1.Clinical study of super-early operation combined with traditional Chuanxiongqin on treatment of hypertensive intracerebral hemorrhage

Objective To explore an effective treatment for hypertensive intracerebral hemorrhage.Methods By the method of random and control,patients with hypertensive intracerebral hemorrhage were randomly divided into two groups:the treatment group(32 patients)was treated with integrated traditional Chinese and western medicine therapy,including super-early operation,conventional western medicine and Chuanxiongqin injection treatment.The control group(40 patients)was treated with operation and conventional western medicine treatment.The effect was evaluated on 28 th day after treatment.Results The effect of the treatment group was superior to that of the control group(χ~2=4.15,3.26,P<0.05).The treatment group had lower mortality rate(χ~2=8.04,P<0.05)and lower morbidity of complications(pulmonary infection:6/16 cases,χ~2=11.37,P=0.01;upper-congestive hemorrhage:8/16 cases,χ~2=4.10,P=0.04)statistical data indicated that there was significant difference between treatment group and control group.Conclusion Super-early operation with traditional Chinese medicine in treatment of hypertensive intracerebral henorrhage has a better effect than the treatment without traditional Chinese medicine.
2.Detection of Chlamydia trachomatis phage Vp1 gene in clinical swab specimens as well as anti-Vp1 antibodies in serum specimens

Objective To detect Chlamydia trachomatis phage Vp1 gene in clinical swab specimens and anti-Vp1 antibodies in serum specimens.MethodsCervical and urethral swab as well as serum specimens were collected from attendees to the sexually transmitted disease(STD) clinic in the Tianjin Institute of STD,during March 2008 to March 2011.PCR was conducted to detect chlamydial phage Vp1 gene in swab samples,enzyme linked immunosorbent assay(ELISA) and Western blot to detect anti-Vp1 antibody in sera.The swab specimens positive for Vp1 gene were subjected to cell culture followed by the detection of Vp1 protein with an immunofluorescence-based method.ResultsTotally,36 out of 1542 swab specimens turned out to be positive for Vp1 gene,and 23 out of 453 serum specimens for anti-Vp1 antibody.No positive results were obtained in the Vp1 gene-positive swab specimens by cell culture and immunofluorescence-based assay.ConclusionThe Vp1 gene of Chlamydial trachomatis phage and anti-Vp1 antibody are successfully detected from clinical swab and serum specimens respectively.
3.Effect of radiofrequency ablation combined with trans arterial chemoembolization and high-frequency hyperthermia on apoptosis in rabbit liver VX2 carcinoma

Objective To study the effects of apoptosis of the tumor cells in rabbit liver VX2 carcinoma after treatment by radiofrequency ablation(RFA) combined with trans arterial chemoembolization(TACE) and high‐frequency hyperthermia(HFH) . Methods Rabbit liver VX2 carcinoma model was established .Rabbit liver VX2 tumor models were divided into the following group:group A ,RFA+ TACE;group B ,RFA + TACE + HFH ;group C ,RFA + HFH ;group D ,TACE+ HFH .The changes of serum ALT was detected to realize the safety of the treatment .Cell apoptosis were detected by DNA agarose gel electrophoresis and terminal deoxynucleotidyltransferase‐mediated Dutp nick end‐labeling(TUNEL) assay ;SP immunohistochemistry ,Western blot and Real‐time quantitative PCR(RT‐PCR) were used to detect Caspase‐3 protein and mRNA expression levels .Results The changes of serum ALT in group B was significantly higher .Compared with other groups ,the apoptosis index in group B was increased marked‐ly(P<0 .05) .Western blot and RT‐PCR Caspase‐3 protein and mRNA levels in group B were higher than the other groups(P<0 .05) .Conclusion RFA+ TACE+ HFH can effectively kill tumor cells and promote apoptosis of tumor cells ,but ,at the same time ,can damage liver function .
4.The cross immune responses of the rhesus monkey induced by the E style Chlamydia trachomatis recombinant major outer membrane protein

Objective To test cross immune responses induced in rhesus monkeys immunized with the recombinant major outer membrane protein(rMOMP).Methods Six rhesus monkeys were divided into three groups:the group vaccinated with purified rMOMP and Freund's adjutants,the group vaccinated with Freund's adjutants only and the control group vaccinated with PBS.All of the rhesus monkeys vaccinated intramuscularly at 0,2,4 weeks.Two weeks after the last time,The IFN-γand Chlamydia-specific antibody titers in sera,which were determined by ELISA,lymphocyte proliferation assay were performed by MTT,and observ the delayed hypersensitivity and in vitro neutralization assays.Results The result of the monkeys immunized with rMOMP and Freund's adjuvant:the specific immune responses can be observed.The in vitro neutralization and lymphocyte proliferation assays were observed better in the same group.Conclusion After being vaccinated with rMOMP,the monkeys can develop strong and effective Chlamydia-specific cross immune responses.
5.Impact of Coronavirus Disease 2019 on long-term renal function in patients with chronic kidney disease undergoing no renal replacement therapy

Objective To investigate the impact of Coronavirus Disease 2019(COVID-19)on long-term renal function in patients with chronic kidney disease(CKD)who received no renal re-placement therapy.Methods Thirty-nine CKD patients with an estimated glomerular filtration rate(eGFR)<60 mL/(min·1.73 m2)at the time of COVID-19 diagnosis,admitted to Guizhou Aero-space Hospital from December 2022 to March 2023,were enrolled in the COVID-19 group.Addition-ally,40 CKD patients without COVID-19 from September to December 2021,were included in control group.Changes in eGFR were compared between the two groups,and the temporal trend of eGFR in the COVID-19 group was analyzed using a mixed-effects linear model.Results No statistically sig-nificant differences were observed in eGFR levels at baseline,1-and 3-month of follow-up between the COVID-19 and control groups(P>0.05).However,at 1 year of follow-up,the eGFR level in the COVID-19 group was significantly lower than that in the control group(P<0.05).The mixed-effects linear model analysis revealed a decrease in eGFR by 1.84 mL/(min·1.73 m2)within 1 year of COVID-19 onset in the COVID-19 group,which inclined 4.62%from baseline,with the most significant decline(7.45%)observed among diabetic patients.Conclusion Patients with CKD who have not undergone renal replacement therapy experience a substantial decline in eGFR and worsened renal function within 1 year of COVID-19,necessitating close long-term renal function mo-nitoring and timely interventions.

