1.The species of pathogenic microorganism and the analysis of its antibiotic resistance in emergency department patients

Objective Explore the species of pathogenic microorganism and analysis the antibiotic resistance in Emergency department patients in West China Hospital, finding out the evidence for clinical therapy. Methods Select the patients from May 2013 to April 2015 in Emergency department, analyses the species of pathogenic microorganism and the result of drug sensitivity test.Results In the 1 622 cases, there are 1 237 cases of gram-negative bacilli, 324 cases of gram positive coccus, 60 cases of fungus and 1cases of gram-negative coccus, accounted for 76.3%, 19.9%, 3.7%and 0.1%respectively.The top five is E.coli, acinetobacter baumannⅡ, klebsiella pneumoniae, pseudomonas aeruginosa and staphylococcus aureus, accounted for 21.4%, 16.7%, 12.4%, 11.5% and 6.5%; according to the sample species, the detection rate of sputum is highest, accounted for 46.6%, next is blood, urine, secretion; in regard to multiple resistant bacteria, detection rate of ESBL-E.coil, ESBL-KPN , CRAB and MRSA is 62.0% , 26.0%, 80.5% and 26.5%; the detection rate of VREF is 9.4% and does not find Vancomycin resistant enterococcus bacteria. Conclusions The species of pathogenic microorganism in emergency department is rich, especially gram-negative bacilli as E.coli, acinetobacter baumannⅡ, and the drug resistance situation is not optimistic. Emergency department shall communicate with microbiology laboratory timely, pay attention to the situation of the species and the drug resistance result, take rational use of antibiotic according to the results of the analysis of drug resistance.

Objective To compare the immune protective effect of different antigens of Trichinella spiralis:excretory-secretory(ES) antigen of adult worms, ES antigen of muscle larva and their mixed antigens in mice. Methods ES antigens of Trichinella spiralis adult worms and muscle larva were produced by the physiological saline culture methods. The ES antigens of Trichinella spiralis adult worms, muscle larva and the mix were used to immunize mice 3 times at 7 day interval, adjuvant control and normal control were set up. Seven day after the final immunization, each mouse was orally challenged with 200 Trichinella spiralis larva. Intestinal adult worms and muscle larva of Trichinella spiralis of each group were recoveried and examined on Day 7 and Day 30 post-challenge,respectively. Results The intestinal adult worms of Trichinella spiralis reduced by 87.95%, 69.48% and 84.34% in the adult worm ES antigen group, muscle larval ES antigen group and their mixed antigen group, respectively, while muscle larva reduced by 74.79%, 87.97% and 86.87% respectively. 4.Immune hyporesponsiveness induced by immature dendritic cells prolonged rat kidney allograft survival

Objective To study the effect of immature dendritic cells (imDC) transfusion in combination with bone marrow transplantation (BMT) on rat kidney allograft survival and its possible mechanism. Methods Renal allograft from DA rat was transplanted to Lewis rat. Forty recipient rats were randomized into 5 groups: (1) Negative control group; (2) imDC group: Lewis rats accepted imDC transfusion in the amount of 2? 107 only; (3) BMT group: Lewis rats accepted bone marrow transplantation in the amount of 2?108 only; (4) imDC + BMT group: Lewis rats accepted both imDC and BMC; (5)Third party donor group: Lewis rats accepted renal allograft from Wistar rats. One-way MLR was performed to assay splenic cell proliferation to allogeneic T cells. Exogenous IL-2 was added at the outset of another group as the former one-way MLR. Normal Lewis rat accepted splenic cells from tolerant rat in the amount of 1?108. DTH was assayed in the trans-tolerance model. Cells from spleen and thymus were harvested from recipient rats for detecting chimerism by flow cytometry. Objective To explore the treatment effect of human growth factor (NGF) on vascular dementia. Method 47 cases with vascular dementia were randomly divided into two groups (study and control). The study group (24 cases) were treated with NGF, the control group (23 cases) were treated with ordinary medicines.Results The intelligence and cognitive ability in the study group were obviously higher than those in the control group (P<0.01). Conclusion Compared with other methods, NGF has a significant curative effect.
7.Effects of magnesium valprote on behavior and lipid peroxidation of brain in chronic stress depression mice

