1.Clinical study on 45 cases with endogenous caesarean scar pregnancy

Objective To study the effect of ultrasound guided curettage after methotrexate and mifepristone in the treatment of endogenous caesarean scar pregnancy.Methods A retrospective analysis was performed on 43 patients of endogenous caesarean scar pregnancy undergoing treatment of ultrasound guided curettage after methotrexate and mifepristone.Results 39 cases were successful,4 cases were transformed to laparotomy because of intraoperative blood loss,7 cases of bleeding after curettage were successful by uterine carity placed double lumina Foley catheter. Conclusion Ultrasound guided curettage after methotrexate and mifepristone is a safe,effective,little trauma and low cost method in the treatment of endogenous caesarean scar pregnancy,it especially adapts to primary hospital.
2.Immune hyporesponsiveness induced by immature dendritic cells prolonged rat kidney allograft survival

Objective To study the effect of immature dendritic cells (imDC) transfusion in combination with bone marrow transplantation (BMT) on rat kidney allograft survival and its possible mechanism. Methods Renal allograft from DA rat was transplanted to Lewis rat. Forty recipient rats were randomized into 5 groups: (1) Negative control group; (2) imDC group: Lewis rats accepted imDC transfusion in the amount of 2? 107 only; (3) BMT group: Lewis rats accepted bone marrow transplantation in the amount of 2?108 only; (4) imDC + BMT group: Lewis rats accepted both imDC and BMC; (5)Third party donor group: Lewis rats accepted renal allograft from Wistar rats. One-way MLR was performed to assay splenic cell proliferation to allogeneic T cells. Exogenous IL-2 was added at the outset of another group as the former one-way MLR. Normal Lewis rat accepted splenic cells from tolerant rat in the amount of 1?108. DTH was assayed in the trans-tolerance model. Cells from spleen and thymus were harvested from recipient rats for detecting chimerism by flow cytometry. Results The median survival time (MST) of the renal allografts was (7. 12 ? 1. 25) days in negative control group, (24. 36 ? 3. 20)days in imDCs group and (7. 87 ? 2. 10)days in BMT group, respectively. Objective To investigate the regulative effect of interferon gamma (INF?) on the expression of platelet-derived endothelial cell growth factor / thymidine phosphorylase (PD-ECGF/TP) and its relation with the anti-cancer effect of 5′-deoxy-5-fluorouridine (5'DFUR) and 5-fluorouracil (5FU) in T24 bladder cancer cells. 5.SELDI-TOF-MS ProteinChip technology for screening of urine markers of bladder carcinoma

Objective To search for new tumor markers from urine of bladder transitional cell carcinomas. Methods Urine samples from 61 bladder cancer patients who were histologically diagnosed and 53 healthy volunteers and 42 cases with other urogenital diseases were analyzed using surface-enhanced laser desorption/ionization time of flight mass spectrometry (SELDI-TOF-MS) IMAC-Cu-3 ProteinChip technology,which can specifically bind the metal-combining-proteins.Proteomic spectra were generated by mass spectrometry.A blinded test set was used to determine the sensitivity and specificity of the differentially expressed markers. Results Four differentialy expressed potential markers were identified.Their corresponding molecular weights were 3445,3703,3896 and 5932, respectively.The 3445 protein was identified as ?-defensin,with its sensitivity for diagnosing bladder cancer being 80% and specificity being 75%.The sensitivity of 3703 protein was 32%,while its specificity was 94%.97% of the patients who were positive for 3703 protein were with high grade and invasive bladder cancer.The sensitivity for 3896 and 5932 proteins were 78% and 55%;the specificity for them were 40% and 35%,respectively. Conclusions SELDI-TOF-MS ProteinChip technology is a quick,easy and practical,high throughput analytic method.It can screen out several relatively specific,potential markers from urine of bladder cancer patients,so it has better clinical feasibility.
6.A study of polymorphism of STR D13S325 locus in Chinese Han poplulation in Wuhan

Population genetic data of the STR D13S325 locus in a sample of 216 unrelated individuals of Han population in Wuhan was studied with the method of PCR. 11 alleles and 30 different genotypes were observed.There is no significant deviation from the Hardy Weinberg equilibrium.The DP value was 0 9292,PE 0 6137 and PIC 0 7755.The results of study of 200 meiosis showed no mutation.The results prove that this STR Locus is useful in the forensic sicence practice in Han population in Wuhan.

