1.Effect of different dose ofsustained-release theophyline on prealbumin, C-reactive protein and white blood cell count in patients with pediatric asthma

Objective To analyse effect of different dose of sustained-release theophyline on prealbumin, C-reactive protein and white blood cell count in patients with pediatric asthma.Methods 58 children with acute asthma attack in our hospital were collected.All children were randomly divided into high dose group and low dose group,29 cases in each group.On the basis of conventional treatment, high dose group were treated with high dose theophylline sustained release tablets 5 mg/kg, one times per eight hours,orally,and low dose group was treated with low dose theophylline sustained release tablets,2 mg/kg, one times per eight hours,orally.Two groups were treatment for two weeks.After the treatment, the serum levels of pre albumin, C-reactive protein and veinal blood level of white blood cell count were detected in all children.Results Compared with high dose group post-treatment, the serum level PA was higher in low dose group (P<0.05);the serum level of CRP was lower in low dose group (P<0.05);the veinal blood level of WBC, percentage of neutrophils and percentage of eosinophils was lower in low dose group (P<0.05) .Conclusion Compared with high dose of sustained-release theophyline,the low dose of sustained-release theophyline can significantly reduce the serum CRP level and veinal blood level of WBC, percentage of neutrophils and percentage of eosinophils, improve serum PA level in patients with pediatric asthma, reduce inflammation in patients.

BACKGROUND:Mesenchymal stem cel s as potential seeded cel s have been widely used in tissue engineering and clinic therapy;thus, the precise, safe, effective isolation of mesenchymal stem cel s is the particular important premise to build culture system.
OBJECTIVE:To review the methods of isolating mesenchymal stem cel s and to compare the merit and demerit of different methods, thereby providing theoretical basis for safe and high-effective isolation of mesenchymal stem cel s.
METHODS:A computer-based online research of CNKI and PubMed databases was performed to col ect articles, which included reviews, clinical trials and experiments, published between 1965 and 2014 with the key words of“mesenchymal stem cel s (MSCs), isolation methods”in Chinese and English. A total of 52 articles were included according inclusion and exclusion criteria
RESULTS AND CONCLUSION:(1) The whole bone marrow culture method can derive a mass of mesenchymal stem cel s, which need to be purified. (2) The density gradient centrifugation method which uses the media with the density of 1.073 g/mL can be used to harvest more purified cel s. (3) The tissue digestion method is suitable for digestion and isolation of adipose tissue and umbilical cord tissue. Type II col agenase digestion is better, but they are both limited by a high demand for operative techniques. (4) Immunomagnetic bead separation is appropriate to study the biological characteristics of a kind of subpopulation of mesenchymal stem cel s which express special surface markers. (5) The combination method is also an optimal way. (6) Some new methods limited by few dates require further studies.
10.Expression of osteopontin in colorectal cancer and hepatic metastatic cancer tissues and its clinical significance

Objective To investigate the expression of osteopontin in colorectal cancer and hepatic metastatic cancer and its clinical significance.Methods The expression of osteopontin in 76 cases of colorectal cancer tissues,30 para carcinoma normal mucosa,liver metastatic tumor tissues in 16 patients was examined by immunohistochemical SP staining method.Results The expression rates of osteopontin in normal mucosa,colorectal cancer tissues and liver metastatic cancer tissues were 6.7% (2/30),69.7% (53/76),75.0% (12/16) respectively.The expression rates of osteopontinin colorectal cancer and liver metastatic cancer was significantly higher than those in the normal mucosa (respectively x2 =34.273,23.014,all P < 0.001).The expression of osteopontin was related to infiltration depth,lymph node metastasis,and distant metastasis (respectively x2 =14.347,6.577,7.278,5.537,all P < 0.05).There was no significant difference in the sex and age.Kaplan-Meier survival analysis showed that patients with osteopontin positive had poor prognosis compared with osteopontin negative patients (P < 0.001).Conclusions The expression of osteopontin is closely related to the invasion and metastasis of colorectal carcinoma,a hopeful indicator for the prognosis of colorectal cancer.
