription in treatment of children apositia.[Methods] To set up the animal model of splenic asthenia with reserpine.Experimental rats were randomly divided into six groups:the control group,the model group,the positive control group.Xiao Shi Jian Er Syrup,and Yun-Pi Prescription treatment groups including high,middle and low dose.Changes of body weight,food intake and swimming time were measured to determine the endurance of rats.[Results] Yun-Pi Prescription could increase body weight and food intake,prolong the swimming time of splenic asthenia rats.[Conclusion] Yun-Pi Prescription could effectively counteract fatigue and improve endurance of rats with splenic asthenia and it is useful in treatment rats with splenic asthenia.

[Objective] Study effect of Yun-Pi Prescription on regulating the immune function,approach the mechanism of Yun-Pi Prescription on how to enhance immune function.[Method]All the mice were divided into groups: the normal group,the model group,the positive control group treated with Xiao Shi Jian Er Syrup,and Yun-Pi Prescription treatment groups including high,middle and low dose;determine the index number of immune organ,phagocytic count and phagocytize activity of normal mice.[Results]The high and middle dose of Yun-Pi Prescription can obviously enhance the phagocytic count and phagocytize activity,Yun-Pi Prescription has an up-regulation effect on immune function.
5.Changes of tear film stability, goblet cell and mucin 5AC expression in conjunctivochalasis patients

Objective To observe the changes of tear film stability,goblet cell and mucin 5AC expression in conjunctivochalasis patients,and explore the mechanism of conjunctivochalasis.Methods Conjunctivochalasis patients (30 cases) and single age-related cataract patients (15 cases) were collected as conjunctivochalasis group and normal control group.Eye symptom assessment (OSDI score),tear break-up time (BUT),Schirmer Ⅰ test,tear fern crystallization tests were performed for all the selected persons.Conjunctival rescent-shaped resections were made for all the conjunctivochalasis patients.Conjunctival tissue samples were stained by HE staining,AB staining,mucin 5AC immunohistochemical staining from the conjunctivochalasis group and norral control group respectively,and then statistical analysis was made.Results The OSDI score in the conjunctivochalasis group (37.80 ± 8.94) was significantly higher than that in the normal control group (11.40 ±4.08) (P <0.01).BUT in the conjunctivochalasis group (6.70 ± 2.76) s was significantly lower than that in the normal control group (13.67 ± 3.48) s (P < 0.01).Schirmer Ⅰ test in the conjunctivochalasis group (6.23 ± 3.13) mum was significantly lower than the normal control group (13.40 ± 3.74)mm (P < 0.01).Tear ferbing crystallization of the conjunctivochalasis group was decreased significantly compared with the normal control group (x2 =14.309,P =0.003).Light microscopic showed that conjunctival thickness was thinned,collagen fibers were less,elastic fiber was reduced,the lamina propria and interstitial were congestion and edema,the number of goblet cells was significantly reduced,and the positive staining of mucin 5AC staining was significantly lower in the conjunctivochalasis group than in normal control group (x2 =9.499,P =0.023).Conclusion For patients with conjunctivochalasis,the tear film function is affected,goblet cells are decreased,tear fern crystallization is decreased,mucin 5AC content is decreased,which finally leads the excessive conjunctival relaxation and abnormal ocular surface and tear.

Background and Objectives:
p21, an important member of the Cip/Kip family, is involved in inhibitory effects of RUNX1b overexpression during the early stage of human hematopoiesis.
Methods:
and Results: We established a human embryonic stem cell (hESC) line with inducible expression of p21 (p21/hESCs). Overexpression of p21 did not influence either mesoderm induction or emergence of CD34+ cells, but it significantly decreased the production of CD43+ cells and changed the expression profile of hematopoiesis-related factors, leading to the negative effects of p21 on hematopoiesis.
Conclusions
In RUNX1b/hESC co-cultures when RUNX1b was induced from D0, perturbation of the cell cycle caused by upregulation of p21 probably prevented the appearance of CD43+ cells, but not CD34+ cells. The mechanisms via which CD34+ cells are blocked by RUNX1b overexpression remain to be elucidated.
7. Facial lipo-filling using high-density fat combined with SVF-GEL

