1.The application of a revised Chinese version of the state-trait anxiety inventory in migrant children

Objective To conduct a preliminary examination of the factor structure and the reliability and validity of a revised Chinese version of the State-Trait Anxiety Inventory-Form Y (STAI-Y) by using the large sample of migrant children.Methods Perform Exploratory Factor Analysis with half of the data (n =5477) from a large-scale questionnaire survey of students in Grades 4-9 at 58 schools for migrants in Beijing,then conduct Confirmatory Factor Analysis with the other half (n =5476).Results The results of state anxiety and trait anxiety both showed two factors.However,the indicators of trait anxiety were not ideal as seen from the following:the total explained variance was 39.22%,the relationship between the factors and items was not in accordance with the original English version,and the correlation between the two factors was unstable in the different samples (r =-0.17,P < 0.001 ; r =-0.06,P < 0.001).In addition,the factor loading of items 24 ( I wish I could be as happy as others) was low.Conclusion The state anxiety portion of this revised Chinese version of the STAI-Y is structured by the factors state anxiety present and state anxiety absent,and with good reliability and validity as well.Nevertheless,the trait anxiety portion of this revised Chinese version of the STAI-Y is structured by the factors trait anxiety present and trait anxiety absent,it shows unstable reliability and validity.
2.Effect of blood purification on extravascular lung water and permeability index in extrapulmonary acute respiratory distress syndrome

Objective To observe the effects of continuous blood purification(CBP) on extravascular lung water and permeability index in patients with severe extrapulmonary acute respiratory distress syndrome (ARDSexp).Methods Fifteen patients with severe ARDSexp were treated with CBP.The level of extravascular lung water index (EVLWI) and pulmonary vascular permeability index (PVPI) was measured before and after treatment,and at the same time the changes of inflammatory indicators were recorded.Results Compared with that before treatment,EVLWI and PVPI was decreased obviously after treatment (P < 0.01),the level of C-reactive protein and interleukin 6 (IL-6),calcitonin and systemic inflammatory response syndrome score and the acute physiology and chronic health Ⅱ score was significantly decreased (P < 0.01).Conclusion CBP can improve the level of EVLWI,PVPI and inflammatory indicators and also the prognosis of severe ARDSexp.
3.Influence of the disinfection effect of acidic electrolyzed oxidizing water on laryngeal endoscope

[Objective] To compare the disinfection effect of different disinfectants on disinfection effect of laryngeal endoscope,and seek for a more safe and effective disinfectant for laryngeal endoscope.[Methods] 180 laryngeal endoscope were divided into three groups with 60 of them in each group.Acidic electrolyzed oxidizing water(AEOW),2% glutaraldehyde and orthophthalaldehyde were used for disinfection.Three minutes and five minutes after disinfection,the samples were got and underwent bacterial detection.The disinfection effect after three minutes and five minutes was compared.[Results] The qualified rate of disinfection and cost between AEOW,2% glutaraldehyde and orthophthalaldehyde showed distinct difference.[Conclusions] Using acidic electrolyzed oxidizing water in the disinfection of laryngeal endoscope has advantages of fast,effective,low-cost,which can be applied in the clinical work.
4.Effect of the mixture of radix sophorae flavescentis on cellular immunologic response in rats with immunosuppressant-induced pneumocystis carinii pneumonia induced

AIM: To study the effect of the radix sophorae flavescentis on cellular immunity in rats with Pneumocystis Carinii Pneumonia (PCP) induced by long-term use of immunosuppressant, and explore the action of traditional Chinese medicine for the immunological regulation and infectious prevention after organ transplantation. METHODS: The experiment was conducted at Department of Pathobiology, Jilin Medical College from May 2005 to March 2006. Forty adult healthy female SD rats were selected from Harbin Medical University (Certification: 02473146) and randomly divided into experiment group and control group, with 20 rats in each. The model of PCP was set up by glucocorticoid injection subcutaneously to SD rats (25 mg once, 2 times/week). The mixture of sophorae flavescentis was given to stomach with tube in experiment group (3 mL/kg, 2 times/day), and was consisted of radix sophorae flavescentis, ash bark, amur cork-tree, malt, milkvetch root and danshen root. Six weeks later, all the rats were anesthetized and broncholveolar lavage fluids were collected.①Alveolar washing fluid was concentrated 10 times and the levels of the soluble interleukin-2 receptor (sIL-2R) were examined by double antigen sandwich ELISA.②Blood was sampled from rat eyes and the count of lymphocytes in peripheral blood were detected.③The percentages of CD3+ and CD4+ subgroups were assessed with erythrocyte chaplet kit sensitized by antigen. 5.Effects of magnesium valprote on behavior and lipid peroxidation of brain in chronic stress depression mice