7.Pancreatic melatonin enhances anti-tumor immunity in pancreatic adenocarcinoma through regulating tumor-associated neutrophils infiltration and NETosis.

Tumor microenvironment contributes to poor prognosis of pancreatic adenocarcinoma (PAAD) patients. Proper regulation could improve survival. Melatonin is an endogenous hormone that delivers multiple bioactivities. Here we showed that pancreatic melatonin level is associated with patients' survival. In PAAD mice models, melatonin supplementation suppressed tumor growth, while blockade of melatonin pathway exacerbated tumor progression. This anti-tumor effect was independent of cytotoxicity but associated with tumor-associated neutrophils (TANs), and TANs depletion reversed effects of melatonin. Melatonin induced TANs infiltration and activation, therefore induced cell apoptosis of PAAD cells. Cytokine arrays revealed that melatonin had minimal impact on neutrophils but induced secretion of Cxcl2 from tumor cells. Knockdown of Cxcl2 in tumor cells abolished neutrophil migration and activation. Melatonin-induced neutrophils presented an N1-like anti-tumor phenotype, with increased neutrophil extracellular traps (NETs) causing tumor cell apoptosis through cell-to-cell contact. Proteomics analysis revealed that this reactive oxygen species (ROS)-mediated inhibition was fueled by fatty acid oxidation (FAO) in neutrophils, while FAO inhibitor abolished the anti-tumor effect. Analysis of PAAD patient specimens revealed that CXCL2 expression was associated with neutrophil infiltration. CXCL2, or TANs, combined with NET marker, can better predict patients' prognosis. Collectively, we discovered an anti-tumor mechanism of melatonin through recruiting N1-neutrophils and beneficial NET formation.
8.A study on the biological effects of electromagnetic pulse with different high⁃intensity on condylar cartilage in rats

Objective:
To explore the biological effects of electromagnetic pulse (EMP) with different high intensities on condylar cartilage in rats.
Methods:
SD rats were randomly divided into a sham group (Sham) and an irradiation group (EMP1: 500 kV/m, 10 Hz; EMP2: 270 kV/m, 10 Hz). Then, they were sacrificed at 1 h, 3 h, 12 h, 24 h and 3 d after irradiation. The degree of cartilage degeneration was evaluated by HE, safranine O-fast green, type Ⅱ collagen immunohistochemistry and TUNEL staining. Immunohistochemistry and western blot were performed to detect the expression of the matrix degradation factors: matrix metalloproteinase-13 (MMP-13), a disintegrin and metalloproteinase with thrombospondin motifs (ADAMTS-5) and the apoptosis key factor cleaved-cysteinyl aspartate specific proteinase (cleaved-Caspase3) in condylar cartilage.
Results :
HE staining showed that, compared with the Sham group, a small amount of exfoliation was found on the fibrous surface layer of the cartilage after irradiation in the EMP1 and EMP2 groups. Compared with the Sham group, the percentage of safranine O-fast green-positive area decreased significantly at 12 h and 24 h (both P<0.01) in the EMP1 group and 12 h and 24 h in the EMP2 group (both P<0.05); the percentage of type Ⅱ collagen-positive area decreased significantly at 3 h and 12 h (P<0.05, P<0.001) in the EMP1 group. In addition, the number of TUNEL-positive apoptotic cells increased significantly at 1 h, 3 h, 12 h, and 24 h in the EMP1 group and 1 h, 3 h, and 12 h in the EMP2 group (P<0.05). Moreover, at different timepoints (except at 3 d) in the EMP1 group and EMP2 group, the percentage of MMP-13, ADAMTS-5- and cleaved Caspase3-positive chondrocytes and their protein levels in condylar cartilage increased significantly after irradiation (P<0.05).
Conclusion
EMP with a certain degree of high-intensity can induce early transient damage to condylar cartilage. This effect is dose-and time-dependent.

Objective:
To analyze the epidemiological history, clinical manifestations, treatment and the short-term prognosis of 31 cases of 2019 novel coronavirus(2019-nCoV) infection in children from six provinces (autonomous region) in northern China.
Methods:
A retrospective analysis of the epidemiological history, clinical symptoms, signs, laboratory examinations, chest imaging, treatment and the short-term prognosis of 31 cases of 2019-nCoV was conducted. The patients were diagnosed between January 25th, 2020 and February 21st, 2020 in 21 hospitals in 17 cities of six provinces(autonomous region) of Shaanxi, Gansu, Ningxia, Hebei, Henan and Shandong.
Results:
The age of the 31 children with 2019-nCoV infection was 7 years and 1 month (6 months -17 years). Nine cases (29%) were imported cases. Other 21 cases (68%) had contact with confirmed infected adults. One case (3%) had contact with asymptomatic returnees from Wuhan. Among the 31 children, 28 patients (90%) were family cluster cases. The clinical types were asymptomatic type in 4 cases (13%), mild type in 13 cases (42%), and common type in 14 cases (45%). No severe or critical type existed. The most common symptom was fever (