Objective:
To discuss the effect of high-density fat-binding SVF-GEL in female facial lipofilling.
Methods:
This is a retrospective study including 32 female patients, received facial fat transplantation during June 2017 to June 2018 in Yichun College. Each patient underwent high-density fat-binding SVF-GEL transplantation for facial surgery. Patients′satisfaction with the surgery and the rate of secondary surgery was evaluated. Fat was harvested from the inner thigh, centrifuged at 1200 g for 3 min, and the liquid was removed. The upper 2/3 part is prepared for SVF-GEL, for further used in delicate lipofilling in eyelid, tear groove and nasolabial groove. The lower 1/3 high density fat was used for volume restoration, such as forehead, temporal area and cheek.
Results:
All patients had significant improvements in facial contours with mild swelling and short recovery time. The satisfaction rate was 68.8%(22/32), and the second operation rate was 15.6%(5/32).
Conclusions
High-density fat-binding SVF-GEL transplantation can achieve good results in correcting facial volume loss.
8.The effects of acrylonitrile on T lymphocyte subsets and expression of toll-like receptor 4 in rats.

OBJECTIVETo explore the effects of acrylonitrile on T lymphocyte subsets, expression of toll-like receptor 4 and related cytokines in rats.
METHODSSixty-four Sprague-Dawley rats were randomly divided into 4 female groups and 4 male groups, and there were 8 rats in each group. Rats in each group were respectively given a single dose of 0, 5, 10 and 20 mg/kg acrylonitrile by gavage, once a day, 5 days a week, for 13 weeks. Blood and spleen T lymphocyte subsets was detected by flow cytometry, the mRNA expression of TLR4, IL-1β and TNF-α was analyzed by real-time quantitative PCR, the protein expression of TLR4 was evaluated by Western blot.
RESULTSCompared with control group, the percentages of blood CD3, CD4 T cells in 20 mg/kg female group and CD4/CD8 ratio in 5, 10 and 20 mg/kg female groups was significantly decreased, CD8 T cells in 20 mg/kg group was significantly increased (P < 0.05 or P < 0.01), blood CD3 T cells in 5 mg/kg male group, CD4 T cells and CD4/CD8 ratio in 20 mg/kg male groups were lower than that of control group, CD8 T cells in 20 mg/kg make group was significantly in oreased (P < 0.05 or P < 0.01). Spleen CD4, CD8 T lymphocyte percentages and CD4/CD8 ratio in 20 mg/kg female group decreased significantly, CD8 T cells in 20 mg/kg male group was significantly increased (P < 0.05 or P < 0.01), spleen CD3, CD4, CD8 T cells in 20 mg/kg male group and CD4/CD8 ratio in 10, 20 mg/kg male groups was also significantly decreased, CD3 T cells in 20 mg/kg and CD8 T cells in 10, 20 mg/kg male groups were significantly increased (P < 0.05 or P < 0.01) (TLR4 mRNA was lower expressed in 5, 10 and 20 mg/kg male groups and 10 mg/kg female group (P < 0.05 or P < 0.01), and TLR4 protein in 5 mg/kg female group and 20 mg/kg male group was significantly lower than control group (P < 0.05). The expression level of IL-1β mRNA was significantly decreased in 5, 10 and 20 mg/kg female group and 5, 10 mg/kg male group (P < 0.05 or P < 0.01), TNF-α mRNA was lower expressed in 10, 20 mg/kg female groups and 5, 10 mg/kg male groups (P < 0.01).
CONCLUSIONAcrylonitrile may lead to the changes of CD3, CD4, CD8 T lymphocyte percentages and CD4/CD8 ratio in rat blood and spleen, and also significantly effected the expression level of TLR4 mRNA and protein together with the secretion of IL-1β, TNF-α. This may cause effects on the cellular immune function.