Objective To explore the effects of magnesium valprote on behaviors and lipid peroxidation of brain in chronic stress depression mice.Methods 40 male mice were randomly divided into normal control group,model group, amitriptyline control group and magnesium valprote group, and.10 mice each group.Except normal control group,other groups were separated one mouse in each box and exposed to 21 days chronic unpredictable stressors, and the mice of normal control group were fed in a box and stress-free.During the stress, the mice of amitriptyline control group were applied amitriptyline(0.2 mg/10g) by intragastric every day,magnesium valprote group were given intragastrically magnesium valprote (0.8 mg/10g), and the mice of model and normal control group were administrated with normal sodium of the same volume by the same method.On 7th, 14th,21st day, all mice weight were weighed and weight gain was calculated.The ethology examination was performed by using jigglecage connected with Biology Function Experiment System and experiment of sucrose solution consumption after the chronic stress.At the end of experiment, all mice were killed, and then their brains were removed and made into brain homogenate.The activities of superoxide dismutase (SOD) and glutathione peroxidase (GSH-Px) and the content of malondialdehyde(MDA) in brain homogenate were analyzed by spectrophotometry.Results Compared with model group, magnesium valprote group were increased on locomotor activity number(97.50 ± 20.54,302.30± 39.04, t = 31.1432, P < 0.01 ), sucrose solution consumption ( ( 15.30 ± 3.60 ) ml, ( 23.70 ± 4.45 ) ml, t =9.8446, P<0.01) ,body weight gain(( 10.56 ± 1.53)g,( 12.63 ±2.12)g, t=5.3113, P=0.007) ,the activity of SOD( t =3.8428, P=0.004) ,and GSH-Px( t= 3.4143, P=0.007), but decreased on irritation number( t =17.1370, P < 0.01 ) and the content of MDA( t= 11.0778, P = 0.002).Conclusion Magnesium valprote can ameliorate behavior impairment of chronic stress-induced depression mice, and the observed antidepressant-like effects may be related to inhibition lipid peroxidation of mice brain.
6.The role of DR5 in TRAIL-induced apoptosis for Jurkat cell line

Objective:To study the role of DR5 in TRAIL apoptotic signal transduction. Methods:BALB/C mice were immunized with recombinant DR5 that contains the full-length extracellular domain of the human DR5. Anti-DR5 mAb was generated by hybridoma. The level of DR5 expression on Jurkat cell line was examined by flow cytometry. The rates of TRAIL-induced apoptosis and anti-DR5 mAb blocking on Jurkat cells were tested by flow cytometry with TRAIL apoptosis kit.Results:The percentage of DR5 expression on Jurkat cells was 94 83%. TRAIL and anti-TRAIL mAb could kill Jurkat cells on dose-dependent, and the killing rate was 90% in the concentration of 50~100 ng/ml. The killing role of TRAIL could be blocked on Jurkat cells pretreated with anti-DR5 mAb. The average percentage of blocking was 90 49%.Conclusion:DR5 plays a very key role in TRAIL induced apoptosis in Jurkat cells.
7.Diagnostic efficacy and complication of transperineal prostate biopsy: less than 10 cores vs more than 10cores

Objective To compare the diagnostic efficacy and complications of transperineal prostate biopsy for<10 cores biopsy vs≥10 cores biopsy. Methods Nine hundred transrectal ul-trasound-guided transperineal prostate biopsies were performed. Patients were divided into 2 groups, <10 cores group and ≥10 cores group. Patient numbers of the 2 groups were 759 and 141, respec-tively. Cancer positive rate and complications were compared between the 2 groups retrospectively. Results Cancer positive rates were 41.6%(316/759) and 51.8%(73/141) in 2 groups (P<0.05). In patients of PSA≤10.0 ng/ml, cancer positive rates were 6.8% (16/235) and 17.8% (8/45) in 2 groups (P<0.05). Gross hematuria was the most common complication associated with biopsy. There was no statistical difference between the 2 groups in post-biopsy gross hematuria rate. Conclu-sions The diagnostic efficacy is higher in≥10 cores prostate biopsy than that in <10 cores prostate biopsy, There is no difference in biopsy related complications regarding biopsy core numbers.
8.The effects of premature atrial contractions on hemodynamics with coronary heart disease and hypertension without basic cardiovascular disease