The factor of antiapoptosis in cancer is recently identified as survivin. Survivin is prominently overexpressed in embryonic and fetal tissues, and various human cancers. It is potentially involved in both inhibition of apoptosis and control of cell division. Recently, survivin is found to be correlated with the genesis, progress, and prognosis of urological cancer. Manipulation of the antiapoptotic pathway maintained by survivin may be beneficial for cancer therapy. Progressive research of survivin in urological cancer.
8.Effect of blood purification on extravascular lung water and permeability index in extrapulmonary acute respiratory distress syndrome

Objective To observe the effects of continuous blood purification(CBP) on extravascular lung water and permeability index in patients with severe extrapulmonary acute respiratory distress syndrome (ARDSexp).Methods Fifteen patients with severe ARDSexp were treated with CBP.The level of extravascular lung water index (EVLWI) and pulmonary vascular permeability index (PVPI) was measured before and after treatment,and at the same time the changes of inflammatory indicators were recorded.Results Compared with that before treatment,EVLWI and PVPI was decreased obviously after treatment (P < 0.01),the level of C-reactive protein and interleukin 6 (IL-6),calcitonin and systemic inflammatory response syndrome score and the acute physiology and chronic health Ⅱ score was significantly decreased (P < 0.01).Conclusion CBP can improve the level of EVLWI,PVPI and inflammatory indicators and also the prognosis of severe ARDSexp.
9.Synergistic effect of anti-human DR5 monoclonal antibody and adriamycin on SMMC-7721 cells

Objective:To determine the sensitivity of SMMC-7721 cells to mDRA-6,the synergistic damage effect of mDRA-6 and adriamycin on human hepatocellular carcimoma cell lines and its possible mechanism.Methods:SMMC-7721 cells were cultured with RPMI1640 medium in regular condition.The morphology was observed by microscope.Cytotoxicity was examined by MTT assay. Apoptosis were detected by flow cytometry.Results:(1)The apoptosis of SMMC-7721 cells could be induced by mDRA-6,1.89 ?g/ml of mDRA-6 cound kill 36% of the cells;(2)Concentration-dependent cytotoxicity of adriamycin was exhibited in SMMC-7721 cells;(3)The combination of mDRA-6 and adriamycin exhibited synergistic effect on SMMC-7721 cells.2 ?g/ml of mDRA6 and 40 ng/ml of adriamycin killed 60%SMMC-7721.Conclusion:MDRA-6 can induce SMMC-7721 cell apoptosis.The combination of mDRA-6 and adriamycin exhibit synergistic effect on SMMC-7721 cells.
10.Extraperitoneal laparoscopic radical prostatectomy for prostate cancer and functional recovery of urinary continence

Objective To evaluate the surgical technique,urinary continence and clinical efficacy of extraperitoneal laparoscopic radical prostatectomy for prostate cancer. Methods This series included 28 cases of prostate cancer aged 60 -75 years (mean,68 years). The PSA level ranged from 0. 7 ng/ml to 23. 6 ng/ml. TNM classification showed T1N0M0 tumor in 11 cases, T2N0M0 tumor in 15 and T3aN0M0 tumor in 2. All cases underwent extraperitoneal laparoscopic radical prostatectomy. The functional reconstructive technique consisted of preservation of the urethral and bladder outlet sphincter muscles, and reconstruction of the bladder neck. Results All the operations were successful. The operative time was 180 - 380 min( mean, 240 min) ,and the blood loss was 400 - 1200 ml( mean,800 ml) . All the catheter were removed 2 weeks after surgery. Mild urinary incontinence occurred in 3 cases, and it improved after supportive treatment. The PSA level was 0. 02 -0. 10 ng/ml 3 months after operation. No recurrence of prostate cancer was found during the follow-up of 1 month-2 years. Conclusions Extraperitoneal laparoscopic radical prostatectomy is safe and effiective for the treatment of prostate cancer. It can be widely applied in clinical practice.
