Objective To study the protective effects of bcl-2 gene against lung ischemia-reperfusion injury in vitro. Methods The lungs of rabbits were harvested respectively and preserved according to the way of lung transplantation. Then the lungs were reperfused using an ex vivo model. The rabbits were divided into 3 groups: control group, 0.9%NaCl was given; group2, Ad/Lac Z and group 3, receiving Ad/s-bcl-2. Agents were given to the rabbits by continuous intravenous infusion 48 hours before operations. Results Ad/s-bcl-2 increased the contents of bcl-2 mRNA in lung tissue, increased lung tissue SOD contents, decreased lung tissue MDA contents, decreased PAP, improved lung edema, reduced incidence of apoptosis and improved lung histological manifestation. These indexes in the Ad/s-bcl-2 group were better than in the groups of 0.9%NaCl and Ad/LacZ. Conclusion The bcl-2 gene amy provide significant protective effects on ischemia-reperfusion injury of the lung.

Objective To study the abdominal perineal resection for rectal cancer and clinical application of pelvic floor recon-struction .Methods 30 cases of rectal cancer patients from 2008-2011 were randomly divided into experimental group and the con-trol group ,30 cases in each group .Experimental group patients by columnar abdominal perineal resection of rectal cancer treatment , postoperative pelvic floor defect for dermal matrix reconstruction .The control group patients for colorectal cancer resection by lapa-rotomy .Results Compared with the traditional open resection ,the operation time ,intraoperative blood loss ,postoperative patholog-ical column T3 N0 M0 positive rate ,incidence of complications and incision aspect of cylindrical abdominal perineal resection were ob-vious advantages ,the difference was statistically significant (P< 0 .05) .Conclusion Compared with Miles surgery ,cylindrical ab-dominal perineal resection for rectal cancer can reduce the incidence of postoperative incisal edge positive rate and bowel perfora -tion .Human decellularized dermal matrix basin redevelopment can significantly reduce the surgical risk and difficulty of surgical op-erations .

Objective To investigate the effect of miR-145 on human lung adenocarcinaoma A549 cell proliferation and FSCN1 expression. Methods pmR-mcherry/miR-145 was constructed and transfected into A549 cell,then the expression of miR-145 and the proliferation of A549 cell were verified by QPCR and MTS assay, respectively.The situation of FSCN1 expression in A549 cell was detected by Western blot.Results pmR-mcherry/miR-145 vector was constructed successfully,and QPCR results indicated that miR-145 expressed effectively.Western blot results showed that FSCN1 was one of the targets of miR-145 in A549 cell.MTS assay results indicated that miR-145 inhibited the proliferation of A549 cell.Conclusion Overexpression of miR-145 can inhibit the proliferation of A549 cell, and FSCN1 was one of its target.