Objective To explore the effects of premature atrial contractions on hemodynamics combined with coronary heart disease and hypertension .Methods According to the advance rate ,62 patients with premature atrial contractions were invided into 3 groups :< 20% group(n= 18) ,20% - 40% group(n= 20) and > 40% group(n= 24) .All of the patients were performed imped‐ance cardiography .The hemodynamics indexes at different times were compared ,and the variation rates in hemodynamics of the three groups combined with basic diseases were analyzed .Results Stroke volume(SV) ,stroke index(SI) ,aortic compliance(AC) , stroke work(SW) ,stroke work index(SWI) ,and heather index(HI) were smallest when premature atrial contractions ,which were middle when the first sinus beats before premature atrial contractions ,and largest when the first sinus beats after premature atrial contractions(P< 0 .05) .The variation rates in hemodynamics were smallest in < 20% group ,which were middle in 20% - 40%group ,and largest in > 40% group(P< 0 .05) .The percentage of patients with basic heart diseases was lowest in < 20% group , which was middle in 20% - 40% group ,and highest in > 40% group(P< 0 .05) .Conclusion The advance rate is relatively large in patients with basic cardiovascular disease .Premature atrial contractions has certain effects on hemodynamics ,and the effects are positively correlated with advance rate .
9.Expression, purification and detection of activities of AprA and its inhibitor AprI from Pseudomonas aeruginosa

Objective To clone and express the alkaline protease AprA , one important virulence factor secreted by Pseudomonas aeruginosa(PAE)in Escherichia coli, to clone and express the inhibitor of AprA (AprI) and its substrate flagellin , and to detect the function of AprA and the inhibitory function of AprI .Methods The genes encoding AprA ,AprI and flagellin gene were amplified respectively by PCR using PAE PAO 1 genome DNA as the template .The expression vec-tors (pET-28a-AprA, pET-28a-AprI and pET-28a-Flagellin) were constructed and transformed into E.coli BL21(DE3) respectively.The recombinant AprA protein was expressed by IPTG induction and purified via denaturing and renaturation. The recombinant AprI and flagellin were expressed and purified by Ni 2+affinity chromatography .The cleavage activities of AprA on flagellin were detected by SDS-PAGE.Results Recombinant AprA , AprI and flagellin protein were expressed and purified .It was demonstrated that AprA cleaved flagellin , which was blocked by AprI .Conclusion Recombinant AprA could cleave its substrates as an alkaline protease , and its inhibitor AprI inhibits the activities of AprA .This study will contribute to further investigations on the role of AprA in the pathogenesis of PAE .
10.Osteoclast inhibitory lectin related protein 2 protects mice from endotoxemia

Objecitv e To investigate the effects of a recombinant protein osteoclast inhibitory lectin related protein 2( OCILRP2)-Fc on LPS-induced endotoxemia by blocking OCILRP 2 signaling pathway and to in-vestigate the roles of OCILRP2 during inflammation.Methods Real-time PCR was used to detect OCILRP2 ex-pression at mRNA level in RAW264.7cells before and after in vitro stimulation with LPS.A mouse model of en-dotoxemia was established by intraperitoneal injection of BALB /c mice with a median lethal dose of LPS .Two hours prior to LPS treatment, mice were intraperitoneally injected with OCILRP2-Fc, human IgG or PBS, re-spectively .Several parameters including the survival rate of BALB/c mice with and without LPS treatment , spleen weight for arterial hyperemia analyzing , histopathological changes of lung and liver by HE staining , serum levels of inflammatory cytokines (IL-6, IL-12, TNF-αand IFN-γ)by ELISA , NF-κB activity by Western blot, were analyzed .Results Real-time PCR showed that LPS elevated in vitro OCILRP2 expression at mRNA level in macrophages (P<0.05).Upon the treatment of OCILRP2-Fc, BALB/c mice suffered from endotoxemia showed obviously increased survival rate , decreased spleen hyperemia , attenuated pathological injury of lung and liver, reduced levels of IL-6, IL-12, TNF-αand IFN-γin serum samples (P <0.05) as compared with mice treated with human IgG and PBS .LPS induced NF-κB p65 phosphorylation and IκB degradation were inhibited by OCILRP2-Fc treatment.Conclusion OCILRP2-Fc protects mice from endotoxemia by blocking OCILRP 2 signaling, which suggests that OCILRP2 plays an important role in LPS induced inflammation.