Objective To explore the effects of magnesium valprote on behaviors and lipid peroxidation of brain in chronic stress depression mice.Methods 40 male mice were randomly divided into normal control group,model group, amitriptyline control group and magnesium valprote group, and.10 mice each group.Except normal control group,other groups were separated one mouse in each box and exposed to 21 days chronic unpredictable stressors, and the mice of normal control group were fed in a box and stress-free.During the stress, the mice of amitriptyline control group were applied amitriptyline(0.2 mg/10g) by intragastric every day,magnesium valprote group were given intragastrically magnesium valprote (0.8 mg/10g), and the mice of model and normal control group were administrated with normal sodium of the same volume by the same method.On 7th, 14th,21st day, all mice weight were weighed and weight gain was calculated.The ethology examination was performed by using jigglecage connected with Biology Function Experiment System and experiment of sucrose solution consumption after the chronic stress.At the end of experiment, all mice were killed, and then their brains were removed and made into brain homogenate.The activities of superoxide dismutase (SOD) and glutathione peroxidase (GSH-Px) and the content of malondialdehyde(MDA) in brain homogenate were analyzed by spectrophotometry.Results Compared with model group, magnesium valprote group were increased on locomotor activity number(97.50 ± 20.54,302.30± 39.04, t = 31.1432, P < 0.01 ), sucrose solution consumption ( ( 15.30 ± 3.60 ) ml, ( 23.70 ± 4.45 ) ml, t =9.8446, P<0.01) ,body weight gain(( 10.56 ± 1.53)g,( 12.63 ±2.12)g, t=5.3113, P=0.007) ,the activity of SOD( t =3.8428, P=0.004) ,and GSH-Px( t= 3.4143, P=0.007), but decreased on irritation number( t =17.1370, P < 0.01 ) and the content of MDA( t= 11.0778, P = 0.002).Conclusion Magnesium valprote can ameliorate behavior impairment of chronic stress-induced depression mice, and the observed antidepressant-like effects may be related to inhibition lipid peroxidation of mice brain.
8.Effect of blood purification on extravascular lung water and permeability index in extrapulmonary acute respiratory distress syndrome

Objective To observe the effects of continuous blood purification(CBP) on extravascular lung water and permeability index in patients with severe extrapulmonary acute respiratory distress syndrome (ARDSexp).Methods Fifteen patients with severe ARDSexp were treated with CBP.The level of extravascular lung water index (EVLWI) and pulmonary vascular permeability index (PVPI) was measured before and after treatment,and at the same time the changes of inflammatory indicators were recorded.Results Compared with that before treatment,EVLWI and PVPI was decreased obviously after treatment (P < 0.01),the level of C-reactive protein and interleukin 6 (IL-6),calcitonin and systemic inflammatory response syndrome score and the acute physiology and chronic health Ⅱ score was significantly decreased (P < 0.01).Conclusion CBP can improve the level of EVLWI,PVPI and inflammatory indicators and also the prognosis of severe ARDSexp.
9.IKKαregulates ultraviolet radiation-induced activation of p53 in a p38K-dependent manner

Objective To explore the signal transduction mechanism of inhibitor kappa B kinase α( IKKα) , one of the catalytic subunits of IKK complex , for regulating p53 transactivation in the cellular ultraviolet radiation ( UVB) repsonse. Methods The transactivation of p53 was determined by dual-luciferase reporter gene analysis system while the expression and activation of IKKα, IKKβ, p53 and p38K was detected by Western blotting assay .Results UVB exposure induced activation and transactivation of p 53 in the wild type mouse fibroblasts ,but the effect was blocked by IKKa deficiency and recovered by reconstitution of IKKαexpression.Under the same conditions , IKKαregulated p38K activation, while inhibi-ting p38K activation down-regulated p53 transactivation under UVB exposure .Conclusion IKKαregulates UVB-induced phosphorylation and activation of p 53 in a p38K-dependent manner .
10.PD-1/PD-L1 signaling pathway and its application in tumor

PD-1/PD-L1 signaling pathway as a T cell immune response co-stimulatory signaling pathway plays an important role in adaptive immunity. PD-1 is a major co-receptor expressing on T cells, binding with its ligands(PD-L1 and PD-L2), PD-1 can inhibit T cell activation and protect the body against the attacks from its own immune system. In addition to adjusting and maintaining autoimmune tolerance, in tumor cells PD-L1 expression is up-regulated, while in the virus-infected T cells PD-L1 expression is also upregulated. PD-1 / PD-L1 are involved in the tumor and infectious pathogen immune evasion, thus blocking the PD-1 / PD-L1 signaling pathway has become a hot research of cancer and chronic diseases. Currently, there are several anti-PD-1 or PD-L1 monoclonal antibodies approved by the FDA to enter clinical studies, which have shown significant anti-cancer effect.