BACKGROUND: The stomach is usually used to substitute the diseased esophagus, which will change the physical digestive passage and results in some complications, such as back-streaming, poor food intake, and dyspepsia. If there is an artificial esophagus to replace the diseased esophagus, then the complications would be greatly reduced, and surgical procedures would be simplified. OBJECTIVE: To investigate the effects of artificial prosthesis of titanium-nickel alloy and silicone on repairing esophageal defect after part excision of esophagus. DESIGN, TIME AND SETTING: An in vivo observation experiment based on animals was performed at the animal laboratory of Second Affiliated Hospital of Guangzhou Medical College between May 1999 and May 2001. MATERIALS: Sixteen pigs of either gender, weighting 30 35 kg, were included. The artificial esophagus constructed by titanium-nickel alloy and silicone were provided by General Research Institute for Nonferrous Metals. Its length was 10 cm and its internal diameter was 20 mm. Its inner layer was made of silicone and its outer layer was titanium-nickel alloy net. METHODS: A segment of 7-cm thoracic esophagus was resected and was replaced by an artificial prosthesis constructed by titanium-nickel alloy and silicone. At 1, 2, 3, 4, 5, 6, 7, and 8 weeks, as well as 3, 4, 6, 8, 10, and 12 months after surgery, animals were sacrificed to take specimens. Sedal slices were stained by hematoxylin-eosin for pathological examination. MAIN OUTCOME MEASURES: Autopsy and histopathological findings of neo-esophagus. RESULTS: Of initial 16 pigs, 1 died owing to shock caused by hemorrhee, 1 died of excessive anesthesia, and the remaining 14 pigs survived a period from 7 days to 1 year. Following sacrifice, some vomicas containing yellow and white liquor purls on the chest wall of 4 pigs were observed, which were wrapped but did not communicate the thoracic cavity. One artificial esophagus was not in place but found in the stomach. One artificial esophagus was twisted and formed an esophagus diverticulum. The false passage around the prosthesis formed so long as the animals survived more than 1 week, which was called as neo-esophagus. The neo-esophagus was constructed primadly by granulation tissue within approximately 2 weeks, and then fibrous connective tissue replaced it. Four weeks later, esophageal epithelial cells covered the internal cavity of the "nee-esophagus". At this time, neonatal smooth muscle cells could be observed, but gland regeneration was not found. Following artificial esophagus displacement, the middle segment of "neo-esophagus" presented with stenosis to different degrees. The stenosis segment tended to stabilize with time (approximately 6 months later). CONCLUSION: Esophageal defect within a certain range of length can be repaired by an artificial esophagus constructed by titanium-nickel alloy and silicone.

Objective: To investigate the necessity of artificial kidney seeper which made through inserting a ureteral tube in the ureter to the ultrasonography-guided percutaneous nephrolithotomy (PCNL). Methods: This was a randomized prospective study. Patients who conformed to the inclusion and exclusion criteria were enrolled at Department of Urology, Frist Affiliated Hospital of Wenzhou Medical University from January 2016 to May 2017. Totally 291 patients were included in the study. Patients were randomly assigned into two groups (artificial kidney seeper group and non-artificial kidney seeper group) in different kidney seeper level (5 to <10 mm, 10 to 20 mm) respectively. The artificial kidney seeper group was inserted by a ureteral cathedral, then were underwent the ultrasonography-guided PCNL in prone position. The non-artificial kidney seeper group were underwent the ultrasonography-guided PCNL in prone position directly. The t test, χ² test, repeated measure analysis was used to data measurement, respectively. Results: Four patients who diagnosed pyonephrosis were excluded. On the 5 to <10 mm level, fever rate (14.6% vs. 4.8%, χ²=5.07, P=0.03), operation time ((65.7±9.9)min vs. (50.3±7.4)min, t=11.47, P=0.00), cost ((18 327±903) yuan vs. (14 583±784) yuan, t=24.50, P=0.00) about artificial kidney seeper group and non-artificial kidney seeper group had statistical differences. And on the 10 to 20 mm level, fever rate (14.5% vs. 3.39%, χ²=4.53, P=0.03), operation time ((66.0±9.9)min vs. (52.4±8.9)min, t=8.30, P=0.00), cost ((16 548±537) yuan vs. (13 102±629) yuan, t=32.10, P=0.00) about artificial kidney seeper group and non-artificial kidney seeper group had statistical differences. And there were no statistical differences in the success rate of puncturing, the failures of the treatment to the stone pieces falling into the ureter and clearance rate of the stone and so on. In this study, 2 cases recovered after received transfusion and digital subtraction angiography artery embolization treatments; D-J tube was indwelled into 4 cases guiding by super smooth thread; 4 cases were finished the surgeries with the help of ureteroscopy, because the stone pieces fell into the ureter during the surgeries. And there was no patients developing septic shock, adjacent viscera injury or other serious complications. Conclusion For seasoned doctors, there is no necessity of regularly artificial kidney seeper for PCNL when the pre-operation seeper ranging from 5 to 20 mm.
