Sixty cases of acute lumbar sprain were treated by puncturing Yintang(Ex-HN 3) toward Shenting(GV 24), among which, 50 cases were cured, 9 were improved and only one failed, and the total effective rate was 98.3%.

BACKGROUND: Changes of leukocytes are related with some postoperative complications after renal transplantation. OBJECTIVE: To observe dynamically the level of white blood cell (WBC) and its morphological changes using automatic blood cell analyzer to evaluate its relationship with infection, immunosuppressive intoxation and acute rejection. DESIGN, TIME AND SETTING: Observation and controlled trial was performed at Second Xiangya Hospital, Central South University from June 2004 to October 2005. PARTICIPANTS: Sixty-eight patients who received renal transplantation were selected. According to clinical symptoms and laboratory biochemical indexes, X-ray, B ultrasound, and renal puncture biopsy, the patients were divided into stable group (n=36), infection group (n=9), intoxation group (n=6) and acute rejection group (n=13). METHODS: The blood routine was tested with Sysmex XE-2100 automatic blood analyzer following renal transplantation and WBC manual differentiation was done to observe peripheral blood morphology, their liver function. In addition, renal and liver functions were detected by automatic biochemical system 7600A. MAIN OUTCOME MEASURES: WBC count and its morphological changes in peripheral blood and liver and renal function after renal transplantation. Objective The aim of this study was to measure quantitatively the TSP-4 mRNA expression and its significance in gastric carcinoma was evaluated by correlating its expression with clinicopathological features, microvessel density(MVD) and MMP-9.Methods Eighty-two patients with gastric carcinoma were recruited in this study. TSP-4 mRNA expression in gastric carcinoma and adjacent tissue was detected by real-time PCR. CD34 and MMP-9 protein expression were examined by immunohistochemistry. MVD was determined according to CD34-positive tubular structures. Results The expression level of TSP-4mRNA in gastric carcinoma was obviously higher than those of adjacent tissue(P=0.03) and it was associated with tumor size, histological type, lymph node metastasis, MVD and MMP-9(P=0.002, P=0.031, P=0.014,r=0.67 P<0.01, P=0.008),but not sex, age and depth of invasion. (P=0.53,P=0.57,P=0.15).Conclusion TSP-4 may play an important role in occurrence and progression of gastric carcinoma and that the effect of TSP-4 on tumor growth and metastasis may be exerted through regulation of angiogenesis and MMP-9 expression in gastric carcinoma.
5.Effects of Rifampicin on Rotenone-induced Oxidative Stress in Differentiated PC12 Cells

[Objective] To explore the effects of rifampicin on cell morphology,intracellular reactive oxygen species (ROS),reduced glutathione (GSH),and cell apoptosis in rotenone-induced differentiated pheochromocytoma (PC12) cells.[Methods] Rotenone was added in rat PC12 cells to develop a model of Parkinson's disease in vitro.Cell morphology was observed by microscope.Intracellular GSH was determined by a microplate reader.The intracellular ROS and the apoptosis rate were measured by flow cytometry.[Results] Compared with control group and rifampicin control group,GSH significantly decreased but ROS and apoptosis rate significantly increased in rotenone group.Compared with rotenone group,GSH significantly increased but ROS and apoptosis rate significantly decreased in a dose-dependent manner in rifampicin control group (100,200,and 300 μmol/L).[Conclusion] Rifampicin may reduce the damage of rotenone-induced differentiated PC12 cells through inhibiting oxidative injury in a dose-dependent manner.
6.Construction of recombinant plasmid siRNA of XIAP and its biological role

Objective To construct siRNA expression vector of XIAP,and study its effect on XIAP expression in Hep3B cells. MethodsThree XIAP siRNA sequences were designed,synthesized,and cloned to pRNAT-U6.1/Neo.The successfully constructed recombinant plasmid was determined by sequence analysis,and will be transfected into Hep3B.The best interference plasmid were analyzed by RTPCR,Western blot,and immunohistochemistry.Results The plasmid of pRNAT-U6.1/Neo-XIAP was constructed successfully,the trans-fected with different plasmid of siRNA XIAP can lower significantly XIAP.Conclusions The siRNA vector of XIAP gene was constructed successfully.It will be a basis for the study of XIAP function in apoptosis regulation of the Hepatoma cells.

8.Transfusion-transmitted Infections Markers in Hospitatized Patients

OBJECTIVE To investigate the distribution of serum markers of transfusion-transmitted infections in hospital,and analyze the clinical values in the control of nosocomical infection. METHODS The serum markers of transfusion-transmitted infections from 21 605 hospitalized cases,such as HBsAg,anti-HCV,anti-HIV,and RPR were detected. RESULTS The positive rate of HBsAg and anti-HCV was 9.78% and 1.70%,respectively.The positive anti-HIV was detected in 13 cases by the primary screening test for HIV,and the 13 cases were reconfirmed by the reconfirmation laboratory of CDC in Hunan Province.The positive RPR was in 4 cases,the same as of TPHA.There were 2 582 positive cases of the four indexes(HBsAg,anti-HCV,anti-HIV and RPR),the total positively detectable rate was 11.95%,which was obviously higher than 7.6% of normals(P

Objective:To investigate the effect ofLeech Capsule on stability ofcarotid plaque and expression ofplatelet membrane glycoprotein in patients with cerebral arteriosclerosis.Methods:203 patients with cerebral arteriosclerosis which with unstable carotid plaque were divided into two groups.There were 101 patients in observation group which were treated by conventional therapy and Leech Capsule, and 102 patients in control group which were treated only by conventional therapy.Then comparison analysis ofIMT, plaque area, unstable plaque number and positive rates ofPAC-1 and CD62P were made before and after treatment with two groups.Results:After treatment, the IMT, plaque area, unstable plaque number and positive rates ofPAC-1 and CD62P in observation group were significantly lesser than those in control group.In observation group, all parameters after treatment were significantly lesser than those before treatment, but in control group there were no significant difference in IMT and positive rates ofCD62P between after treatment and before treatment.In addition, there were no obvious adverse reactions after treatment by leech capsule.Conclusion:Leech Capsule was an effective drug for stabilizing carotid plaque and depressing the level ofplatelet activation, and it had few adverse reaction.It was worth further spreading.

Objective To explore the role and clinical significance of GSTM 3 ( glutathione S-trans-ferase mu 3) expression in prostate cancer (PCa). Methods We had used the two-dimensional fluores-cence difference gel electrophoresis ( 2D-DIGE) and mass spectral analysis to further verify the microarray data of mRNA expression profiling discovered .GSTM3 mRNA level was detected by Rael-time Quantitative PCR ( RT-QPCR) in 28 pairs of prostate cancer tissue and benign tissue .The relationship of GSTM 3 level with the serum PSA level and the clinical feature of PCa were analyzed . Results In 2D-DIGE study, we found that the expression of GSTM 3 protein in adjacent tissues was significantly higher than that in PCa tis-sues (P<0.05).RT-QPCR results showed that GSTM3 in adjacent tissues (8.12±0.51) was significantly higher than that in PCa tissues (7.18±0.54) (P<0.05).There was no significant difference of GSTM3 ex-pression in different serum PSA packets ( P>0.05) and prostate cancer clinical pathological parameters ( P>0.05). Conclusions GSTM3 expression is down-regulated in PCa tissues, and we may identify PCa by detecting the GSTM 3 expression .
